Altered mitochondrial RNA production in adipocytes from HIV-infected individuals with lipodystrophy.

BACKGROUND: Damage to mitochondria (mt) is a major side effect of highly active antiretroviral therapy (HAART) that includes a nucleoside reverse transcriptase inhibitor (NRTI). Such damage is associated with the onset of lipodystrophy in HAART-treated HIV+ patients. To further investigate mt changes during this syndrome, we analysed the expression of mtRNA in adipocytes from lipodystrophic HIV+ patients taking NRTI-containing HAART and compared it with similar cells from healthy individuals. MATERIALS AND METHODS: Total RNA was extracted from adipocytes collected from different anatomical lo

Interaction of phosphoinositolglycan(-peptides) with plasma membrane lipid rafts of rat adipocytes.

Insulin receptor-independent activation of the insulin signal transduction cascade in insulin-responsive target cells by phosphoinositolglycans (PIG) and PIG-peptides (PIG-P) is accompanied by redistribution of glycosylphosphatidylinositol (GPI)-anchored plasma membrane proteins (GPI proteins) and dually acylated nonreceptor tyrosine kinases from detergent/carbonate-resistant glycolipid-enriched plasma membrane raft domains of high-cholesterol content (hcDIGs) to rafts of lower cholesterol content (lcDIGs). Here we studied the nature and localization of the primary target of PIG(-P) in isolate

Growth factor regulation of uncoupling protein-1 mRNA expression in brown adipocytes.

To study the effect of the mitogens epidermal growth factor (EGF), acidic and basic fibroblast growth factors (aFGF and bFGF), and vasopressin on brown adipocyte differentiation, we analyzed the expression of uncoupling protein-1 (UCP-1) mRNA. Quiescent brown preadipocytes express high levels of UCP-1 mRNA in response to triiodothyronine (T3) and norepinephrine (NE). The addition of serum or the mitogenic condition aFGF + vasopressin + NE or EGF + vasopressin + NE decreases UCP-1 mRNA. A second addition of mitogens further decreases UCP-1 mRNA. Treatment with aFGF or bFGF alone increases UCP-1

Resistin gene expression in human adipocytes is not related to insulin resistance.

OBJECTIVES: Obesity is an important risk factor for the development of insulin resistance and type 2 diabetes. Recently, a newly described circulating hormone resistin, which is expressed primarily in adipocytes, has been shown to antagonize insulin action in mice. Resistin, therefore, has been suggested to play a role in the pathogenesis of insulin resistance. RESEARCH METHODS AND PROCEDURES: We studied the expression of the resistin gene in primary cultured human adipocytes and preadipocytes. We also examined resistin gene expression in subcutaneous abdominal adipocytes in women (n = 24) ov

Zinc deficiency reduces leptin gene expression and leptin secretion in rat adipocytes.

The present study was conducted to measure ob mRNA abundance in the zinc-deficient (ZD) rats and the secretion of leptin from adipose tissue obtained from ZD, zinc-adequate (ZA), and pair-fed (PF) rats. It was found that ob mRNA abundance was greatest (P < 0.05) in adipose tissue obtained from ZA and PF rats. Ob mRNA abundance was similar in PF and ZD rats. To study leptin secretion from adipose tissue in a cell culture model, a method was developed to use excised epididymal adipose tissue from ZD, ZA, and PF rats. Tissue was incubated in Opti-modified Eagle's medium (MEM) cell culture medium

Mineralocorticoid and glucocorticoid receptors inhibit UCP expression and function in brown adipocytes.

Uncoupling proteins (UCP), specific mitochondrial proton transporters that function by uncoupling oxidative metabolism from ATP synthesis, are involved in thermoregulation and control of energy expenditure. The hibernoma-derived T37i cells, which possess functional endogenous mineralocorticoid receptors (MR), can undergo differentiation into brown adipocytes. In differentiated T37i cells, UCP1 mRNA levels increased 10- to 20-fold after retinoic acid or beta-adrenergic treatment. Interestingly, UCP2 and UCP3 mRNA was also detected. Aldosterone treatment induced a drastic decrease in isoproteren

Insulin-like action of chromate on glucose transport in isolated rat adipocytes.

