[Adenosine in treatment of rats with spinal cord injury].

OBJECTIVE: To observe the changes of endogenous extracellular adenosine after spinal cord injury (SCI) to rats and the effect of exogenous adenosine on extracellular calcium after SCI and post-injury neurological function. METHODS: A ventral compression injury model of T13 spinal cord was used, and the extracellular fluids were collected consecutively every 20 minutes after injury by using microdialysis. Adenosine in the samples was analyzed using high-pressure liquid chromatography (HPLC) with u.v. detection. The rats received different doses of 2-chloroadenosine (2-CADO), a nonspecific agon…

Function of murine adenosine deaminase in the gastrointestinal tract.

Adenosine deaminase (ADA) deficiency in humans leads to a combined immunodeficiency characterized by severe T and B cell lymphopenia. ADA-deficient humans also display defective development of gut-associated lymphoid tissues (GALT). They lack lymphoid cells, and the Peyer's patches are without germinal centers. In mice, ADA-deficient fetuses die perinatally due to liver damage, but they also exhibit pathology in the thymus, spleen, and the small intestine. The GI phenotype associated with ADA-deficient humans prompted us to examine the effect of ADA-deficiency on mouse small intestine tissue. …

Adenosine-triphosphate in trauma-related and elective hypothermia.

BACKGROUND: In trauma patients, hypothermia is a frequent event. According to the literature, the majority of trauma patients are presenting a core temperature of less than 34 degrees C at admission. In contrast to the benefit of hypothermia in elective surgery, clinical experience with hypothermia in trauma patients has identified hypothermia to be one major cause of severe posttraumatic complications. It was hypothesized that this diverse effect of hypothermia is related to depletion of high-energy phosphates like adenosine triphosphate (ATP) in trauma patients. To verify this hypothesis, th…

Adenosine: an effective and safe antiarrhythmic drug in pediatrics.

Adenosine is an effective, safe drug for the diagnosis and treatment of paroxysmal tachycardias in adult and pediatric patients. A starting dose of 0.05-0.10 mg/kg as a rapid bolus injection is recommended for infants and children. An electrophysiologic effect can be expected within 20 seconds after injection. Dosage may be increased up to 0.3 mg/kg in steps of 0.05-0.10 mg/kg or until conversion to sinus rhythm is reached. Due to its basic electrophysiologic properties of slowing conduction in the atrioventricular (AV) node, which may result in transient AV block, adenosine is almost always e…

Characterization of ocular hypertension induced by adenosine agonists.

PURPOSE: Previous studies have shown that adenosine agonists may induce a rise in intraocular pressure (IOP), a reduction in IOP, or both. Although the reduction in IOP results from the activation of adenosine A1 receptors, the mechanisms responsible for the rise in IOP have not been investigated. This study examines the receptors and mechanisms responsible for the adenosine agonist-induced rise in IOP. METHODS: The ocular effects of the nonselective adenosine agonist NECA, the relatively selective adenosine A2 agonist CV-1808, the A2a agonist CGS-21680, and the A1 agonist R-PIA were evaluate…

Intraosseous adenosine for the termination of supraventricular tachycardia in an infant.

In recent years, the intraosseous technique has dramatically improved the speed at which vascular access may be achieved in critically ill children, and adenosine has been shown to rapidly convert supraventricular tachycardia to normal sinus rhythm. Until now, no report has demonstrated that this drug may be effectively given by this route in human beings. This case report describes the successful termination of supraventricular tachycardia in an infant with the intraosseous administration of adenosine.

Diagnostic use of adenosine in two pediatric patients.

We describe the successful diagnostic use of adenosine in two pediatric patients. In the first case, adenosine infusion enabled exclusion of accessory pathway conduction in a patient who had previous evidence of Wolff-Parkinson-White syndrome. In the second case, adenosine infusion aided in the diagnosis of a nonreciprocating reentrant atrial tachycardia in a postoperative Fontan patient. The transient atrioventricular nodal blocking effect of adenosine added diagnostic certainty which was not apparent from the surface 12-lead electrocardiograms or rhythm recording in both patients. As the the…

Mechanism-specific effects of adenosine on atrial tachycardia.

BACKGROUND: Recent reports suggest that adenosine, in addition to terminating supraventricular tachycardia involving the atrioventricular (AV) node, may have antiarrhythmic effects on atrial tachycardia. The electrophysiological effects of adenosine on supraventricular tissue include shortening of action potential duration in atrial myocytes mediated by the potassium current, IKACh,Ado; shortening of action potential duration and hyperpolarization in sinus node cells; and anti-adrenergic electrophysiological effects resulting from inhibition of adenylyl cyclase. We therefore hypothesized that …

Adenosine use in pregnant women with supraventricular tachycardia.

