Evaluation of seven experimental phages for inclusion in the international phage set for the epidemiological typing of Listeria monocytogenes.

The purpose of our study was to evaluate the inclusion of seven experimental phages into the international phage set for subtyping Listeria monocytogenes. The seven additional phages included the broad-host-range virulent Myoviridae phage A511 (M. J. Loessner, Appl. Environ. Microbiol. 57:1912-1918, 1991), three temperate phages from the Danish subsystem for typing serotype 1/2 strains (12682, 6223, and 5775) (P. Gerner-Smidt, V.T. Rosdahl, and W. Frederiksen, APMIS 101:160-167, 1993), and three temperate phages isolated by this laboratory in France (9425, 1313, and 197). A panel of 395 Lister

Phage types and drug susceptibility patterns of Staphylococcus aureus from two hospitals in northwest Ethiopia.

Fifty-eight Staphylococcus aureus strains from Gonder College of Medical Sciences Hospital, (43 isolates from patients and 15 from carriers) and 28 from Debre Tabor Hospital (14 each from patients and carriers), both in northwest Ethiopia, were phage typed in 1988. Eighty-one of the 86 strains were recognized by their phage types. A multiresistant strain of type 29 was frequent among Gonder patients'. This strain was also recovered from the hospital personnel. Other well known multiresistant strains of phage type NT/A994 and 85/A994 were identified among patients from Gonder and phage type 85/

Further results and experiences with phage-typing of Listeria.

Results of phage-typing of 22 strains from 16 listeriosis cases, which were isolated during an epidemic outbreak in the district of Magdeburg, showed that most strains were determined by the octal codes 73611, 73615, 73655, 73645. We suppose that this was the epidemic strain, although also other phage-patterns (40000, 00600, 03200) were found. The source of the epidemic outbreak could not cleared up. The purpose of further investigations was to establish how far phage-patterns of Listeria from the same patients from different materials might be identified as identical, similar or different. We

Salmonella typhi R-plasmids in Hungary.

The antibiotic sensitivity of 4095 Salmonella typhi strains isolated in Hungary from January, 1974 to June 1979, was tested. Twelve strains derived from one patient and seven chronic carriers were resistant to antibiotics due to R-plasmids. One of the S. typhi strains carried two R-plasmids and the change in the phage type of this strain was caused by one of the plasmids. R-plasmid of the same restrictive property was found in an Escherichia coli strain isolated from the faeces of the corresponding carrier. All of the R-plasmids examined were fi-, E. coli and Shigella flexneri phages were rest

[Phage-typing and tetracycline resistance in Salmonella panama strains of animal origin in the Federal Republic of Germany (1969-1975) (author's transl)].

505 out of 1369 S. panama strains isolated mainly from animals, foods, and feedstuffs from 1969 to 1975 were phage-typed. 41% (= 207) of the S. panama isolates examined originated from swine, 26% from cattle and calves, 15% from dogs, and 13% were from other animal species and materials. 465 strains (=92%) showed typical reactions. 23% were specified as phage-type A, 24% as phage-type B and 36% as phage-type G. Phage-types D, E, and F did not occur. In swine, being the most frequent carriers of S. panama, phage-types A, B, and G had nearly the same shares. In dogs, phage-type G predominated to

Combinatorially Screened Peptide as Targeted Covalent Binder: Alteration of Bait-Conjugated Peptide to Reactive Modifier.

A peptide-type covalent binder for a target protein was obtained by combinatorial screening of fluoroprobe-conjugated peptide libraries on bacteriophage T7. The solvatochromic fluoroprobe works as a bait during the affinity selection process of phage display. To obtain the targeted covalent binder, the bait in the selected consensus peptide was altered into a reactive warhead possessing a sulfonyl fluoride. The reaction efficiency and site/position specificity of the covalent conjugation between the binder and the target protein were evaluated by liquid chromatography-tandem mass spectrometry

Development of an ex vivo model to study Staphylococcus aureus invasion of the osteocyte lacuno-canalicular network.

Staphylococcus aureus has multiple mechanisms to evade the host's immune system and antibiotic treatment. One such mechanism is the invasion of the osteocyte lacuno-canalicular network (OLCN), which may be particularly important in recurrence of infection after debridement and antibiotic therapy. The aim of this study was to develop an ex vivo model to facilitate further study of S. aureus invasion of the OLCN and early-stage testing of antibacterial strategies against bacteria in this niche. The diameter of the canaliculi of non-infected human, sheep, and mouse bones was measured microscopica

Bacteriophage Sf14 survival under simulated gastrointestinal conditions.

