Cognition-enhancing drugs: current landscape, mechanisms, and future prospects.

Cognition-enhancing drugs (CEDs) are pharmacological agents aimed at improving memory, attention, and executive function. This review explores both established and emerging CEDs that target neurotransmitter systems, neuroinflammation, and neuroplasticity. It discusses innovative design strategies, such as multi-target agents and prodrugs, and examines clinical challenges in Alzheimer's disease and other conditions. We address ethical, societal, and regulatory concerns, especially those relating to non-medical use, highlighting the importance of responsible development and precision-based appro

Biological cognition as viability-constrained expansion of controllable futures.

Biological cognition is often described through homeostasis, prediction, exploration, or control. However, none of these principles alone specifies how a living system should act while preserving viability and enlarging its future possibilities. We tested whether living-cognition-like policy selection can be operationalized as viability-constrained expansion of controllable futures. A minimal grid-world model was implemented in which agents evaluated finite-horizon policies under internal viability dynamics, hazards, resources, movement costs, and reachable-endpoint controllability. Seven redu

Resting-state intracranial high-frequency oscillations and human cognition.

In 59 drug-resistant epilepsy patients undergoing stereo-electroencephalography, we evaluated relationships between rates of 80-250 Hz high-frequency oscillations (HFO-Rs) and standardized neuropsychological scores. We also quantified how local HFO-Rs modulate functional connectivity among brain regions showing significant HFO-R-cognitive score associations. We hypothesized that regional variation in cortical HFO-Rs relates to domain-specific neuropsychological performance. We analyzed HFO-Rs in 6048 gray matter channels excluding epileptogenic and lesional cortices. Resting-state HFO-Rs were

Clozapine-induced human microglial exosomes impair neurites and cognition.

BACKGROUND: Clozapine is the most effective treatment for treatment-resistant schizophrenia but has been linked to cognitive impairment and brain volume reductions. The potential mechanisms underlying these effects remain unclear. Microglial exosomes, which carry microRNAs (miRNAs) and other cargo, act as immune-neuron communication vectors capable of modulating neuronal function and cognition. METHODS: We compared cognitive performance and inflammatory markers across clozapine-treated individuals, haloperidol-treated individuals, and healthy controls. Human microglial cells were treated with

The Mind From Within: Visceral Roots of Human Cognition.

Humans think and act not only with the brain but also with the heart, lungs, gut, and other viscera, such as the bladder, long regarded as primarily vegetative. A fresh wave of experiments is revealing the role of cardiac-cycle phase and baroreceptor firing in perception and social behavior; respiration-locked neural dynamics in attention and learning; and gastrointestinal rhythms and vagal afferents in affect and decision-making. These discoveries are made possible by emerging methods, including ingestible sensors that record gastrointestinal signals in situ during emotional and cognitive tas

Association of sleep duration with Alzheimer's disease and cognition.

INTRODUCTION: The association of sleep duration with Alzheimer's disease (AD) - related brain atrophy and cognition remains unclear. METHODS: Among 38,816 participants in the UK Biobank, we examined the association between sleep duration and a validated machine-learning-derived magnetic resonance imaging (MRI) signature of AD-related atrophy (Spatial Pattern of Abnormality for Recognition of Early AD [SPARE-AD]) and three cognitive test scores using generalized additive models. Independently, electronic health records (EHR) from TriNetX were used to examine the 10-year AD risk associated with

Positive Childhood Experiences, Cognition, and Biomarkers of Alzheimer's Disease.

Positive childhood experiences (PCEs) have unknown effects on late life cognition and Alzheimer's Disease biomarkers. We examined 406 Asian, 1179 Black, 349 Latinx, and 498 White KHANDLE and STAR study participants with data on PCEs, longitudinal cognitive measures, MRI (n = 560), and amyloid PET (n = 281). We conducted mediation and multigroup models within the structural equation modeling framework allowing us to examine the direct association of PCEs with episodic memory level and change as well as the indirect effects of PCEs through education. We additionally conducted linear regressions

Connecting to Cognition: A Methodological Journey With Urban Dementia.

Contemporary sociological developments spanning the ontological turn, post-cognitivism, biosocial transdisciplinarity, post-qualitative inquiry, and new empiricism are opening up novel sites of methodological contention regarding the nature of cognition as a research object. While these developments are conceptually stimulating, their (potential) overlapping methodological implications are under-explicated. Outlining a moving ethnography of urban transport use by passengers with dementia, in this article I delve into the more pragmatic methodological contingencies of attempts to engage with co

Sleep, cognition, and electrophysiology: Intersecting pathways in childhood cancer.

