Axl-inhibitor bemcentinib alleviates mitochondrial dysfunction in the unilateral ureter obstruction murine model.

Renal fibrosis is a progressive histological manifestation leading to chronic kidney disease (CKD) and associated with mitochondrial dysfunction. In previous work, we showed that Bemcentinib, an Axl receptor tyrosine kinase inhibitor, reduced fibrosis development. In this study, to investigate its effects on mitochondrial dysfunction in renal fibrosis, we analysed genome-wide transcriptomics data from a unilateral ureter obstruction (UUO) murine model in the presence or absence of bemcentinib (n = 6 per group) and SHAM-operated (n = 4) mice. Kidney ligation resulted in dysregulation of mitocho

Tricarboxylic acid cycle activity measured by 13C magnetic resonance spectroscopy in rats subjected to the kaolin model of obstructed hydrocephalus.

Evaluating early changes in cerebral metabolism in hydrocephalus can help in the decision making and the timing of surgical intervention. This study was aimed at examining the tricarboxylic acid (TCA) cycle rate and (13)C label incorporation into neurotransmitter amino acids and other compounds 2 weeks after rats were subjected to kaolin-induced progressive hydrocephalus. In vivo and ex vivo magnetic resonance spectroscopy (MRS), combined with the infusion of [1,6-(13)C]glucose, was used to monitor the time courses of (13)C label incorporation into the different carbon positions of glutamate i

Preventive Effect of Butyrate in Colon Cancer Cell Metabolism.

Butyrate, a short-chain fatty acid produced by the fermentation of soluble dietary fiber by gut bacteria, also functions as a histone deacetylase inhibitor known to induce apoptosis and promote differentiation in colon tumor cells. During tumorigenesis, cancer cells undergo metabolic reprogramming to meet energetic and biosynthetic demands, increasing glycolytic metabolism and reducing oxidative metabolism-a phenomenon known as the Warburg effect. This study aimed to evaluate the impact of butyrate on the aggressiveness-related metabolic phenotype of three colon cancer cell lines (LS1034, C2BB

Engineering Escherichia coli for Anaerobic Succinate Fermentation Using Corn Stover Hydrolysate as a Substrate.

Succinic acid is regarded as one of the most important platform chemicals used in materials science, chemistry, and food industrial applications. Currently, the main bottlenecks in the microbial succinate synthesis lie in the low titer, cofactor imbalance, and high production costs. To overcome these challenges, the reductive tricarboxylic acid cycle (TCA) and glucose uptake pathway were enhanced, increasing the titer of succinate to 4.31 g/l, 2.06-fold of the original strain. Furthermore, formate dehydrogenase from Candida boidinii was simultaneously overexpressed to increase the regeneration

Bacterial assay for quantitative measurement of nanomolar concentrations of a metabolite.

A mutant of Pseudomonas fluorescens was used to develop a radioligand, competitive binding assay to quantitatively measure gamma-aminobutyric acid. The highly reliable and reproducible assay was sensitive (nM detection), rapid, and easy to perform. Nonspecific activity and scatter were insignificant. Radiolabel was irreversibly fixed by the cells in an energy-dependent reaction. The finding that a bacterium was effective in quantitatively detecting nanomolar amounts of a metabolite suggests that other bacteria or their mutants might be used in competitive binding assays to detect and quantify

Final report on the safety assessment of Malic Acid and Sodium Malate.

Malic Acid functions in cosmetic formulations as a pH adjuster, and Sodium Malate functions as a skin conditioning agent-humectant. Malic Acid is reportedly used in almost 50 cosmetic formulations across a range of product types at low concentrations, whereas Sodium Malate is used in only one. As a pH adjuster, Malic Acid is used at low concentrations. One commercial method of preparing Malic Acid is hydration of fumaric acid or maleic acid, and then purified to limit the amount of the starting material present. Because Malic Acid is a component of the Kreb's cycle, another method is fermentat

Cholera toxin promotes pathogen acquisition of host-derived nutrients.

Vibrio cholerae is the causative agent of cholera, a potentially lethal enteric bacterial infection1. Cholera toxin (CTX), a protein complex that is secreted by V. cholerae, is required for V. cholerae to cause severe disease. CTX is also thought to promote transmission of the organism, as infected individuals shed many litres of diarrhoeal fluid that typically contains in excess of 1011 organisms per litre. How the pathogen is able to reach such high concentrations in the intestine during infection remains poorly understood. Here we show that CTX increases pathogen growth and induces a distin

Substrate-enzyme competition attenuates upregulated anaplerotic flux through malic enzyme in hypertrophied rat heart and restores triacylglyceride content: attenuating upregulated anaplerosis in hypertrophy.