The effects of chromium compounds on 3-O-methylglucose (3-O-MG) transport were studied in isolated rat adipocytes. Sodium chromate significantly stimulated 3-O-MG uptake into adipocytes in a dose-dependent manner without altering the equilibrium space of 3-O-MG in adipocytes. The stimulatory effect reached the maximum at 300 microM, and the effect was 60-70% of the maximal insulin effect that was obtained with 20 nM insulin. The chromate concentration achieving a half-maximal effect was estimated at 50 microM. The effect of the combination of 1 mM chromate and 20 nM insulin was equipotent to t

Adrenalectomy-induced alterations in glucagon binding and lipolysis in isolated rat adipocytes.

Adipocytes from adrenalectomized rats nearly lost their lipolytic response to glucagon concomitant with a 90% decrease in the number of glucagon receptors per cell. Quantitative analysis of the relation between amount of cell-bound glucagon and hormone-stimulated lipolysis revealed that the ability of the remaining 10% of glucagon receptors to induce lipolysis was not impaired. Binding of the beta-adrenergic antagonist [3H]dihydroalprenolol and maximal lipolysis induced by (-)-isoproterenol, (Bu)2cAMP, 3-isobutyl-1-methylxanthine, and adenosine deaminase were reduced only 10 to 20% after adren

Regulation of lipid metabolism by dipyridamole and adenosine antagonists in rat adipocytes.

1. Acute effects of dipyridamole, an inhibitor of adenosine transport, direct activators of adenylate cyclase and thirteen adenosine antagonist analogs on fatty acid synthesis have been examined in terms of the control of [1-14C]acetate incorporation into labeled fatty acids in the presence of glucose. 2. This monosaccharide acts as a stimulator of lipogenesis by generating NADPH for the lipid synthesis. 3. The relationship between lipogenesis and lipolysis was compared with a variety of adenylate cyclase stimulators. 4. The data obtained reveals that dipyridamole potentiated the inhibitory or

[Season variations in lipolysis activity of adipocytes from fox, badger and hedgehog].

1. We have studied the seasonal variations of the lipolytic response to a beta agonist, isoproterenol, and to theophylline of the isolated adipocyte from fox, badger and hedgehog, related to the circannual variations of testes and thyroid activities. 2. With the fox, we find a coincidence, in spring, between the maximum lipolytic sensitivity of the adipocyte, the thyroid function and the return to sexual rest. 3. With the badger, in winter, the maximum reaction of the adipocyte is contemporary with testes activity and with thyroid rest. 4. With the hedgehog, we observe two maximums in the sens

Glycosylated extracellular vesicles drive a metabolic interplay between gastric cancer cells and adipocytes.

Aim: Tumor cells and extracellular vesicles (EVs) dynamically interact with adipocytes, contributing to tumor progression and systemic metabolic dysfunctions. While the role of glycosylation in this process remains unclear, glycans facilitate communication between tumors and their surrounding environment. Here, we aimed to elucidate the pivotal role of sialyl-Tn (STn), a tumor-associated glycan, in driving a bidirectional metabolic reprogramming between gastric cancer cells and adipocytes. Methods: EVs were isolated from gastric cancer cells with distinct glycosylation profiles by ultracentrif

Spatial Plasticity and Multidirectional Crosstalk of Cancer-Associated Adipocytes in Therapy-Remodeled Niches.

Cancer-associated adipocytes (CAAs) are increasingly recognized as active and heterogeneous components of the tumor microenvironment. Initially defined as tumor-adjacent adipocytes characterized by lipid depletion, reduced expression of mature adipocyte markers, and inflammatory activation, CAAs are now understood to encompass a spectrum of context-dependent states. These states are shaped by anatomical location, preexisting adipocyte properties, tumor-derived signals, and therapeutic exposure. Through metabolic exchange, paracrine signaling, direct cell contact, extracellular-vesicle communic

Bone Marrow Adipocytes in Multiple Myeloma: Emerging Immunometabolic Interactions and Implications for Immunotherapy.

Immunotherapy has revolutionized the treatment of multiple myeloma (MM), but durable immune control remains limited, increasing interest in how the bone marrow (BM) microenvironment influences antitumor immunity. BM adipocytes (BMAds) are increasingly recognized as metabolically active components of the BM. This review summarizes the current evidence linking BMAds to myeloma cell survival and immune cell function, particularly T cell activity. It also discusses their potential relevance to monoclonal antibodies, bispecific antibodies, and chimeric antigen receptor (CAR) T-cell therapy. Preclin

PPAR-δ is a potential modulator of central carbon metabolism in human adipocytes.

