The in vitro interaction of neutrophils and virulent axenic Entamoeba histolytica trophozoites on Chang liver cells was studied to determine if the presence of neutrophils enhanced destruction of liver cell monolayers. Axenic amoebae (strain HM1:IMSS) destroy liver cell monolayers in a dose-dependent manner (P less than .001). Human neutrophils had no effect on the liver cell monolayers; however, when neutrophils were added to amoebae, destruction of monolayers was enhanced (P less than .05 compared with amoebae alone). Similar results were obtained when Chinese hamster ovary (CHO) cells were

Groups of athymic rnu rats were found to be susceptible to SAW 408 Entamoeba histolytica introduced intra-intestinally. The same strain of rat could be infected orally with cultured trophozoites using a gastric secretion inhibitor, cimetidine, as a pretreatment. Groups of Lac:P and Wistar rats similarly treated were also found to be susceptible although the parasite population growth was slow. A similar regimen attempted on susceptible mice was unsuccessful.

Eighty-nine patients infected with Entamoeba histolytica, non-pathogenic amoebae or Trichuris trichiura were studied prospectively to determine the safety and efficacy of diphetarsone therapy. An additional 75 patients were studied retrospectively to assess further the efficacy of diphetarsone in the treatment of E. histolytica cyst passers. Side effects were noted in 9% and included gastrointestinal upset, lightheadedness and headache. Transient liver function abnormalities were recorded in 5.6%. diphetarsone was completely effective in the treatment of Dientamoeba fragilis, Entamoeba hartman

Results of testing 298 sera from hamsters inoculated in the liver with Entamoeba histolytica and from 25 uninoculated controls by indirect hemagglutination (IHA) and enzyme linked immunosorbent assay (ELISA) showed that measurable amounts of antibody appeared as early as 5 days after inoculation, generally increased to a high level within 20 days, and persisted for the duration of the experiments (58 days). There was fair correlation between size of liver lesion and titers. Strain differences in amebae used for antigen and for inoculation could not be detected by either test. The IHA test appe

BACKGROUND: Enterocolitis and gastroenteritis remain major health problems, particularly in children living in developing countries. Intestinal protozoa, such as Entamoeba histolytica, Blastocystis, and Cyclospora, are frequently associated with these conditions. Amebic colitis can cause serious complications, including fulminant necrotizing colitis, toxic megacolon, extraintestinal amebiasis, and stunting in children. The diagnosis of amoebiasis is challenging, relying on microscopic examination, which cannot distinguish E. histolytica from the nonpathogenic E. dispar and E. moshkovskii. Ther…

Balamuthia mandrillaris, a free-living amoeba, can cause Balamuthia amebic encephalitis (BAE), a rare and often fatal cerebral infection. The reported mortality rate is >90%, largely attributable to the absence of specific clinical manifestations, sensitive diagnostic methods, and effective therapeutic interventions. We herein describe a middle-aged, male patient diagnosed with BAE using Metagenomic Capture sequencing (MetaCAP) who achieved full recovery following early medical therapy without neurosurgical treatment. Our findings indicate that MetaCAP serves as a rapid and sensitive diagnosti

Free-living amoebae of the genus Acanthamoeba are the etiological agents of cutaneous lesions, granulomatous amoebic encephalitis (GAE) and amoebic keratitis (AK), which are chronic infections with poor prognosis if not diagnosed promptly. Currently, there is no optimal therapeutic scheme to eradicate the pathologies these protozoa cause. In this study we report the morphological and molecular identification of three species of the genus Acanthamoeba, belonging to T4 group; A. polyphaga isolated from the corneal ulcer of a patient sample of AK case; A. castellanii isolated from the contact len

Recently, the search for novel therapeutic agents against Acanthamoeba species has been focused on the evaluation of natural resources. Among them, marine microorganisms have risen as a source of bioactive compounds with the advantage of the ability to obtain unlimited and constant amounts of the compounds in contrast to other natural sources such as plants. Furthermore, marine actinomycetes have recently been reported as highly rich in bioactive agents including salinosporamides, xiamycines, indolocarbazoles, naphtyridines, phenols, dilactones such as antimycines and macrolides among others.

We report on an HIV-negative but immunocompromised patient with disseminated acanthamoebiasis, granulomatous, amoebic encephalitis and underlying miliary tuberculosis and tuberculous meningitis. The patient responded favorably to treatment with miltefosine, an alkylphosphocholine. The patient remained well with no signs of infection 2 years after treatment cessation.

Naegleria fowleri causes an acute and rapidly fatal central nervous system infection called primary amebic meningoencephalitis (PAM) in healthy children and young adults. We describe here the identification of N. fowleri isolated from the brain of one of several cows that died of PAM based on sequencing of the internal transcribed spacers, including the 5.8S rRNA genes.

We describe a fatal case of amebic encephalitis caused by Balamuthia mandrillaris in a 3-year-old Czech boy who had never traveled abroad. This is the first such infection reported in Europe. The diagnosis was established by brain biopsy, in which abundant trophozoites and a few round amebic cysts were identified. The presence of multiple nucleoli in some trophozoites suggested the organism to be Balamuthia mandrillaris and this was confirmed by indirect immunofluorescence. The amebae invaded brain tissue, including neurons, and blood vessel walls, causing thrombovasculitis. The tissue reactio

Using the indirect fluorescent antibody test and sera from 10 proved cases of invasive amoebiasis, the effects of absorption of IgG on observed titres of amoeba-specific IgM and IgA were investigated. In addition, results of cellulose acetate precipitin tests were compared with anti-amoebic antibody levels. Antiamoebic IgM was found at titres of 1:14 to 1:112 after IgG absorption in four cases. Anti-amoebic IgA was detected in sera from four cases, but the maximum titre was 1:28. There was no relationship between the presence of amoeba-specific IgM or IgA and the result of the precipitin test,
