Presence and significance of Bacillus thuringiensis Cry proteins associated with the Andean weevil Premnotrypes vorax (Coleoptera: Curculionidae).

The Andean weevil Premnotrypes vorax represents an important cause of damage to Colombian potato crops. Due to the impact of this plague on the economy of the country, we searched for new alternatives for its biological control, based on the entomopathogenic bacteria Bacillus thuringiensis. A total of 300 B. thuringiensis strains obtained from potato plantations infested with P. vorax were analyzed through crystal morphology, SDS-PAGE, PCR and bioassays. We used site-directed mutagenesis to modify the Cry3Aa protein. Most of the B. thuringiensis isolates had a bipyramidal crystal morphology. S

Susceptibility and behavioral responses of the dampwood termite Zootermopsis angusticollis to the entomopathogenic nematode Steinernema carpocapsae.

Termites exploit microbially rich resources such as decayed wood and soil that are colonized by potentially pathogenic and parasitic fungi, bacteria, viruses, and nematodes. In colonies composed of thousands of individuals, the risk of infection among nestmates is significant, and individual and social behavior could involve various adaptations to resist disease and parasitism. Here we show that the dampwood termite Zootermopsis angusticollis (Hagen) exhibits a dosage dependent susceptibility to the soil nematode Steinernema carpocapsae (Weiser) (Mexican strain) and that this social insect sig

Genetic engineering of Periplaneta fuliginosa densovirus as an improved biopesticide.

The smoky-brown cockroach (Periplaneta fuliginosa) densovirus (PfDNV) has previously shown potential in urban pest control. To improve its efficacy as a biopesticide, the genome of PfDNV was engineered by inserting the insect-specific toxin gene BmKIT1 in the open reading frame encoding the major structural proteins. A green fluorescent protein (GFP) marker was tagged to the BmKIT1 at its C-terminus for in vivo imaging using Confocal laser scanning microscopy (CFSM). Using a virion rescue strategy, the genomes of recombinant and wild-type (wt) PfDNV were then cotransfected in P. fuliginosa nym

[Use of satellites for public health purposes in tropical areas].

The epidemiological hallmark of the new millennium has been the emergence or recrudescence of transmissible diseases with high epidemic potential. Disease tracking is becoming an increasingly global task requiring implementation of more and more sophisticated control strategies and facilities for sustainable development. A promising initiative involves the use of satellite technology to monitor and forecast the spread of disease. The Health Early Warning System (HEWS) was designed based on successful application of satellite data in food programs as well as in other areas (e.g. weather, farmin

Absorption of pathogenic autoantibodies by the extracellular domain of pemphigus vulgaris antigen (Dsg3) produced by baculovirus.

Pemphigus vulgaris (PV) is an autoimmune blistering disease, in which autoantibodies against PV antigen (PVA or Dsg3) play a pathogenic role in inducing blister formation. Bacterial fusion proteins of PVA failed to absorb pathogenic autoantibodies from PV patients' sera probably because they did not represent the proper conformation. Therefore, a chimeric protein, PVIg, consisting of the whole extracellular domain of PVA and the constant region of human IgG1, was produced in either in COS7 or in insect Sf9 eucaryotic cells. Both PVIg-COS7 and PVIg-Sf9 were recognized by all of the 35 PV sera t

Evidence for induced radioresistance from survival and other end points: an introduction.

A substantial body of data published during the past 30 years makes a strong case for the existence of cellular radioprotective mechanisms that can be up-regulated in response to exposure to small doses of ionizing radiation. Either these "induced" mechanisms can protect against a subsequent exposure to radiation that may be substantially larger than the initial "priming" or "conditioning" dose, or they may influence the shape of the survival response to single doses so that small radiation exposures are more effective per unit dose than larger exposures above a threshold where the induced rad

St Louis encephalitis virus establishes a productive, cytopathic and persistent infection of Sf9 cells.

