Neomycin-induced deafness in neonatal mice.

BACKGROUND: Hearing impairment is a rising public health issue, and current therapeutics fail to restore normal auditory sensation. Animal models are essential to a better understanding of the pathophysiology of deafness and developing therapeutics to restore hearing. NEW METHODS: Wild-type CBA/CaJ neonatal mice P2-5 were used in this study. Neomycin suspension (500 nl of 50 or 100 mg/ml) was micro-injected into the endolymphatic space. Cochlear morphology was examined 3 and 7 days after injection; hair cell (HC) loss, supporting cell morphology, and neurite denervation pattern were assessed …

Butylphthalide enhances recovery from sudden deafness.

OBJECTIVES: Cochlear microcirculation disturbance caused by vasculopathy is a common cause of sudden deafness (SD). Reactive oxygen species (ROS) plays an important role in cochlear injury during ischemia-reperfusion. Butylphthalide can improve microcirculation, reduce ROS formation and inhibit apoptosis. The aim of this study was to investigate the therapeutic effect of butylphthalide on patients with SD. PATIENTS AND METHODS: The hearing gains from 32 ears treated with butylphthalide were compared with that of 32 ears treated with non-butylphthalide. Butylphthalide capsules was administrate…

PMCA2 pump mutations and hereditary deafness.

Hair cells of the inner ear detect sound stimuli, inertial or gravitational forces by deflection of their apical stereocilia. A small number … ariation of the cytoplasmic free Ca2+ concentration ([Ca2+]c). Missense mutations of PMCA2w/a cause deafness and loss of balance in humans. Mouse models in which the pump is genetically ablated or mutated show hearing and balance impairment, which correlates with defects in homeostatic regulation of stereociliary [Ca2+]c, decreased sensitivity of mechanotr …

[Sensorineural deafness in Guillain-Barré syndrome].

The patient was a 31-year-old woman in whom facial paralysis occurred 1 week after the onset of pyelonephritis. Peripheral facial paralysis, sensorineural deafness, dysarthria, weakness of the four limbs, loss of tendon reflex, and peripheral sensory disturbance of the four limbs were noted during the initial examination. These symptoms were dominant in left side. The number of cells and protein level were increased in the cerebrospinal fluid, and reduced motor nerve action potential was detected with peripheral nerve conduction test. Based on these findings, axonal Guillain-Barré syndrome (GB…

Mitral valve prolapse and sudden deafness.

To examine the association between MVP and Idiopathic Sudden Sensorineural Hearing loss (ISSNHL). 349 subjects, 86 with ISSNHL and 263 controls underwent a 2D-echocardiography. Patients with ISSNHL had higher rates of MVP (29.1% vs 2.7%, p<0.001), mitral leaflet thickening (15.1% vs 2.3%, p<0.001), mitral regurgitation (16.3% vs 6.5%, p=0.02) and left atrial enlargement (11.6% vs 3.8%, p=0.01). Our results support the hypothesis that MVP could be one of the etiological factors of ISSNHL.

Familial mastocytosis associated with neurosensory deafness.

Mastocytosis is a disease characterized by excessive accumulation of mast cells in different tissues and symptoms caused by the release of mast cell mediators. The skin is frequently directly involved in mastocytosis. The disease is rarely seen in other members of the subjects' family; only 49 cases of familial mastocytosis have been reported. Familial mastocytosis associated with hearing loss may represent a newly described inherited entity. We describe a brother and sister exhibiting skin mastocytosis and neurosensory deafness, associated with a history of hearing loss in their father's fami…

Autosomal dominant ectrodactyly with sensorineural deafness.

Ectrodactyly or split hand/split foot malformation as it is now known, is a rare developmental disorder sometimes associated with other systemic malformations. Very few cases of congenital hearing loss coexisting with ectrodactyly have been reported world-wide. We profile one such family with the father and daughter being affected with bilateral foot ectrodactyly minor abnormalities of the 5th fingers and severe sensorineural hearing loss (SNHL). The inheritance in this family was likely to be autosomal dominant. There were no cytogenetically demonstrable structural aberrations in either case.

Cochlear implantation for symptomatic hereditary deafness.

Recently, the effectiveness of cochlear implantation for hereditary deafness has been reported. We performed cochlear implantation for two patients with symptomatic hereditary deafness. Deafness in one patient was thought to be a result of albinism-deafness syndrome and in the other patient, a result of chronic progressive external ophthalmoplegia syndrome. Since their speech perception abilities improved dramatically, we believe that cochlear implantation should be actively performed for these two syndromes.
