RNA-Seq analysis of the Sclerotinia homoeocarpa--creeping bentgrass pathosystem.

Sclerotinia homoeocarpa causes dollar spot disease, the predominate disease on highly-maintained turfgrass. Currently, there are major gaps in our understanding of the molecular interactions between S. homoeocarpa and creeping bentgrass. In this study, 454 sequencing technology was used in the de novo assembly of S. homoeocarpa and creeping bentgrass transcriptomes. Transcript sequence data obtained using Illumina's first generation sequencing-by-synthesis (SBS) were mapped to the transcriptome assemblies to estimate transcript representation in different SBS libraries. SBS libraries included

A repurposing approach identifies off-patent drugs with fungicidal cryptococcal activity, a common structural chemotype, and pharmacological properties relevant to the treatment of cryptococcosis.

New, more accessible therapies for cryptococcosis represent an unmet clinical need of global importance. We took a repurposing approach to identify previously developed drugs with fungicidal activity toward Cryptococcus neoformans, using a high-throughput screening assay designed to detect drugs that directly kill fungi. From a set of 1,120 off-patent medications and bioactive molecules, we identified 31 drugs/molecules with fungicidal activity, including 15 drugs for which direct antifungal activity had not previously been reported. A significant portion of the drugs are orally bioavailable a

Negative regulation of yeast WASp by two SH3 domain-containing proteins.

BACKGROUND: WASp family proteins promote actin filament assembly by activating Arp2/3 complex and are regulated spatially and temporally to assemble specialized actin structures used in diverse cellular processes. Some WASp family members are autoinhibited until bound by activating ligands; however, regulation of the budding yeast WASp homolog (Las17/Bee1) has not yet been explored. RESULTS: We isolated full-length Las17 and characterized its biochemical activities on yeast Arp2/3 complex. Purified Las17 was not autoinhibited; in this respect, it is more similar to SCAR/WAVE than to WASp prot

Two verprolin homology domains increase the Arp2/3 complex-mediated actin polymerization activities of N-WASP and WAVE1 C-terminal regions.

WASP family proteins induce actin polymerization through a C-terminal verprolin homology, cofilin homology, and acidic (VCA) region by activating the Arp2/3 complex. The N-WASP VCA region is the most potent activator of the Arp2/3 complex. In addition, full-length WAVE1 and a WAVE1 VCA fragment show differential activity. The mechanisms underlying these differences are poorly understood. We examined the activities of various N-WASP and WAVE1 VCA mutant proteins with several types of fusion moieties. When fused to GST, maltose-binding protein, or the WAVE1 proline-rich domain, N-WASP VCA and WA

The interplay of Hrd3 and the molecular chaperone system ensures efficient degradation of malfolded secretory proteins.

Misfolded proteins of the secretory pathway are extracted from the endoplasmic reticulum (ER), polyubiquitylated by a protein complex termed the Hmg-CoA reductase degradation ligase (HRD-ligase), and degraded by cytosolic 26S proteasomes. This process is termed ER-associated protein degradation (ERAD). We previously showed that the membrane protein Der1, which is a subunit of the HRD-ligase, is involved in the export of aberrant polypeptides from the ER. Unexpectedly, we also uncovered a close spatial proximity of Der1 and the substrate receptor Hrd3 in the ER lumen. We report here on a mutant

Carbohydrate recognition domain of surfactant protein D mediates interactions with Pneumocystis carinii glycoprotein A.

Pneumocystis carinii continues to cause severe pneumonia in immunocompromised patients. Surfactant protein D (SP-D), a lung collectin, markedly accumulates during P. carinii pneumonia and binds to glycoprotein A (gpA) on the surface of P. carinii, thereby enhancing interactions with alveolar macrophages. Herein, we report the structural basis of the interaction of SP-D with gpA. We demonstrate that natural SP-D binds to purified gpA in the presence of 2 mM calcium in a saturable, concentration-dependent manner, which is abolished by 10 mM ethylenediaminetetraacetic acid. Impact of salivary media on sustained silver release and the structural stability of silver nanomodified polymethyl methacrylate denture base resin.

