Macrophage inflammatory proteins in cytomegalovirus-related inner ear injury.

OBJECTIVE: Inner ear inflammation triggered by CMV infection may play a role in CMV-related auditory pathogenesis. The purpose of the study was to determine if a virally encoded macrophage inflammatory protein played a role in CMV-related hearing loss. DESIGN: Mutagenesis was performed with deletion of a guinea pig CMV macrophage inflammatory protein. Intracochlear inoculations were performed on three groups of animals (n = 18). Group 1 received sterile viral media, Group 2 received wild-type CMV virus, and Group 3 received "knockout" (KO) virus with a deleted immunomodulation gene. Baseline

Immunosuppressive treatment in bilateral vestibulopathy with inner ear antibodies.

CONCLUSIONS: Although vestibular recovery was observed after steroid treatment, it remains uncertain whether this improvement was spontaneous or due to medication. These data do not allow us to generally recommend corticosteroid treatment in patients with BVF and inner ear antibodies. OBJECTIVE: A retrospective study was performed based on the observation of two patients with suspected autoimmune bilateral vestibular failure (BVF) with normal hearing and antilabyrinthine or nervous tissue-specific serum antibodies who showed vestibular recovery after corticosteroid treatment. Clinical and experimental studies of autoimmune inner ear disease.

Otolaryngologists have long sought to identify causes of sensorineural hearing loss that could be reversed by medical treatment. An increasing amount of clinical and experimental evidence indicates that this postulated entity is related to autoimmune inner ear disease. Because of the lack of well-defined detection methods for identifying autoimmune processes within the human inner ear and the fact that it is one of the few organs of the body not amenable to diagnostic biopsy there has been great interest in developing animal models that mimic clinical entities. Different models will be present

Characterization of an experimentally induced inner ear immune response.

OBJECTIVE: To determine the effects of a sterile immune response on the structure and function of the cochlea. METHODS: An immune response was created in guinea pigs by systemically sensitizing the animals to keyhole limpet hemocyanin and subsequently challenging the inner ear with the protein. Animals were allowed to survive for 1 to 5 weeks, after which the cochlea was evaluated histologically. Hearing was measured by auditory brainstem response before the inner ear challenge, during the survival period, and prior to sacrifice. RESULTS: Inflammatory cells infiltrated the cochlea from the c

Non-insulin-dependent diabetic microangiopathy in the inner ear.

UNLABELLED: Hearing loss has long been associated with diabetes mellitus. Microangiopathy, associated with thickening of the basement membranes of small vessels, has been implicated as a major source of multiple system organ disease. OBJECTIVE: This study was designed to evaluate changes in basement membrane thickness in the inner ear of laboratory animals suffering from non-insulin-dependent diabetes mellitus (NIDDM) with, and without, exposure to moderate intensity noise exposure in an attempt to extrapolate the same disease process in humans. DESIGN: Spontaneously hypertensive-corpulent n

Steady-state free precession imaging of the inner ear.

The authors describe a steady-state free precession technique for imaging the inner ear. Although the signal-to-noise ratio is not as great as with other three-dimensional MR techniques, the inherent high contrast of inner ear structures makes this a valuable technique for patients with sensorineural hearing loss and for those who refuse paramagnetic contrast material.

[Correlation between inner ear disorders and temporomandibular joint diseases].

Acute and chronic inner ear diseases involve many etiological factors, some as yet unknown. ENT-specific, orthopedic, hemorrheological, immunological and neurological disorders can affect the cochleovestibular system and induce hearing loss, vertigo and/ortinnitus. We performed a prospective study to analyze factors of the dentognathological system and of the temporomandibular joint that can influence acute and chronic inner ear dysfunctions. A total of 138 patients (49.3% female, 50.9% male) receiving clinical treatment for inner ear dysfunctions (12.3% chronic sensorineural hearing loss, 15.

Recurrent meningitis in children due to inner ear abnormalities.

Recurrent meningitis in children is not only a potentially life threatening condition, but often involves the child in the trauma of repeated hospital admissions and multiple and invasive investigations to try and find an underlying cause. Symptoms and signs of CSF rhinorrhoea or otorrhoea are infrequent in these patients. Unilateral deafness may be difficult to diagnose in the young child. Full ENT examination may be normal. We report seven cases of children with recurrent meningitis in whom inner ear abnormalities were only indicated as the site of entry of infection by hypocycloidal tomogra

Experimental cytomegalovirus infection: viremic spread to the inner ear.

The entry route of viruses into the inner ear is still controversial. Guinea pig cytomegalovirus (CMV) has been used to create an animal model of systemic infection as well as labyrinthitis. In this study, seronegative guinea pigs were administered intracardiac inoculations of varying doses of guinea pig CMV. After three or eight days, the animals were killed and necropsy specimens examined for viral antigen. In the specimens examined three days after inoculation, no label was seen in the cochlea. However, label was found in the spleen with doses of 100 microL, and also in the liver in animals

Contralateral hearing loss following inner ear injury: sympathetic cochleolabyrinthitis?

