Multiple lymphomatous polyposis (MLP), characterized by multiple polyps involving long segments of the gastrointestinal (GI) tract, is believed to represent GI involvement by mantle cell lymphoma (MCL), primarily based on its histologic and immunophenotypic similarities with MCL. However, rearrangement of the bcl-1 locus, the molecular lesion characteristic of MCL, has not been investigated in this group of patients. The authors evaluated the morphologic, immunophenotypic, and molecular features of 18 cases of MLP and 8 B-cell lymphomas involving the GI tract (including 6 MALT lymphomas). All

Centrocytic lymphoma, or mantle cell lymphoma (MCL), is characterized by a chromosomal translocation t(11;14) (q13;q32) involving the bcl-1 locus on chromosome 11. Cyclin D1 is a cell-cycle regulatory protein essential for G1-S transition and has been identified as a potential transforming gene affected by the translocation. In this study, 32 cases of MCL were analysed for the bcl-1 rearrangement and cyclin D1 protein expression. In 17 cases, a rearrangement at the major translocation cluster of bcl-1 could be detected. Twenty-four cases exhibited nuclear cyclin D1 expression that was not dete

BACKGROUND: In the normal cell cycle, tumor suppressor gene products (p53) and cyclin (cyclin D1) cooperate. Abnormalities in the cooperation of these factors may result in malignant transformation of the cell. Mutant p53 protein overexpression is defined in many human cancers, including endometrial carcinoma. This study investigated the role of cyclin D1 in the development of human uterine endometrial carcinoma.
METHODS: Seventy-four patients whose pathology slides contained either normal or hyperplastic endometrium adjacent to endometrial carcinoma were studied. Immunohistochemical staining

We have previously shown that the p57KIP2 gene, which encodes a cyclin-dependent kinase inhibitor, undergoes genomic imprinting and lies within a 700-kb domain of imprinted genes on 11p15, including IGF2 and H19. Loss of heterozygosity and loss of imprinting (LOI) of this region are frequently observed in Wilms' tumor (WT) and other embryonal malignancies. Although LOI of p57KIP2 was observed in some WTs (approximately 10%), allele-specific expression was preserved in most tumors examined. Because our initial studies were inconclusive concerning the absolute expression level of p57KIP2 in WT,

The growth arrest mediated by p53 is caused at least in part by the p53 mediated expression of p21 (p21waf1/Cip1). Since only one-third of primary Burkitt's lymphomas (BL) demonstrate mutations in the p53 gene, we examined the structural integrity of the p21 coding region by single-strand conformational polymorphism and DNA sequence analysis to determine the extent to which this gene is mutated in BL. Of 34 BLs analyzed, a frequent change (38%) at codon 31 that replaced Ser with Arg was found in 13 samples, 10 of which were from Africa. This change at codon 31 is also detected in peripheral bl

The expressions and significance of proliferating cell nuclear antigen (PCNA) were studied by immunohistochemical method in 40 cases of gallbladder carcinoma and 42 cases of cholangiocarcinoma. The counting score of positive cells was significantly lower in cases of well-differentiation, histologic grade I and without metastasis than that in cases of poor-differentiation, histologic grade III and with metastasis. Close correlations were found between the survival time and the counting score of PCNA. These results suggested the cholangiocarcinoma and gallbladder carcinoma with highly positive c

The major locus for autosomal dominant polycystic kidney disease (PKD1) is located in a gene-rich region on chromosome 16p13.3. Recently the identification of the gene responsible for PKD1 has been described. While searching for candidate genes in this region, we isolated a new member of the cyclin family. We have characterized the transcript by sequencing, determination of the exon intron boundaries, and Northern blot analysis. Cyclin F is related to A- and B-type cyclins by sequence, but its function is unknown.

Dysregulated cell cycle progression drives uncontrolled proliferation and is a fundamental hallmark of cancer, making cyclin-dependent kinases (CDKs), which largely govern cell-cycle progression and cell division, attractive therapeutic targets. Currently, CDK inhibitors (CDKIs), particularly agents targeting CDK4/6, have shown clinical activity in HR+/HER2-metastatic breast cancer. Recent structural elucidation of the CDK2 catalytic domain and its allosteric sites has enabled the development of several selective CDK2 inhibitors, many of which have exhibited encouraging antitumor activity. BACKGROUND: Cyclin-dependent kinase 4/6 (CDK4/6) inhibitors are widely used in the treatment of advanced breast cancer. While a reversible increase in serum creatinine is a recognized class effect, the spectrum and severity of renal adverse events (AE) associated with these agents remain incompletely characterized.
METHODS: We conducted a retrospective analysis of the French national pharmacovigilance database from 2016 to January 2024 to identify reports of renal AE associated with the CDK4/6 inhibitors, palbociclib, abemaciclib, and ribociclib. Cases were classified as pseudo-renal failure,

