Mercury, lead, and cadmium in umbilical cord blood.

The study described in this article aimed to determine if measurable levels of mercury, lead, and cadmium are detected in the umbilical cord blood specimens collected in a community hospital in Rhode Island and if prenatal exposure correlates with prematurity or fetal growth indicators. Total mercury, lead, and cadmium concentrations were measured in 538 specimens of cord blood and correlated with demographic characteristics and pregnancy outcomes for each mother-infant pair. Lead concentrations determined in the cord blood of Rhode Island women (geometric mean 0.99 microg/dL) were similar to …

Preparation and characterization of artificial antigens for cadmium and lead.

Cadmium and lead were conjugated to two carrier proteins using a bifunctional chelator [2-(4-aminobenzyl)-diethylenetriaminepentaacetic acid] to synthesize artificial antigens for cadmium and lead. The techniques, including ultraviolet spectrometry, circular dichroism, sodium dodecyl sulfate polyacrylamide gel electrophoresis, and inductively coupled plasma optical emission spectroscopy, were utilized for characterizing the artificial antigens. The results of ultraviolet spectrometry showed characteristic absorption peak shifts between conjugates and carrier proteins. Circular dichroism result…

Cadmium is a catalytic inhibitor of DNA topoisomerase II.

Cadmium (Cd(2+)) is a highly toxic and carcinogenic metal that is an environmental and occupational hazard. DNA topoisomerase II is an essential nuclear enzyme and its inhibition can result in the formation of genotoxic and recombinogenic DNA double strand breaks. In this study we showed that cadmium chloride strongly inhibited the DNA decatenation activity of human topoisomerase IIα in the low micromolar concentration range and that its inhibitory effects were reduced by glutathione. Because the activity of topoisomerase II is strongly inhibited by thiol-reactive compounds this result suggest…

Adverse effects of cadmium exposure on mouse sperm.

The effects of cadmium chloride exposure on sperm functional parameters were evaluated on eight-week-old ICR-CD1 male mice administered with a single s.c. injection of 1, 2 and 3 mg CdCl(2)/kg bw. Groups of animals treated with each dose, as well as their respective controls, were sacrificed after 24h to detect short-term (acute) effects and after 35 days. Sperm cells were collected from the epididymis and several parameters of sperm quality and function were evaluated, namely density, morphology, motility, viability, mitochondrial function, acrosome integrity, together with DNA fragmentation …

Effect of cadmium chloride on metallothionein levels in carp.

Due to anthropogenic activities, heavy metals still represent a threat for various trophic levels. If aquatic animals are exposed to heavy metals, we can obviously observe considerable toxicity. It is well known that organisms treated with heavy metals synthesize low molecular mass compounds rich in cysteine. In this work the effects of cadmium chloride (2.5, 5, 7.5, 10 and 12.5 mg/L) on common carp (Cyprinus carpio) was investigated. We determined cadmium content in tissue of muscle, liver and kidney by atomic absorption spectrometry with electrothermal atomization and content of metallothion…

Cadmium(II) complex formation with cysteine and penicillamine.

The complex formation between cadmium(II) and the ligands cysteine (H(2)Cys) and penicillamine (H(2)Pen = 3,3'-dimethylcysteine) in aqueous solutions, having C(Cd(II)) approximately 0.1 mol dm(-3) and C(H(2)L) = 0.2-2 mol dm(-3), was studied at pH = 7.5 and 11.0 by means of (113)Cd NMR and Cd K- and L(3)-edge X-ray absorption spectroscopy. For all cadmium(II)-cysteine molar ratios, the mean Cd-S and Cd-(N/O) bond distances were found in the ranges 2.52-2.54 and 2.27-2.35 A, respectively. The corresponding cadmium(II)-penicillamine complexes showed slightly shorter Cd-S bonds, 2.50-2.53 A, but …

Current status of cadmium as an environmental health problem.

Cadmium is a toxic metal occurring in the environment naturally and as a pollutant emanating from industrial and agricultural sources. Food is the main source of cadmium intake in the non-smoking population. The bioavailability, retention and toxicity are affected by several factors including nutritional status such as low iron status. Cadmium is efficiently retained in the kidney (half-time 10-30 years) and the concentration is proportional to that in urine (U-Cd). Cadmium is nephrotoxic, initially causing kidney tubular damage. Cadmium can also cause bone damage, either via a direct effect o…

Cadmium toxicity in glutathione mutants of Escherichia coli.

The higher affinity of Cd(2+) for sulfur compounds than for nitrogen and oxygen led to the theoretical consideration that cadmium toxicity should result mainly from the binding of Cd(2+) to sulfide, thiol groups, and sulfur-rich complex compounds rather than from Cd(2+) replacement of transition-metal cations from nitrogen- or oxygen-rich biological compounds. This hypothesis was tested by using Escherichia coli for a global transcriptome analysis of cells synthesizing glutathione (GSH; wild type), gamma-glutamylcysteine (DeltagshB mutant), or neither of the two cellular thiols (DeltagshA muta…

A structural study of cadmium interaction with aquatic microorganisms.

