The Role of GCN2 Kinase in Mediating the Effects of Amino Acids on Longevity and Feeding Behaviour in Drosophila.

Restriction of amino acids in the diet can extend lifespan in diverse species ranging from flies to mammals. However, the role of individual amino acids and the underlying molecular mechanisms are only partially understood. The evolutionarily conserved serine/threonine kinase General Control Nonderepressible 2 (GCN2) is a key sensor of amino acid deficiency and has been implicated in the response of lifespan to dietary restriction (DR). Here, we generated a novel Drosophila GCN2 null mutant and analyzed its response to individual amino acid deficiency. We show that GCN2 function is essential f

Tripeptidyl peptidases: enzymes that count.

Protein degradation is essential for the life and death of every cell. Proteins are broken down to their constitutive amino acids by a succession of peptidases, both in lysosomes and in the cytosol. Tripeptidyl-peptidases I and II are enzymes that can 'count to three' and release N-terminal tripeptides from oligopeptides generated by different endopeptidases. The tripeptides are then degraded by other exopeptidases to release amino acids and dipeptides. Mutations in tripeptidyl-peptidase I have recently been associated with a lysosomal storage disease, late infantile neuronal ceroid lipofuscin

[Chemical composition of a protein concentrate from Saccharomyces and its effect on immunologic response].

The content of the main nutrients, as well as amino-acid, fatty-acid and mineral composition of a protein concentrate prepared from yeast-Saccharomyces grown in molasses were studied. It was found that the product contained a significant amount of protein, all essential amino acids, insignificant quantity of lipids in which saturated fatty acids prevailed. The concentrate ash was rich in calcium, phosphorus, magnesium, silicon, iron and other elements. The protein product included into experimental rations during a month produced no significant effect on the B-system immunity and did not induc

Electrosynthesis of α-Amino Acids from NO and other NOx species over CoFe alloy-decorated Self-standing Carbon Fiber Membranes.

The conversion of industrial exhaust gases of nitrogen oxides into high-value products is significantly meaningful for global environment and human health. And green synthesis of amino acids is vital for biomedical research and sustainable development of mankind. Herein, we demonstrate an innovative approach for converting nitric oxide (NO) to a series of α-amino acids (over 13 kinds) through electrosynthesis with α-keto acids over self-standing carbon fiber membrane with CoFe alloy. The essential leucine exhibits a high yield of 115.4 μmol h-1 corresponding a Faradaic efficiency of 32.4 %, an

Fast label-free identification of bacteria by synchronous fluorescence of amino acids.

Fast identification of pathogenic bacteria is an essential need for patient's diagnostic in hospitals and environmental monitoring of water and air quality. Bacterial cells consist of a very high amount of biological molecules whose content changes in response to different environmental conditions. The similarity between the molecular compositions of different bacterial cells limits the possibility to find unique markers to enable differentiation among species. Although many biological molecules in the cells absorb at the UV-Vis region, only a few of them can be detected in whole cells by thei

Nutrient content of earthworms consumed by Ye'Kuana Amerindians of the Alto Orinoco of Venezuela.

For the Makiritare (Ye'Kuana) native people of the Alto Orinoco (Venezuela), earthworms (Anellida: Glossoscolecidae) are an important component of the diet. Two species in particular are widely consumed: 'kuru' (Andiorrhinus kuru n. sp.) and 'motto' (Andiorrhinus motto). We analysed eviscerated kuru body proper, and whole and smoked preparations of motto for their content of protein and amino acids, fatty acids and 20 minerals and trace elements. The samples contained large amounts of protein (64.5-72.9% of dry weight), essential amino acids, calcium and iron together with notable quantities o

Free amino acids in plasma and muscle of high yielding dairy cows in early lactation.

Free AA in plasma and muscle were monitored in 36 dairy cows from 2 wk before until 15 wk after parturition. Cows were kept indoors and fed individually. The AA concentrations in plasma from 6 to 15 wk of lactation were compared with precalving concentrations. Plasma concentrations of Met, Phe, Glu, and Gln decreased by 16, 24, 25, and 25%, respectively. The ranking of the essential AA according to their decrease in plasma corresponded to the order in which they generally appear to be limiting for milk protein synthesis. The decrease of Glu and Gln in plasma exceeded that of the essential AA a

Roles of Nutrients in the Brain Development, Cognitive Function, and Mood of Dogs and Cats.

The brain is the central commander of all physical activities and the expression of emotions in animals. Its development and cognitive health critically depend on the neural network that consists of neurons, glial cells (namely, non-neuronal cells), and neurotransmitters (communicators between neurons). The latter include proteinogenic amino acids (e.g., L-glutamate, L-aspartate, and glycine) and their metabolites [e.g., γ-aminobutyrate, D-aspartate, D-serine, nitric oxide, carbon monoxide, hydrogen sulfide, and monoamines (e.g., dopamine, norepinephrine, epinephrine, and serotonin)]. Impact of cooking and food matrix structure on the nutritional profile of quinoa leaf protein extracts assessed by the INFOGEST static in vitro digestion method.

Proteins are essential macronutrients with increasing global demand. Green leaves have been proposed as a promising source of alternative proteins. Chenopodium quinoa leaves were used in this work to produce quinoa leaf protein concentrates (QPC) containing RuBisCO. The protein nutritional quality was assessed using the INFOGEST static in vitro digestion method, and the effects of cooking and gelling on protein digestibility determined. The protein extract showed high protein digestibility (>95%), regardless of the applied cooking treatment or the formulation of the solid gels. Additionally, t

Lysine catabolism, amino acid transport, and systemic acquired resistance: what is the link?

