Angiotensin II stimulates canonical TGF-β signaling pathway through angiotensin type 1 receptor to induce granulation tissue contraction.

UNLABELLED: Hypertrophic scar contraction (HSc) is caused by granulation tissue contraction propagated by myofibroblast and fibroblast migration and contractility. Identifying the stimulants that promote migration and contractility is key to mitigating HSc. Angiotensin II (AngII) promotes migration and contractility of heart, liver, and lung fibroblasts; thus, we investigated the mechanisms of AngII in HSc. Human scar and unwounded dermis were immunostained for AngII receptors angiotensin type 1 receptor (AT1 receptor) and angiotensin type 2 receptor (AT2 receptor) and analyzed for AT1 recepto

660 nm LED preconditioning programs mitochondrial signaling in BMSCs for functional tracheal cartilage regeneration.

Stem-cell-based cartilage regeneration is limited not only by cell delivery, but also by the difficulty of establishing a stable and functionally competent regenerative cell state before implantation. Here, we report a photonic mitochondrial programming strategy in which 660 nm light-emitting diode (LED) irradiation preconditions bone marrow-derived mesenchymal stem cells (BMSCs) to enhance functional tracheal cartilage regeneration. BMSCs were seeded onto egg white-derived ring-shaped porous scaffolds and exposed to defined 660 nm LED irradiation before implantation. Among the tested irradiat

Interleukin-17A Orchestrates Lung Injury and Remodeling Through p53 and uPA System Crosstalk.

Alveolar inflammation, elevated interleukin-17A (IL-17A), and fibrin deposition are common features in all forms of lung injury followed by fibrotic repair. Type II alveolar epithelial cell (AEC) viability, regulated by tumor suppressor protein p53 and changes in uPA-mediated fibrinolysis, has been linked to lung injury and pulmonary fibrosis (PF). Nevertheless, mechanistic details linking increased IL-17A with p53 and PAI-1 to lung injury and remodeling remain unclear. We found that IL-17A and its receptor (IL-17RA) are induced during various lung injuries. IL-17A augments IL-17RA, p53 and do

Inhibition of elastin degradation alleviates joint degeneration in aging mice, dogs, and human models.

Extracellular matrix degradation is a fundamental pathological feature of osteoarthritis, while the roles of degraded matrix remain largely unknown. We previously showed that serum elastin fragments were a systemic aging driver. Here, we found that elastin fragments were upregulated in synovial fluid in dual-center osteoarthritis patients. Elastin fragments actively impaired joint tissue in mice and human explants. Mechanistically, a specific elastin motif containing Valine-Glycine-Valine-Alanine-Proline-Glycine (VGVAPG) oligopeptide (E-motif) promoted macrophage secretion of inflammatory fact

Microneedle patches deliver targeted GLP-1RAs-loaded nanoparticles for the treatment of rheumatoid arthritis.

Despite the strong immune-modulatory benefits, there has been a lag in using glucagon-like peptide-1 receptor agonists (GLP-1RAs) in rheumatoid arthritis (RA) due to inefficient articular delivery and poorly understood direct cartilage protection. In this paper, we addressed these critical issues through the mechanism-informed design of a hierarchical drug delivery system. We elucidated previously unnoted ways in which GLP-1RAs curbed RA pathology via direct suppression of the cGAS-STING signaling pathway to inhibit chondrocyte ferroptosis and concomitantly induced macrophage M2 polarization.

Cartilage targeted grape skin-derived extracellular vesicles ameliorate osteoarthritis by attenuating chondrocyte senescence.

Osteoarthritis (OA) is predominantly driven by chondrocyte senescence, accompanied by progressive cartilage degradation. Facing the substantial worldwide impact of this condition, creating sustainable and economical therapies has become increasingly urgent. Plant-derived extracellular vesicles (PEVs) present a highly attractive option, notably because they are readily sourced, exhibit minimal immunogenicity, boast excellent biocompatibility, and can be produced at scale. Crucially, these nanovesicles facilitate cell-to-cell signaling by shuttling inherent bioactive cargo. In our study, we prop

Cartilage-targeted siMMP13 delivery using cap-modified tetrahedral framework nucleic acids for Osteoarthritis therapy.

Osteoarthritis (OA) is a degenerative joint disorder characterized by cartilage deterioration, with limited therapeutic options available to halt disease progression. A key driver of this pathology is the chondrocyte overexpression of matrix metalloproteinase 13 (MMP13), an enzyme that degrades type II collagen (Col2). Although small interfering RNA (siRNA) mediated silencing of MMP13 has shown therapeutic potential, its clinical translation is hindered by poor stability, inadequate cartilage infiltration, and a short joint residence time. To overcome these barriers, we engineered a cartilage-

Cascade-Responsive MXene@Cu-MOF Heterostructure Integrates Antioxidant Activity, Infection Control, and Vascularization for Tracheal Repair.

Extensive tracheal repair is limited by oxidative injury, infection, and insufficient vascularization with delayed cartilage maturation. We developed cascade-responsive MXene@Cu-MOF heterostructures within gelatin methacryloyl (GelMA) hydrogels to enable staged tracheal repair. Copper-based metal-organic framework (Cu-MOF) nanocrystals grown in situ on ultrathin Ti3C2Tx MXene form an integrated 2D/3D heterointerface, preventing restacking and ensuring uniform "backpack" distribution on a photothermal skeleton. The platform acts in three stages: 1) MXene scavenges radicals, reducing oxidative s

Long-acting triamcinolone acetonide prodrug microcrystals for sustained intra-articular treatment of osteoarthritis.

