Biochar reinforced the populations of cbbL-containing autotrophic microbes and humic substance formation via sequestrating CO2 in composting process.

The quality of compost is drastically reduced due to the loss of carbon, which negatively impacts the environment. Carbon emission reduction and carbon dioxide (CO2) fixation have attracted much attention in composting research. In this study, the relationship between CO2 emission, humic substances (HS) formation and cbbL-containing autotrophic microbes (CCAM) was analyzed by adding biochar during cow manure composting. The results showed that biochar can facilitate the degradation of organic matter (OM) and formation of HS, as well as reinforce the diversity and abundance of CCAM community, t…

Combined addition of biochar and garbage enzyme improving the humification and succession of fungal community during sewage sludge composting.

The influences of combination of garbage enzyme and biochar on total organic carbon (TOC) degradation, humification and the fungal succession during sewage sludge (SS) composting were established. Results showed that the GE and BC + GE treatments significantly increased the enzyme activity of fluorescein diacetate hydrolase (FDA) and increased the TOC degradation rate by 9.8% and 21.9% relative to control. The excitation-emission matrix (EEM) combined with the percentage fluorescence response (Pi, n) also proved that the combination of BC and GE promoted fulvic acid-like and humic-like substan…

Effects of chlortetracycline on the fate of multi-antibiotic resistance genes and the microbial community during swine manure composting.

Excessive use of antibiotics in breeding industry leads to accumulation of antibiotic residuals and antibiotic resistance genes (ARGs) in environment from improperly treated livestock excrements. Four commonly used veterinary antibiotics including chlortetracycline (CTC), sulfamerazine (SMZ), enrofloxacin (ENR) and erythromycin (ERY) were monitored in the swine manure composting. Co-resistance and cross-resistance effects among relative ARGs, correlations between ARGs and bacterial community under multiple antibiotics residual during the composting were investigated in this research. With CTC …

Heat and desiccation are the predominant factors affecting inactivation of Bacillus licheniformis and Bacillus thuringiensis spores during simulated composting.

AIMS: The suitability of composting for disposal of livestock mortalities due to Bacillus anthracis was assessed by measuring viability of surrogate spores from two strains each of Bacillus licheniformis and Bacillus thuringiensis after a heating cycle modelled on a cattle composting study. METHODS AND RESULTS: Sporulation was attempted from 10 to 37°C, but poor yields at lower temperatures resulted in 25, 30 and 37°C being selected to generate sufficient spores (8 log10  CFU ml(-1) ) for experiments. Spores were inoculated into 3 g autoclaved dried-ground compost rehydrated with 6 ml water o…

Real-time determination of swine manure liquid compost maturity by oxidation-reduction potential and pH monitoring.

The aim of this study was to develop a real-time diagnostic method for assessing the maturity of liquid compost from swine manure by monitoring the oxidation-reduction potential (ORP) and pH, two key indicators of the oxidative status of organic matter and nitrogen compounds. The experimental system comprised a cylindrical bioreactor equipped with an aeration and mixing device. During the liquid composting process of swine manure in the bioreactor, the ORP and pH were continuously monitored, while changes in the liquid compost maturity and concentrations of soluble nitrogen and organic matter …

Compost application more strongly shapes soil fungal than bacterial communities across arable fields.

Organic amendments such as compost are widely promoted for enriching soil microbial communities and supporting key soil functions, yet their … xa from both bacterial (Thermoactinomycetaceae) and fungal (Thermomyces) lineages characteristic of composting processes, suggesting establishment of compost-derived microbes in agricultural soils. In contrast, solid digestate from biogas plants had weaker and more variable effects on soil microbial communities. Structural equation modelling suggested that c …

Amoxicillin and copper affect humification during compost: Insight into dissolved organic matter and bacterial community succession.

