Dexmedetomidine Inhibition of GABA Neurons in the Ventral Tegmental Area Attenuates the Depth of Sedation in Mice.

INTRODUCTION: Dexmedetomidine is a sedative anesthetic with a unique ability to produce an arousable state of sedation. However, the neural mechanisms underlying this arousable sedation remain incompletely understood. Previous studies have implicated dopaminergic neurons in the ventral tegmental area (VTA) in mediating this effect, but the upstream neural circuit mechanisms remain unclear. Furthermore, although VTA GABAergic neurons exert robust inhibitory effects on dopaminergic neurons, their specific role in dexmedetomidine-induced arousable sedation remains unclear. METHODS: Vgat-Cre tran

Electroacupuncture Alleviates Pain and Anxiety in CFA-Induced Mice by Suppressing Glutamatergic Neurons in the Spinoparabrachial Pathway.

The spinoparabrachial tract is a major ascending pathway that transmits nociceptive signals from the spinal cord to the brain, but whether electroacupuncture (EA) relieves pain-anxiety comorbidity by modulating this pathway remains unclear. Here, we investigated the effects of EA on mechanical allodynia and anxiety-like behaviors in Complete Freund's adjuvant (CFA)-induced inflammatory pain mice using von Frey testing, the open field test, and the elevated plus maze, combined with chemogenetic manipulation of glutamatergic projections from the spinal dorsal horn (SDH) to the lateral parabrachi

Excitatory paraventricular thalamus-posterior insular cortex circuit mediates pain-anxiety comorbidity in diabetic neuropathy.

Painful diabetic neuropathy (PDN) is frequently accompanied by anxiety, yet the neural circuit mechanisms associated with nociceptive hypersensitivity to affective dysfunction remain unclear. Here we combined brain-wide c-Fos mapping, viral tracing, in vivo fiber photometry, whole-cell recordings, and projection-specific opto/chemogenetics in a mouse model of streptozotocin-induced PDN to define a thalamo-cortex circuit for pain-anxiety comorbidity. Within the posterior insular cortex (pIC), CaMKIIα⁺ excitatory neurons were recruited in PDN mice with comorbid anxiety, exhibiting heightened neu…

Medial Preoptic Nucleus to Periaqueductal Gray GABAergic Pathway Underlies Individual Differences in Consolation-Like Behavior in Mice.

INTRODUCTION: Consolation is a complex empathic behavior that varies considerably across individuals, yet the neural basis of this heterogeneity remains poorly understood. The medial preoptic area is a key hub for instinctive social behaviors, but it remains unclear which of its subregions - the medial preoptic nucleus (MPN) versus the surrounding area (MPA) - or which neuronal subtypes (GABAergic, glutamatergic, or galanin-expressing) contribute to individual differences in consolation. METHODS: We used a social defeat-induced consolation paradigm in male mice and categorized them into low-

A dorsolateral BNST-parabrachial nucleus pathway regulates adolescent alcohol-induced negative affect in females.

Adolescent alcohol use is a major predictor of alcohol use disorder in adulthood and coincides with a critical developmental window for the emergence of affective disorders. These comorbidities may heighten relapse vulnerability, particularly in females. Negative reinforcement mechanisms, in which alcohol alleviates negative emotional states, are thought to drive continued drinking. We previously demonstrated that chemogenetic stimulation of the dorsolateral bed nucleus of the stria terminalis (dlBNST) induces negative affect-like behavior in adult female mice with a history of adolescent inte

Parabrachial neuropeptide Y Y1 receptor-expressing neurons govern the sensory dimension of neuropathic pain.

Pain perception involves complex sensory and emotional processes, yet the supraspinal circuits that sustain neuropathic pain remain incompletely understood. Here, we demonstrate that parabrachial nucleus (PBN) neurons expressing the inhibitory G-protein-coupled neuropeptide Y Y1 receptor (Npy1r) are critical modulators of neuropathic pain. Approximately 19% of lateral PBN neurons express Npy1r. Stimulus-evoked Fos activation and calcium dynamics of these neurons increases in mice with spared nerve injury (SNI) compared to sham controls. Pharmacological activation of Y1 receptors or chemogeneti

Parvalbumin neurons in inferior colliculus mediate maladaptive inhibitory compensation in age-related hearing loss.