Survivors of pediatric cancer are at high risk of long-term cognitive dysfunction. This problem is exacerbated as this population often reports poor sleep quality, which can negatively affect well-being. Sleep-related brain activity is known to influence cognitive development by regulating memory consolidation and cognitive functioning. However, the differences in sleep-related brain activity between healthy controls and pediatric cancer survivors are unclear. This review examines how sleep disturbances and cognitive impairments converge in survivors of childhood cancer, and proposes electroph

Perinatal fluoxetine exposure does not impair cognition in offspring.

Selective serotonin reuptake inhibitors (SSRIs) such as fluoxetine (FLX) are widely prescribed to treat depression during pregnancy, however their lasting effects on both maternal and offspring behavior remains uncertain. Long-term outcomes on cognitive performance in the offspring are mixed, and in addition the role of maternal care is understudied. In humans it is impossible to study the effect of SSRIs without the underlying maternal illness, therefore, our aim was to investigate the long-term effects of perinatal FLX exposure on cognitive performance of dams and their offspring in Wistar r

Research progress on platelet factor 4 in improving cognition.

Platelet Factor 4 (PF4) is a chemokine that plays a crucial role in coagulation and immune functions. Previous studies have identified numerous biological functions of PF4, including its involvement in hematopoiesis, inhibition of angiogenesis, modulation of platelet coagulation, promotion of the host inflammatory response, vascular inhibition, and anti-tumor properties. Surprisingly, recent research has revealed that PF4 also contributes to improving cognitive abilities and restoring neural aging. Mice treated with PF4 demonstrated enhanced performance in behavioral experiments, indicating th

Carotid revascularization improves cognition in patients with carotid stenosis.

Patients with atherosclerosis in the carotid arteries are at risk of ischemic stroke and cognitive decline due to emboli and chronic hypoperfusion of brain tissue. Revascularization procedures improve cerebral hemodynamics, which has some effect on cognitive function. Some authors suggest that the presence of stenosis in the carotid arteries is an independent factor influencing cognitive decline. The aim of this study was to investigate the relationship between the degree of stenosis of a stented carotid artery and attentional performance in individuals with carotid atherosclerosis. A prospect

Effects of ketamine on postoperative cognition: a scoping review.

Postoperative delirium and other forms of perioperative neurocognitive deficits occur commonly in older adult patients. Ketamine, administered either before or during general anaesthesia, may have cognitive-sparing properties owing to its ability to reduce neuroinflammation, increase neurotrophin levels, or allow reductions in the doses of other general anaesthetic drugs. However, the efficacy of ketamine in preventing perioperative neurocognitive disorders remains highly controversial. This scoping review summarises clinical studies that examined the cognitive-sparing properties of ketamine i

Social cognition, psychosocial development and well-being in galactosemia.

BACKGROUND: Classic galactosemia is a rare inherited metabolic disease with long-term complications, particularly in the psychosocial domain. Patients report a lower quality of social life, difficulties in interactions and social relationships, and a lower mental health. We hypothesised that social cognition deficits could partially explain this psychological symptomatology. Eleven adults with galactosemia and 31 control adults participated in the study. We measured social cognition skills in cognitive and affective theory of Mind, and in basic and complex emotion recognition. We explored psyc

Social isolation impairs cognition via Aβ-mediated synaptic dysfunction.

Social isolation (SI) is a common phenomenon in the modern world, especially during the coronavirus disease 2019 pandemic, and causes lasting cognitive impairments and mental disorders. However, it is still unclear how SI alters molecules in the brain and induces behavioural dysfunctions. Here, we report that SI impairs cognitive function and induces depressive-like behaviours in C57BL/6 J mice, in addition to impairing synaptic plasticity and increasing the levels of APP cleavage-related enzymes, thereby promoting Aβ production. Moreover, we show that in APP/PS1 transgenic mice, SI accelerate

Cerebral amyloid angiopathy impacts neurofibrillary tangle burden and cognition.

Cerebral amyloid angiopathy commonly co-occurs with amyloid β plaques and neurofibrillary degeneration and is proposed to contribute to cognitive impairment. However, the interplay among these pathologic changes of Alzheimer disease is not well understood. Here we replicate and extend findings of a recent study that suggested the association of cerebral amyloid angiopathy and cognitive impairment is mediated by neurofibrillary degeneration. We employed similar approaches but in a larger, clinical-based (as opposed to community-based) set of 4915 autopsied National Alzheimer's Coordinating Cent
