Non-alcoholic fatty liver disease (NAFLD) has emerged as the most common chronic liver disease. NLRP3 inflammasome activation has been widely studied in the pathogenesis of NAFLD. Cathepsin B (CTSB) is a ubiquitous cysteine cathepsin, and the role of CTSB in the progression and development of NAFLD has received extensive concern. However, the exact roles of CTSB in the NAFLD development and NLRP3 inflammasome activation are yet to be evaluated. In the present study, we used methionine choline-deficient (MCD) diet to establish mice NASH model. CTSB inhibitor (CA-074) was used to suppress the ex

Autophagy, a conservative degradation process for long-lived and damaged proteins, participates in a variety of biological processes including obesity. However, the precise mechanism of action behind obesity-induced changes in autophagy still remains elusive. This study was designed to examine the role of the antioxidant catalase in high fat diet-induced changes in cardiac geometry and function as well as the underlying mechanism of action involved with a focus on autophagy. Wild-type (WT) and transgenic mice with cardiac overexpression of catalase were fed low or high fat diet for 20 weeks pr

Because metformin affects glucose and free fatty acid (FFA) metabolism in peripheral insulin target tissues, we investigated the effect of this drug in restoring a normal secretory pattern in rat pancreatic islets whose function has been impaired by chronic exposure to elevated FFA or glucose concentrations. We cultured rat pancreatic islets with or without FFA (2 mmol/l oleate/palmitate 2:1) or high glucose (16.7 mmol/l) concentrations in the presence or absence of metformin (0.25-12.5 microg/ml) and then measured insulin release, glucose utilization, glucose, and FFA oxidation. When compared

An infant died at 8 months of age with a history of developmental regression, hypotonia, severe weakness, cardiomegaly, congestive heart failure, and hepatomegaly. A diagnosis of Pompe's disease (glycogenosis type II) was established by muscle biopsy at 5 months of age. Vacuolar myopathy involved muscle fibers of histochemical type I more than type II. Many vacuoles were filled with glycogen. In addition, increased amounts of neutral lipid were demonstrated by oil red O stain, electron microscopy, and quantitative analysis. Acid alpha-1,4-glucosidase activity was demonstrated to be deficient.

Insulin resistance (IR) is a pathogenic mechanism for type 2 diabetes mellitus (T2DM), and receptor-interacting serine-threonine kinase 2 (RIPK2) participates in mediating IR. To investigate the specific role of RIPK2 in IR, this study utilized bovine serum albumin-conjugated palmitic acid (0.5 mmol/L) to create an IR model in rat L6 myotubes, followed by transfection with/without short hairpin RNA against RIPK2 (shRIPK2)/RIPK2 overexpression plasmid, and X-linked inhibitor of apoptosis (XIAP) overexpression plasmids. The expressions of RIPK2 and XIAP were evaluated using quantitative real-tim

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) stands as the most widespread chronic liver disorder globally. Histone β-hydroxybutyrylation (Kbhb)-a novel post-translational modification of histones driven by β-hydroxybutyrate (BHB)-has recently been recognized as a key epigenetic modulator. Our study aimed to explore how BHB influences the expression of hepatic lipid metabolism-associated genes in MASLD, and to determine whether histone Kbhb acts as the mechanistic mediator underlying these effects.
METHODS: For in vivo experiments, db/db mice fed a high-fat die

Background and Objectives: Pro-inflammatory diet and high-fat diet (HFD) often coexist in real-world, but their combined impact on the gut-liver axis and potential nutritional countermeasures remain insufficiently studied. This study aimed to evaluate a pro-inflammatory-high-fat composite dietary pattern on the intestine and liver in the population, and to further evaluate the protective potential of astaxanthin (ATX) in complementary experimental systems. Methods: Data from the NHANES 2005-2010 were used to construct four composite exposure groups based on the dietary inflammation index (DII)

BACKGROUND: l-type calcium channels (LTCCs) play an important role in tumorigenesis, but their expression profile, functional significance, and therapeutic potential in gastric adenocarcinoma (GAC) remain unclear. Concurrently, aberrant lipid metabolism is widely acknowledged as a key driver of GAC progression, but its upstream regulatory mechanisms have yet to be fully elucidated. This study was designed to explore the association between LTCCs and lipid metabolic reprogramming in GAC and to investigate its influence on GAC progression and clinical outcomes.
METHODS: The expression profiles

Obesity-induced osteoporosis is a growing global health challenge, necessitating more therapeutic strategies. Emerging evidence links the reduction of bone-specific type H vessels to osteoporosis due to their crucial role in angiogenesis-osteogenesis coupling. Nicotinamide Mononucleotide (NMN) is known to improve metabolic diseases, but its role in regulating type H vessels in obesity-induced osteoporosis remains unclear. An obesity-induced osteoporosis mouse model was established using a high-fat diet (HFD). In vitro, primary bone marrow mesenchymal stem cells (BMSCs) and human umbilical vein

Aims: Type 2 diabetes mellitus (T2DM) significantly elevates the likelihood of atrial fibrillation (AF); However, the precise mechanisms remain incompletely elucidated. Mitochondrial dysfunction is a hallmark of diabetic cardiomyopathy, and recent evidence suggests that activation of the cGAS-STING signaling pathway may contribute to metabolic inflammation in the atria. This study aims to investigate the role of mitochondrial DNA (mtDNA)-mediated cGAS-STING activation in promoting diabetes-associated atrial fibrillation (AF) through cardiomyocyte-macrophage crosstalk. Methods and results: Usin

Previous studies have shown that baicalein (BAI) can reduce pyroptosis of pancreatic acinar cells (PACs) in hyperlipidemic acute pancreatitis (HAP). This study aimed to elucidate the potential molecular mechanism of PAC pyroptosis mediated by BAI in HAP. A HAP rat model was established via a high-fat diet supplemented with 5% sodium taurocholate. Macrophages were treated with palmitic acid (PA). The rats and cells were treated with BAI. Molecular docking and DARTS assay were used to analyze BAI binding to HMGB1. Co-IP revealed that HMGB1 interacted with TLR4 and NLRP3 and that TLR4 interacted

BACKGROUND: The "gut-islets axis" is an important endocrine signaling axis that regulates islets function by modulating the gut microbiota and endocrine metabolism within the gut. However, the specific mechanisms and roles of the intestine in islets regulation remain unclear. Recent studies investigated that exosomes derived from gut microbiota can transport signals to remotely regulate islets β-cell function, suggesting the possibility of novel signaling pathways mediated by gut exosomes in the regulation of the "gut-islet axis.".
METHODS: The exosomes were isolated from the intestinal enter
