Bioinformatics analyses of comorbid mechanisms between psoriasis and type 2 diabetes mellitus.

Epidemiological association between psoriasis and T2DM suggests shared pathophysiology that are to be explored. Microarray expression profiles for psoriasis and T2DM were obtained from the Gene Expression Omnibus (GEO) database. The "limma" package in R software was used to screen the differentially expressed genes (DEG). GO and KEGG enrichment analysis were further conducted to explore the functions of co-DEGs. By intesecting genes of the key disease-related modules from WGCNA with co-DEGs, candidate co-driver genes were identified and their PPI network was constructed. Hub genes with good di

From bugs to brain: unravelling the GABA signalling networks in the brain-gut-microbiome axis.

Convergent data across species paint a compelling picture of the critical role of the gut and its resident microbiota in several brain functions and disorders. The chemicals mediating communication along these sophisticated highways of the brain-gut-microbiome (BGM) axis include both microbiota metabolites and classical neurotransmitters. Amongst the latter, GABA is fundamental to brain function, mediating most neuronal inhibition. Until recently, GABA's role and specific molecular targets in the periphery within the BGM axis had received limited attention. Yet, GABA is produced by neuronal an

Predicting five-year comorbid bipolar disorder after attention-deficit/hyperactivity disorder diagnosis: a population-based machine learning approach.

BACKGROUND: Early detection and accurate prediction of bipolar disorders (BDs) comorbidity in individuals with attention-deficit/hyperactivity disorder (ADHD) are clinically critical. This study used machine-learning methods to identify features predictive of subsequent BD among patients initially diagnosed with ADHD. METHODS: We analyzed claims from the Taiwan National Health Insurance Research Database (2000–2013) and included patients aged ≥ 12 years with at least two diagnoses of ADHD. Predictor features included demographics (sex, age at ADHD onset), healthcare utilization (psychiatric ou

Mapping the Level of Evidence of Prenatal, Childhood, and Adolescent Exposure to Volatile Organic Compounds and Health Outcomes: Protocol for a Scoping Review.

BACKGROUND: Volatile organic compounds (VOCs) are a diverse group of organic chemicals with widespread presence in daily life. VOCs may have detrimental health effects on humans, particularly during critical periods such as pregnancy, childhood, and adolescence. Although the body of evidence is growing, significant gaps remain in the current understanding of the associations of VOCs on health across these developmental stages. Moreover, existing reviews focused on specific VOCs or individual health conditions, highlighting the need for a comprehensive summary of the current literature to stimu

Are outcomes of giant ventral hernia repair inferior? A propensity-matched analysis.

BACKGROUND: Ventral hernia repair (VHR) of large defects poses significant technical challenges due to the size of the hernia and complexity of operative techniques required for fascial closure. This study examined clinical outcomes in "giant" open VHR (GVHR), with hernia defect size (HDS) ≥ 200 cm2, versus "non-giant" open VHR (NGVHR) with HDS < 200 cm2 using a propensity-matched approach. METHODS: A prospectively-maintained database from a tertiary hernia center was reviewed for patients undergoing open VHR. 1:1 propensity-score matching was performed for GVHR versus NGVHR based on age, BMI

Shenqi Pill alleviates acetaminophen-induced liver injury: a comprehensive strategy of network pharmacology and spectrum-effect relationship reveals mechanisms and active components.

BACKGROUND: Acetaminophen (APAP), commonly used for its antipyretic and analgesic properties, can cause severe liver injury or even acute liver failure when overdosed. However, the options for treating APAP-induced liver toxicity are limited. Shenqi Pill (SQP), a traditional Chinese herbal formula, has shown effectiveness in treating various liver ailments. SQP consists of cinnamon, aconite, rehmannia, cornus, peony bark, Chinese yam, poria, and alisma in a ratio of 1:1:8:4:3:4:3:3. However, the mechanisms and active components of SQP that counteract drug-induced liver injury (DILI) are not we

Piperine: an emerging biofactor with anticancer efficacy and therapeutic potential.

Anticancer drug discovery needs serious attention to overcome the high mortality rate caused by cancer. There are still many obstacles to treating this disease, such as the high cost of chemotherapeutic drugs, the resulting side effects from the drug, and the occurrence of multidrug resistance. Herbaceous plants are a reservoir of natural compounds that can be anticancer drugs with novel mechanisms of action. Piperine, a bioactive compound derived from Piper species, is gaining attention due to its unique dual role in directly inhibiting tumor growth and enhancing the bioavailability of chemot

Research models of sulfur mustard- and nitrogen mustard-induced ocular injuries and potential therapeutics.

Sulfur mustard (SM) is a notorious, bifunctional alkylating vesicant that was first used in warfare during World War I in 1917 and since then has been deployed in numerous skirmishes with its most recent documented use being during the Middle Eastern conflicts. Apart from its use in combat and terrorist activities, continual threat of accidental exposure from old stockpiles and improperly discarded munitions is ever present, especially to the innocent and unassuming civilian populations. SM can cause devastating injuries, depending on the dosage of SM exposure, route of exposure, as well as th

Dreams, False Starts, Dead Ends, and Redemption: A Chronicle of the Evolution of a Chemoinformatic Workflow for the Optimization of Enantioselective Catalysts.

Catalyst design in enantioselective catalysis has historically been driven by empiricism. In this endeavor, experimentalists attempt to qualitatively identify trends in structure that lead to a desired catalyst function. In this body of work, we lay the groundwork for an improved, alternative workflow that uses quantitative methods to inform decision making at every step of the process. At the outset, we define a library of synthetically accessible permutations of a catalyst scaffold with the philosophy that the library contains every potential catalyst we are willing to make. To represent the

p21-activated kinase 4 phosphorylates peroxisome proliferator-activated receptor Υ and suppresses skeletal muscle regeneration.

BACKGROUND: Skeletal muscle regeneration is an adaptive response to injury that is crucial to the maintenance of muscle mass and function. A p21-activated kinase 4 (PAK4) serine/threonine kinase is critical to the regulation of cytoskeletal changes, cell proliferation, and growth. However, PAK4's role in myoblast differentiation and regenerative myogenesis remains to be determined. METHODS: We used a mouse model of myotoxin (notexin)-induced muscle regeneration. In vitro myogenesis was performed in the C2C12 myoblast cell line, primary myoblasts, and primary satellite cells. In vivo overexpre

Initial prospective study of ambulatory mPCNL on upper urinary tract calculi.

BACKGROUND: exposure to dioxins has been associated with an increased risk of soft-tissue sarcoma (STS). In a study relating to 1989-1998, a significant excess of STS was found in the population living within a 2-kilometre radius of an industrial incinerator located inside a Mantua industrial site. In the years 1974-1991, the incinerator burned hazardous chlorine-containing waste that resulted in the emission of dioxins. After 1991, it was exclusively fed with organic synthetic products not containing chlorine. OBJECTIVES: the aim of this case-control study was to estimate the STS risk, for 1
