Enhancement of the cyclic motor activity of the ovine small intestine by lysergic acid derivatives. Mechanism and significance.

The effects of a variety of substances influencing the initiation of cyclic motor events at the gastroduodenal junction were studied in conscious sheep. A significant and long-lasting decrease in the frequency of cyclic events from approximately 100 min to 30-40 min was elicited by the intraduodenal bulb administration of the lysergic acid derivatives methylergonovine and methysergide. This effect was dose-dependent and occurred with smaller dosages than those effective when administered parenterally or more distal in the jejunum. Intraluminal administration of cyproheptadine, fonazine, and me

Specific binding of the muscarinic antagonist [3H]quinuclidinyl benzilate is not associated with preganglionic motor neurons in the dorsal motor nucleus of the vagus.

The present study evaluates the binding of [3H]quinuclidinyl benzilate, [3H]QNB, as a measure of cholinergic muscarinic binding in six areas of the rat medulla oblongata associated with the cranial nerves. In an experimental group, the right vagus nerve was severed in the neck in order to determine whether the specific muscarinic binding sites might be located on cells that contribute efferent fibers to the vagus nerve. The level of activity of choline acetyltransferase (ChAT) also was determined in the same six areas. Additional experiments utilizing the retrograde transport of toxic ricin, a

The effect on breathing of abruptly stopping carotid body discharge.

In bilaterally vagotomized, decerebrate or pentobarbitone anaesthetized cats, intense carotid body discharge (FET O2 ca. 0.075) was abruptly removed by injections centrally of 100% O2-equilibrated Ringer into both external carotid arteries. In an inspiration the injections usually shortened that inspiration, reduced its volume and prolonged the immediately following expiration. Early in expiration they prolonged that expiration, but later in expiration they shortened it. The inspiratory results can be reconciled with von Euler's model of the inspiratory off-switch if the off-switch acts early

The effects of positive end-expiratory pressure on right and left ventricular performance.

The cardiovascular effects of positive end-expiratory pressure (PEEP) were studied in mechanically ventilated, vagotomized, Beta-blocked, anesthetized dogs. To compensate for the effect of PEEP on decreasing systemic venous return, acute plasma volume expansion was accomplished returning stroke volume and cardiac output to control values. Left and right ventricular filling pressures (LVFP and RVFP) and aortic pressure were measured relative to pressure (transmural pressure). Ventricular performance was assessed by comparing the transmural ventricular filling pressures at similar stroke volumes

Central neural stimulation of respiration in unanesthetized decerebrate cats.

A previously reported central neural respiratory control process was restudied in unanesthetized decerebrate cats during spontaneous breathing, and during conditions of constant chemical stimulation where phrenic nerve activity was used to quantitate respiratory output. Respiration was increased by carotid sinus nerve stimulation. The pattern of respiration was examined at the cessation of such stimulation. In spontaneously breathing animals, active hyperventilation (HV) was followed by hyperpnea for up to 30 s and never by apnea. Passive HV was always followed by apnea. In animals with contro

Controlled trial of metoclopramide in the treatment of flatulent dyspepsia.

A double-blind cross-over trial of metoclopramide (Maxolon) against placebo was undertaken in 42 patients with flatulent dyspepsia. A highly significant difference was found in favour of the active drug (P<0.01). The time at which the drug is given in relation to the meal and onset of symptoms is probably important. It is concluded that metoclopramide is effective in the short-term treatment of these symptoms but should be started only after thorough investigations have excluded more serious disease.

Case Report: Recurrent peptic ulcer perforations-a possible role of median arcuate ligament syndrome.

INTRODUCTION: Median arcuate ligament syndrome (MALS), also known as Dunbar syndrome, is a rare vascular compression disorder caused by extr … trunk, consistent with MALS. Surgical release of the median arcuate ligament combined with truncal vagotomy was performed. At 18-month follow-up, the patient remained free of abdominal pain, dyspeptic symptoms, and further ulcer-related complications. CONCLUSION: This case highlights MALS as a potential and often overlooked cause of refractory or recurre …

Spleen-targeted ultrasound restores gut-brain homeostasis in colitis via vagal neuroimmune modulation.

Inflammatory bowel disease (IBD) is increasingly recognized as a systemic disorder involving gut-brain axis dysfunction and neuropsychiatric … ed ultrasound stimulation of the spleen (sFUS) at intensities of 0.5 or 1.0 W/cm2. Subdiaphragmatic vagotomy (SDV) or sham surgery was performed before DSS induction and subsequent sFUS treatment to examine the involvement of vagal signaling. Therapeutic outcomes were evaluated using histological, molecular, microbiome, transcriptomic, and behavioral analy …

Camizestrant-induced heart rate reduction is mediated via a sinoatrial node pacemaker channel mechanism.

Camizestrant, a next-generation selective estrogen receptor (ER) degrader and complete ER antagonist, has been associated with a reversible … owing combined sympathetic (atenolol) and parasympathetic (atropine) inhibition in dogs, as well as vagotomy in rats. Camizestrant reduced hHCN4 current density in Chinese hamster ovary cells, as well as beat rate and "funny" pacemaker (If) current activity in hESC-derived SAN cardiomyocytes. Camizestrant at 75 mg/kg for 7 days significantly reduced If cur …
