Effects of pesticide exposure on oxidative stress and DNA methylation urinary biomarkers in Czech adults and children from the CELSPAC-SPECIMEn cohort.

Current-use pesticide (CUP) exposure occurs mainly through diet and environmental application in both agricultural and urban settings. While pesticide exposure has been associated with many adverse health outcomes, the intermediary molecular mechanisms are still not completely elucidated. Among others, their roles in epigenetics (DNA methylation) and DNA damage due to oxidative stress are presumed. Scientific evidence on urinary biomarkers of such body response in general population is limited, especially in children. A total of 440 urine samples (n = 110 parent-child pairs) were collected dur

Aberrant DNA methylation profile exacerbates inflammation and neurodegeneration in multiple sclerosis patients.

Multiple sclerosis (MS) is an autoimmune and demyelinating disease of the central nervous system characterised by incoordination, sensory loss, weakness, changes in bladder capacity and bowel function, fatigue and cognitive impairment, creating a significant socioeconomic burden. The pathogenesis of MS involves both genetic susceptibility and exposure to distinct environmental risk factors. The gene x environment interaction is regulated by epigenetic mechanisms. Epigenetics refers to a complex system that modifies gene expression without altering the DNA sequence. The most studied epigenetic

Life style factors and acquired susceptibility to environmental disease.

Multifactorial risk factors are responsible for many diseases. They can be broadly categorized as environmental, genetic and life style factors. Much attention has been focused on the first two categories, e.g. the identification of environmental toxicants/carcinogens and the elucidation of genetic susceptibility to disease. Life style risk factors such as aging, poor nutrition, infection and exposure to toxicants can also increase susceptibility to illnesses. These life style factors can therefore be considered to cause acquired susceptibility for increased risk for environmental disease. Amo

Detection of Brucella abortus B19 strain DNA in seminal plasma by polymerase chain reaction in Brazil.

A total of 27 seminal plasma samples from cattle-breeding farms or semen centres located in Minas Gerais, Brazil, previously negative by serological and tested positive for Brucella spp. with primer specific for the amplification of the gene virb5 by polymerase chain reaction (PCR) were analysed for the detection of Brucella abortus DNA by PCR. It was found that nine samples (33.33%) contained B. abortus B19 strain DNA, two (7.40%) contained B. abortus DNA and five (18.51%) contained both DNA. The larger number of samples with B. abortus B19 strain DNA would explained by the environmental cont

Sperm as a multilayered epigenetic information system: encoding and transmission of paternal environmental signals.

Spermatozoa are increasingly recognized as carriers of non-genetic paternal information, rather than passive vehicles for the haploid genome. During spermatogenesis and post-testicular epididymal maturation, the paternal germline establishes a compact but functionally organized epigenome composed of DNA methylation, retained histones and histone modifications, chromatin-associated factors, and diverse small RNAs. These layers are environmentally responsive. In animal models, paternal diet, stress, toxicant exposure, and inflammatory or microbiome-related challenges can alter sperm DNA methylat

Epigenetic inheritance of DNA methylation changes in fish living in hydrogen sulfide-rich springs.

Environmental factors can promote phenotypic variation through alterations in the epigenome and facilitate adaptation of an organism to the environment. Although hydrogen sulfide is toxic to most organisms, the fish Poecilia mexicana has adapted to survive in environments with high levels that exceed toxicity thresholds by orders of magnitude. Epigenetic changes in response to this environmental stressor were examined by assessing DNA methylation alterations in red blood cells, which are nucleated in fish. Males and females were sampled from sulfidic and nonsulfidic natural environments; indiv

Genotoxicity and DNA Damage in Long-Term SSRI Therapy: A Review Across SSRIs With Citalopram as a Case Study.

