[Effect of indirect endolymphatic antibiotic therapy on clinical and immunological indicators in patients with erysipelas of the lower extremities].

Two groups of patients with erysipelas of the lower extremities underwent indirect endolymphatic therapy with bicillin-3 (58 patients) and routine penicillin therapy (79 patients). Comparative clinicoimmunological examinations in the two groups revealed that lymphotropic administration of the antibiotic had a more favourable effect on the disease process, as evidenced by more rapid reverse time courses of erysipelas clinical signs, less frequent incidence of early relapses and normalization of the majority of immunological parameters by the end of the treatment. For estimation of the anti-recu

CD3 hyporesponsiveness and in vitro apoptosis are features of T cells from both malignant and nonmalignant secondary lymphoid organs.

There is a dogma in tumor immunology that tumor-infiltrating lymphocytes (TIL) are defective based on their lack of antitumoral efficacy in vivo and on impaired response to in vitro functional tests. However, TIL have been compared usually with peripheral blood T lymphocytes, raising doubts on the conclusions drawn. Therefore, we compared TIL from B cell non-Hodgkin's lymphomas (NHL) with T cells from nonmalignant secondary lymphoid organs. NHL-TIL were unresponsive to activation by immobilized anti-CD3 mAb, although bypassing T cell receptor (TCR)/CD3 signaling led to proliferation. The poor

The CD4 transmembrane GGXXG and juxtamembrane (C/F)CV+C motifs mediate pMHCII-specific signaling independently of CD4-LCK interactions.

CD4+ T cell activation is driven by five-module receptor complexes. The T cell receptor (TCR) is the receptor module that binds composite surfaces of peptide antigens embedded within MHCII molecules (pMHCII). It associates with three signaling modules (CD3γε, CD3δε, and CD3ζζ) to form TCR-CD3 complexes. CD4 is the coreceptor module. It reciprocally associates with TCR-CD3-pMHCII assemblies on the outside of a CD4+ T cells and with the Src kinase, LCK, on the inside. Previously, we reported that the CD4 transmembrane GGXXG and cytoplasmic juxtamembrane (C/F)CV+C motifs found in eutherian (place

Possible role of natural killer cells in pemphigus vulgaris - preliminary observations.

Pemphigus vulgaris (PV) is an autoimmune blistering disease that affects the skin and multiple mucous membranes, and is caused by antibodies to desmoglein (Dsg) 1 and 3. Natural killer (NK) cells have a role in autoimmunity, but their role in PV is not known. NK cells in the peripheral blood leucocytes (PBL) of 15 untreated Caucasian patients with active PV were studied and compared with healthy controls for the expression of major histocompatibility complex (MHC) class II and co-stimulatory molecules. CD56+ CD16- CD3- NK or CD56+ CD16+ CD3- NK cells from the PBL of PV patients co-express MHC

Immunohistopathogenesis of persistent generalized lymphadenopathy in HIV-positive patients.

Persistent generalized lymphadenopathy (PGL) is a reactive lymphadenitis affecting HIV-positive patients; furthermore, PGL is often a prodrome of AIDS-related complex and AIDS. In the present review the authors describe the histology and the immunohistochemistry of lymph nodes of patients affected by PGL. Histologic alterations of lymph nodes with PGL are classified according to three main types: follicular hyperplasia without or with follicular fragmentation, follicular involution and follicular depletion. Immunohistology demonstrates a peculiar infiltration of CD3+/CD4+ and CD3+/CD8+ lymphoc

Immunohistochemical characterization of the lesions of feline progressive lymphocytic cholangitis/cholangiohepatitis.

The histopathological features of liver biopsies from 20 cats with progressive lymphocytic cholangitis/cholangiohepatitis are reported. These biopsies were subject to immunohistochemical investigation for expression of CD3, CD79. Major Histocompatibility Complex (MHC) Class II molecules, and feline IgG, IgM and IgA. Livers from five normal cats, which were also examined showed constitutive expression of MHC Class II by sinusoidal Kupffer cells and bile duct epithelium, in addition to a population of portal, and bile duct inter-epithelial, CD3+ T lymphocytes. Signaling via the CD2 receptor enhances HTLV-1 replication in T lymphocytes.