BACKGROUND: Age-related hearing loss (ARHL) may be caused by the abnormal excitability of inferior colliculus (IC) neurons, and this mechanism may be related to the inhibitory compensation of parvalbumin (PV) neurons; however, its specific mechanism remains unclear. This study focused on PV neurons and investigated the inhibitory compensatory mechanism in the IC during ARHL. METHODS: We investigated hearing changes in C57BL/6J mice from young to middle-aged using auditory brainstem response and acoustic startle reflex tests, followed by analysis of cluster activity in excitatory and inhibitor

GPR30 in spinal cholecystokinin-positive neurons modulates neuropathic pain.

Neuropathic pain, a major health problem affecting 7-10% of the global population, lacks effective treatment due to its elusive mechanisms. Cholecystokinin-positive (CCK+) neurons in the spinal dorsal horn (SDH) are critical for neuropathic pain, yet the underlying molecular mechanisms remain unclear. Here, we show that the membrane estrogen receptor G-protein coupled estrogen receptor (GPER/GPR30) in spinal neurons was significantly upregulated in chronic constriction injury (CCI) mice and that inhibition of GPR30 in CCK+ neurons reversed CCI-induced neuropathic pain. Furthermore, GPR30 in sp

The Crucial Role of Cerebellar Crus I in Electroacupuncture Pretreatment for Attenuating Myocardial Ischemia-Reperfusion Injury.

BACKGROUND: Electroacupuncture pretreatment (EA-pre) has been shown to help reduce myocardial ischemia-reperfusion injury (MIRI), but the underlying mechanism remains unclear. Our previous studies indicated that EA activates the cerebellar cortex, specifically the Crus Ⅰ. However, whether activation of the Crus Ⅰ contributes to the attenuation of MIRI induced by EA-pre remains unclear. This study investigated the possible relationship between EA-induced relief of MIRI and the activation of Crus Ⅰ. METHODS: Electrocardiogram recording, echocardiography, and cardiac histology staining were used

Hippocampal ensembles regulate circuit-induced relapse of extinguished fear.

Extinction learning is central to behavioral therapies for post-traumatic stress disorder (PTSD), but relapse poses a major challenge to this approach. Recent work has revealed a critical role for the thalamic nucleus reuniens (RE) in the suppression of extinguished fear memories. Silencing the RE yields a relapse of extinguished fear (i.e., "circuit-induced relapse"). Considerable work suggests that RE may contribute to extinction by inhibiting the retrieval of hippocampal (HPC)-dependent fear memories. To test this hypothesis, we first examined whether undermining the formation of contextual

Oxytocin Receptor Expression and Activation in Parasympathetic Brainstem Cardiac Vagal Neurons.

Autonomic imbalance, particularly reduced activity from brainstem parasympathetic cardiac vagal neurons (CVNs) is a major characteristic of many cardiorespiratory diseases. Therapeutic approaches to selectively increase CVN activity have been limited by lack of identified selective translational targets. Recent work has shown that there is an important excitatory synaptic pathway from oxytocin (OXT) neurons in the paraventricular nucleus of the hypothalamus (PVN) to brainstem CVNs, and that OXT could provide a key selective excitation of CVNs. In clinical studies, intranasal OXT increases para

Intranasal LAG3 antibody infusion induces microglia-dependent antidepressant effect by mobilizing astrocytic P2Y1R-mediated BDNF synthesis in the hippocampus.

Intranasal infusion of lymphocyte-activating gene-3 antibody (In-LAG3-Ab) has microglia-dependent antidepressant effects, but the underlying mechanism remains unclear. Since microglia can interact with astrocytes through purinergic signaling, we hypothesize that microglia-driven purinergic signaling may mediate the antidepressant effect of In-LAG3-Ab. The results showed that a single In-LAG3-Ab infusion in chronically stressed mice produced both an antidepressant effect and increased adenosine triphosphate (ATP) levels in the dentate gyrus, both of which were suppressed by chemogenetic inhibit

Inhibition of ventral tegmental area projections to the paraventricular thalamus attenuates conditioned cues-induced heroin seeking behavior in rats.

Ventral tegmental area (VTA) is typically involved in heroin reward, but it is still unclear whether the VTA or neural projections containing VTA are also involved in conditioned cues (CS)-induced heroin-seeking behavior. In present study, rats were first trained to self-administer heroin for 14 days, then undergone withdrawal for another 14 days. Subsequently, part of rats were assessed CS-induced heroin-seeking behaviors and tested c-Fos protein expression in brain areas after microinjection with saline, neostigmine or tetrodotoxin (TTX) into the VTA. Part of rats were tested CS-induced hero

Nociceptor Neurons Facilitate Orthodontic Tooth Movement via Piezo2 in Mice.