Amoxicillin (AMX) and copper (Cu) often coexist in manure, while their combined effects at varying concentrations and the associated mechanisms influencing compost humification remain unclear. The present study combined multiple spectroscopic characterization and microbial analysis methods to elucidate the effects of AMX and Cu on the humification during aerobic composting of dairy manure. The results revealed that both low and high levels of combined AMX and Cu impeded dissolved organic matter degradation and led to a reduction in humic acid (HA) content by 3.8%-5.0% relative to the control t…

Application of solutions obtained from tomato waste compost and wood ash as organic fertilizer in tomato cultivation.

BACKGROUND: The present study investigates the capacity of solutions derived from organic fertilisers to enhance the yield and quality of tomatoes (both grafted and ungrafted) cultivated in greenhouses. Three fundamental fertilisation applications were conducted: an organic tomato waste compost solution (CS), a wood ash solution (WAS), and an inorganic (NPK; 20-20-20 + ME) fertilisation control. The administration of organic fertilisers was conducted at a rate of 0.5 L per plant per week, formulated as a solution with an EC value of 2 dS m- 1, which is widely regarded as being optimal for the

Enhancing compost safety: Simultaneous reduction of antibiotic resistance genes and bioavailable heavy metals by microbial additives.

The continuous accumulation of antibiotic resistance genes (ARGs) and heavy metals (HMs) during sludge composting poses increasing environmental risks, highlighting the urgent need to explore effective remediation strategies. However, the mechanisms by which microbial additives simultaneously mitigate these two types of pollutants during sludge composting remain unclear. Different doses (0, 0.03 %, 0.09 %, and 0.15 %) of a commercial microbial agent were added during co-composting of sludge and garden waste to evaluate their impacts on the bioavailability of HMs and the abundance of ARGs. The …

Effects of composting on the fate of doxycycline, microbial community, and antibiotic resistance genes in swine manure and broiler manure.

Aerobic composting is an economical and effective technology that is widely used to treat animal manure. To study the fate of doxycycline (DOX), the microbial community, and antibiotic resistance genes (ARGs) during composting, aerobic composting of broiler manure and swine manure was carried out under natural environmental conditions. Aerobic composting effectively removed DOX (with a removal rate > 97%) and most ARGs from animal manure. The microbial diversity and the numbers of ARGs were higher in composted swine manure compared with composted broiler manure. The microbial community structu…

A novel fast-vegetative propagation technique of the pioneer shrub Baccharis linearis on mine tailings by adding compost.

Interest in Baccharis linearis has increased as an alternative for assisted phytostabilization due to its spontaneous colonization of tailings dumps. The search for a novel fast-vegetative propagation technique to accelerate its coverage on mine tailings is a promising research area for sustainable mine closure plans. In this study, we determined the optimal proportion of compost and tailings as growing media to promote fast B. linearis propagation through a compound layering technique. The assessed growing substrates were: 100% tailings, 70% tailings + 30% compost, and 50% tailings + 50% comp

Manure Compost Is a Potential Source of Tetracycline-Resistant Escherichia coli and Tetracycline Resistance Genes in Japanese Farms.

Manure compost has been thought of as a potential important route of transmission of antimicrobial-resistant bacteria (ARB) and antimicrobial resistance genes (ARGs) from livestock to humans. To clarify the abundance of ARB and ARGs, ARB and ARGs were quantitatively determined in tetracycline-resistant Escherichia coli (harboring the tetA gene)-spiked feces in simulated composts. In the simulated composts, the concentration of spiked E. coli decreased below the detection limit at day 7. The tetA gene remained in manure compost for 20 days, although the levels of the gene decreased. Next, to cl

Development and application of EEAST: a life cycle based model for use of harvested rainwater and composting toilets in buildings.

Harvested rainwater systems and composting toilets are expected to be an important part of sustainable solutions in buildings. Yet, to this date, a model evaluating their economic and environmental impact has been missing. To address this need, a life cycle based model, EEAST was developed. EEAST was designed to compare the business as usual (BAU) case of using potable water for toilet flushing and irrigation to alternative scenarios of rainwater harvesting and composting toilet based technologies. In EEAST, building characteristics, occupancy, and precipitation are used to size the harvested …

Purification of polymerase chain reaction (PCR)-amplifiable DNA from compost piles containing bovine mortalities.

