Podocyte Shape Regulation by Semaphorin 3A and MICAL-1.

Podocytes are complex epithelial cells with foot processes that are essential for the integrity and function of the kidney glomerular filters. Podocyte foot processes linked by slit diaphragms constitute signaling platforms that tightly regulate the cell shape and the function of the filtration barrier. Semaphorin (Sema) 3A is a class 3 semaphorin secreted by podocytes that has autocrine and paracrine functions in the kidney. We have shown that Sema3A regulates podocyte shape and that excess Sema3A signaling induces glomerular disease and aggravates diabetic nephropathy. MICAL-1 is an actin-bi

Characterization of the cystic cavities of canine invasive micropapillary mammary carcinoma reveals a complex tumor microenvironment.

Invasive micropapillary carcinoma (IMC) of the mammary gland is an aggressive mammary tumor affecting humans, dogs, and other species. This study characterized the cystic cavities and tumor microenvironment in in situ, transitional, and invasive areas of canine IMC, comparing findings to human IMC. Methods included histopathology; histochemistry (PAS, Alcian Blue, Oil Red) for basement membrane and secretion assessment; immunohistochemistry and immunofluorescence for myoepithelial markers (p63, calponin-1, smooth muscle myosin, smooth muscle alpha-actin, cytokeratin 14, vimentin) and other pro

Porcine submandibular glands as potential salivary gland experimental models: histological, immunohistochemical, and ultrastructural characterization.

Salivary glands play an essential role in oral homeostasis by producing saliva, which protects oral tissues and maintains the oral environment. Despite growing interest in porcine models for translational biomedical research, the immunophenotypic characterization of porcine salivary glands remains limited in the literature, with few studies addressing their cytokeratin and contractile protein expression profiles. This gap constrains the ability to directly compare porcine and human glandular phenotypes and hinders the establishment of the pig as a validated salivary gland experimental model. T

A Case of Infantile Epileptic Spasms Syndrome with the SPTBN1 Mutation and Review of βII-Spectrin Variants.

Background: Spectrin proteins are critical cytoskeleton components that maintain cellular structure and mediate intracellular transport. Pathogenic variants in SPTBN1, encoding βII-spectrin, have been associated with various neurodevelopmental disorders, including developmental delay, intellectual disability, autism spectrum disorder, and epilepsy. Here we report a Korean infant with infantile epileptic spasms syndrome (IESS) and an SPTBN1 mutation and provide a review of this mutation. Methods: The genomic data of the patient were analyzed by whole exome sequencing. A comprehensive literature

Recent Treatment Options for Primary Colonic Leiomyosarcoma: A Case Report and a Review of the Literature.

INTRODUCTION: Primary colonic leiomyosarcoma (LMS) accounts for only 0.12% of colonic malignancies. However, it is highly aggressive and associated with a poor prognosis. While radical surgical resection is considered the mainstay of treatment, the optimal surgical approach and the role of adjuvant chemotherapy remain unclear due to the small number of reported cases. CASE PRESENTATION: A 45-year-old, male patient presented with hematochezia. Colonoscopy revealed a 40-mm, type 1 tumor in the sigmoid colon. Immunohistochemical analysis of a biopsy specimen found atypical spindle cell prolifera

CRTC1::MAML2-Positive Hidradenoma With Sebaceous Differentiation.

Hidradenoma can exhibit several cell types, including clear cells, polyhedral eosinophilic cells, squamoid cells, mucinous cells, oxyphilic (oncocytic) cells, and transitional (intermediate) cells. However, sebocytes have not yet been described in hidradenoma. Here, we present a case of CRTC1::MAML2-positive hidradenoma with sebaceous differentiation. In addition, this tumor had two cell-type layers that were morphologically similar to apocrine glandular cells and myoepithelial cells throughout the lesion. A 66-year-old male presented with a 20 mm nodule on his upper lip with a 3-year history.

Histone deacetylase inhibition enhances extracellular vesicles from muscle to promote osteogenesis via miR-873-3p.

Regular physical activity is widely recognized for reducing the risk of various disorders, with skeletal muscles playing a key role by releasing biomolecules that benefit multiple organs and tissues. However, many individuals, particularly the elderly and those with clinical conditions, are unable to engage in physical exercise, necessitating alternative strategies to stimulate muscle cells to secrete beneficial biomolecules. Histone acetylation and deacetylation significantly influence exercise-induced gene expression, suggesting that targeting histone deacetylases (HDACs) could mimic some ex

Low-grade adenosquamous carcinoma of the breast: A rare case presentation.

