Modulatory actions of Echinococcus granulosus antigen B on macrophage inflammatory activation.

Cestodes use own lipid-binding proteins to capture and transport hydrophobic ligands, including lipids that they cannot synthesise as fatty acids and cholesterol. In E. granulosus s.l., one of these lipoproteins is antigen B (EgAgB), codified by a multigenic and polymorphic family that gives rise to five gene products (EgAgB8/1-5 subunits) assembled as a 230 kDa macromolecule. EgAgB has a diagnostic value for cystic echinococcosis, but its putative role in the immunobiology of this infection is still poorly understood. Accumulating research suggests that EgAgB has immunomodulatory properties,

Improved cotton transformation protocol mediated by Agrobacterium and biolistic combined-methods.

The combined Agrobacterium- and biolistic-mediated methods of cotton transformation provide a straightforward and highly efficient protocol for obtaining transgenic cotton. Cotton (Gossypium spp.) is the most important crop for natural textile fiber production worldwide. Nonetheless, one of the main challenges in cotton production are the losses resulting from insect pests, pathogens, and abiotic stresses. One effective way to solve these issues is to use genetically modified (GM) varieties. Herein, we describe an improved protocol for straightforward and cost-effective genetic transformation

Characterization of a novel cathepsin L-like protease from Taenia solium metacestodes for the immunodiagnosis of porcine cysticercosis.

Porcine cysticercosis is an endemic parasitic disease caused by infection with Taenia solium that is found predominantly in developing countries. In order to aid in the development of simple diagnostic approaches, identification and characterization of potential new antigens for immunodiagnostic purposes is desired. The cysteine protease family has previously been found to have important immunodiagnostic properties. These proteases are expressed as zymogens which contain a signal peptide, pro-peptide, and an active domain. Subsequent catalytic cleavage of the pro-peptide converts these zymogen

Hessian fly larval feeding triggers enhanced polyamine levels in susceptible but not resistant wheat.

BACKGROUND: Hessian fly (Mayetiola destructor), a member of the gall midge family, is one of the most destructive pests of wheat (Triticum aestivum) worldwide. Probing of wheat plants by the larvae results in either an incompatible (avirulent larvae, resistant plant) or a compatible (virulent larvae, susceptible plant) interaction. Virulent larvae induce the formation of a nutritive tissue, resembling the inside surface of a gall, in susceptible wheat. These nutritive cells are a rich source of proteins and sugars that sustain the developing virulent Hessian fly larvae. In addition, on suscept

Matrix expansion and syncytial aggregation of syndecan-1+ cells underpin villous atrophy in coeliac disease.

BACKGROUND: We studied the expression of sulphated glycosaminoglycans (GAGs) in coeliac disease (CD) mucosa, as they are critical determinants of tissue volume, which increases in active disease. We also examined mucosal expression of IL-6, which stimulates excess GAG synthesis in disorders such as Grave's ophthalmopathy. METHODS: We stained archival jejunal biopsies from 5 children with CD at diagnosis, on gluten-free diet and challenge for sulphated GAGs. We then examined duodenal biopsies from 9 children with CD compared to 9 histological normal controls, staining for sulphated GAGs, hepar

Effects of decreased O2 and elevated CO2 on survival, development, and gene expression in cowpea bruchids.

Use of modified atmospheres with depleted O(2) and/or elevated CO(2) is an environmentally friendly alternative to currently used fumigants for control of stored grain insect pests. In the present study, we examined the impact of hypoxia and hypercapnia on cowpea bruchids (Callosobruchus maculatus), a storage pest of cowpea and other legumes. Two O(2)/CO(2) combinations were used; (i) 10% O(2)+10% CO(2), (ii) 2% O(2)+18% CO(2). In both cases, N(2) was maintained at 80%, equivalent to normal atmospheric concentration. In ambient atmosphere, the rate of O(2) consumption and CO(2) output at diffe

Cross-clade protective immune responses to influenza viruses with H5N1 HA and NA elicited by an influenza virus-like particle.

