Camel milk fat-based human milk fat substitute.

Human milk is the gold standard for infant nutrition. This study aimed to develop a camel milk fat-based human milk lipid mimetic whose fatty acid (FA) composition, structural properties, and in vitro digestibility more closely resemble those of human milk fat than conventional vegetable oil-based formulas. Camel milk (CM) fat was physically blended with sunflower, rapeseed, palm kernel, fish oils, and 1,3-dioleoyl-2-palmitoylglycerol using a computational optimization model to approximate the total and sn-2 FA profiles of Egyptian human milk. The resulting formulation, together with three com

The isoflavone metabolites, O-desmethylangolensin and (S)-equol, relax isolated arteries ex vivo and decrease arterial blood pressure in vivo.

Dietary isoflavones are suggested to have positive cardiovascular effects. Their intestinal absorption is low, but they undergo extensive gastrointestinal metabolism by gut bacteria, resulting in metabolites with higher bioavailability. However, there is limited data on the effect of individual pure isoflavone metabolites on the vascular system. Therefore, four of these metabolites were first screened ex vivo for vasorelaxant properties on isolated rat aortic rings. The most potent of these metabolites, namely O-desmethylangolensin (O-DMA) and (S)-equol, had EC50 values of 49.3 and 59.1 μΜ, re

Nephrotoxicity and innovative risk assessment of cinnabar-containing An-Gong-Niu-Huang Wan based on bioavailability and speciation using on an IPBET/small intestinal organ-on-a-chip.

An-Gong-Niu-Huang Wan (AGNHW) is a traditional Chinese medicine preparation containing cinnabar (HgS). However, its clinical use has been controversial due to concerns regarding mercury nephrotoxicity. To evaluate the renal toxicity, mice were administered AGNHW, cinnabar, methylmercury (MeHg), and mercuric chloride (HgCl₂) for 28 days. Blood biochemistry, histopathology, and the expression of nephrotoxicity-sensitive genes were assessed. To assess the health risks associated with AGNHW, we established an innovative Physiologically Based Extraction Test (IPBET) /small intestinal organ-on-a-chi

Chitosan oligosaccharide-coated carrier-free evodiamine-curcumin nanoassemblies enhance oral delivery and anti-atherosclerotic efficacy.

Oxidative stress and dysregulated vascular smooth muscle cell (VSMC) proliferation drive atherosclerotic (AS) progression and plaque development. Natural phytochemicals, including evodiamine (Evo) and curcumin (Cur), show anti-atherosclerotic potential, but poor aqueous solubility and insufficient intestinal absorption limit their oral use. Here, Evo and Cur were co-assembled into carrier-free Evo-Cur nanoparticles (EC NPs) and coated with chitosan oligosaccharide (COS) to obtain COS-coated EC nanoparticles (COS@EC NPs). Spectroscopic analyses and molecular dynamics simulations indicated that

Real-world use of intravenous ferric derisomaltose in Brazilian patients with iron deficiency anemia.

BACKGROUND: Iron deficiency anemia remains a major global health concern, especially among women, children, and people with chronic illnesses. Oral iron therapy is the standard treatment, but it is often limited by gastrointestinal side effects or poor absorption. Intravenous ferric derisomaltose is a newer formulation that allows high-dose, rapid iron replenishment with a favorable safety profile. This study aimed to assess the effectiveness and safety of intravenous ferric derisomaltose in patients with iron deficiency anemia who could not tolerate or did not respond to oral iron. METHODS:

Pediococcus acidilactici PA-1 Alleviates Diet-Induced Obesity by Modulating Hepatic Amino Acid Metabolism and Gut Microbiota in Mice.

Obesity and type 2 diabetes mellitus are global health challenges often associated with disrupted lipid and amino acid metabolism, as well as gut microbiota dysbiosis. Probiotic interventions offer promising potential in ameliorating metabolic disorders, yet strain-specific mechanisms remain to be elucidated. In this study, a canine-derived Pediococcus acidilactici PA-1 was subjected to in vitro evaluation for probiotic properties, and its anti-obesity effects were investigated in high-fat diet (HFD)-induced obese mice. In vitro, PA-1 exhibited safety and favorable probiotic activity including

Pharmacokinetic Interaction between Imatinib and Tacrolimus in Rats.

