[Metabolic syndrome: from insulin resistance to adipose tissue dysfunction].

Recently the definition, the pathophysiology and even the clinical utility of metabolic syndrome (MS) have been discussed. The risk induced … eogenesis. The belief that insulin-resistance is the main cause of MS could change in favour of the adipose tissue dysfunction. The most common cause of a reduced capacity of the adipose tissue to store fats is the increased dietary intake, also present in lipodistrophy, type 1 diabetes mellitus and polycystic ovarian syndrome. The adipose tissue production of a …

Remodelling of adipose tissue during experimental omental adhesion formation.

BACKGROUND: Peritoneal adhesions are fibrous bands of tissue connecting normally separated organs and frequently involve the fat-laden greater omentum. Remodelling of fibrin-rich exudate under reduced fibrinolytic conditions is thought to initiate adhesion formation following surgery. It is unclear whether adhesions that involve the omentum develop in a similar manner. To improve understanding of omental adhesion formation, adipose tissue distribution, cell proliferation and procollagen type I gene expression were investigated in a murine surgical model and in established omental adhesions fro…

Depot-specific steroidogenic gene transcription in human adipose tissue.

CONTEXT: Sex steroids (androgens and oestrogens) and corticosteroids (glucocorticoids and mineralocorticoids) have a major impact on fat distribution. Several genes involved in steroid synthesis and metabolism, such as 11beta-hydroxysteroid dehydrogenase type 1 and aromatase, are known to be expressed within adipose tissue, thus modulating local steroid levels; however, our knowledge of which genes are expressed and at what level is incomplete. OBJECTIVE: To detect by real-time quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) which of 13 key steroidogenic genes are trans…

Regulation of inflammation-related genes in human adipose tissue.

The identification of a moderate increase in circulating inflammatory factors in obese subjects, the description of changes in inflammatory gene expression in adipose tissue (AT) and the discovery that macrophage cells infiltrate AT are observations contributing to the concept that human obesity is a chronic inflammatory illness. This concept has led to some revision of the physiopathology of obesity and of its related metabolic and cardiovascular co-morbidities. Low-grade inflammation in the AT and the subsequent production of specific biomarkers could actually link expanded fat mass to obesi…

[Lipodystrophic syndromes: congenital or acquired diseases of adipose tissue].

Lipodystrophic syndromes regroup a heterogeneous group of genetic or acquired diseases. Lipodystrophy, an altered development and/or repartition of body fat, is associated with alterations of lipid and glucose metabolism with insulin resistance. Genetic forms, rare, can be generalized and recessive resulting from mutations in the seipin or AGPAT2 gene. Partial lipodystrophies are dominant and observed in patients mutated in the gene encoding PPAR-gamma or lamin A/C, a gene seen also mutated in patients with syndromes of premature aging. Acquired forms are common and regroup the highly prevalen

Sympathetic and parasympathetic innervation of adipose tissue: metabolic implications.

PURPOSE OF REVIEW: Adipose tissue is of fundamental importance for the integrative physiology of the body by serving metabolic and endocrine functions. The purpose of this review is to evaluate the functions of the autonomic innervation of adipose tissue. RECENT FINDINGS: Different lines of evidence from studies on the sympathetic or parasympathetic innervation of adipose tissue have indicated that the autonomic nervous system modulates the fundamental properties of adipose tissue function and biology at the cellular and molecular level. This is reflected in the modulation of lipolysis/lipoge…

[Insulin resistance and adipose tissue gene expression in humans].

Obesity is a risk factor for type 2 diabetes and cardiovascular diseases. The hypothesis that cytokines could play a role in the pathophysiology of obesity and insulin resistance is suggested in the last few years. We showed a positive correlation between circulating interleukin (IL-6) levels and obesity and insulin resistance suggesting that IL-6 could be involved in insulin resistance in humans. We showed a decrease of both circulating and adipose tissue IL-6 levels in non-diabetic obese subjects after a very low calorie diet program inducing weight loss. This suggests that adipose tissue co…

Adipose tissue as a buffer for daily lipid flux.

Insulin resistance occurs in obesity and Type II (non-insulin-dependent) diabetes mellitus, but it is also a prominent feature of lipodystrophy. Adipose tissue could play a crucial part in buffering the flux of fatty acids in the circulation in the postprandial period, analogous to the roles of the liver and skeletal muscle in buffering postprandial glucose fluxes. Adipose tissue provides its buffering action by suppressing the release of non-esterified fatty acids into the circulation and by increasing triacylglycerol clearance. In particular, the pathway of 'fatty acid trapping' (adipocyte u…

Larger amounts of visceral adipose tissue in Asian Americans.

OBJECTIVE: Excess visceral adipose tissue (VAT) is recognized as an important risk factor for the development of coronary heart disease and type 2 diabetes. Several studies have reported less VAT in African Americans compared with whites. As little is known about the levels of VAT in Asians, we compared whole-body VAT in Asian Americans with European Americans. RESEARCH METHODS AND PROCEDURES: VAT was measured using whole-body multislice magnetic resonance imaging in 54 women (18 Asian Americans, 36 European Americans) and 53 men (19 Asian Americans, 34 European Americans) with body mass inde…

Identification of progesterone receptor in human subcutaneous adipose tissue.