Livestock production systems utilize composting as a method of disposal of livestock mortalities, but there is limited information on the rate and extent of carcass decomposition. Detection of specific DNA fragments by PCR offers a method for investigating the degradation of carcasses and other biological materials during composting. However, the purity of extracted DNA is critical for successful PCR analysis. We applied a method to purify DNA from compost samples and have tested the method by analyzing bovine and plant DNA targets after 0, 4, and 12 month of composting. The concentration of o…

Clostridium botulinum and bio-compost. A contribution to the analysis of potential health hazards caused by bio-waste recycling.

Bio-waste recycling and the production and use of bio-compost are politically encouraged in Europe. Quality control takes no consideration of pathogenic anaerobic spore formers, e.g. Clostridium botulinum. A protocol for health hazard analysis concerning this pathogen has been developed. Samples of marketed bio-compost were tested and results showed that about 50% of the tested samples contained C. botulinum. For the first time it has been shown that the use of bio-compost represents a health hazard to humans and animals, especially in the future when spores will have accumulated in the enviro

Cd, Cr, and Pb concentrations increase during composting: implications of uncertain mass balance and analytical recovery for compliance sampling.

Metal concentrations during composting reflect changes in dry mass, metal inputs or losses, and the fraction recovered by digestion. Concentrations of Zn, Cu, Fe, Mn, Ni, Cr, Cd, and Pb were measured over 90 days in five poultry litter (PL): vegetable waste blends and analyzed under three assumptions about dry matter loss. Cd in pure-PL compost increased from 0.84 to 3.34 mg kg-1 dry matter. After correction for the loss inferred from Zn and Mn, mean apparent recovery factors were 1.03 for Zn, Cu, Mn, and Ni and 5.11 for Cr, Cd, and Pb (95% bootstrap interval 3.05-8.23). The latter group remai…

Integrated metagenomic and 16S rRNA analysis reveals temporal associations between resistance genes and microbial communities during dairy manure composting.

Dairy manure composting is widely applied to stabilize organic waste and reduce environmental pollution, yet the behavior of resistance determinants during this process remains insufficiently resolved. In this study, shotgun metagenomic sequencing was used to characterize temporal changes in antibiotic resistance genes (ARGs), metal resistance genes (MRGs), biocide resistance genes (BRGs), mobile genetic elements (MGEs), and microbial community composition during dairy manure composting. Rather than inferring direct mechanistic causation, our analyses focused on identifying statistically suppo…

Comparative effects of siderite, goethite, and hematite on nitrogen loss and nitrogenous gases release during municipal sludge aerobic composting.

Despite iron ores exhibiting the potential to regulate microbial nitrogen transformation, their roles in municipal sludge aerobic composting remain unclear. This study evaluated the effects of siderite, goethite, and hematite on nitrogen loss and nitrogenous gases emissions during sludge aerobic composting. Results showed that all three iron ores increased ROS and Fe(II) concentrations, with goethite and hematite increasing nitrogen retention by 13.13% and 18.18%. Goethite and hematite regulated pH and Fe(II) dynamics and increased the abundance of hao and nxrA genes, thereby enhancing nitrifi…

Greenhouse gas emissions in a microbial fuel cell-assisted composting system: Microbial insights into bacterial structures and genetic functions.

Greenhouse gas emissions, including methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O), have significant environmental impacts during the composting process. Herein, a microbial fuel cell (MFC)-assisted composting system was employed to investigate the emission profiles of these gases. CH4 and CO2 emissions were peaked during the temperature-heating phase, while N2O emission showed maximum levels during the temperature-heating and maturation phases. Compared to the control (without MFC), MFC increased the abundance of Proteobacteria, Bacteroidota, and Gemmatimonadota in the compostin…
