Reduction of aflatoxin contamination in maize kernels mediated by host-induced gene silencing.

Aflatoxins produced by some species of Aspergillus pose a significant threat to global food safety, because they contaminate a variety of crops including maize. In this study, we used host-induced gene silencing (HIGS) to reduce aflatoxin accumulation in maize upon infection with Aspergillus flavus and Aspergillus parasiticus. Two RNAi constructs were designed and introduced into maize, one targeting two essential fungal genes involved in ergosterol biosynthesis (CYP51A and CYP51B) and the other targeting three aflatoxin biosynthesis genes (AflC, AflD, and AflR). The presence of RNAi transgene…

Mechanistic and Structural Analysis of Aflatoxin B1 Degradation by Bacillus safensis Multicopper Oxidase.

Aflatoxin B1 (AFB1) is a potent mycotoxin threatening food and feed safety. Here, we report the identification and characterization of a Bacillus safensis-derived multicopper oxidase (BsaMCO) capable of efficient AFB1 detoxification. Recombinant BsaMCO exhibited robust in vitro activity, achieving >78% degradation of AFB1 under 24 h incubation at 37 °C. Optimization experiments revealed that enzyme concentration, pH, temperature, metal ions, and electron acceptors significantly influenced degradation efficiency, defining an operational window suitable for practical applications. Assessment of near-infrared hyperspectral imaging for the discrimination of naturally aflatoxin-contaminated nuts.

Aflatoxins (AFs) and ochratoxin A (OTA) are major safety concerns in nuts due to their toxicity and uneven distribution. To address this, near-infrared hyperspectral imaging (NIR-HSI) was evaluated as a rapid, non-destructive tool for single-kernel classification, complemented by a preliminary HPLC-FLD screening. In this study, 52 peanut and 33 pistachio samples of commercial origin were analyzed for AFs and OTA using an HPLC-FLD method. The occurrence of AFB1 detected in peanut samples was 7.70%, whereas OTA was detected in pistachio samples with an occurrence of 3.03%. The maximum concentrat…

Safe natural antifungal and anticancer agents are increasingly needed. This study investigated β-conglycinin glycoproteins from chickpea, soybean, and lupin seeds for their dual antifungal and anticancer potential. Proteins were isolated and tested against Aspergillus spp. (A. flavus, A. niger, A. parasiticus) and colon cancer cell lines (DLD-1, SW620, HCT8, HT29). MIC values, sporulation, aflatoxin inhibition, and cell wall integrity assays (β-1,3-glucan, chitin, SEM imaging, and leakage) reflected the antifungal activity of the studied proteins, while cell viability, apoptosis markers, migra

Unravelling the Mechanistic Approach of Aflatoxin Contaminated Food on Neurodegenerative Diseases-A Novel Approach.

Aflatoxins (AFs) are a group of toxic secondary metabolites and a dietary toxin produced predominantly by Aspergillus species such as Aspergillus flavus and Aspergillus parasiticus. The four most common and harmful forms of AFs include Aflatoxin B1 (AFB1), Aflatoxin B2 (AFB2), Aflatoxin G1 (AFG1), and Aflatoxin G2 (AFG2), which pose a significant health threat due to their widespread contamination of food and feed products. Particularly, AFB1 has raised a major global health concern. Noxious neurological outcomes have been associated with chronic exposure to AF-contaminated food, contributing …

Aflatoxin B1: Challenges and Strategies for the Intestinal Microbiota and Intestinal Health of Monogastric Animals.

The objective of this review is to investigate the impacts of aflatoxins, particularly aflatoxin B1 (AFB1), on intestinal microbiota, intestinal health, and growth performance in monogastric animals, primarily chickens and pigs, as well as dietary interventions to mitigate these effects. Aflatoxin B1 contamination in feeds disrupts intestinal microbiota, induces immune responses and oxidative damage, increases antioxidant activity, and impairs jejunal cell viability, barrier function, and morphology in the small intestine. These changes compromise nutrient digestion and reduce growth performan…

Key factors influencing aflatoxin M1 concentration in dairy products: A comprehensive review.