Selective serotonin reuptake inhibitors (SSRIs) are first-line therapies for depressive and anxiety disorders and are often used for years, including in reproductive-age patients and during pregnancy. This has intensified concern about possible genotoxicity and DNA damage. Citalopram is emphasized because it has the most extensive and consistently positive in vivo genotoxicity evidence among SSRIs, particularly in rodent comet and micronucleus endpoints. This review summarizes genotoxicity data for SSRIs, focusing on citalopram, across in vitro assays, animal studies, human biomonitoring, and

Introduction to epigenetic mechanisms in the brain.

This chapter provides an introductory overview of epigenetic mechanisms, focusing on the molecular processes that regulate gene expression without altering the DNA sequence. It outlines the foundational concepts of DNA methylation, histone modifications, chromatin remodeling, and noncoding RNAs, with emphasis on their roles in cell identity and transcriptional regulation. The interactions between these epigenetic mechanisms, such as the crosstalk of histone acetylation and DNA methylation, are also discussed. Key mechanisms of long-term gene inactivation, like X-chromosome inactivation and gen

An Overview of How Epigenetics, MicroRNA-21, and Endocrine Disrupting Compounds Affect Oocyte Maturation and Pre-Implantation Embryo Development.

Epigenetic regulation is pivotal in reproductive processes, such as oocyte maturation and pre-implantation embryonic development, and it impacts gene expression without altering DNA sequence through mechanisms including DNA methylation, histone modifications, and non-coding RNAs. Primarily, microRNA-21 is involved in meiotic progression, apoptosis, and cumulus cell function, which are necessary for oocyte competency. miR-21 dysregulation can lead to improper oocyte maturation and poor embryonic development, ultimately causing developmental defects. During pre-implantation embryonic development

Mitochondria-Targeting microRNAs (mitomiRs): Potential Mediators of Environmental Mitoepigenetics in Mammalian Spermatogenesis.

The field of mitoepigenetics involves the investigation of modifications in mitochondrial DNA (mtDNA), genomic DNA that encodes mitochondrial proteins, and the expression of RNAs that regulate mitochondrial gene expression but do not alter the DNA sequence. This area of research is growing rapidly and has substantial relevance to male mammalian fertility. Among the known mitoepigenetic mechanisms, mitochondrial microRNAs (mitomiRs) have attracted substantial attention due to their potential roles in modulating mitochondrial gene expression in response to environmental stressors. Since many pro

Epigenetic Changes in the Pathogenesis of Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a systemic autoimmune disease that affects about 1% of the world's population. The etiology of RA remains unknown. It is considered to occur in the presence of genetic and environmental factors. An increasing body of evidence pinpoints that epigenetic modifications play an important role in the regulation of RA pathogenesis. Epigenetics causes heritable phenotype changes that are not determined by changes in the DNA sequence. The major epigenetic mechanisms include DNA methylation, histone proteins modifications and changes in gene expression caused by microRNAs an

A CpG Epigenetic Switch Reverts PFAS ToxicityAn In Vitro Study.

Perfluorooctanesulfonic acid (PFOS) is a persistent environmental pollutant widely present in ecosystems and humans, linked to numerous diseases, including cancers, due to its potent toxicity. Despite its harmful effects, effective strategies for mitigating PFOS-induced toxicity remain undeveloped or at best underdeveloped. In this effort, we explore the concept of toxicity reversal by loci-specific DNA methylation editing to reverse PFOS-induced toxicity, whereby key cellular processes such as proliferation, migration, and apoptosis are restored. To demonstrate our concept, we employed CRISPR

m6A-methylated Lonp1 drives mitochondrial proteostasis stress to induce testicular pyroptosis upon environmental cadmium exposure.

Cadmium (Cd) is a widely distributed typical environmental pollutant and one of the most toxic heavy metals. It is well-known that environmental Cd causes testicular damage by inducing classic types of cell death such as cell apoptosis and necrosis. However, as a new type of cell death, the role and mechanism of pyroptosis in Cd-induced testicular injury remain unclear. In the current study, we used environmental Cd to generate a murine model with testicular injury and AIM2-dependent pyroptosis. Based on the model, we found that increased cytoplasmic mitochondrial DNA (mtDNA), activated mitoch

The neuroendocrine modulation of global DNA methylation in neuropsychiatric disorders.