Glomus tumors of the female genital tract are rare, and to our knowledge, malignant glomus tumors (MGTs) of the female genital tract have not been previously reported. The diagnosis of MGTs is challenging, given their morphologic and immunophenotypic overlap with other common uterine mesenchymal tumors, especially in the absence of classic benign glomus tumor components. Here, we report a case involving a 34-year-old woman with uterine MGT that was positive for smooth muscle actin, h-caldesmon, cyclin D1, and synaptophysin, and negative for desmin. CARMN::NOTCH2 fusion was identified using hyb

Cyclin E1 is a critical mediator of cell cycle dynamics, controlling G1/S phase transition in complex with CDK2. Overexpression of cyclin E1 can lead to replication stress, DNA damage, and heightened genomic instability. This is associated with oncogenesis, and is frequently observed pan-cancer; but is particularly common in high grade serous ovarian cancer (HGSOC). HGSOC exhibits a genomic landscape dominated by structural variation and instability, with high cyclin E1 expression in up to 58 % and CCNE1 amplification in up to 30 % of cases. HGSOC therefore represents an exemplar setting for i

Cyclin dependent kinase 2 (CDK2) is responsible for enforcing progression through the G1-S phase transition. Mutations and alterations in the CDK2 signaling pathway are associated with various cancers, most commonly breast, ovarian, prostate, leukemia, and lymphoma. CDK2 inhibitors have shown promising preclinical and early clinical results, and this class of agents may be most effective against cancers with cyclin E overactivity. Common side effects observed include nausea, vomiting, diarrhea, anemia, and fatigue. This clinical review summarizes past and current CDK2 inhibitors in clinical tr

BACKGROUND AND OBJECTIVES: Numerous studies have demonstrated that differentially expressed microRNA (miRNA) play a crucial role in the pathophysiology of preeclampsia (PE). These miRNA are central to ongoing investigations into the pathogenesis of PE. This study aims to investigate the relationship between changes in miR-377-3p and cell cyclin G2 (CCNG2) expression and the clinical features of PE, as well as to further analyze the effect of miR-377-3p on CCNG2.
METHODS: A total of 120 pregnant subjects were recruited, consisting of 70 PE cases and 50 healthy pregnancies. The expression level

Proper action of the female sex steroids 17β-estradiol (E2) and progesterone (P4) on the endometrium is essential for fertility. Beyond its role in regulating the cell cycle, cyclin A2 (CCNA2) also mediates E2 and P4 signaling in vitro, but a potential role in modulating steroid action for proper endometrial tissue development and function is unknown. To fill this gap in our knowledge, we examined human endometrial tissue from fertile and infertile cisgender women for CCNA2 expression and correlated this with pregnancy outcome. Functional assessment of CCNA2 was validated in vivo using a condi

INTRODUCTION: Rheumatoid arthritis (RA) is a chronic inflammatory disease associated with synovial proliferation and bone erosion, which leads to the structural and functional impairment of the joints. Immune cells, together with synoviocytes, induce a pro-inflammatory environment and novel treatment agents target inflammatory cytokines. Psoriasis is a chronic immune-mediated skin disease, and several cytokines are considered as typical mediators in the progression of the disease, including IL-23, IL-22, and IL-17, among others.
AREA COVERED: In this review, we try to evaluate whether cyclin-

BACKGROUND: Pulmonary hypertension (PH) is a chronic vascular disease characterized, among other abnormalities, by hyperproliferative smooth muscle cells and a perturbed cellular redox and metabolic balance. Oxidants induce cell cycle arrest to halt proliferation; however, little is known about the redox-regulated effector proteins that mediate these processes. Here, we report a novel kinase-inhibitory disulfide bond in cyclin D-CDK4 (cyclin-dependent kinase 4) and investigate its role in cell proliferation and PH.
METHODS: Oxidative modifications of cyclin D-CDK4 were detected in human pulmo

Mantle cell lymphoma (MCL) is a rare B-cell malignancy with a predominantly aggressive clinical course and poor prognosis. Abnormal expression of Ambra1 is closely related to the occurrence and development of various tumors. However, the role of Ambra1 in MCL remains unknown. Here, we performed both in vitro and in vivo experiments to investigate how Ambra1 regulates MCL progression and whether Ambra1 modulates the sensitivity of MCL cells to the CDK4/6 inhibitor palbociclib. We discovered that MCL cells had decreased levels of Ambra1 expression relative to normal B cells. Overexpression of Am
