Artificial Intelligence-Based Assessment of Pathologic Response to Neoadjuvant Chemoimmunotherapy in Non-Small Cell Lung Cancer: An International Multicenter Cohort Study.

BACKGROUND: Pathologic response (PR), expressed as the percentage of residual viable tumor (%RVT), has emerged as a surrogate endpoint after neoadjuvant chemoimmunotherapy in non-small cell lung cancer (NSCLC). However, PR assessment remains subject to interobserver variability and is insufficiently standardized across institutions and clinical trials. We hypothesized that artificial intelligence (AI)-based %RVT scoring would be concordant with expert manual assessment and consistent across real-world, multi-scanner cohorts. MATERIALS AND METHODS: Four retrospective NSCLC cohorts treated with

Construction of artificial periosteum with methacrylamide gelatin hydrogel-wharton's jelly based on stem cell recruitment and its application in bone tissue engineering.

The presence of periosteum can greatly affect the repair of a bone fracture. An artificial periosteum imitates the biological function of natural periosteum, which is capable of protecting and maintaining bone tissue structure and promoting bone repair. In our artificial periosteum, biocompatible methacrylate gelatin was used to provide the mechanical support of the membrane, E7 peptide added bioactivity, and Wharton's jelly provided the biological activity support of the membrane, resulting in a hydrogel membrane (G-E-W) for the chemotactic recruitment of bone marrow mesenchymal stem cells (B

Personalized Cell Therapy for Patients with Peripheral Arterial Diseases in the Context of Genetic Alterations: Artificial Intelligence-Based Responder and Non-Responder Prediction.

Stem/progenitor cell transplantation is a potential novel therapeutic strategy to induce angiogenesis in ischemic tissue, which can prevent major amputation in patients with advanced peripheral artery disease (PAD). Thus, clinicians can use cell therapies worldwide to treat PAD. However, some cell therapy studies did not report beneficial outcomes. Clinical researchers have suggested that classical risk factors and comorbidities may adversely affect the efficacy of cell therapy. Some studies have indicated that the response to stem cell therapy varies among patients, even in those harboring li

Polyurethane foam/spheroid culture system using human hepatoblastoma cell line (Hep G2) as a possible new hybrid artificial liver.

The risk of xenozoonosis infections poses the greatest obstacle against the clinical application of hybrid artificial liver support system (HALSS). Primary human hepatocytes are an ideal source for HALSS, but the shortage of human livers available for hepatocyte isolation limits this modality. To resolve this issue, we used human hepatocytes with replication capacity (fetal hepatocytes, Hep G2, and Huh 7) in a polyurethane foam (PUF)/spheroid culture system in vitro, and analyzed liver functions such as ammonia removal and albumin synthesis capacity; results were compared to those of porcine h

Detection of 14q32 translocations in B-cell malignancies by in situ hybridization with yeast artificial chromosome clones containing the human IgH gene locus.

Partner sites of 14q32 translocations found in B-cell malignancies were detected by fluorescence in situ hybridization (FISH) using yeast artificial chromosome (YAC) clones, Y20 and Y6, containing the human Ig heavy chain (IgH) gene locus. Y20 spans a 160-kb upstream and 40-kb downstream region of the JH segments on chromosome band 14q32.33. Y6 is 300-kb upstream of Y20, and spans a further 320-kb telomeric region. The human DNA sequences amplified by Alu polymerase chain reaction of the YAC clones were used as probes for FISH to study six patients with non-Hodgkin's lymphoma (NHL), one patien

Development and validation of an artificial intelligence model for basal cell carcinoma detection and section completeness assessment in Mohs micrographic surgery in an Asian cohort.

BACKGROUND: Mohs micrographic surgery for basal cell carcinoma (BCC) requires rapid intraoperative histologic interpretation. This is labor-intensive and incomplete tissue sections compromise margin assessment. Artificial intelligence has shown promise in BCC detection but section completeness assessment remains underexplored. OBJECTIVE: Develop and validate an artificial intelligence model for BCC detection and section completeness assessment on Mohs frozen-section histology. METHODS: We conducted a retrospective single-center cohort study using histology slides from 130 patients who underw

Integrated transcriptomic and single cell analysis combined with artificial neural network identifies a robust gene signature for early discrimination of MASL and MASH.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease, ranging from simple steatosis (MASL) to metabolic dysfunction-associated steatohepatitis (MASH). However, reliable noninvasive strategies for accurately distinguishing MASL from MASH at an early stage remain limited. We therefore aimed to develop a robust molecular model to improve early identification of disease progression and subtype discrimination. METHODS: Five datasets from the Gene Expression Omnibus were integrated as a training cohort comprising 149 MASL and 158 M

Senescent Cell Derived Artificial Vesicle-Based Senolytic Sonovaccine Platform with Augmented Lymph Node Delivery and Antigen Cross-Presentation Efficacy.

The efficacy of senolytic vaccines aiming to clear senescent cells is limited by narrow antigen coverage and inefficient CD8+ T cell priming due to poor lymph node (LN) accumulation and inefficient antigen cross-presentation. Here, we report SenoVac, a modular dual-component nanovaccine platform explicitly consisting of two sequentially administered key components: an "albumin hitchhiking" DSPE-PEG-DBCO conjugate, and azide-functionalized senescent cell-derived artificial vesicles (SCAVs) that co-encapsulate the sonosensitizer hematoporphyrin monomethyl ether (HMME) and the TLR7/8 agonist adju

Influencing factors and prediction of growth heterogeneity in solid nodule non-small cell lung cancer based on artificial intelligence: a prospective study.

BACKGROUND: The identification of rapidly growing solid nodules (SNs) through various preliminary examinations and their prompt removal can significantly improve the prognosis of patients with solid nodular lung cancer. However, previous studies have mostly focused on determining the nature of the solid nodules, with limited research on their growth heterogeneity. This study aimed to identify multi-dimensional factors influencing rapid nodule growth based on clinical, imaging, pathological, and genetic characteristics and provide a predictive model for solid nodule lung cancer growth. METHODS

Comparison of artificial intelligence image processing with manual leucocyte differential to score immune cell infiltration in a mouse infection model of cystic fibrosis.

Immune cell differentials are most commonly performed manually or with the use of automated cell sorting devices. However, manual review by research personnel can be both subjective and time consuming, and cell sorting approaches consume samples and demand additional reagents to perform the differential. We have created an artificial intelligence (AI) image processing pipeline using the Biodock.ai platform to classify immune cell types from Giemsa-stained cytospins of mouse bronchoalveolar lavage fluid. Through multiple rounds of training and refinement, we have created a tool that is as accur