OBJECTIVE: Tacrolimus, a calcineurin inhibitor (CNI), is the first-line treatment for chronic myeloid leukemia (CML) and advanced gastrointestinal stromal tumors (GIST). Imatinib and tacrolimus are both substrates of the hepatic enzymes CYP3A4/5 and efflux transporter P-gp, so drug-drug interactions may occur during their co-administration treatment. Therefore, this study aimed to evaluate the pharmacokinetic interaction between imatinib and tacrolimus in rats. METHODS: Rats were divided into groups I (30 mg/kg imatinib administered for 14 days), II (1.89 mg/kg tacrolimus and 30 mg/kg imatini

Bulk and surface engineered drug delivery system of repurposed drug enhances low-dose gemcitabine efficacy in pancreatic cancer.

The clinical use of gemcitabine (GEM), a frontline chemotherapeutic agent for pancreatic ductal adenocarcinoma (PDAC), is limited by its short half-life, rapid systemic clearance, associated dose-limiting toxicities and a faster development of resistance in pancreatic cancer. Aspirin (ASP), a repurposed NSAID, has been shown to sensitize PDAC cells to GEM through modulation of multiple oncogenic and inflammatory pathways. However, its clinical use is restricted by dose-dependent gastrointestinal toxicity. To overcome these challenges, we present a bulk and surface engineered oral ASP delivery

Pharmacokinetics and tolerability of liposomal synthetic cannabidiol subcutaneous depot in Holstein dairy calves.

As animal welfare concerns and pain management awareness in cattle increase, long-term analgesics can become a solution for the treatment of painful husbandry procedures. Cannabidiol (CBD) has anti-inflammatory and analgesic properties. Nevertheless, poor gastrointestinal absorption and first-pass liver metabolism results in low oral bioavailability in humans. The use of injected liposomal-CBD facilitates slow-drug-release, providing prolonged CBD plasma concentrations, with increased bioavailability. Study objectives were to determine preliminary pharmacokinetics and tolerability of a single …

Effect of 2 Forms of Tenofovir on Duodenal Enterocytes-A Hypothesis for Different Effect of Tenofovir Disoproxil Fumarate and Tenofovir Alafenamide on Body Weight and Plasma Lipids.

BACKGROUND: Tenofovir disoproxil fumarate (TDF), compared to tenofovir alafenamide (TAF), leads to lower body weight and plasma lipids by an unknown mechanism. We hypothesize that TDF, when absorbed, may damage enterocytes of the proximal duodenum, leading to reduced absorption of nutrients. METHODS: People with human immunodeficiency virus, without significant gastrointestinal symptoms, receiving a regimen containing TDF (n = 12) or TAF (n = 12), underwent esophagogastroduodenoscopies. Plasma/serum concentrations of nutrients absorbed from proximal duodenum and serum intestinal fatty acid-bi

Population pharmacokinetics and exposure-response relationship for temsavir following fostemsavir administration in treatment-experienced HIV patients.

Fostemsavir (FTR, GSK3684934; formerly BMS-663068) is a human immunodeficiency virus type 1 (HIV-1) attachment inhibitor approved for the treatment of multidrug-resistant HIV-1 infection in heavily treatment-experienced (HTE) patients failing their current antiretroviral regimen. FTR is an extended-release prodrug and is hydrolyzed by alkaline phosphatase in the gastrointestinal lumen to its active moiety, temsavir (TMR). TMR is primarily metabolized by esterase-mediated hydrolysis with contributions from cytochrome P450 (CYP) 3A4. A population pharmacokinetic (PK) analysis was performed using

Severe Impact of Omeprazole Timing on pH-Sensitive Dasatinib Absorption: Unveiling Substantial Drug-Drug Interaction.

The absorption and bioavailability of most tyrosine kinase inhibitors are affected by gastrointestinal pH as they are weak basic lipophilic drugs. Hence, concomitant use of acid reducing agents (ARAs) is frequently restricted. Particularly comedication of crystalline dasatinib (Sprycel) and proton-pump inhibitors (PPIs) should be avoided. Drug-drug interaction (DDI) studies with PPIs report approximately 40%-80% bioavailability reduction of dasatinib. Limitations in the design of these studies do not allow for assessing the near maximum DDI as timing of PPI dosing was either not reported or 22

Natural products can be potential inhibitors of metalloproteinase II from Bacteroides fragilis to intervene colorectal cancer.

Bacteroides fragilis, a gram negative and obligate anaerobe bacterium, is a member of normal gut microbiota and facilitates many essential roles being performed in human body in normal circumstances specifically in Gastrointestinal or GI tract. Sometimes, due to genetics, epigenetics, and environmental factors, Bacteroides fragilis and their protein(s) start interacting with intestinal epithelium thus damaging the lining leading to colorectal cancers (CRC). To identify these protein(s), we incorporated a novel subtractive proteomics approach in the study. Metalloproteinase II (MPII), a Bactero