The Sf9 cell line, commonly used for gene expression by recombinant baculovirus, has been productively infected by St Louis encephalitis (SLE) virus, a flavivirus. SLE viral infection produced a c.p.e. in the Sf9 cells characterized by giant cells and the presence of 10-fold fewer cells in the infected cultures after the first week of infection compared with uninoculated control cultures. Infected Sf9 cells expressed SLE viral antigens, and intracellular virus particles were observed by electron microscopy. Titres of cell-associated SLE virus rose slightly over an 8 week period, whereas titres

Identification of filarial larvae in vectors by DNA hybridization.

Infectivity rates of insect vectors are the best criteria by which to assess the transmission of filarial parasites and the efficacy of filariasis control programs. Currently available DNA probes can be used to estimate the proportion of vectors containing larvae of a given filarial species but provide no information on three other important variables in the transmission dynamics of filarial nematodes: the developmental stage, the location and the actual number of larvae that are present in the vector, all of which can be reliably determined by microscopy. However, species identification is of

SIFamide Signaling regulates energy mobilization during phototactic behavior in the Asian Citrus psyllid, Diaphorina citri.

Phototactic behavior is an energy-intensive visually guided behavior critical for insect survival and dispersal. The central neuromodulatory mechanisms underlying this oriented response remain poorly understood, particularly with respect to peptidergic regulation. To address this knowledge gap, we conducted a study on the Asian citrus psyllid, Diaphorina citri, a vector of citrus huanglongbing, whose adults exhibit robust positive phototaxis. The SIFamide (SIFa) neuropeptide (DcSIFa) and its cognate receptor (DcSIFaR) were identified, both of which showed the highest expression levels in the h

Exploitation of host mating behaviour by a parasitoid fly.

Egg-laying decisions are crucial in insect parasitoids because the host constitutes the only resource available for immature development. We estimated the extent to which host sex, size and mating behaviour are involved and interact in shaping host selection in the association between the Japanese beetle, Popillia japonica, and a parasitoid fly, Istocheta aldrichi. Under field conditions, we found that females and larger individuals of the Japanese beetle, both males and females, are associated with the highest levels of parasitism, regardless of seasonal variations in host size and sex ratio.

Dietary American Cockroach (Periplaneta americana) Chitin Influences Growth Performance, Nutrient Utilization, Gut Microbiota, and Riboflavin-Related Metabolism in Juvenile Grass Carp (Ctenopharyngodon idella).

The American cockroach (Periplaneta americana) is an emerging sustainable source of insect-based chitin, yet its physiological effects and optimal inclusion levels in herbivorous fish diets remain undefined. This study evaluated the dose-dependent effects of dietary P. americana chitin (0%, 2.5%, 5%, 10%, and 20%) on juvenile grass carp (Ctenopharyngodon idella) over a 63-day trial. Results revealed a distinct biphasic response in growth and nutrient utilization. Fish fed the 5% chitin diet exhibited the highest weight gain and specific growth rate (SGR), coinciding with the highest apparent d

A Plastivore Gut System Enables In Vivo Biotransformation and Partial Defluorination of Perfluorooctanoic Acid.

Per- and polyfluoroalkyl substances (PFAS) are highly persistent contaminants that resist biological degradation, hindering the development of sustainable remediation strategies. Here we reveal that plastivorous Tenebrio molitor larvae mediate gut-driven biotransformation and partial defluorination of perfluorooctanoic acid (PFOA) in vivo. Over 15 days, the apparent loss of extractable parent PFOA was 47.9% and 41.2% in the 0.5 g kg-1 and 20 g kg-1 PFOA dietary treatments, respectively. Control-corrected inorganic fluoride recovered from larval tissues and frass, together with the identificati

Climate-stressed crop mycobiomes as pre-harvest sentinels of human mycotoxin exposure: an AI-enabled One Health framework linking dysbiosis, exposomics, and toxicopathology.

Climate change is altering the conditions under which crop-associated fungi colonize plants and produce mycotoxins. Current control relies on both pre-harvest management and post-harvest testing; however, field-level fungal-community changes are rarely used to guide where and when testing should be intensified. This Review critically compares evidence linking weather and crop stress, fungal-community composition, toxigenic strain activity, contamination during harvest and storage, human biomarkers, and organ-specific effects. On this basis, we propose the Crop Mycobiome-Exposome-Toxicopatholog

Integration of Morphology, Histology, and Ultrastructure of the Gall-Inducing Andricus quercustozae (Cynipidae) Larval Alimentary Canal.

