Gut Microbiota-Derived Butyrate Strengthens Blood-Brain Barrier Integrity and Attenuates Streptococcus suis SC19-Induced Meningitis: Involvement of the Gut-Brain Axis.

Streptococcus suis serotype 2 (SS2) is a major zoonotic pathogen that causes severe meningitis and high mortality in both humans and swine. Growing evidence shows that metabolites generated along the gut-brain axis regulate neuroinflammation. In this study, we demonstrate that oral administration of sodium butyrate (NaB) (300 or 600 mg/kg/day) for 28 days lowered mortality and lessened neuropathological signs in mice infected with SS2 (SC19 strain). Butyrate prophylaxis was associated with lower bacterial loads in the blood and brain tissue, preserved expression of tight junction proteins (ZO-

The gut microbiota influences neurodegenerative diseases through the gut-brain axis: molecular mechanisms and effects on immune function.

The pathogenesis of neurodegenerative diseases (NDDs), including Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS), is complex and multifactorial. Recent studies indicate that the microbiota-gut-brain axis (MGBA) plays a crucial role in the development and progression of NDDs. The MGBA concept reveals a complex bidirectional regulatory network between the gut microbiota and the central nervous system (CNS), linking them through immune, neural, endocrine, and metabolic pathways. This review summarizes the components of the MGBA, communication pathways b

Gut Dysbiosis and Microbiota-Derived Metabolites in Neurodegenerative Diseases: Molecular and Biochemical Mechanisms Along the Gut-Brain Axis.

Neurodegenerative disorders such as Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS) share key molecular features, including neuroinflammation, oxidative stress, mitochondrial dysfunction, and progressive neuronal loss. Increasing evidence indicates that gut dysbiosis and alterations in microbiota-derived metabolites are involved in these processes through multiple pathways along the gut-brain axis. However, while broad compositional changes are well-documented, a critical knowledge gap remains regarding the specific biochemical signal transduction pa

From gut leakage to memory loss: environmental 2,4-DTBP triggers neuroinflammation through the gut-brain axis in zebrafish.

Despite widespread environmental detection of 2,4-di-tert-butylphenol (2,4-DTBP), its neurotoxic impacts on cognitive behavior remain largely unexplored. In this study, adult zebrafish were exposed to 0.01, 0.1, and 1 μM 2,4-DTBP for 28 days. T-maze testing revealed dose-dependently longer path lengths to the food-baited target zone, indicative of impaired spatial memory. Evans blue extravasation demonstrated that 2,4-DTBP increased blood-brain barrier permeability and bioaccumulated in the zebrafish brain. Ultrastructural analysis confirmed impaired synaptic plasticity and neuron injury. Targ

Branched-chain amino acids and gut microbiota: coregulation and impact on neurological function via the gut-brain axis.

Diseases that cause neurological dysfunction, such as Parkinson's disease (PD), Alzheimer's disease (AD), and maple syrup urine disease (MSUD), among others, are characterized by complex and multifaceted etiologies. There is growing evidence that branched-chain amino acids (BCAAs), regulated by the gut microbiota, play a critical role in the development of the central nervous system (CNS) disorders. This review focuses on the potential role of branched-chain amino acid metabolism in regulating brain function and gut microbiota. First, we summarize the current understanding of BCAAs, encompassi

Xinqingning tablet attenuates ischemic stroke complicated by gut dysbiosis through regulating the miR-126-driven gut-brain axis.

BACKGROUND: Ischemic stroke (IS), the predominant clinical stroke subtype, is increasingly linked to dysregulation of the gut-brain axis (GBA)-a bidirectional neuroendocrine-immune interface connecting intestinal homeostasis with cerebrovascular pathophysiology. Xinqingning Tablet (XQNT) demonstrates neuroprotective potential in IS complicated by gut dysbiosis (GD), yet its mechanisms of GBA modulation remain unclear. METHODS: A dual-hit IS-GD mouse model was established via fecal slurry transplantation and permanent middle cerebral artery occlusion (pMCAO) surgery. Gut function was evaluated

Puerarin's multidimensional antidepressant action: decoding the gut-liver-brain axis through gut microbiota, hepatic homeostasis, and neuroimmune regulation.

BACKGROUND: Puerarin is a flavonoid bioactive component extracted from the Chinese herb radix puerariae, which has been reported to have anti-inflammatory and neuroprotective effects and is a potential drug for the treatment of neuroinflammatory diseases. There is increasing evidence that the gut-liver-brain axis is closely related to neurological disorders. However, studies on the use of puerarin for the treatment of depression based on gut-liver-brain axis-mediated inflammatory injury have not been reported. METHODS: In the present study, a 4-week chronic restraint stress (CRS) mouse depres

Gut Microbiota as Neuroimmune Modulators in Myasthenia Gravis: Mechanistic Insights from the Gut-Brain Axis to Therapeutic Innovations.

Myasthenia gravis (MG) is a chronic autoimmune disorder characterized by an immune-mediated attack on neuromuscular junction acetylcholine receptors (AChRs), and its pathogenesis is closely linked to immune dysregulation. Emerging evidence has highlighted the pivotal role of the gut microbiota in the pathophysiology of MG through immunomodulation, microbial metabolite signaling, and gut-brain axis interactions. This review combines 16S rRNA sequencing, metagenomic, and metabolomic data to reveal distinct gut microbial signatures in patients with MG. These signatures include reduced α-diversity

Multi-level analysis of gut microbiome extracellular vesicles-host interaction reveals a connection to gut-brain axis signaling.