Lysine is an essential amino acid for human nutrition, which is generally low in cereal diets. Its biosynthesis via the aspartate-pathway and catabolism is controlled by complex feedback mechanisms. Recently, aspartate-derived amino acids were found to be elevated during pathogen infection in Arabidopsis and a lysine catabolite, pipecolic acid, was identified as critical regulator of systemic acquired resistance. Pipecolic acid is mobile in plants, functions as an intensifier of defense responses and mediates systemic acquired resistance establishment via signal amplification. The altered path

Essential Roles of Nutrient Factors in Learning and Memory.

Essential nutrient factors, including water- and fat-soluble vitamins, minerals, and essential amino acids, play an important role in brain function. We have investigated the roles of these nutrients in learning and memory in mice. Interestingly, we found that dietary deficiency of vitamin B1 or magnesium, inhibition of the vitamin A signaling pathway, and restricted intake of tryptophan impair hippocampus-dependent memory. Furthermore, magnesium deficiency causes neuroinflammation in the hippocampus. Conversely, dietary heat-killed Lactobacillus species enhance hippocampus-dependent memory. T

Trophoblast-specific overexpression of Slc7a5 increases fetal amino acid availability and hepatic mTOR signalling and causes increased fetal growth.

Abnormal fetal growth is linked to perinatal complications and increased risk of metabolic and cardiovascular diseases, yet the underlying mechanisms remain unclear. Placental System L amino acid transport is decreased in human pregnancies complicated by fetal growth restriction and increased in fetal overgrowth. We previously demonstrated that trophoblast-specific overexpression (OX) of Slc7a5/LAT1 enhances transplacental leucine transport and fetal growth. However, the mechanistic link between increased placental transfer of leucine and stimulation of fetal growth remains unclear. We hypothe

Ligand-independent role of the beta 4 integrin subunit in the formation of hemidesmosomes.

Recently, we have shown that a region within the beta4 cytoplasmic domain, encompassing the second fibronectin type III (FNIII) repeat and the first 27 amino acids of the connecting segment, is critical for the localization of alpha6 beta4 in hemidesmosomes. In addition, this region was shown to regulate the distribution of HD1/plectin in transfected cells. In order to investigate the function of the beta4 extracellular and cytoplasmic domains in the assembly and integrity of hemidesmosomes, we have constructed chimeric receptors consisting of the extracellular and transmembrane domains of the

The regulation of the sulfur amino acid biosynthetic pathway in Cryptococcus neoformans: the relationship of Cys3, Calcineurin, and Gpp2 phosphatases.

Cryptococcosis is a fungal disease caused by C. neoformans. To adapt and survive in diverse ecological niches, including the animal host, this opportunistic pathogen relies on its ability to uptake nutrients, such as carbon, nitrogen, iron, phosphate, sulfur, and amino acids. Genetic circuits play a role in the response to environmental changes, modulating gene expression and adjusting the microbial metabolism to the nutrients available for the best energy usage and survival. We studied the sulfur amino acid biosynthesis and its implications on C. neoformans biology and virulence. CNAG_04798 e

Amino Acids Supplementation in Cancer: What Do We Feed, the Patient or the Tumor?

BACKGROUND/OBJECTIVES: Diet and obesity contribute to approximately 50% of tumor development. Therefore, nutrition plays a key role not only in cancer prevention but also in determining prognosis. Notably, between 30% and 90% of cancer patients experience malnutrition. Furthermore, the hypercatabolic state induced by tumors leads to widespread protein degradation, clinically manifesting as sarcopenia or cachexia, and ultimately accelerating mortality. This narrative review examines the potential role of amino acids (AAs) in inhibiting tumor growth and counteracting protein-energy malnutrition-

Nonenzyme browning and its effect on protein nutrition.

The nonenzyme browning involves the thermal decomposition of sugars, the caramelization, the decomposition of oxi-acids, the so called "Maillard reaction" between amino acids and carbohydrates, the reaction between oxidized fats and proteins, and those alterations which take place by the alkaline treatment of proteins. The Maillard reaction is of secondary importance in the case of foodstuffs and fodders with low carbohydrate contents (meats, meat meal, fish meal). By the heat treatment, the sulphur-containing amino acids of proteins (cystine, methionine) are damaged primarily because of oxida

General and specialized parenteral amino acid formulations for nutrition support.

Advances in the understanding of amino acid metabolism and of the interaction of amino acids with skeletal muscle, liver, brain, and other tissues have led to refinements of parenteral amino acid solutions. Clinical situations may dictate the use of specific amino acid formulations. Branched-chain amino acid (BCAA) solutions may normalize aberrant amino acid profiles in patients with hepatic encephalopathy; however, controlled trials demonstrate little effect on clinical outcome, and the effectiveness in patients with acute liver failure or undergoing orthotopic liver transplantation is unprov

Primary structure and processing of lysosomal alpha-glucosidase; homology with the intestinal sucrase-isomaltase complex.

Lysosomal alpha-glucosidase (acid maltase) is essential for degradation of glycogen in lysosomes. Enzyme deficiency results in glycogenosis type II. The amino acid sequence of the entire enzyme was derived from the nucleotide sequence of cloned cDNA. The cDNA comprises 3636 nt, and hybridizes with a messenger RNA of approximately 3.6 kb, which is absent in fibroblasts of two patients with glycogenosis type II. The encoded protein has a molecular mass of 104.645 kd and starts with a signal peptide. Sites of proteolytic processing are established by identification of N-terminal amino acid sequen