The transition from simple herbivory to specialised gall induction in Cynipidae marks an evolutionary peak in insect-plant interactions. This structural and ultrastructural study suggests that the gall wasp larva has evolved from a passive consumer into a biological architect, utilising a highly specialised body plan. The structure of the large midgut, the midgut epithelium reinforced by septate junctions, and the cuboidal cells of the hindgut allow for the possibility that the larva of Andricus quercustozae may control itself independently from its host, working as an active producer rather t

Development and Immunogenicity Evaluation of Baculovirus-Expressed Feline Bocavirus VP2 Virus-like Particles Vaccine in a Mouse Model.

Feline bocavirus (FBoV) is an emerging enteric virus associated with gastrointestinal diseases in cats and has attracted increasing attention in feline health. However, no commercial vaccine is currently available for the prevention and control of FBoV infection. VP2 is the major capsid protein of FBoV and represents a promising target for vaccine development. In this study, the FBoV VP2 protein was expressed using the insect baculovirus expression system. The purified VP2 protein self-assembled into virus-like particles (VLPs), which were formulated with Alum, ISA 206, or GEL 02 adjuvants to

A Molecular and Cellular Mechanism for Bitter Taste in the mosquito Aedes aegypti.

The yellow fever mosquito, Aedes aegypti, is a vector of Zika, Dengue, Chikungunya and yellow fever, disease-causing viruses impacting millions of people annually across the globe. Female mosquitoes transmit pathogens through serial blood-feeding, while both male and female mosquitoes feed on plant resources. Contact chemosensation (taste) guides these two feeding modes in both positive ways, feeding readily on sugar in nectar or ATP in blood, and negative ways, demonstrating aversion to chemically diverse bitter compounds in blood or sugar meals. Here, we identify a receptor, AaegGr14, that i

Hydrophilic and lipophilic diblock polymer-based nanoplatform improves the bioactivity of insect growth regulator via inhibiting chitin synthesis.

Insect growth regulators (IGRs) have shown great potential in green pest management, but their low control efficacy and delayed action limit their widespread application. In recent years, various nanodelivery systems have revolutionized pesticides by enhancing bioavailability, reducing dosage requirements, improving environmental compatibility, etc. Herein, a hydrophilic and lipophilic diblock polymer (HLDP) was designed and developed to construct an efficient nanoplatform for an IGR diflubenzuron (DFB). The HLDP could spontaneously assemble with DFB into DFB/HLDP complex via electrostatic int

Antibody functionalized thiolated chitosan stabilized silver nanoparticles as theranostics toward visceral leishmaniasis.

Leishmaniasis (VL) is a neglected disease caused by Leishmania protozoa, transmitted through insect vectors, and can be fatal without adequate treatment. Current therapies are hindered by issues such as drug resistance and side effects, making rapid diagnosis essential. Biosensors are being explored to enhance detection, and silver nanoparticles (AgNPs) have emerged as promising tools for both diagnosis and treatment. To improve their stability and functionality, AgNPs can be coated with thiolated chitosan (TCh), which is synthesized by introducing thiol groups to chitosan using 3-mercaptoprop

Enterovirus-like particles encapsidate RNA and exhibit decreased stability due to lack of maturation.

To counteract hand, foot, and mouth disease-causing viruses such as enterovirus A71 and coxsackievirus A6, virus-like particles (VLPs) have emerged as a leading contender for the development of a multivalent vaccine. However, VLPs have shown rapid conversion from a highly immunogenic state to a less immunogenic state and low particle integrity lifetimes compared to inactivated virus vaccines, thus raising concerns about their overall stability. Here, we produce VLPs to investigate capsid stability using cryogenic electron microscopy (cryo-EM), mass spectrometry (MS), biochemical assays, and at