Acoustic tumor surgery provided an ideal model in which to study possible contralateral hearing loss following a destructive surgical procedure on the inner ear. Follow-up audiometric studies were performed on patients with unilateral acoustic tumors who had undergone resection of their tumors. Patients who had obvious causes for contralateral hearing loss, such as chronic otitis media, unrelated otologic surgery, and noise-induced hearing loss, were excluded from this patient population. A total of 380 patients had available pre- and postoperative audiograms. A mean air-conduction threshold (

[Inner ear and diabetes mellitus studies on 660 patients].

The purpose of this study was to examine the existence of any hearing loss typical of diabetes mellitus. 660 patients were interviewed by means of a questionnaire with regard to case history and the course of their disease. Internal and ophthalmological findings were registered which were compiled during clinical treatment for metabolic stabilisation. ENT examination was performed including vestibular and auditory tests. The evaluation of data showed that there was no median hearing loss which could be attributed to the diabetic metabolic disorder. The hypothesis that diabetes causes premature

Inner ear disease characterized by rolling in C3H mice.

A neurologic disease characterized by rolling developed in C3H mice. Pseudomonas aeruginosa was isolated from the middle and internal ears or meninges of affected mice. The principal pathologic finding was aggressive, primary, purulent otitis media, with extension to the inner ear, acoustic nerve, and meninges. Stresses that may have contributed to induction of the disease were not delineated; however, acidification of the drinking water resulted in near elimination of the disease.

Fetal respiratory distress causing CNS and inner ear hemorrhage.

Fifty-two consecutive temporal bones of infants who died neonatally, or in utero of natural causes, were studied. Complete autopsies were performed. Twenty-eight infants had a variety of pulmonary disorders which resulted in severe respiratory distress prior to their death. Of these, the majority had bleeding intracranially and into the inner ear. There were five major pathways of central nervous system and subarachnoid hemorrhage involvement of the inner ear: 1) the modiolus, 2) cochlear aqueduct, 3) retrograde via the cochlear vein, 4) episodes of spontaneous bleeding into various compartmen

Experimental alteration of physiological state of inner ear fluids.

Mechanisms of endolymphatic hydrops formation in the inner ear are not yet fully understood. The available knowledge of the composition and the function of inner ear fluids, of mechanisms for maintenance of microhomeostasis, and the possible factors responsible for hydrops formation were reviewed. The nature of the functional barrier systems of the inner ear was investigated by measuring the amount of intravenously or intracisternally injected test substances which entered the perilymph. Based on studies of the osmotic relationship between the blood and the perilymph, an hypothesis of hydrops

Protocol for canalostomy in rats to enable inner ear injection.

Here, we present a protocol for precise inner ear access in rats using a refined canalostomy technique. We describe steps for surgical exposure of the mastoid bone, localization of the target semicircular canal, fenestration using a cranial drill, and quantitative microinjection of agents. We further validate this approach by presenting fluorescence-based assessment results. This standardized approach enables reproducible delivery of chemicals or viruses into the inner ear.

Schwann cells in the inner ear: development, disease, and regeneration.

Schwann cells are classically known as the constituent supporting cells of the peripheral nervous system. Beyond the scope of merely myelinating axons of the more saliently known neurons, Schwann cells comprise the majority of peripheral nervous system tissue. Through the lens of the inner ear, additional properties of Schwann cells are becoming elucidated. Therein, the process of myelin formation in development is more aptly understood as a homeostatic oscillation of differentiation status. Perpetual interaction between neural and non-neural cells of the inner ear maintains an intricate balan

The Role of Pericytes in Inner Ear Disorders: A Comprehensive Review.

Inner ear disorders, including sensorineural hearing loss, Meniere's disease, and vestibular neuritis, are prevalent conditions that significantly impact the quality of life. Despite their high incidence, the underlying pathophysiology of these disorders remains elusive, and current treatment options are often inadequate. Emerging evidence suggests that pericytes, a type of vascular mural cell specialized to maintain the integrity and function of the microvasculature, may play a crucial role in the development and progression of inner ear disorders. The pericytes are present in the microvascul

[Rehabilitation with cochlear implants in children with malformations of the inner ear].

Cochlear implants (CIs) are the treatment of choice for hearing rehabilitation in children with congenital or acquired profound hearing loss or deafness in order to ensure appropriate speech development and avoid social deprivation. However, in the case of a radiologically detectable malformation of the inner ear structures and potentially associated hypo- or aplasia of the vestibulocochlear nerve, application of a CI is either not possible, or the functional outcome may be of limited predictability. In addition, the risk of surgical complications is also increased in these patients. Counselin