Recent work identifies the recruitment of alternate routes for carbohydrate oxidation, other than pyruvate dehydrogenase (PDH), in hypertrophied heart. Increased carboxylation of pyruvate via cytosolic malic enzyme (ME), producing malate, enables "anaplerotic" influx of carbon into the citric acid cycle. In addition to inefficient NADH production from pyruvate fueling this anaplerosis, ME also consumes NADPH necessary for lipogenesis. Thus, we tested the balance between PDH and ME fluxes in hypertrophied hearts and examined whether low triacylglyceride (TAG) was linked to ME-catalyzed anaplero

Mycobacterium tuberculosis (Mtb) isocitrate dehydrogenases show strong B cell response and distinguish vaccinated controls from TB patients.

Proteins released from Mycobacterium tuberculosis (Mtb) during late logarithmic growth phase are often considered candidate components of immunogenic or autolysis markers. One such protein is isocitrate dehydrogenase (ICD), a key regulatory enzyme in the citric acid cycle. We have evaluated the immunogenic properties of two isoforms of Mtb ICD and compared them with the control antigens heat-shock protein 60 and purified protein derivative (PPD). PPD lacks the sensitivity to distinguish between bacillus Calmette-Guérin (BCG)-vaccinated and tuberculosis (TB)-infected populations, and, therefore

Metabonomic Strategy for the Evaluation of Chinese Medicine Salvia miltiorrhiza and Dalbergia odorifera Interfering with Myocardial Ischemia/Reperfusion Injury in Rats.

Extract of Salvia miltiorrhiza and Dalbergia Odorifera (SM-DOO) has been traditionally used for the prevention and treatment of cardiovascular diseases. However, information regarding the pharmacodyamic material basis and potential mechanism remain unknown. Male Sprague-Dawley rats were divided into four groups: Sham, Model, Diltiazem, and SM-DOO group, n = 6. Rats were pretreated with homologous drugs for 7 days, and then subjected to 30 minutes of ischemia followed by 180 minutes of reperfusion. Cardioprotection effects of SM-DOO on myocardial ischemia/reperfusion (MI/R) injury rats were exa

Succinate as a metabolic-signaling dual-function hub in skeletal muscle: linking exercise adaptation to myopathic dysfunction.

Skeletal muscle plasticity is shaped by mechanical loading, metabolic flux, and intercellular signaling. Beyond its canonical role as a tricarboxylic acid (TCA) cycle intermediate, succinate is increasingly viewed as a potential extracellular signaling metabolite that may link mitochondrial metabolism to tissue remodeling. However, whether succinate acts as a causal mediator rather than a correlate of metabolic stress remains incompletely defined. In this hypothesis-driven narrative review, we propose the Succinate Dual-Function Hub framework, in which succinate is considered a context-depende

Stable Isotope-Resolved Metabolomics Studies on Corticosteroid-Induced PC12 Cells: A Strategy for Evaluating Glucose Catabolism in an in Vitro Model of Depression.

Depression is a common psychopathological state or mood disorder syndrome. The serious risks to human life and the inadequacy of the existing antidepressant drugs have driven us to understand the pathogenesis of depression from a new perspective. Our research group has found disturbances in glucose catabolism in both depression and nephrotic syndrome. What are the specific metabolic pathways and specificities of glucose catabolism disorders caused by depression? To address the above scientific questions, we creatively combined traditional metabolomics technology with stable isotope-resolved me

Effects of intermediary metabolites and electron transport inhibitors on action of chloroquine on Brugia pahangi and Onchocerca volvulus.

We examined the possibility that chloroquine is interfering with aerobic energy-generating processes in the adult filarial parasites, Brugia pahangi and Onchocerca volvulus. Using motility of these parasites as an assay of drug effect, we found that micromolar concentrations of chloroquine caused significant paralysis, but only in alkaline medium (pH 8.4). The addition of 12 mM glutamine or 10 mM albizziin to the medium completely antagonized drug-induced paralysis. In addition, in B. pahangi, all of the tricarboxylic acid cycle intermediates (10 mM) except citrate and pyruvate antagonized the

Complex interplay of riboregulatory mechanisms at the gcvT-NGFG_01513-gcvH operon of Neisseria gonorrhoeae.

UNLABELLED: The sibling sRNAs NgncR_162 and NgncR_163 are global post-transcriptional regulators of Neisseria gonorrhoeae metabolism, which affect amino acid uptake and biosynthesis, the citric acid and methyl citric acid cycle, and in particular pathways contributing to serine-glycine metabolism. Here, we show that gcvT, gcvP, and gcvH, constituting specific components of the glycine cleavage system which converts glycine to carbon dioxide, ammonia, and 5,10-methylene tetrahydrofolate, are negatively regulated by the sibling sRNAs, while the fourth component of the GCV multienzyme complex, th
