Single-locus chromatin memory enables flexible spatial fate specification in the Drosophila visual system.

Spatial patterning generates neuronal diversity by compartmentalizing progenitors into domains with distinct molecular identities. However, these patterning cues are often transient in neurogenic domains, raising the question of how spatial information can be preserved and how rigidly it constrains neuronal fates. Here we show that a single-locus chromatin memory in the medulla of the Drosophila optic lobe faithfully preserves spatial identity while conferring the flexibility for specific neuronal fates to bypass spatial restriction. Medulla progenitors are partitioned into three spatial domai

Spontaneous coronary artery dissection as a threshold disorder of coronary wall integrity.

Spontaneous coronary artery dissection (SCAD) is an increasingly recognized cause of non-atherosclerotic acute myocardial infarction, predominantly affecting young and middle-aged women and patients during pregnancy or the postpartum period. Although major progress has been made in clinical recognition, angiographic diagnosis, conservative management, and extracoronary vascular screening, the molecular basis of SCAD remains incompletely understood. Here, we develop a provisional, evidence-integrating framework that conceptualizes SCAD as a threshold disorder of coronary wall integrity rather t

Spatial instruction of tissue immunity.

Tissue immunity must meet the architectural and physiological demands of each organ, from viral entry in the respiratory tract to immune surveillance in the gastrointestinal mucosa. Recent advances in spatial technologies and computational biology now allow us to map entire immune communities in situ, capturing not only their composition but their positional logic, connectivity patterns, and local transcriptional landscapes. These tools are revealing that immune function is not evenly distributed but is patterned along regionalized cytokine gradients, anatomical landmarks, and physical niches

Utilising Single Cell Analysis and Spatial Transcriptomics to Elucidate Molecular Mechanisms of Wound Healing: A Literature Review.

The use of single-cell RNA sequencing and spatial transcriptomics has recently challenged and refined concepts in the field of wound healing by enabling detailed characterisation of cellular heterogeneity while providing spatial context. Previously used separately, these methods are now increasingly applied together due to the advent of newer platforms, enabling delineation of discrete subpopulations of cutaneous cells, including epidermal basal stem cells, fibroblasts, neutrophils, macrophages, and T cells. Detailed transcriptional profiling of these subpopulations has revealed upregulation o

Liquid Biopsy and Nonendoscopic Biomarkers for Precision Early Detection of Esophageal Cancer: Evidence, Risk Stratification and Translational Challenges.

Esophageal cancer remains one of the most lethal malignancies worldwide, largely because most tumors are detected after symptoms develop. Esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC) differ in epidemiology, precursor lesions and molecular biology, and therefore require subtype-specific early-detection pathways. This narrative critical review used a structured PubMed/MEDLINE search and classifies evidence by intended use: population risk triage for ESCC in high-incidence regions; targeted case finding for previously undiagnosed Barrett esophagus; surveillance an

Distinguishing Meniere's disease from vestibular migraine: A comprehensive review of symptoms and diagnostic insights.

Meniere's disease and vestibular migraine are common episodic vestibular syndromes whose overlapping vertigo, auditory, and migraine-associated features can complicate differential diagnosis. In this narrative review, we compared the diagnostic criteria, symptom patterns, pathophysiology, and complementary test findings of these syndromes. Vestibular migraine is diagnosed clinically based on recurrent vestibular symptoms, migraine history, temporal association with migraine features, and exclusion of alternative causes; audiological and vestibular evaluations are not required. In contrast, dia

Single-Cell Insights into Medicinal Plant Development and Metabolism.

Medicinal plants are major sources of therapeutic natural products, yet the cell-type-specific organization that governs metabolite biosynthesis, transport, and storage remains imperfectly resolved by organ-level omics. This review synthesizes studies published up to June 2026 that used single-cell, single-nucleus, spatial, metabolomic, and epigenomic approaches to medicinal plant systems, following a PRISMA-guided literature search across PubMed, Web of Science, Scopus, and CNKI. Emerging evidence shows that specialized metabolism is organized through discrete and often rare cell populations,

Platelet-neutrophil niches associate with epidermal immune activation and systemic inflammation in psoriatic disease.