BACKGROUND: Vaccination is a cost-effective counter-measure to the threat of seasonal or pandemic outbreaks of influenza. To address the need for improved influenza vaccines and alternatives to egg-based manufacturing, we have engineered an influenza virus-like particle (VLP) as a new generation of non-egg or non-mammalian cell culture-based candidate vaccine. METHODOLOGY/PRINCIPAL FINDINGS: We generated from a baculovirus expression system using insect cells, a non-infectious recombinant VLP vaccine from both influenza A H5N1 clade 1 and clade 2 isolates with pandemic potential. Environmental signals generate a differential and coordinated expression of the heme receptor gene family of Bartonella quintana.

Of all bacteria, Bartonella quintana has the highest reported in vitro hemin requirement, yet an explanation for this remains elusive. To produce diseases such as trench fever, endocarditis, and bacillary angiomatosis, B. quintana must survive and replicate in the disparate environments of the Pediculus humanus corporis (body louse) gut and the human vasculature. We previously identified a five-member family of hemin binding proteins (Hbps) synthesized by B. quintana that bind hemin on the outer surface but share no similarity to known bacterial heme receptors. In the present study, we examine

The bereft gene, a potential target of the neural selector gene cut, contributes to bristle morphogenesis.

The neural selector gene cut, a homeobox transcription factor, is required for the specification of the correct identity of external (bristle-type) sensory organs in Drosophila. Targets of cut function, however, have not been described. Here, we study bereft (bft) mutants, which exhibit loss or malformation of a majority of the interommatidial bristles of the eye and cause defects in other external sensory organs. These mutants were generated by excising a P element located at chromosomal location 33AB, the enhancer trap line E8-2-46, indicating that a gene near the insertion site is responsib

Expression of the S-coded genes of lymphocytic choriomeningitis arenavirus using a baculovirus vector.

A DNA copy of the lymphocytic choriomeningitis virus (LCMV, WE strain)S RNA species has been inserted in both orientations into plasmids containing a 7.1 kb DNA sequence of the baculovirus Autographa californica nuclear polyhedrosis virus (AcNPV). The inserts were placed behind the polyhedrin gene promoter of AcNPV and in the place of the nucleotide sequences including and flanking the polyhedrin gene translation initiation codon. The derived plasmids were used to obtain recombinant AcNPV viruses after transfection of Spodoptera frugiperda cells in the presence of infectious AcNPV DNA and the

Isolation and characterization of a cDNA encoding for a lysozyme from the gut of the reduviid bug Triatoma infestans.

We have isolated and characterised a Triatoma infestans cDNA encoding a lysozyme. A 174-bp fragment was amplified by PCR using degenerate oligodeoxyribonucleotide primers derived from the known amino acid sequences of lysozyme from other insects. This PCR fragment was used to screen a cDNA gut library of T. infestans. A clone containing the 3'-end of the lysozyme cDNA (219 bp) was isolated and sequenced. RACE was used to amplify the 5'-end of the lysozyme cDNA. After sequencing the complete lysozyme cDNA, the deduced 417 amino acid sequence showed high identity (40-50%) with other chicken-type

Pathogen promotes insect ovarian development by inducing a dual role of host ATPSyn-β.

Candidatus Liberibacter asiaticus (CLas), a pathogenic bacterium causing citrus Huanglongbing, forms a mutualistic relationship with the Asian citrus psyllid, Diaphorina citri. However, the underlying molecular mechanisms remain unclear. We find that CLas enhances the development of testes, ovaries, and mating behavior in D. citri. Transcriptome analysis of ovaries reveals that CLas infection upregulates ATP synthesis genes, increasing ATP levels in D. citri. Silencing the ATPSyn-β gene expression reduces ATP content in the ovary and triglyceride and glycogen content in the fat body, thus decr

Cardiac neurons expressing a glucagon-like receptor mediate cardiac arrhythmia induced by high-fat diet in Drosophila.

Cardiac arrhythmia leads to increased risks for stroke, heart failure, and cardiac arrest. Arrhythmic pathology is often rooted in the cardiac conduction system, but the mechanism is complex and not fully understood. For example, how metabolic diseases, like obesity and diabetes, increase the risk for cardiac arrhythmia. Glucagon regulates glucose production, mobilizes lipids from the fat body, and affects cardiac rate and rhythm, attributes of a likely key player. Drosophila is an established model to study metabolic diseases and cardiac arrhythmias. Since glucagon signaling is highly conserv

Biochemical and molecular responses of Spodoptera frugiperda to insecticide exposure: detoxification enzymes, gene expression, and genotoxic effects.