Human T lymphotropic virus type 1 (HTLV-1) is considered the etiologic agent of adult T cell leukemia/lymphoma and several chronic progressive immune-mediated diseases. Approximately 1-4% of infected individuals develop disease, generally decades following infection. Increased proviral transcription, mediated by the viral 40-kDa trans-activating protein, Tax, has been implicated in the pathogenesis of HTLV-1-associated diseases. Since the HTLV-1 promoter contains sequences responsive to cyclic AMP and protein kinase C, we hypothesized that lymphocyte activation signals initiated through the TC

T cell receptor-zeta and granzyme B expression in mononuclear cell infiltrates in normal colon mucosa and colon carcinoma.

BACKGROUND: Whereas the presence of a lymphoid infiltrate has been associated with a favourable prognosis in colorectal carcinoma, the proliferative and cytotoxic responses of freshly isolated tumour infiltrating lymphocytes are frequently impaired. In mice, tumour induced immune suppression has been associated with a decreased expression of the zeta-chain of the T cell receptor (TCR)-CD3 complex, and loss of mRNA for granzyme B. AIM: To compare the expression of TCR-zeta and granzyme B in lymphocytes infiltrating normal colonic mucosa and Duke's A and D colorectal carcinomas. SPECIMENS: Par

Lymphocyte disturbance and functional assessment of the [Asp521Asn] ZAP70 mutation.

Activated zeta-chain-associated protein kinase 70 (ZAP70) phosphorylates the TCRαβ:CD3:zeta complex to diversify and amplify TCR signaling. Patients with ZAP70 mutations can present with phenotypes of immune dysregulation as well as infection. We identified the first Taiwanese boy with the [Asp521Asn] ZAP70 mutation who presented with recurrent pneumonia, inflammatory bowel disease-like diarrhea, transient hematuria and autoimmune hepatitis. He had isolated CD8 lymphopenia, eosinophilia, hypogammaglobulinemia, and impaired lymphocyte proliferation. Downstream CD3/CD28 signaling, phosphorylatio

T-cell-engaging antibodies for the treatment of solid tumors: challenges and opportunities.

Purpose of reviewT-cell-engaging antibodies or T-cell engagers (TCEs) can connect a patient's cytotoxic T cells with cancer cells, leading to potent redirected lysis. Until very recently, only one TCE was approved, the CD19/CD3-bispecific blinatumomab. Many new TCEs in late-stage clinical development target various hematopoietic lineage markers like CD20, BCMA, or CD123. Although very compelling single-agent activity of TCEs was observed with various blood-borne cancers, therapy of solid tumor indications has thus far been less successful.Recent findingsThe approval in 2022 of the gp100 peptid

Immune-effector and infection-associated reporting patterns of CD20×CD3 bispecific antibodies in B-cell lymphoma: a label-anchored FAERS analysis with descriptive JADER contextualization.

BACKGROUND: CD20×CD3 bispecific antibodies are associated with acute immune-effector toxicities and infections, but drug-specific postmarketing co-reporting and report-level onset patterns remain incompletely characterized in B-cell lymphoma. METHODS: We performed a label-anchored FAERS pharmacovigilance analysis with descriptive JADER contextualization. Deduplicated FAERS reports from 2022 Q4 through 2026 Q1 were restricted to primary-suspect epcoritamab, glofitamab, or mosunetuzumab reports in B-cell lymphoma, yielding 2,879 exposed reports. Discovery of co-stimulatory anti-CD28 VHHs for developing cancer immune therapeutic anti-tumor/CD3/CD28 trispecific T cell engager.

INTRODUCTION: CD28 is a critical co-stimulatory receptor expressed on T cells. Alongside the T-cell receptor (TCR), it provides a dual-signal requirement that is necessary for the complete activation of T cells. Developing co-stimulatory agonists targeting CD28 therefore represents a promising strategy to enhance the efficacy of T cell based anti-tumor immunotherapy. RESULTS: In this study, we isolated 34 VHHs (VHHs) from a phage-display immune library generated using PBMCs of a Bactrian camel immunized with recombinant human CD28 protein. These VHHs bound to cell surface CD28 with EC50 value

Dual bispecific protein engager-armed T cells targeting BCMA and B7-H3 overcome antigen heterogeneity in multiple myeloma.

Multiple myeloma (MM) remains an incurable plasma cell malignancy, with most patients ultimately progressing to relapsed or refractory MM (RRMM) despite therapeutic advances. Antigen heterogeneity and immune escape, particularly the loss of B-cell maturation antigen (BCMA), pose major challenges to effective immunotherapy. Notably, BCMA loss driven by alterations in TNFRSF17 occurs in a substantial subset of RRMM, thereby limiting the durability of BCMA-targeted therapies. In parallel, B7-homolog H3 (B7-H3), frequently overexpressed in MM, has been implicated in tumor progression, immune evasi

Design and preclinical characterization of an affinity-tuned BCMA×CD3 bispecific T-cell engager for broad B-cell depletion.

