Enhanced whole-cell biotransformation of carboxylic acid reductase enzymes via tandem expression of phosphopantetheinyl transferase.

Carboxylic acid reductases (CARs) function as versatile biocatalysts with a broad substrate range. However, CARs still struggled with relatively low activity in many enzyme reactions. While extensive efforts have focused on engineering CAR itself, the influence of phosphopantetheinyl transferase (Sfp), which is essential for CAR activation, has received significantly less attention. Consequently, information regarding the optimal ratio of CAR to Sfp for efficient biotransformation remains limited. To touch this issue, we investigated the effect of tandem sfp genes to CAR by introducing CAR-med…

Biotransformation of enrofloxacin-copper combined pollutant in aqueous environments by fungus Cladosporium cladosporioides (CGMCC 40504).

The combined pollution of antibiotics and heavy metals in aqueous environments increases the risk of aquatic ecosystem disruption and the complication of pollutant management. Here, a fungus Cladosporium cladosporioides 11 (CC11) isolated from aquaculture pond sediments possessed effective capacities to biotransform the combined pollution of enrofloxacin (ENR) and copper ion (Cu). ENR and Cu were considerably abated by CC11, and the presence of Cu (30 mg/L) promoted the biotransformation efficiency of ENR. The biotranformation of ENR in ENR-Cu co-contamination was associated to ligninolytic en…

Redox-Activated Contrast-Enhanced T1-Weighted Imaging Visualizes Glutathione-Mediated Biotransformation Dynamics in the Liver.

GSH-mediated liver biotransformation is a crucial physiological process demanding efficient research tools. Here, we report a type of amorphous FexMnyO nanoparticles (AFMO-ZDS NPs) as redox-activated probes for in vivo visualization of the dynamics of GSH-mediated biotransformation in liver with T1-weighted magnetic resonance imaging (MRI). This imaging technique reveals the periodic variations in GSH concentration during the degradation of AFMO-ZDS NPs due to the limited transportation capacity of GSH carriers in the course of GSH efflux from hepatocytes to perisinusoidal space, providing dir…

Biotransformation Capacity of Zebrafish (Danio rerio) Early Life Stages: Functionality of the Mercapturic Acid Pathway.

Zebrafish (Danio rerio) early life stages offer a versatile model system to study the efficacy and safety of drugs or other chemicals with regard to human and environmental health. This is because, aside from the well-characterized genome of zebrafish and the availability of a broad range of experimental and computational research tools, they are exceptionally well suited for high-throughput approaches. Yet, one important pharmacokinetic aspect is thus far only poorly understood in zebrafish embryo and early larvae: their biotransformation capacity. Especially, biotransformation of electrophil…

Granule size-dependent biotransformation of organic micropollutants in aerobic granular sludge under different bioconversion conditions.

Organic micropollutants (OMPs) are commonly detected in municipal wastewater. Conventional activated sludge processes partially remove these compounds, allowing them to enter receiving waters and pose ecological risks. Biotransformation, governed by microbial community composition and activity, is the main pathway for OMP removal. Aerobic granular sludge (AGS), with its distinct structure and microbial communities compared to conventional activated sludge, has emerged as a promising alternative. Full-scale AGS reactors contain predominately large granules (>1 mm), alongside medium (0.2-1 mm), …

Enhancing ginsenoside biotransformation by engineering a thermophilic enzyme: Disulfide bond-induced activity-thermostability trade-off.

The thermophilic β-glycosidase from Sulfolobus solfataricus (SS-Bgl) has great potential for producing rare ginsenoside CK from ginseng stems and leaves, which are rich in main ginsenosides. To enhance its thermal stability, we designed disulfide bonds based on MD simulations and the Disulfide by Design 2.0 (DbD2) tool. Unexpectedly, introducing disulfide bonds in flexible loops (mutants P95C/R178C and F98C/R178C) significantly increased catalytic activity but decreased stability. The CK productivity from ginsenosides Rb1, Rb2, and Rd of the mutant P95C/R178C increased by 83.51%, 10.90%, and 1

BHT and Its Metabolites Disrupt β2-Adrenergic Signaling: A Biotransformation-Dependent Cardiotoxicity Mechanism in Zebrafish.

Butylated hydroxytoluene (BHT) is a widely used synthetic phenolic antioxidant, ubiquitously detected in aquatic environments. While traditionally considered safe based on in vitro data, BHT adversely affects aquatic organisms at environmentally relevant concentrations. Here, we demonstrate that BHT-induced cardiotoxicity is primarily mediated by its metabolites through disruption of β2-adrenergic receptor (β2AR) signaling. In zebrafish larvae exposed to 0.01-1 μM BHT for 6 days, metabolites BHT-CHO, BHT-COOH, and BHT-Q accumulated prominently, inducing cardiac morphological and histological a

Relationship between biotransformation enzyme genes in polycyclic aromatic hydrocarbon metabolism, telomere length and missed abortion.

The mechanism underlying the effects of Polycyclic aromatic hydrocarbons (PAHs) on missed abortion (MA) remains unclear. This study explored the relationship between PAHs exposure, telomere length (TL), metabolizing enzyme gene polymorphism, and MA in a case-control study with 253 pregnant women. A competitive enzyme-linked immunosorbent assay (ELISA) was used to quantify PAH-DNA adducts. The DNA was extracted from villi tissue for the analysis of TL using the real-time quantitative polymerase chain reaction (qPCR) method. Single nucleotide polymorphism (SNP) genotyping was performed using mul

Tacrine First-Phase Biotransformation and Associated Hepatotoxicity: A Possible Way to Avoid Quinone Methide Formation.

Tacrine was withdrawn from clinical use as a drug against Alzheimer's disease in 2013, mainly due to drug-induced liver injury. The culprit of tacrine-associated hepatotoxicity is believed to be the 7-OH-tacrine metabolite, a possible precursor of quinone methide (Qmeth), which binds to intracellular -SH proteins. In our study, several different animal and human models (liver microsomes, primary hepatocytes, and liver slices) were used to investigate the biotransformation and hepatotoxicity of tacrine and its 7-substituted analogues (7-methoxy-, 7-phenoxy-, and 7-OH-tacrine). Our goal was to f…

Detection of dynorphin 1-17 biotransformation fragments in human nasal polyps by UPLC-QTOF-MS.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a persistent inflammation of the sinonasal mucosa. CRSwNP treatments are associated with inconsistent efficacy and recurrence of symptoms. Dynorphin 1-17 (DYN 1-17) and its fragments have been shown to modulate the immune response in various inflammatory conditions. This study aimed to investigate the effect of different pH and degrees of inflammation on DYN 1-17 metabolism in human CRSwNP tissues. DYN 1-17 was incubated with grade 3 and grade 4 inflamed tissues of CRSwNP patients at pH 5.5 and pH 7.4 over a range of incubation periods. The

Biotransformation of sulfamethoxazole (SMX) by aerobic granular sludge: Removal performance, degradation mechanism and microbial response.

Aerobic granular sludge (AGS) is a promising biotechnology for the treatment of antibiotic-rich wastewater. However, little is known about the antibiotics degradation mechanism and microbial response in a sulfamethoxazole (SMX)-loaded AGS system. Herein, the results of a continuous 240 days test suggested that 0.5-5 mg/L of SMX could be thoroughly removed by AGS via adsorption and degradation. The degradation pathway of SMX involved the hydrolysis of the sulfonamide bond and cleavage of NS or CS bonds, subsequently leading to the production of small molecular substances (e.g. benzene and 5-met