There is an increasing body of knowledge on the influence of differential DNA methylation of specific genomic regions in psychiatric disorders. However, fewer studies have addressed global DNA methylation (GMe) levels. GMe is an estimative of biological functioning that is regulated by pervasive mechanisms able to capture the big picture of metabolic and environmental influences upon gene expression. In the present perspective article, we highlighted evidence for the relationships between cortisol and sex hormones and GMe in psychiatric disorders. We argue that the far-reaching effects of cort

Highly Sensitive and Real-Time Detection of Zinc Oxide Nanoparticles Using Quartz Crystal Microbalance via DNA Induced Conjugation.

With the development of nanotechnology, nanomaterials have been widely used in the development of commercial products. In particular, zinc oxide nanoparticles (ZnONPs) have been of great interest due to their extraordinary properties, such as semiconductive, piezoelectric, and absorbance properties in UVA and UVB (280-400 nm) spectra. However, recent studies have investigated the toxicity of these ZnONPs; therefore, a ZnONP screening tool is required for human health and environmental problems. In this study, we propose a detection method for ZnONPs using quartz crystal microbalance (QCM) and

DNA methylation differences associated with social anxiety disorder and early life adversity.

Social anxiety disorder (SAD) is a psychiatric disorder characterized by extensive fear in social situations. Multiple genetic and environmental factors are known to contribute to its pathogenesis. One of the main environmental risk factors is early life adversity (ELA). Evidence is emerging that epigenetic mechanisms such as DNA methylation might play an important role in the biological mechanisms underlying SAD and ELA. To investigate the relationship between ELA, DNA methylation, and SAD, we performed an epigenome-wide association study for SAD and ELA examining DNA from whole blood of a co

Multiomic characterization of DNA released after airway exposure to allergens and nanoparticles reveals selective export of repetitive and fragile site DNA.

Allergen and nanoparticle exposure generate reactive oxygen species (ROS) that endanger genome integrity, yet airway epithelial cells survive repeated exposures. Here we show that environmental allergens from Alternaria alternata and polyethyleneimine (PEI) nanoparticles induce oxidative stress, triggering the rapid release of double-stranded DNA fragments enriched for repetitive elements (SINEs, LINEs, and LTRs), centromeric sequences, and late-replicating, origin-poor fragile site-containing genes. Nanopore sequencing, along with a multiomics approach, was used to analyze fragments of extrac

Human formalin-fixed paraffin-embedded tissues: an untapped specimen for biomonitoring of carcinogen DNA adducts by mass spectrometry.

DNA adducts represent internal dosimeters to measure exposure to environmental and endogenous genotoxicants. Unfortunately, in molecular epidemiologic studies, measurements of DNA adducts often are precluded by the unavailability of fresh tissue. In contrast, formalin-fixed paraffin embedded (FFPE) tissues frequently are accessible for biomarker discovery. We report here that DNA adducts of aristolochic acids (AAs) can be measured in FFPE tissues at a level of sensitivity comparable to freshly frozen tissue. AAs are nephrotoxic and carcinogenic compounds found in Aristolochia herbaceous plants

Characterization of DNA methylation in PBMCs and donor-matched iPSCs shows age-related methylation is reset during stem cell reprogramming.

DNA methylation is an important epigenetic mechanism that helps define and maintain cellular functions. It is influenced by many factors, including environmental exposures, genotype, cell type, sex, and aging. Since age is the primary risk factor for developing neurodegenerative diseases, it is important to determine if age-related DNA methylation is retained when cells are reprogrammed to an induced Pluripotent Stem Cell (iPSC) state. Here, we selected peripheral blood mononuclear cells (PBMCs; n = 99) from a cohort of diverse and healthy individuals enrolled in the Genetic and Epigenetic Sig