UNLABELLED: Microbiota-released extracellular vesicles (MEVs) have emerged as a key player in intercellular signaling. However, their involvement in the gut-brain axis has been poorly investigated. We hypothesize that MEVs cross host cellular barriers and deliver their cargoes of bioactive compounds to the brain. In this study, we aimed to investigate the cargo capacity of MEVs for bioactive metabolites and their interactions with the host cellular barriers. First, we conducted a multi-omics profiling of MEVs' contents from ex vivo and stool samples. Metabolomics analysis identified various ne

Parkinson's Disease and the Microbiota-Gut-Brain Axis: Metabolites, Mechanisms, and Innovative Therapeutic Strategies Targeting the Gut Microbiota.

The human gut microbiota is diverse and abundant and plays important roles in regulating health by participating in metabolism and controlling physiological activities. The gut microbiota and its metabolites have been shown to affect the functioning of the gut and central nervous system through the microbiota-gut-brain axis. It is well established that microbiota play significant roles in the pathogenesis and progression of Parkinson's disease (PD). Disorders of the intestinal microbiota and altered metabolite levels are closely associated with PD. Here, the changes in intestinal microbiota an

D-mannose alleviates rotenone-induced PD mouse model through microbiota-gut-brain axis.

Parkinson’s disease (PD) is the second most common neurological degenerative disease in the elderly. Disruption of gut-brain axis crosstalk has been shown to be associated with the pathogenesis of PD. Although drug treatments for PD have been in development for decades, there is currently no drug or therapy that can completely cure PD. Therefore, there is an urgent need to discover new drugs to develop effective PD treatment strategies. D-mannose is a natural, bioactive monosaccharide that is a popular nutritional and health-beneficial food supplement worldwide. However, whether it has a neuro

The Gut-Brain Axis and Neurodegenerative Diseases: From Mechanistic Pathways to Therapeutic Interventions.

As the global population ages, the incidence of neurodegenerative diseases (NDDs) has increased. Neurodegenerative diseases have complex pathogenesis and currently lack fundamental treatment methods. The gut-brain axis (GBA) participates in the pathogenesis of NDDs through bidirectional communication between the gut microbiota and the central nervous system (CNS). This review systematically introduces the pathogenic characteristics of representative NDDs such as Alzheimer's disease (AD), Parkinson's disease (PD), and multiple sclerosis (MS), as well as the association mechanisms between these

From mechanisms to therapy: Targeting the gut-brain axis in chronic gastrointestinal pain.

Chronic gastrointestinal pain (CGP) is a common and often difficult-to-manage symptom in disorders of gut-brain interaction (DGBI). Owing to the limited efficacy of current therapeutic approaches in a subset of patients, a better understanding of gut-brain axis (GBA) dysfunction may facilitate the development of improved treatment strategies. This review summarizes the pathophysiological mechanisms underlying CGP, focusing on the contributions of microbial dysbiosis, mucosal immune activation, and neuroendocrine disturbances to peripheral and central nociceptive sensitization. Current therapeu

Dietary energy levels regulate feed intake of broilers through the brain-gut axis.

BACKGROUND: Feed intake (FI) in broilers is significantly influenced by dietary energy levels through the brain-gut axis (BGA), profoundly affecting growth performance. The mechanisms underlying dietary energy regulation of FI and intestinal health via BGA remain unclear. AIM: This study investigated the effects of low- (LED, 2,900 kcal/kg), medium- (MED, 3,200 kcal/kg), and high-energy diets (HED, 3,500 kcal/kg) on BGA function over 21 days. RESULTS: We found that increasing dietary energy reduced FI but increased body weight gain and improved feed conversion ratio. MED elevated anorexigeni…

From dysbiosis to postoperative neurocognitive disorders: The anesthesia-disrupted microbiota-gut-brain axis.

BACKGROUND: Postoperative neurocognitive disorders (PND) remains a prevalent and debilitating complication in older surgical patients, with accumulating evidence implicating gut microbiota dysbiosis as a contributing factor. Anesthesia and surgical stress are known to disrupt intestinal microbial ecosystems, but the precise mechanisms by which these perturbations translate into central nervous system pathology are not fully understood. The gut-brain axis has emerged as a critical biological conduit, yet a comprehensive synthesis of the microbial, metabolic, and neuroinflammatory pathways linki

Cognitive Impairment Associated with Chemotherapy: Neuroimmunological Interactions, Gut-Brain Axis, and Therapeutic Approaches.

Chemotherapy is a treatment designed to contain or eradicate neoplastic cells; however, patients may experience various treatment-related adverse effects. Chemotherapy-related cognitive impairment (CRCI), clinically referred to as "chemobrain," is a frequent complication with a duration ranging from months to years, affecting between 17% and 70% of cancer patients. These cognitive deficits not only impair social, educational, and occupational functioning but may also impact survival outcomes, possibly by interfering with medication adherence and health-related behaviors. Emerging evidence has