Platelets are increasingly recognised as inflammatory mediators that influence leukocyte behaviour, yet their spatial organisation and contribution to psoriatic skin inflammation remain incompletely understood. Here, spatial transcriptomics was used to map platelet-leukocyte niches (PLNi) across inflammatory skin diseases, contrasting psoriasis (PsO) with atopic dermatitis (AD). PLNi were selectively expanded in PsO lesions, where platelet-neutrophil co-localisation defined transcriptionally active regions enriched for stress and inflammatory mediators. PsO and psoriatic arthritis (PsA) shared

Receptor tyrosine kinase signaling in mammalian eye development.

Receptor tyrosine kinase (RTK) signaling is a central mechanism by which local tissue interactions are translated into coordinated morphogenesis during eye development. The vertebrate eye provides a uniquely tractable system for studying this logic because it is assembled from neural epithelium, surface ectoderm, neural crest-derived mesenchyme, vascular endothelium, and specialized epithelial and endothelial derivatives across a prolonged developmental timeline. In this review, we synthesize how major RTK families, including FGF-FGFR, EGF-ErbB, PDGF-PDGFR, VEGF-VEGFR, Eph-ephrin, ANGPT-TEK/Ti

Multiscale profiling of tyrosine kinase inhibitor cardiotoxicity reveals mechanosensitive ion channel PIEZO1 as cardioprotective.

Tyrosine kinase inhibitors (TKIs) have improved cancer outcomes but are limited by cardiovascular toxicity, most notably hypertension and heart failure. The underlying mechanisms remain poorly understood, hindering the development of protective strategies. Here, we investigated the role of endothelial mechanotransduction in mediating vascular and cardiac injury caused by the vascular endothelial growth factor receptor-targeting TKI sunitinib. Using patient-specific induced pluripotent stem cell-derived endothelial cells (iPSC-ECs) and a mouse model of TKI-induced hypertension, we identified do

Genetic Crosstalk Between Type 1 Diabetes and Sjögren's Syndrome: A Systematic Exploration of Risk Genes and Common Pathways.

Sjögren's Syndrome (SS) and Type 1 Diabetes (T1D) are autoimmune disorders that can co-occur in patients, leading to complex clinical presentations. Despite observational evidence of their co-occurrence, the underlying genetic mechanisms remain poorly understood. To investigate the shared genetic factors and pathways between SS and T1D, we conducted a comprehensive analysis using multiomic approaches. Conditional and conjunctional false discovery rate analyses were performed to identify genetic polygenicity and overlap between the two diseases. Functional annotation and pathway analysis identi

Cognitive Decline in Chronic Inflammatory Conditions: Exploring Links Between Systemic Inflammation and Neurodegeneration.

Chronic inflammatory diseases (CIDs), such as rheumatoid arthritis (RA), obesity, type 2 diabetes mellitus (T2DM), systemic lupus erythematosus (SLE), fibromyalgia (FM), and chronic infection, are risk factors for neurodegenerative diseases, such as Alzheimer's disease (AD) and Parkinson's disease (PD). This review synthesizes evidence from longitudinal cohort studies and clinical trials, highlighting the contrasting evidence to explain the complex association between systemic inflammation and neurodegeneration. These important mechanisms are disruption of the blood-brain barrier, microglial a

IKAROS Gene Regulatory Network Reveal ERG as a Vulnerability in B-cell Acute Lymphoblastic Leukemia.

