Interpretable multimodal learning for integrating neuroimaging and genetic data in Alzheimer's disease.

INTRODUCTION: Early detection of Alzheimer's disease (AD) requires models that combine brain structure changes with genetic risk, but existing methods struggle to align these different data types. METHODS: We present R-GenIMA, an interpretable multimodal large language model that pairs a region-of-interest vision transformer with genetic prompting to jointly analyze structural MRI and single nucleotide polymorphisms (SNPs). Each brain region becomes a visual token and SNP profiles are encoded as structured text, letting the model link regional atrophy to genetic factors through cross-modal at

Entropy Regularization in Deep Reinforcement Learning: A Structured Review Across Classical Control, Generative Policies, and Reasoning Language Models.

Entropy regularization is a recurring mechanism in reinforcement learning (RL), but its meaning changes across algorithmic settings. In classical online RL, entropy encourages exploration and smooths policy improvement; in inverse RL and imitation learning, maximum-entropy resolves ambiguity among expert-consistent behaviors; in offline RL, entropy must be balanced against data support; in generative policies, entropy becomes a tractability problem; and in reinforcement learning with verifiable rewards (RLVR) for large language models (LLMs), token entropy is tied to reasoning diversity, calib…

Prot-ΔΔG: Prediction of protein-protein binding affinity changes upon mutations with pre-training strategies.

Protein-protein interactions are essential for diverse biological activities, but amino acid mutations can disrupt these interactions, leading to dysfunction and disease. Mutation impacts can be quantified via change in protein-protein binding affinity (ΔΔG) before and after mutation. Accurate prediction of ΔΔG is critical for understanding disease mechanisms, guiding drug discovery, and advancing protein engineering. Existing computational methods often exhibit reduced accuracy in the absence of high-resolution protein structural data, failing to fully capture sequence-embedded patterns and e

Evaluation of an Artificial Intelligence Conversational Chatbot to Enhance HIV Preexposure Prophylaxis Uptake: Development and Usability Internal Testing.

BACKGROUND: The HIV epidemic in the United States disproportionately impacts gay, bisexual, and other men who have sex with men (MSM). Despite the effectiveness of HIV preexposure prophylaxis (PrEP) in preventing HIV acquisition, uptake among MSM remains suboptimal. Motivational interviewing (MI) has demonstrated efficacy at increasing PrEP uptake among MSM but is resource-intensive, limiting scalability. The use of artificial intelligence, particularly large language models with conversational agents (ie, "chatbots") such as ChatGPT, may offer a scalable approach to delivering MI-based counse…

DAVA: Decoding Art With Visual Analytics Through Feature Modeling and Multi-Agent Collaboration.

Figurative art, as a culturally embedded medium, encodes narrative, symbolic, and emotional meanings that reflect artistic choices and historical realities. The recent availability of large-scale digital collections of figurative artworks creates opportunities for computational analysis, but existing methods mostly focus on classification or style detection, lacking structured modeling of high-level figurative elements and integration of cultural context. We present DAVA, a visual analytics system that supports interdisciplinary exploration of figurative art. First, we model paintings across t

Gravity-awareness: a deep learning and LLM framework for predicting human adaptation to altered gravitational environments.

Earth's gravity fundamentally shapes human behavior. The brain encodes this force as an internal model of gravity, enabling the prediction and interpretation of gravitational effects during perception and action. Understanding how this model adapts to altered gravity is critical for predicting human performance in spaceflight. We present a computational framework for modelling neurophysiological adaptation across diverse gravitational environments. The framework has two components trained on open-access data from altered-gravity studies, particularly parabolic flights. The first component (Cor

Application of pharmacokinetic modelling for 2,3,7,8-tetrachlorodibenzo-p-dioxin exposure assessment.

Polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans, and mono- and non-ortho polychlorinated biphenyls (dioxin-like PCBs) are identified as a family or group of organic compounds known as 'dioxins' or dioxin-like chemicals (DLCs). The most toxic member of this group is 2,3,7,8-tetrachlorodibenzo-(p)-dioxin (TCDD). Historically, DLCs have caused a variety of negative human health effects, but a disfiguring skin condition known as chloracne is the only health effect reported consistently. As part of translational research to make computerized models accessible to health risk assesso

HyLnc: a hybrid deep learning and feature-based approach for long non-coding RNA prediction.

Long non-coding RNAs (lncRNAs) play important roles in gene regulation, development and disease, yet accurate identification of lncRNAs from transcriptomic data remains a major computational challenge. Existing methods often rely either on handcrafted sequence features or deep learning approaches, each with their inherent limitations in capturing the full complexity of RNA sequences. In this study, we proposed HyLnc, a computational framework that integrates transformer-based contextual embeddings with biologically meaningful sequence features for improved lncRNA prediction. A custom BERT-base

Locating Youth Exposed to Parental Justice Involvement in the Electronic Health Record: Development of a Natural Language Processing Model.

BACKGROUND: Parental justice involvement (eg, prison, jail, parole, or probation) is an unfortunately common and disruptive household adversity for many US youths, disproportionately affecting families of color and rural families. Data on this adversity has not been captured routinely in pediatric health care settings, and if it is, it is not discrete nor able to be readily analyzed for purposes of research. OBJECTIVE: In this study, we outline our process training a state-of-the-art natural language processing model using unstructured clinician notes of one large pediatric health system to i

Big data in U.S. neuro-oncology: trends and translational priorities.

PURPOSE: Neuro-oncology generates complex clinical, imaging, and molecular data, yet datasets remain relatively small and fragmented across modalities and institutions. While "big data" is traditionally defined by large sample size, neuro-oncology datasets are often characterized instead by high dimensionality. This study aims to provide an overview of the landscape of major U.S. neuro-oncology data resources and evaluate how these datasets are used in contemporary research. METHODS: A selection of neuro-oncology datasets was evaluated, including population registries, clinical data networks,