Intra-articular injection of triamcinolone acetonide (TA) is widely used to treat osteoarthritis (OA), a prevalent chronic joint disease imposing substantial clinical and socioeconomic burdens. However, current long-acting TA formulations suffer from limited therapeutic duration or require invasive administration, compromising patient compliance. To address this challenge, we developed two TA prodrug microcrystals (MCs)-PKTA and PETA-by conjugating hydrophobic palmitoyl groups via acid-sensitive ketal or enzymatically cleavable ester linkages, respectively. Both formulations exhibited high dru

WIPI1-mediated mitophagy dysfunction in ventricular remodeling associated with long-term diabetes mellitus.

BACKGROUND: WIPI1 is a member of the WD-repeat protein family that interacts with phosphoinositides and plays a crucial role in autophagy. This study investigated how WIPI1-mediated mitophagy dysfunction contributes to ventricular remodeling in rat and mouse models of diabetes mellitus. METHODS: The study utilized a 32-weeks diabetic animal model to simulate long-term diabetic conditions. AAV9-cTNT-WIPI1 vectors were employed to overexpress WIPI1 in the myocardium. Cardiac function was assessed by echocardiography. Mitochondrial membrane potential was assessed using JC-1 dye. Oxygen consumpti

Lactoferrin Stimulates Chondrogenesis and Promotes Healing of the Auricular Elastic Cartilage.

Ear reconstruction surgeries for congenital deformities and trauma are common, highlighting the need for improved cartilage regeneration. Lactoferrin (LF), a natural and cost-effective protein, is promising due to its anti-inflammatory, antimicrobial, and prochondrogenic properties. This study investigates the effects of LF on the viability, proliferation, and chondrogenesis of rabbit auricular chondrocytes. For in vitro studies, auricular chondrocytes were cultured for three passages, after which 3D pellets were formed. LF significantly increased chondrocyte metabolic activity by 1.5 times at

The dual effects of iron deficiency on intervertebral disc development and on acute injury-induced disc degeneration.

Clinical studies have reported that anemic patients with lower back pain exhibit significantly more severe lumbar intervertebral disc (IVD) degeneration compared to non-anemic individuals. Given that iron deficiency (ID) is the leading cause of anemia, we hypothesized that ID may disrupt IVD metabolism-particularly extracellular matrix synthesis-thereby contributing to aberrant disc development and degeneration. To investigate this, we established an iron-deficiency anemia (IDA) rat model via an iron-deficient diet and induced IVD degeneration through needle puncture. Degenerative changes were

Remodeling Cellular Uptake via Inducing Clustering of Urokinase Receptors in Hydrogel Microspheres for Alleviating Cartilage Senescence.

Cellular senescence impairs clathrin-mediated endocytosis (CME), partly due to reduced membrane fluidity and disrupted lipid raft composition, which hinder receptor clustering and limit drug uptake. To overcome this, a urokinase receptor (µPAR) antibody-modified cationic liposome was developed loaded with plasmid of DNA-repairing enzyme Sirt6 (anti-µPAR@Lipo-Sirt6), targeting the overexpressed µPAR on senescent chondrocytes to promote receptor clustering and enhance CME. Compared to Lipo-Sirt6, anti-µPAR@Lipo-Sirt6 induced µPAR clustering and enhanced cellular uptake via the CME pathway. The r

TEA domain transcription factor 1(TEAD1) induces cardiac fibroblasts cells remodeling through BRD4/Wnt4 pathway.

Cardiac fibroblasts (CFs) are the primary cells tasked with depositing and remodeling collagen and significantly associated with heart failure (HF). TEAD1 has been shown to be essential for heart development and homeostasis. However, fibroblast endogenous TEAD1 in cardiac remodeling remains incompletely understood. Transcriptomic analyses revealed consistently upregulated cardiac TEAD1 expression in mice 4 weeks after transverse aortic constriction (TAC) and Ang-II infusion. Further investigation revealed that CFs were the primary cell type expressing elevated TEAD1 levels in response to press

7α,25-Dihydroxycholesterol-Induced Oxiapoptophagic Chondrocyte Death via the Modulation of p53-Akt-mTOR Axis in Osteoarthritis Pathogenesis.

This study aimed to exploring the pathophysiological mechanism of 7α,25-dihydroxycholesterol (7α,25-DHC) in osteoarthritis (OA) pathogenesis. 7α,25-DHC accelerated the proteoglycan loss in ex vivo organ-cultured articular cartilage explant. It was mediated by the decreasing extracellular matrix major components, including aggrecan and type II collagen, and the increasing expression and activation of degenerative enzymes, including matrix metalloproteinase (MMP)-3 and -13, in chondrocytes cultured with 7α,25-DHC. Furthermore, 7α,25-DHC promoted caspase dependent chondrocytes death via extrinsic

Bio-3D printing iPSC-derived human chondrocytes for articular cartilage regeneration.

Osteoarthritis is a leading cause of pain and joint immobility, the incidence of which is increasing worldwide. Currently, total joint replacement is the only treatment for end-stage disease. Scaffold-based tissue engineering is a promising alternative approach for joint repair but is subject to limitations such as poor cytocompatibility and degradation-associated toxicity. To overcome these limitations, a completely scaffold-free Kenzan method for bio-3D printing was used to fabricate cartilage constructs feasible for repairing large chondral defects. Human induced pluripotent stem cell (iPSC