Spodoptera frugiperda (fall armyworm) is a highly destructive agricultural pest with a remarkable ability to develop resistance to insecticides, posing a severe threat to global food security. Understanding the physiological and molecular responses of this species to insecticide exposure is crucial for improving resistance monitoring and developing effective management strategies. In this study, the toxicity and the associated biochemical, molecular, and genotoxic responses of third-instar S. frugiperda larvae were investigated following exposure to four commonly used insecticides: emamectin b

Synergistic action of different molecular mechanisms causes striking levels of insecticide resistance in the malaria vector Anopheles gambiae.

Intensifying insecticide resistance in the malaria vector Anopheles gambiae poses a serious threat to the progress achieved the last decades in reducing malaria deaths in Africa. The genetic basis of insecticide resistance is often complex, involving multiple genes and mutations. However, we still lack a clear understanding of how each mechanism contributes to overall resistance and how highly resistant phenotypes arise. In this study, we generated a suite of transgenic An. gambiae strains carrying either individual mechanisms or combinations that frequently co-occur in nature. We show that co

Integrating electrophysiology, behavioral response, and docking studies to decipher odorant-binding protein function in the brinjal shoot and fruit borer, Leucinodes orbonalis Guenee.

The brinjal shoot and fruit borer, Leucinodes orbonalis Guenée, inflicts significant yield losses in brinjal, often resulting in the extensive use of insecticides. Development of insecticide resistance and ecological concerns demand safer and species-specific alternatives. This study examines plant-derived volatile organic compounds (VOCs) as potential semiochemicals for its management. The present study employed electroantennography (EAG) to examine the summated neuronal response in the antennal of unmated male and female L. orbonalis moths to host plant VOCs. Both male and female antennae sh

Discovery and taste mechanisms of novel umami peptides from edible grasshopper (Oxya chinensis sinuosa) based on machine learning, molecular docking and dynamics simulation.

This study aimed to reveal umami peptides from grasshopper, and then explored their umami perception mechanism and umami enhancement. Four potential umami peptides (GDRFL, NGWGDY, NVTY and VVNY) were isolated, characterized and screened from grasshopper protein hydrolysate through machine learning and in silico approaches. Subsequently, sensory evaluation and electronic tongue confirmed their umami taste with thresholds of 0.08-0.25 mg/mL, which were lower than that of monosodium glutamate (0.3 mg/mL). The potent umami taste of GDRFL and NGWGDY was mainly driven by their umami amino acid compo

Allelic variation in a cluster of epsilon glutathione S-transferase genes contributes to DDT and pyrethroid resistance in the major African malaria vector Anopheles funestus.

BACKGROUND: Insecticide resistance in malaria vectors is a serious challenge to malaria control and elimination. Elucidation of the role of detoxification genes in resistance is necessary to develop targeted strategies to reduce malaria burden. Glutathione S-transferase epsilon clusters (GSTe genes) are upregulated in DDT- and pyrethroid-resistant Anopheles funestus mosquitoes across Africa. However, except for GSTe2, the molecular mechanisms behind this upregulation remain unclear. Here, we established that overexpression and allelic variation of GSTe genes contribute to insecticide resistanc

Discovery of Sennidin B as a Potent Multitarget Inhibitor of Insect Chitinolytic Enzymes.

Natural products with multitarget inhibitory activity against insect chitinolytic enzymes offer the potential for developing environmentally friendly insecticides. However, most inhibitors show limited efficacy, hindering their practical application. In this study, inspired by the dimeric structure of phlegmacin B1, sennidin B, a rhein analogue, was identified as an effective inhibitor of insect chitinolytic enzymes. Sennidin B exhibited a nanomolar Ki value (80 nM) against OfChi-h from Ostrinia furnacalis, showing at least a 600-fold improvement in potency compared to rhein. Molecular dynamic