Multiple sensory afferents, including mechanosensitive and nociceptive nerves, are projected to the periodontium. Peptidergic afferents expressing transient receptor potential vanilloid 1 (TRPV1), a receptor for capsaicin, mediate pain caused by orthodontic forces. However, their role in orthodontic force-induced alveolar bone remodeling is poorly understood as is the contribution of mechanosensitive ion channels such as Piezo2 in nociceptive nerves. To investigate this role, we studied orthodontic tooth movement and alveolar bone remodeling using neural manipulations and genetic mouse models.

Opposing actions of co-released GABA and neurotensin on the activity of preoptic neurons and on body temperature.

Neurotensin (Nts) is a neuropeptide acting as a neuromodulator in the brain. Pharmacological studies have identified Nts as a potent hypothermic agent. The medial preoptic area, a region that plays an important role in the control of thermoregulation, contains a high density of neurotensinergic neurons and Nts receptors. The conditions in which neurotensinergic neurons play a role in thermoregulation are not known. In this study optogenetic stimulation of preoptic Nts neurons induced a small hyperthermia. In vitro, optogenetic stimulation of preoptic Nts neurons resulted in synaptic release of

Inhibition of dorsal raphe GABAergic neurons blocks hyperalgesia during heroin withdrawal.

Opioid withdrawal signs, such as hyperalgesia, are manifestations of opioid use disorder that may contribute to opioid seeking and taking. We have previously identified an association between dorsal raphe (DR) neurons and the expression of hyperalgesia during spontaneous heroin withdrawal. Here, we found that chemogenetic inhibition of DR neurons decreased hyperalgesia during spontaneous heroin withdrawal in male and female C57/B6 mice. By neuroanatomy, we identified three major subtypes of DR neurons expressing μ-opioid receptors (MOR) that were activated in hyperalgesia during spontaneous wi

Epac2 in midbrain dopamine neurons contributes to cocaine reinforcement via enhancement of dopamine release.

Repeated exposure to drugs of abuse results in an upregulation of cAMP signaling in the mesolimbic dopamine system, a molecular adaptation thought to be critically involved in the development of drug dependence. Exchange protein directly activated by cAMP (Epac2) is a major cAMP effector abundantly expressed in the brain. However, it remains unknown whether Epac2 contributes to cocaine reinforcement. Here, we report that Epac2 in the mesolimbic dopamine system promotes cocaine reinforcement via enhancement of dopamine release. Conditional knockout of Epac2 from midbrain dopamine neurons (Epac2

Rescue of obesity-induced infertility in female mice by silencing AgRP neurons.

Obesity impacts multiple sites of the hypothalamus-pituitary gland-ovary axis (HPO axis) and has become a leading cause of female infertility. However, the critical hypothalamic neurons that participate in the development of obesity-induced infertility have not been well defined yet. Previous studies suggested that metabolic-sensing agouti-related peptide-expressing (AgRP) neurons in the arcuate nucleus (ARC) are hyperactive in diet-induced obesity (DIO) mice. We hypothesize that these neurons may convey metabolic dysfunction onto the HPO axis and contribute to obesity-induced infertility's pa

Activation of ventrolateral orbital cortex improves mouse neuropathic pain-induced anxiodepression.

Depression and anxiety are frequently observed in patients suffering from neuropathic pain. The underlying mechanisms remained unclear. The ventrolateral orbital cortex (VLO) has attracted considerable interest in its role in antidepressive effect in rodents. In the present study, we further investigated the role of the VLO in the anxiodepressive consequences of neuropathic pain in a chronic constriction injury of infraorbital nerve-induced trigeminal neuralgia (TN) mouse model. Elevated plus maze, open field, forced swimming, tail suspension, and sucrose preference tests were used to evaluate

Basolateral amygdala is required for reconsolidation updating of heroin-associated memory after prolonged withdrawal.

Postretrieval extinction procedures are effective nonpharmacological interventions for disrupting drug-associated memories. Nonetheless, the conditioned stimulus (CS) memory retrieval-extinction procedure is ineffective in inhibiting drug craving and relapse after prolonged withdrawal, which significantly undermines its therapeutic potential. In the present study, we showed that, unlike the CS memory retrieval-extinction procedure, noncontingent heroin injections (unconditioned stimulus [UCS]) 1 hour before the extinction sessions decreased the heroin-priming-induced reinstatement, renewal, an