B-cell acute lymphoblastic leukemia (B-ALL) is driven by transcriptional dysregulation that impairs B-cell differentiation and sustains leukemic growth. A defining feature of high-risk B-ALL is mutations in IKZF1, which encodes the tumor suppressor IKAROS. Here, we map IKAROS gene regulatory networks in IKZF1-mutated Ph B-ALL using an inducible IKAROS system and multi-omic profiling. IKAROS restoration reprograms chromatin accessibility and transcriptional control, shifting regulation from an ETS-dominated state to one enriched for B-cell lineage factors. Among repressed transcription factors,

Longitudinal paired liver biopsies and transcriptome profiling in alcohol-associated hepatitis reveal dynamic changes in cellular senescence.

BACKGROUND AND AIMS: Alcohol-associated hepatitis (AH) is an acute form of alcohol-related liver disease (ALD) with high mortality rate. AH is histologically characterised by cellular processes, including steatosis, inflammation and cell death. Apoptosis is the most studied form of cell death in AH; however, the role of cellular senescence, another response to cellular injury, in AH is unknown. Here, we explore the mechanisms of ALD pathophysiology and describe the role of senescence in AH. METHODS: We performed RNA sequencing and bioinformatics analysis of 0- and 28-day transjugular liver bi

Genetic and environmental factors contributing to anophthalmia and microphthalmia: Current understanding and future directions.

Anophthalmia is defined as a complete absence of one eye or both the eyes, while microphthalmia represents the presence of a small eye within the orbit. The estimated birth prevalence for anophthalmia is approximately 3 per 100000 live births, and for microphthalmia, it is around 14 per 100000 live births. However, combined evidence suggests that the prevalence of these malformations could be as high as 30 per 100000 individuals. Microphthalmia is reported to occur in 3.2% to 11.2% of blind children. Anophthalmia and microphthalmia (A/M) are part of a phenotypic spectrum alongside ocular colob

A complex mechanism translating variation of a simple genetic architecture into alternative life histories.

Understanding the processes that link genotype to phenotype is a central challenge in biology. Despite progress in discovering genes associated with ecologically relevant traits, a poor understanding of the processes and functions via which molecules mediate evolutionary differences leaves us critically far from linking proximate and ultimate causes of evolution. This knowledge gap is particularly large in multifaceted phenotypes of ecological relevance such as life histories where multiple traits covary and influence fitness. In Atlantic salmon (Salmo salar), variation in a key life-history t

A trade-off in evolution: the adaptive landscape of spiders without venom glands.

BACKGROUND: Venom glands play a key role in the predation and defense strategies of almost all spider groups. However, the spider family Uloboridae lacks venom glands and has evolved an adaptive strategy: they excessively wrap their prey directly with spider silk instead of paralyzing it first with toxins. This shift in survival strategy is very fascinating, but the genetic underpinnings behind it are poorly understood. RESULTS: Spanning multiple spider groups, we conducted multiomics analyses on Octonoba sinensis and described the adaptive evolution of the Uloboridae family at the genome lev…

Targeting immune-fibroblast cell communication in heart failure.

Inflammation and tissue fibrosis co-exist and are causally linked to organ dysfunction1,2. However, the molecular mechanisms driving immune-fibroblast cell communication in human cardiac disease remain unexplored and there are at present no approved treatments that directly target cardiac fibrosis3,4. Here we performed multiomic single-cell gene expression, epitope mapping and chromatin accessibility profiling in 45 healthy donor, acutely infarcted and chronically failing human hearts. We identified a disease-associated fibroblast trajectory that diverged into distinct populations reminiscent

CD163+ macrophages in mantle cell lymphoma induce activation of prosurvival pathways and immune suppression.

Mantle cell lymphoma (MCL) is dependent on a supportive tumor immune microenvironment (TIME) in which infiltration of CD163+ macrophages has a negative prognostic impact. This study explores how abundance and spatial localization of CD163+ cells are associated with the biology of MCL, using spatial multiomic investigations of tumor and infiltrating CD163+ and CD3+ cells. A total of 63 proteins were measured using GeoMx digital spatial profiling in tissue microarrays from 100 diagnostic MCL tissues. Regions of interest were selected in tumor-rich and tumor-sparse tissue regions. Molecular profi