B cell maturation antigen (BCMA) is a validated target for plasma cell depletion; however, T cell-redirecting approaches must balance potent cytotoxicity with controlled T cell activation. Here, we describe the design and preclinical characterization of gamgertamig, an affinity-tuned BCMA×CD3 bispecific T-cell engager engineered to promote target-dependent activity. Gamgertamig is a humanized IgG4-based 1 + 1 bispecific antibody incorporating Fc-silencing mutations and a reduced-affinity CD3-binding arm, resulting in a >50-fold affinity bias toward BCMA relative to CD3. Gamgertamig exhibited h

Clinical features and management of GPRC5D-associated adverse events in novel GPRC5D × CD3 bispecific talquetamab-treated relapsed/refractory multiple myeloma patients: the MonumenTAL-1 China cohort experience.

OBJECTIVES: Talquetamab (G-protein-coupled receptor class C group 5 member D [GPRC5D] × CD3 bispecific antibody) demonstrated antitumor activity in relapsed/recurrent multiple myeloma (RRMM) in the phase I/II MonumenTAL-1 study. We report the safety profile of talquetamab in Chinese patients from MonumenTAL-1, focusing on GPRC5D-associated on-target/off-tumor adverse events (AEs). METHODS: Adult Chinese patients with heavily pretreated RRMM and measurable disease received subcutaneous talquetamab 0.4 mg/kg once weekly (QW) or 0.8 mg/kg biweekly (Q2W). Leveraging in vitro Tumor Cell Killing and Cytokine Release to Predict Cytokine Release Syndrome Associated with CD3 T-cell Bispecifics in Oncology: a Retrospective Analysis.

While T-cell Bispecifics (TCBs) are promising molecules for cancer treatment, their clinical development remains challenging due to Cytokine Release Syndrome (CRS). There is currently no method to accurately predict the doses expected to trigger CRS from pre-clinical data, resulting in the selection of non optimal First-In-Human (FIH) doses, far from the doses expected to show clinical benefit, to address safety concerns. In this work, a retrospective analysis aiming to identify which in vitro assay (focusing on tumor cell killing and cytokine release) is predictive of CRS at the first adminis

Extracellular vesicle surface display of αPD-L1 and αCD3 antibodies via engineered late domain-based scaffold to activate T-cell anti-tumor immunity.

Extracellular vesicles (EVs) are emerging as promising carriers for the delivery of therapeutic biologics. Genetic engineering represents a robust strategy for loading proteins of interest into EVs. Identification of EV-enriched proteins facilitates protein cargo loading efficiency. Many EV-enriched proteins are sorted into EVs via an endosomal sorting complex required for transport (ESCRT)-dependent pathway. In parallel, viruses hijack this EV biosynthesis machinery via conserved late domain motifs to promote egress from host cells. Inspired by the similarity of biogenesis between EVs and vir

CD3 and CD20 co-expression in a case of canine peripheral T-cell lymphoma with prominent cardiac and peripheral nerve involvement.

An 8-y 9-mo-old male Pug dog was presented because of anorexia, hindlimb ataxia, vomiting, and progressive weight loss. Clinical examinations revealed atrophic hindlimb muscles with decreased postural reaction, enlargement of the cardiac silhouette and megaesophagus on radiograph, and reduced cardiac contractility on ultrasonography. The dog died 10 d after the initial examination, and an autopsy was performed. Grossly, the heart was enlarged, with multifocal-to-coalescing extensive plaque-like areas of discoloration on the epicardial surface. On cross-section, the ventricles were moderately d

Terminal deoxynucleotidyl transferase (TdT)-negative T-cell lymphoblastic lymphoma with loss of the T-cell lineage-specific marker CD3 at relapse: a rare entity with an aggressive outcome.

Terminal deoxynucleotidyl transferase (TdT)-negative T-cell lymphoblastic lymphoma is a variant of T-cell lymphoblastic lymphoma/T-cell lymphoblastic leukaemia. TdT is a marker of immaturity expressed in 90%-95% cases of lymphoblastic lymphoma and useful in differentiating it from other mature lymphomas/leukaemias. It has been associated with poorer response to chemotherapy and a more aggressive outcome. Here we present a case of TdT-negative T-cell lymphoblastic lymphoma in a 28-year-old man who presented with superior vena cava syndrome. The patient was treated with hyper-cyclophosphamide,vi
