Skin autoimmunity and blood coagulation.

Evidence exists that the immune and coagulation systems are simultaneously activated in some systemic autoimmune disorders. Although proinflammatory mediators induce tissue factor (TF) expression, the main initiator of blood coagulation, activated proteases of coagulation may act on protease-activated receptors (PAR) triggering inflammation. Such a cross-talk amplifies and maintains the activation of both systems. This review focuses on the involvement of immune and coagulation system in two skin disorders as chronic urticaria (CU), autoimmune in about 45% of cases, and bullous pemphigoid (BP)

Alterations in the Blood-Brain Barrier in Mood Disorders and Neurodegenerative Diseases.

Depressive disorders and suicidal behaviors represent major causes of health loss. Modifications of brain microvasculature, and specifically alterations of the blood-brain barrier have been supposed to participate in the vulnerability to those disorders along with cognitive impairment, especially in the older adults. In this article, we addressed evidence linking blood-brain barrier impairments with mood disorders and suicide. Secondly, we investigated their relationship with depression in old age, and with neurodegenerative processes. Particular attention was drawn toward the potential intera

Cyclone-induced hematological emergencies: preparedness and response from a hematologist's viewpoint.

Cyclone disasters pose significant health challenges, including severe hematological emergencies resulting from trauma, infections, and systemic inflammatory responses. Injuries sustained during cyclones can lead to trauma-induced coagulopathy, acute anemia, and disseminated intravascular coagulation (DIC), requiring immediate intervention. Additionally, post-disaster conditions such as malnutrition, dehydration, and poor sanitation contribute to increased susceptibility to infections, further exacerbating blood-related disorders. Hematologists play a vital role in diagnosing and managing thes

Chitosan/oxidized cellulose composite nanofiber sponges: a rapid and effective hemostasis strategy for non-compressible hemorrhage.

Non-compressible hemorrhagic wounds, prevalent in combat and mass trauma scenarios, are characterized by arterial injury-induced exsanguination and geometrically complex wound morphology that challenges conventional hemostatic interventions. This paper presents a novel nanofiber hemostatic sponge (CT@GP-HAc), made from 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO) oxidized cellulose nanofibers (TOCN) and chitosan nanofibers (CSNF), crosslinked with genipin (GP) in an acetic acid atmosphere. The sponge features excellent shape memory and rapid hemostatic capabilities, specifically design

[Pre-hospital diagnosis of nosebleed in children].

Early signs of haemorrhages develop in the childhood, nose bleeding being predominant. Recurrent nose bleeding was observed in 130 (89.0%) patients with thrombocytopathies, 37 patients with nasal pathologies, 13 out of 14 children having willebrand's disease, 3 patients suffering from Rendu-Osler disease, in a boy with a factor XIII deficiency, and in a girl with psoriasis. In the case of haemorrhagic diathesis, nose bleeding was, as a rule, combined with haemorrhages of other types (skin, gum, uterine, etc.) whereas in the case of ENT pathology, nose bleeding was the only haemorrhagic symptom

Magnetic coagulometry: towards a new nanotechnological tool for ex vivo monitoring coagulation in human whole blood.

Blood clotting disorders consisting of unwanted blood clot formation or excessive bleeding are some of the main causes of death worldwide. However, there are significant limitations in the current methods used to clinically monitor the dynamics of clot formation in human whole blood ex vivo. Here a new magnetic coagulometry platform for testing ex vivo coagulation is described. This platform exploits the sensitivity of the out-of-phase component of alternating current (AC) magnetic susceptibility (χ'') to variations in mobility and agglomeration of magnetic nanoparticles when trapped during bl

Testing strategies used in the diagnosis of rare inherited bleeding disorders.

INTRODUCTION: Rare Bleeding Disorders have a low population prevalence and may not be recognized by most clinicians. In addition, knowledge gaps of the indicated laboratory tests and their availability add to the potential for delayed diagnosis or misdiagnosis. The lack of widely available commercial, regulatory body approved esoteric tests limits them to reference laboratories, thus limiting easy access for patients. AREAS COVERED: A literature search of PubMed, Medline, and Embase and a review of international society guidelines were performed. Additional references from published articles

Hemostasis assessment in Fontan patients using the new thrombodynamics test.

BACKGROUND: Thromboembolic events are a well-known risk for Fontan patients and often lead to morbidity and mortality in cyanotic patients and patients with a single ventricle. Coagulopathy and thrombophilia, in addition to disturbed Fontan blood flow and endothelial injury, are major contributors to thromboembolic complications. However, there is currently no consensus regarding the optimal medication to prevent or treat these events. Identification of coagulation disorders is therefore crucial for selecting appropriate management strategies and evaluating long-term outcomes in these patients

Monitoring the Formation of Fibrin Clots as Part of the Coagulation Cascade Using Fluorescent Single-Walled Carbon Nanotubes.

Blood coagulation is a critical defense mechanism against bleeding that results in the conversion of liquid blood into a solid clot through a complicated cascade, which involves multiple clotting factors. One of the final steps in the coagulation pathway is the conversion of fibrinogen to insoluble fibrin mediated by thrombin. Because coagulation disorders can be life-threatening, the development of novel methods for monitoring the coagulation cascade dynamics is of high importance. Here, we use near-infrared (NIR)-fluorescent single-walled carbon nanotubes (SWCNTs) to image and monitor fibrin

Genetic diagnosis of haemophilia and other inherited bleeding disorders.

Inherited deficiencies of plasma proteins involved in blood coagulation generally lead to lifelong bleeding disorders, whose severity is inversely proportional to the degree of factor deficiency. Haemophilia A and B, inherited as X-linked recessive traits, are the most common hereditary hemorrhagic disorders caused by a deficiency or dysfunction of blood coagulation factor VIII (FVIII) and factor IX (FIX). Together with von Willebrand's disease, a defect of primary haemostasis, these X-linked disorders include 95% to 97% of all the inherited deficiencies of coagulation factors. The remaining d

The role of peripheral blood smear examination in the evaluation of suspected platelet-related disorders in children: A practical approach and an illustrated review.

Platelets, along with coagulation factors and vasculature, represent the three main compartments of hemostasis. Upon investigation of a suspected hemostasis disorder, platelet count, size and morphology often offer important clues to the diagnosis or help narrow the differential diagnosis. In this review, we describe a general approach to diagnosing platelet disorders, starting with easily obtained data such as findings of complete blood count (CBC) and microscopic review of a stained peripheral blood smear. We discuss general findings that help separate consumptive from underproduction thromb

Clot waveform analysis of prolonged activated partial thromboplastin time in various disorders.

INTRODUCTION: The prolongation in APTT reflects various disorders affecting the blood coagulation system. Automated coagulation analyzers such as ACL TOP, help in evaluating the clotting cascade by displaying clot reaction curves or waveforms. The fibrin formation curve forms S-shaped curve showing the changes in the absorbance on X-axis and time in seconds on Y-axis. The clotting data is processed to obtain derivatives. MATERIALS AND METHODS: This observational study analyzed 80 samples including 20 samples from normal healthy individuals as control. The parameters studied were time (secs),