Aflatoxin B1 (AFB1) contamination in livestock feed, followed by its ingestion by dairy animals, leads to hydroxylation of the toxin in the liver and its conversion into aflatoxin M1 (AFM1), which is subsequently excreted in milk. The occurrence of AFM1 in milk and dairy products is a major global public health concern. This review examines the key factors influencing AFM1 contamination in dairy products. The primary source is the presence of Aspergillus fungi in animal feed, which produce AFB1 during cultivation, harvesting, and storage. Environmental conditions (particularly high temperature

Aflatoxin B1-Induced Hepatic Mutagenesis in Mice Expressing Gene-Edited Neil1.

Hepatocellular carcinoma (HCC) remains one of the leading causes of cancer-associated mortality, correlating with obesity, alcohol consumption, hepatitis B and C infections, and dietary exposure to aflatoxin B1 (AFB1). The etiology of AFB1-induced HCC involves the formation of highly mutagenic guanine lesions that can be repaired by a branch of the base excision repair pathway initiated by the DNA glycosylase, NEIL1. In a murine model, NEIL1 deficiency results in increased AFB1-induced mutagenesis and carcinogenesis. Previous analyses identified several defective NEIL1 variants in human popula

SntB triggers the antioxidant pathways to regulate development and aflatoxin biosynthesis in Aspergillus flavus.

Following the discovery of nociceptin/orphanin FQ (N/OFQ) peptide receptor (NOP) and its endogenous ligand, an extensive search has started to find selective agonists and antagonists targeting this novel receptor-ligand system due to their therapeutic potentials. By the help of the combinatorial chemistry a series of hexapeptides with a general formula of Ac-RYY-R/K-W/I-R/K-NH(2) having high NOP receptor affinity and selectivity were identified. On the basis of this information we developed a number of novel compounds. The detailed structure-activity studies on the partial agonist Ac-RYYRIK-NH

Aflatoxin B1 Induces Inflammatory Liver Injury via Gut Microbiota in Mice.

Aflatoxin B1 (AFB1), a potent food-borne hepatocarcinogen, is the most toxic aflatoxin that induces liver injury in humans and animals. Species-specific sensitivities of aflatoxins cannot be fully explained by differences in the metabolism of AFB1 between animal species. The gut microbiota are critical in inflammatory liver injury, but it remains to reveal the role of gut microbiota in AFB1-induced liver injury. Here, mice were gavaged with AFB1 for 28 days. Then, the modulation of gut microbiota, colonic barrier, and liver pyroptosis and inflammation were analyzed. To further verify the direc…

Berberine-loaded albumin nanoparticles reverse aflatoxin B1-induced liver hyperplasia.

Hepatocellular carcinoma (HCC) can be produced from aflatoxin B1 (AFB1) administration. Although berberine (BER) acts as an anticancer agent and can counteract the AFB1 effect, it has low bioavailability. Nanotechnology can overcome this problem. This research aimed to synthesize berberine nanoparticles (NPs) and then estimate their therapeutic effect compared to that of berberine against aflatoxin-induced hepatotoxicity. The desolvation method was used to prepare BER-NPs. Aflatoxicosis was induced by 5 consecutive intraperitoneal injections (IP) of 200 µg/kg/day AFB dissolved in dimethylsulfo

The membrane mucin Msb2 regulates aflatoxin biosynthesis and pathogenicity in fungus Aspergillus flavus.

As a pathogenic fungus, Aspergillus flavus can produce carcinogenic aflatoxins (AFs), which poses a great threat to crops and animals. Msb2, the signalling mucin protein, is a part of mitogen-activated protein kinase (MAPK) pathway which contributes to a range of physiological processes. In this study, the roles of membrane mucin Msb2 were explored in A. flavus by the application of gene disruption. The deletion of msb2 gene (Δmsb2) caused defects in vegetative growth, sporulation and sclerotia formation when compared to WT and complement strain (Δmsb2C ) in A. flavus. Using thin-layer chromat…