STATEMENT OF PROBLEM: Silver nanoparticle (AgNP)-modified denture base materials can reduce biofilm-induced oral inflammatory lesions such as denture stomatitis because of the release of bioactive silver ions. However, whether the release of silver ions is sustained when exposed to human saliva and their effect on the molecular structure of the denture base resin are unclear. PURPOSE: The purpose of this in vitro study was to assess the influence of salivary media on the sustained release of silver ions from a modified-denture base resin and to determine their effect on the molecular structur

Invasive fungal diseases in immunocompetent adults: emerging patterns and clinical challenges.

BACKGROUND: Invasive fungal diseases (IFDs) have traditionally been regarded as complications of classical immunosuppression. However, IFDs are increasingly recognized in adults who are apparently immunocompetent or non-neutropenic, particularly in the setting of severe viral pneumonia, critical illness, diabetes, structural lung disease, surgery, trauma or burns, injection or inhalational drug use, invasive devices, broad-spectrum antibiotics, and previously unrecognized immune dysregulation. OBJECTIVE: This narrative review summarizes emerging patterns and clinical challenges of IFD in appa

Dual Evidence of Trichoderma pleuroti Antagonism against Pleurotus pulmonarius: Direct Inhibition and Volatile-Mediated Bioassays with Volatilome Fingerprinting.

Trichoderma is known to cause green mold contamination, which severely threatens Pleurotus pulmonarius cultivation in tropical regions, including Malaysia, and can lead to major yield and economic losses. In this study, the contact-dependent and contactless antagonistic interactions between Trichoderma pleuroti and P. pulmonarius were investigated, and associated morphological changes and volatile profiles were characterized. Dual-culture assays recorded rapid overgrowth of P. pulmonarius by T. pleuroti, leading to growth suppression and mycelial discoloration. Light and scanning electron micr

Advances in Targeting BCR-ABLT315I Mutation with Imatinib Derivatives and Hybrid Anti-Leukemic Molecules.

Resistance to imatinib remains a therapeutic challenge, largely driven by point mutations within the kinase domain of the BCR-ABL, among which the T315I substitution constitutes the most clinically significant barrier. Ponatinib effectively inhibits this mutant form but is limited by dose-dependent cardiovascular toxicity, prompting efforts to develop safer and more selective agents. Recent advances highlight aminopyrimidine-derived scaffolds and their evolution into thienopyrimidines, oxadiazoles, and pyrazines with improved activity against BCR-ABLT315I. Further progress has been achieved wi

Effects of salinity on photosynthetic characteristics, rhizosphere microbial community structure, and diversity of Ulmus pumila L.

Salt stress is a key abiotic limiting factor restricting agricultural productivity, making the screening and breeding of salt-tolerant plants essential for alleviating soil salinization. Ulmus pumila, a native Chinese tree with fast growth, good wood properties and high tolerance to salt stress and drought, was investigated to reveal its salt adaptation and underlying mechanisms. This study applied phenotypic identification, physiological determination and high-throughput sequencing to analyze growth traits and rhizosphere microbial communities under low, moderate and high salt stress. GcrR in Streptococcus mutans Inhibits the Morphological Transition of Candida albicans Through Ras1-cAMP/PKA Pathway in the Cross-Kingdom Biofilms.

The dual-species biofilms formed by Streptococcus mutans and Candida albicans exhibit enhanced cariogenic potential due to the production of extracellular polysaccharides (EPSs). The response regulator GcrR in S. mutans negatively regulates EPS synthesis, but its impact on dual-species biofilms remains unclear. The objective of the present study was to investigate the effect of GcrR on the cariogenicity of cross-kingdom biofilms established by S. mutans and C. albicans. The crystal violet (CV) staining was conducted to evaluate the dual-species biofilm formation. The anthrone-sulfuric acid ass

Combined application of organic and microbial amendments reshapes soil microbiome diversity and improves soybean growth performance.

Agricultural practices, including organic amendments and microbial inoculants, strongly influence soil microbial communities, which play a crucial role in maintaining soil function and structure. To identify an effective strategy for improving soil microbiome diversity and crop production, we assessed and compared the soil microbial community in fields treated with cattle manure amendment (MA), microbial inoculant (MI), and a mixture of cattle manure and microbial inoculant (MM), while an untreated field served as the control. We then evaluated treatment effects on soybean production. Microbia

Insights into novel benzo[h]quinoline-based 2-thioxothiazolidin-4-one and thiazol-4-one hybrids as potential antimicrobial, and antibiofilm candidates: Design, synthesis, molecular modeling, and in-silico ADME studies.