Adipogenesis involves adipocyte differentiation and synthesis and storage of fats. PPAR-γ is the master regulator of adipogenesis and regulates genes for adipocyte differentiation, lipogenesis, adipocyte survival and adipokine secretion. Central carbon metabolism (CCM) comprises of three key pathways glycolysis, tricarboxylic acid (TCA) cycle and pentose phosphate pathway (PPP). CCM utilizes carbon sources to provide energy and building blocks for lipogenesis. Several targets of transcription factor PPAR-γ have been identified in CCM pathways. However, the exact mechanism of PPAR-γ modulation

Acute deletion of PLIN5 in brown adipocytes causes mitochondrial dysfunction and cold intolerance.

Cold exposure of mice is associated with adaptive molecular and organellar changes in brown adipose tissue (BAT) that promote thermogenesis to defend body temperature. We previously reported that the lipid droplet protein Perilipin 5 (PLIN5) robustly increases in BAT during acute exposure of mice to cold. We demonstrated that chronic induction of BAT PLIN5 within the physiological range in mice housed at room temperature mimics the effects of cold exposure in terms of increased thermogenic gene expression in BAT, increased BAT mitochondria cristae packing, and increased uncoupled mitochondrial

Danshenol A Mediates the Proliferation and Differentiation of Adipocytes in Thyroid-Associated Ophthalmopathy.

INTRODUCTION: An increase in the intraorbital adipose tissue is the main pathological feature of thyroid-associated ophthalmopathy (TAO). IGF-1R activates PI3K/AKT signaling and accelerates adipogenesis. Pingmu decoction has been demonstrated to promote orbital adipocyte apoptosis; however, less is reported regarding the action mechanism of Danshenol A (DA), a single active ingredient of Salvia miltiorrhiza (Danshen). Accordingly, this study aimed to investigate the role and association of DA and IGF-1R in the proliferation and lipid accumulation of orbital adipocytes. METHODS: Primary human

Alternative Receptor Signaling for the Selective and Multifaceted Regulation of Human Brown Adipocytes.

Brown adipose tissue (BAT) is increasingly recognized as a metabolically active organ in adult humans that contributes to systemic energy homeostasis and represents a potential therapeutic target for obesity-associated metabolic diseases. However, effective strategies to increase BAT mass or thermogenic activity in humans have not yet been established. Although β-adrenergic receptors have traditionally been viewed as the principal drivers of adaptive thermogenesis and adipocyte browning, β-adrenergic stimulation alone may be insufficient to safely enhance BAT thermogenic capacity due to system

Obesity- and Glucose-Dependent Differential Autophagy Marker Expression in Adipose Tissues and Adipocytes.

BACKGROUND: Autophagy is an evolutionarily highly conserved process and plays an important role in cellular homeostasis and metabolism. In obesity and obesity-associated insulin resistance, both enhanced and suppressed autophagy have been observed in adipose tissues (AT). OBJECTIVE AND METHODS: We report on a panel of 7 autophagy markers critically involved in mammalian cellular autophagy in a cohort of patients with obesity (n = 40; mean BMI 54.8 ± 6.7 kg/m2) versus lean individuals (n = 10; mean BMI = 22.7 ± 1.7 kg/m2), during 3T3-L1 adipocyte differentiation in vitro and in mature 3T3-L1 a

Dangerous liaisons: interactions between cancer cells and adipocytes as drivers of cancer progression.

Metabolic plasticity and flexibility are key characteristics that allow cancer cells to adapt and thrive in different environments. Specifically, cancer cells can dynamically change the routing of metabolic pathways in response to environmental changes and adapt their metabolic activity depending on local nutrient availability. The tumor microenvironment (TME) plays crucial roles in cancer development and progression. It is now widely accepted that different stromal cells, as well as soluble factors, including metabolites, derived from the TME support cancer cell proliferation and survival and

GDF3 is an endogenous antagonist of the ActE-ALK7/ACVR2 pathway in adipocytes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) and steatohepatitis (MASH) arise, in part, from excessive free fatty acid flux from adipose tissue to the liver. Activin E (ActE, encoded by INHBE) suppresses adipocyte lipolysis through the type I receptor ALK7 (ACVR1C). Loss-of-function variants in INHBE and ACVR1C reduce waist-to-hip ratio in humans, yet genetic knockouts in mice produce insulin resistance and hepatic steatosis, suggesting discrepancies between human and mouse biology. We identify GDF3, a TGF-β superfamily ligand upregulated in obese adipose tissue, as the pri