The molecular mechanisms of cadmium toxicity for aquatic phototrophic microorganisms, reversible adsorption on the surface, and cellular uptake during growth were investigated by combining batch macroscopic experiments with atomic-level in situ Cd K-edge X-ray absorption fine structure spectroscopy. The following species were examined: marine planktonic (Skeletonema costatum, Thalassiossira weissflogil) and freshwater periphytic (Navicula minima, Achnanthidium minutissum) diatoms, cyanobacteria (Gloeocapsa sp.), anoxygenic phototrophic bacteria (Rhodopseudomonas palustris), and freshwater diat…

Cadmium: toxic effects on the reproductive system and the embryo.

The heavy metal cadmium (Cd) is a pollutant associated with several modern industrial processes. Cd is absorbed in significant quantities from cigarette smoke, and is known to have numerous undesirable effects on health in both experimental animals and humans, targeting the kidneys, liver and vascular systems in particular. However, a wide spectrum of deleterious effects on the reproductive tissues and the developing embryo has also been described. In the testis, changes due to disruption of the blood-testis barrier and oxidative stress have been noted, with onset of widespread necrosis at hig…

Removal of cadmium from aqueous solutions by palygorskite.

The sorption characteristics of palygorskite with respect to cadmium were studied with the aim of assessing its use in water purification systems. Using a batch method the influence of time (0.5-48 h), initial Cd concentration (5-150 mg/l or 0.044-1.34 mmol/l), ionic strength ([Ca(II)]: 0-0.1 mol/l), pH (3-7) and mineral dose (1-20 g/l) on Cd removal was evaluated. The sorption of Cd on palygorskite appeared as a fast process, with equilibrium being attained within the first half an hour of interaction. This process could be described by the Langmuir model and gave a maximum Cd sorption of 4.5…

Omphalocele induction in the chick embryo by administration of cadmium.

BACKGROUND: Ventral body wall (VBW) defects occur in 1:2000 live births. We examined the association of VBW defect with somite abnormality and lordosis in the chick using in vitro and in ovo methods. METHODS: Explanted chick embryos were treated at 60 hours with 50 microL sodium acetate or 0.001% cadmium acetate solution to produce VBW defects. Mortality and abnormality rates were assessed. A further cohort of chicks was treated in ovo by dropping 50 microL 0.001% to 0.01% cadmium acetate onto the embryo and allowing development to 16.5 days for further assessment of the defect and skeletal s…

Nephrotoxicity and the proximal tubule. Insights from cadmium.

Cadmium (Cd(2+)) is a non-essential heavy metal, which is taken up from the environment into the body through pulmonary and enteral pathways. The S1 segment of the kidney proximal tubule (PT) is a major target of chronic Cd(2+) toxicity. Renal dysfunction develops in up to 7% of the general population and in its most severe form displays major features of Fanconi syndrome, such as a defective protein, amino acid, glucose, bicarbonate and phosphate reabsorption. The major pathway for Cd(2+) uptake by PT cells (PTCs) in vivo is apical endocytosis of Cd(2+) complexed to the high-affinity metal-bi…

The initial pathogenesis of cadmium induced renal toxicity.

The novel application of magic angle spinning 1H NMR spectroscopy, coupled with pattern recognition techniques, has identified biochemical changes in lipid and glutamate metabolism that precede classical nephrotoxicity. These changes occurred in the bank vole (Clethrionomys glareolus) after chronic dosing, at a low level of exposure and at a renal Cd(2+) concentration (8.4 microgram/g dry wt) that was nearly two orders of magnitude below the WHO critical organ concentration (200 microg/g wet wt). These early stage effects of Cd(2+) on the biochemistry of renal tissue may reflect adaptation mec

Nephrotoxicity of repeated injections of cadmium-metallothionein in rats.

Cadmium-metallothionein (Cd-MT) may have a role in the pathogenesis and irreversibility of Cd nephrotoxicity. In the present study, rats were injected with 0.3 mg Cd/kg body wt per week as Cd-MT for 5 consecutive weeks and a group of rats (n = 3) was killed 24 hr after each injection. A group of three rats was kept for an additional week after the 5 weeks of Cd-MT injection for recovery. After the first injection, urinary Cd and protein levels and kidney/body wt ratio were increased. The electrophoretic pattern of urinary protein showed increased excretion of low-molecular-weight proteins, esp…

Adverse health effects in workers exposed to cadmium.

One hundred fifty-two workers who had been exposed to different levels of cadmium (Cd) absorption (blood Cd mean 16.34 micrograms/L, median 11.0 micrograms/L) in a factory making nickel-cadmium batteries were studied. A physical evaluation, as well as blood and urine cadmium and N-acetyl-beta-D-glucosaminidase (NAG), and other routine laboratory tests were performed in order to assess the relationship between different levels of exposure to occupational cadmium and potential cadmium-induced renal damage and other adverse health effects. We found significant correlation between levels of cadmiu…