Shigella flexneri is an enteric pathogen responsible for causing shigellosis, a type of bacillary dysentery of significant global health concern. The increasing prevalence of antibiotic resistance has driven the search for alternative treatments, including bacteriophage (phage) therapy. However, the effectiveness of orally administered phages for enteric pathogens depends on their ability to survive passage through the gastrointestinal tract. In this study, the ability of bacteriophage Sf14 to maintain infectivity was evaluated under simulated gastrointestinal conditions using a modified INFOG

Application of Phagotherapy in the Treatment of Burn Patients (Review).

Treatment of patients with a burn injury is a complex process involving multicomponent multidirectional intensive therapy of the majority of organs and systems damaged by thermal effects on the skin, alternating with repeated surgical interventions aimed at removing nonviable tissues with subsequent plastic closure of wound defects. After the recovery from the burn shock, local infectious complications are considered to be the leading problem that decelerates the process of recovery and is the main cause of lethal outcomes. Since the skin integrity is broken, microorganisms penetrate readily i

Genomics of Salmonella phage ΦStp1: candidate bacteriophage for biocontrol.

Multidrug-resistant Salmonella causing Salmonellosis is a food-borne pathogen and hence a public health hazard. Alternatives to antibiotics, such as phages, are possible solutions to this increasing drug resistance. In this context, several Salmonella phages were isolated and characterized. This paper describes the physiochemical and whole genome characterization of one such bacteriophage, ΦStp1, which efficiently infects serovars Salmonella Enteritidis and Salmonella Typhimurium. Morphological observations by transmission electron microscopy and phylogenetic analysis using terminase gene clas

Sequence of a variant Shiga-like toxin type-I operon of Escherichia coli O111:H-.

PCR amplification was used to screen faecal isolates of Escherichia coli from a 12-month-old boy with haemolytic uraemic syndrome for the presence of Shiga-like toxin (SLT)-encoding genes. One isolate, belonging to serotype O111:H-, was positive for SLT-I by this method. UV induction indicated that the strain was lysogenic for a lambdoid bacteriophage, but this did not encode the toxin. Southern hybridization analysis of chromosomal DNA revealed that the SLT-I gene was located on an 8.5-kb EcoRI fragment. SLT-I was further localized to within a 3.0-kb SphI-EcoRI fragment. A separate subclone c

Characterization and therapeutic efficacy of phage p9676 against epidemic ST11-KL64 Klebsiella pneumoniae: Insights from genomic analysis and in vivo studies.

INTRODUCTION: Klebsiella pneumoniae presents a formidable challenge in clinical settings due to its increasing resistance to last-line antibiotics like carbapenem, tigecycline, and colistin, raising global concerns. An emerging alternative strategy involves harnessing bacteriophages to combat infections caused by multidrug-resistant bacteria. METHODS AND RESULTS: In this study, we isolated the bacteriophage p9676 from hospital sewage and demonstrated its ability to lyse K. pneumoniae strains with ST11-KL64 capsular type. Transmission electron microscopy and genomic analysis identified p9676 a

Lowered Abundance of Gut Bacteriophage Species Is Associated With Human Cancer Cachexia.

BACKGROUND: Cancer cachexia exemplifies a high medical need condition without effective treatment. Recent studies implicated bacterial gut microbiome alterations to cancer cachexia. Whether the gut bacteriophage profile, an important microbiome component for health and disease, is also related to cancer cachexia remains unknown. We aimed to profile gut microbiome alterations in human cancer cachexia with attention on bacteriophages. METHODS: We performed shotgun metagenomic sequencing in stool samples from 78 cachectic and 42 noncachectic patients (53% male, mean age 67 ± 8 years) with newly

Pulsed-field gel electrophoresis as an epidemiologic tool in the investigation of laboratory acquired Salmonella typhi infection.

Strains of Salmonella typhi implicated in two separate cases of laboratory acquired infection from patients and the medical laboratory technologists who processed the patients' samples were analysed by pulsed-field gel electrophoresis. Although all four isolates were of bacteriophage type E1, PFGE was able to demonstrate that the strains responsible for the two laboratory acquired cases were not genetically related. The PFGE patterns of the isolates from the MLTs were found to be identical to those of the corresponding patients after digestion with restriction enzyme AvrII. This provided genet