Low-grade adenosquamous carcinoma of the breast is a rare variant of metaplastic mammary carcinoma. It shows indolent behavior contrary to the usual aggressive nature of metaplastic carcinomas and has a good prognosis despite being triple negative. Recurrence rates tend to be high and a consequence of incomplete excision. Although this variant has an infiltrative growth pattern, owing to its bland cytologic features, it is liable to be confused with benign sclerosing adenotic breast lesions. We present here a case of a 55-year-old postmenopausal female, who presented with a painless, mobile, h

Myoepithelioma-like tumors of the vulvar region: A case report and review of the literature.

Myoepithelioma-like tumors of the vulvar region (MELTVR) is a kind of solid tumor newly recognized in recent years, which is characterized by mesenchymal tumors of adult female vulva. The histopathology is similar to myoepithelioma, but the immunohistochemical phenotype and genetic changes are different from myoepithelioma. It usually has clear boundary and partial capsule, mixed with two forms of cells (epithelioid and spindle), the cells are mild, the nucleoli are clear, mitoses are rare, some cases have myxoid differentiation. In this article, a case of MELTVR diagnosed in our hospital is d

The Role of Gap Junctions in the Generation of Smooth Muscle Cells from Bone Marrow Mesenchymal Stem Cells.

BACKGROUND: Studies have shown that stem cell transplantation can improve smooth muscle cell (SMC) regeneration and remodelling. Gap junctions can enhance the cytoprotective effects of neighbouring cells. We investigated the effect of gap junctions on the differentiation of bone marrow mesenchymal stem cells (BMSCs) into SMCs. MATERIALS AND METHODS: Rat BMSCs and SMCs were obtained from the bone marrow and bladder of Sprague-Dawley rats, respectively. Flow cytometry and multilineage differentiation were performed to assess the characteristics of these cells. BMSCs and SMCs were incubated toge

Recurrent borderline phyllodes tumor of the breast submitted to mastectomy and immediate reconstruction: Case report.

INTRODUCTION: Phyllodes tumors are biphasic fibroepithelial tumors that account for fewer than 1% of mammary tumors. They usually appear in middle-aged women, with an average size of 4-7 cm and rapid growth. PRESENTATION OF CASE: A 25-year-old woman sought care after excision of recurrent nodules in the right breast, with a diagnosis of borderline phyllodes tumor. She had no new lesions on physical examination. Imaging studies revealed an anechoic collection and nodular areas in a previous surgical site, correlated with pathology. Immuno-histochemical examination was positive for vimentin, ca

Human Papillomavirus-Related Multiphenotypic Sinonasal Carcinoma with Unique HPV type 52 Association: A Case Report with Review of Literature.

Human papillomavirus (HPV)-related multiphenotypic sinonasal carcinoma (HMSC) is a recently described distinctive clinicopathologic entity defined by association to high risk HPV, localization to sinonasal tract and close histologic resemblance to salivary gland tumors. Lack of awareness of its pathologic features and biology among pathologists and oncologists make this entity susceptible to misdiagnosis and erroneous management. Herein, we illustrate a case of HMSC of the nasal cavity associated with heretofore unreported subtype HPV-52 and discuss the challenges associated with diagnosis and

Low-grade cribriform cystadenocarcinoma of the palatal gland: A case report.

Low-grade cribriform cystadenocarcinoma (LGCCC) is a malignant salivary gland tumor. LGCCC occurs rarely, with the parotid gland being the most commonly afflicted site. Few cases arise in other sites. The present study reports a case of LGCCC that occurred at the palatal gland of the hard palate. A 56-year-old female was referred to Saitama Medical University Hospital (Moroyama, Saitama, Japan) due to an intraoral mass. Since cytological examination and biopsy led to a diagnosis favoring a neoplasm, but with uncertain malignant potential, the tumor was resected with a safe surgical margin. The

Primary male neuroendocrine adenocarcinoma involving the nipple simulating Merkel cell carcinoma - a diagnostic pitfall.

Male breast cancer is a rare entity accounting for < 1% of all breast cancer cases in the United States, but with a rate that has been rising over the last 25 years. Nipple skin/subcutaneous tumors in men are even rarer. Likewise, true neuroendocrine carcinoma of the breast, defined as > 50% of tumor cells staining for either chromogranin or synaptophysin, is not a common entity, usually occurring in older women. We present the case of a 70-year-old man with a slowly growing nipple mass that had enlarged over the previous 1.5 years. The histology consisted of nests, trabeculae and sheets of ba