Sex steroids are postulated to play a role in adipose tissue regulation and distribution, because the amount and location of adipose tissue changes during puberty and menopause. Because of the nature of adipose tissue, receptors for the female sex steroids have been difficult to demonstrate. To date, estrogen receptor messenger RNA and protein have been identified in human subcutaneous adipose tissue, but the presence of progesterone receptor (PR) has not been reported. In this study, we demonstrate PR message by Northern blot analysis in RNA isolated from the abdominal subcutaneous adipose ti…

Childhood obesity, adipose tissue distribution, and the pediatric practitioner.

The prevalence of pediatric obesity is increasing in the United States. Sequelae from pediatric obesity are increasingly being seen, and long-term complications can be anticipated. Obesity is the most common cause of abnormal growth acceleration in childhood. Obesity in females is associated with an early onset of puberty and early menarche. Puberty is now occurring earlier in females than in the past, and this is probably related either directly or indirectly to the population increase in body weight. The effect of obesity on male pubertal maturation is more variable, and obesity can lead to

Postprandial adipose tissue lipoprotein lipase activity in primary hypertriglyceridemia.

The fasting activity of adipose tissue lipoprotein lipase has been previously reported to be either normal or reduced in subjects with a primary form of hypertriglyceridemia. The postprandial activity of adipose tissue lipoprotein lipase has not been previously reported in these subjects. In subjects with primary hypertriglyceridemia the fasting lipoprotein lipase activity eluted from pieces of adipose tissue by heparin and the enzyme activity present in extracts of acetone--ether tissue powders were similar to the level of enzyme activity found in normal subjects. There also was no difference…

Regulation of lipolysis by human adipose tissue in hyperthyroidism.

The effects of noradrenaline (NA) and isopropyl-noradrenaline (ISNA) on glycerol release and cAMP levels in sc adipose tissue were studied in vitro in 27 patients with hyperthyroidism. In 11 patients, the studies were repeated after 6--12 months of treatment for hyperthyroidism. A third group comprised 21 euthyroid patients otherwise healthy except for morbid obesity. The lipolytic response to ISNA, observed in untreated thyrotoxic patients, was found to be reduced by 30% when the patients were reexamined after treatment for thyrotoxicosis. This reduction was attributable to a decrease in the …

Inhibition of soluble epoxide hydrolase ameliorates hyperhomocysteinemia-induced perivascular adipose tissue dysfunction: Revealing epoxyeicosatrienoic acid as a perivascular adipose tissue-derived relaxing factor.

OBJECTIVES: Hyperhomocysteinemia harms vascular endothelium and smooth muscle. We recently reported that homocysteine also impairs perivascular adipose tissue (PVAT) function but the mechanisms remain poorly elucidated. This study aimed to advance mechanistic understanding of hyperhomocysteinemia-induced PVAT dysfunction through revealing the role of epoxyeicosatrienoic acids (EETs) in the anticontractile/vasorelaxing activity of PVAT. Further, the effect of targeting soluble epoxide hydrolase (sEH) against hyperhomocysteinemia-induced PVAT dysfunction was explored. METHODS: Methionine diet-i…

Emodin inhibits lipid accumulation and inflammation in adipose tissue of high-fat diet-fed mice by inducing M2 polarization of adipose tissue macrophages.

Adipose tissue macrophages (ATMs) represent the most abundant leukocytes in adipose tissue (AT). An increase in number and a phenotypical switch of ATMs during the development of obesity contribute to chronic inflammation and metabolic disorders, which have been regarded as potential therapeutic targets to restore AT homeostasis. Emodin has been shown to exert strong anti-inflammatory property via acting on macrophages in a range of disease models. However, whether emodin exerts a beneficial effect on obesity via modulating ATMs has not been reported. In high-fat diet (HFD)-induced obese mice,…

The role of adipose tissue in puberty and reproductive health.

Adipose tissue is an endocrine organ that signals energy status to the hypothalamic-pituitary-gonadal axis to regulate reproductive function. Notably, in mammals, adipose tissue biology-adipose tissue expansion and body fat distribution-is closely linked to the onset of puberty. Some studies showed that early adipose tissue development continues into childhood or adulthood, indicating its potential impact on reproductive function. Factors such as maternal obesity, childhood body mass index gain, and adolescent obesity significantly contribute to early puberty onset and negative reproductive ev…

Role of gremlin-1 in the pathophysiology of the adipose tissues.

Gremlin-1 is a secreted bone morphogenetic protein (BMP) antagonist playing a pivotal role in the regulation of tissue formation and embryon … ans (HSPGs). Growing evidence has highlighted a central role of gremlin-1 in the homeostasis of the adipose tissue (AT). Of note, gremlin-1 is involved in AT dysfunction during type 2 diabetes, obesity and non-alcoholic fatty liver disease (NAFLD) metabolic disorders. In this review we discuss recent findings on gremlin-1 involvement in AT biology, with particul …