The growing crisis of antimicrobial resistance urges the immediate development of new therapeutic pharmaceuticals that can overcome current resistance mechanisms, including biofilm formation. This study included the synthesis and strategic design of a novel set of quinoline-based scaffolds (3-11) to enhance interactions with the bacterial Peptide Deformylase (PDF) enzyme. The compounds underwent extensive in vitro biological assessment, comprising initial inhibition zone (IZ) screening, followed by quantitative minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC)

Second-Order Chemometric Classification of Candida Species Using Excitation-Emission Matrix Fluorescence Spectroscopy: A Proof-of-Concept Study.

Microorganisms of the genus Candida are responsible for a wide range of fungal infections and are associated with increased morbidity and mortality rates. Although several clinical methods are available for the diagnosis of candidiasis, they have limitations related to response time and laboratory complexity. In this study, spectroscopic techniques based on excitation-emission matrix (EEM) fluorescence were explored in combination with higher-order chemometric methods, such as PARAFAC and Tucker3, coupled with classification algorithms, including Quadratic Discriminant Analysis (QDA), for the

Enhanced phytoextraction of manganese and cadmium by Polygonum lapathifolium L. via Enterobacter sp. inoculation: Roles of rhizosphere regulation and endophytic microbiome assembly.

Plant growth-promoting bacteria (PGPB) can enhance phytoremediation, but whether inoculation alters rhizosphere microbial abundance and elicits tissue-specific endophytic responses remains unclear. Here, Enterobacter sp. was applied to improve Mn/Cd phytoextraction by Polygonum lapathifolium L. Inoculation decreased rhizosphere pH and increased acid-extractable Mn and Cd by 11.8% and 45.5%, respectively, accompanied by enhanced nutrient availability and C, N and P cycling enzyme activities. It also altered rhizosphere bacterial and fungal community structure, increased their absolute abundance

The role of the transcription factor FsBrlA in regulating sporulation, cell wall integrity stress response, and pathogenicity in Fusarium solani.

BrlA is a multifunctional transcription factor identified in specific clades of filamentous fungi, notably within the Hyphomycetes. It is widely involved in sporulation, cell wall stress response and pathogenicity, and plays an important role in fungal growth and development. To investigate the biological function of the FsbrlA in Fusarium solani, this study employed a split-marker homologous recombination strategy to directionally construct FsbrlA knockout and complementation strains. Through analyses of growth and development phenotypes, determination of cell wall components and trehalose co

Potential Activity of 6-Pentyl-α-pyrone as an Antiviral for Bovine Coronavirus.

During infection in vitro with the strain 438/06 of bovine coronavirus (BCoV), a β-coronavirus similar to severe acute respiratory syndrome (SARS) CoV-2, treatment with 6-pentyl-α-pyrone (6PP), a fungal metabolite obtained from Trichoderma atroviride, was recently shown to influence viral load by reducing viral entry. Herein, the ability of 6PP to counteract the BCoV infection was further investigated both in vitro and in silico. Following the BCoV (strain 282/23) infection in bovine (MDBK) cells, the 6PP in co-treatment increased cell viability, reduced morphological signs of cell death, and

A tomato MIK2-clade receptor is involved in the perception of a Fusarium-derived elicitor.

Plants continuously monitor their environment to adapt to threats affecting health and fitness. Cell-surface receptors detect conserved microbe-associated molecular patterns (MAMPs) or endogenous immunogenic signals to activate signaling pathways that induce broad-spectrum disease resistance, known as pattern-triggered immunity (PTI). In Arabidopsis thaliana, the leucine-rich repeat receptor kinase MIK2 emerged as a versatile receptor involved in perceiving endogenous SCOOP peptides as well as potential MAMPs derived from Fusarium and related fungi. While SCOOP perception appears restricted to

Blood Culture-Negative Infective Endocarditis: A Review.

Blood culture-negative infective endocarditis (BCNIE) represents a diagnostically challenging subset of infective endocarditis in which routine blood cultures remain negative despite fulfillment of Duke-ISCVID diagnostic criteria. BCNIE arises primarily from prior antibiotic exposure, infection with fastidious or nonculturable organisms, or noninfectious conditions that mimic endocarditis. Common fastidious pathogens include Coxiella burnetii, Bartonella species, Brucella species, Tropheryma whipplei, fungi, and nutritionally variant streptococci. Because delayed pathogen identification may po