OBJECTIVE: To review a case experience and published reports of treatment of supraventricular tachycardia with adenosine in pregnant women. DATA SOURCES: Published reports and clinical experience. DATA SYNTHESIS: Seven pregnant women with supraventricular tachycardia treated with adenosine have been described in the literature. We describe an eighth patient. Treatment has terminated the dysrhythmia in all cases, and no adverse maternal or fetal effects have been reported. CONCLUSIONS: Based on theoretical considerations and on limited published experience, adenosine appears to be safe and e…

Adenosine in the termination of albuterol-induced supraventricular tachycardia.

Five percent of children in the United States have asthma. Status asthmaticus is one of the most common conditions for which children seek care in a pediatric emergency department. beta 2-Agonists such as albuterol are the mainstay of emergency therapy for such children. We present a case of a 4-year-old boy who experienced supraventricular tachycardia (SVT) following albuterol therapy. This is believed to be the first report of adenosine being successfully used to treat a child with albuterol-induced SVT. We also briefly review the recognition and management of SVT in children and the pharmac…

Ionic dependence of adenosine uptake into cultured astrocytes.

Adenosine uptake in cultured astrocytes is dependent on various ions and energy metabolism. The Na(+)-gradient plays an important role, since nigericin, ouabain, amiloride and substitution of Na+ with choline inhibited adenosine uptake. The proton-gradient was of importance, since carbonylcyanide m-chlorophenylhydrozone (CCCP) and omeprazole also inhibited adenosine uptake. Furthermore, adenosine uptake was dependent on Cl- anion. Substitution of Cl- with isethionate, as well as DIDS or furosemide inhibited adenosine uptake. Adenosine uptake was also sensitive to Ca2+ gradient, removal of extr…

[Efficacy of adenosine triphosphate in terminating supraventricular tachycardia].

The efficacy of adenosine triphosphate (ATP) was tested in 23 patients suffering from supraventricular tachycardia; 10 patients had orthodromic circus movement tachycardia and 13 typical av nodal reentrant tachycardia. At a mean dose of 0.18 mg/kg all tachycardias were halted in less than 1 minute due to transient acute av nodal block followed by return to normal av conduction within 8 seconds. All patients had transient minor side effects. One elderly subject had a short episode of atrial fibrillation after the acute av block. In conclusion, ATP is useful and safe for rapid termination of sup…

[The usefulness of adenosine triphosphate in supraventricular paroxysmal tachycardias].

With the objective to evaluate adenosine triphosphate (ATP) usefulness in supraventricular paroxysmal tachycardia (SPT), doses of 0.25 mg/kg, of ATP are administered fastly i.v. in bolus, to fifty consecutive patients with SPT resistant to vagal stimulation maneuvers. The effect achieved allowed to identify two groups: a) 39 patients in whom tachycardia was interrupted; b) 11 patients in whom tachycardia did not cease, but the induced transient provoked atrial-ventricular (A-V) blockage allowed to identify the underlying mechanism of tachycardia (in 6 of them atrial tachycardia and in 5 atrial…

Proarrhythmic effect of adenosine in a patient with atrial flutter.

Adenosine has been proposed as a useful diagnostic agent in patients with narrow complex regular tachycardia of uncertain origin. Its effects are usually transient owing to its extremely short plasma half life and, as a consequence, it is thought to be safer than other drugs used in the acute treatment of such arrhythmias. However, adenosine had a proarrhythmic effect when administered to a patient in order to confirm the diagnosis of atrial flutter. As expected, a transient increase in atrioventricular block was seen but this was followed by a doubling of the ventricular rate and haemodynamic…

Microcirculatory responses to adenosine in the newborn pig retina.

The reactivity of retinal arterioles and venules to exogenous and endogenous adenosine was investigated in the newborn piglet eye in vivo. The retinal microcirculation of isoflurane-anesthetized newborn pigs was observed at 310x using videomicroscopy, and changes in the diameter of arterioles (50-100 microns in diameter) and venules (150-250 microns in diameter) occurring in response to intravitreal topical microsuffusions of various adenosinergic compounds were determined. Dose-dependent dilations of the arterioles and venules resulted from intravitreal adenosine (0.2-200 nmol) and three of i…

[Initial experience with adenosine triphosphate in supraventricular arrhythmia].

The author presents data from the literature pertaining to the possible use of adenine nucleosides in recent years. The main action of adenosine or adenosine triphosphate is focused above all on influencing supraventricular tachycardia which developed as a result of the reentry mechanism. Based on his own observation, the author presents his initial experience with adenosine triphosphate (ATP, Spofa) in different indications: interference with supraventricular tachycardia with slim QRS complexes, interference of tachycardia with wide QRS complexes, termination of atrial flutter in digitalis in…
