Changes in leucocyte cytochrome oxidase activity associated with deficiency of copper in laboratory and farm animals.

The cytochrome oxidase activity of circulating leucocytes was investigated by means of a semiquantitative method using blood films. A significant decrease in activity was found in male rats, cattle and sheep that had been deprived of copper. During copper depletion, leucocyte cytochrome oxidase activity declined more slowly than did plasma copper concentration and plasma ferroxidase I activity, and so was less sensitive as a guide to the copper status of the animal.

Binding of ligands and spectral shifts in cytochrome c oxidase.

1. On addition of reductant (ascorbate plus NNN'N'-tetramethyl-p-phenylenediamine) to isolated cytochrome c oxidase (ox heart cytochrome aa(3)), in the presence of the inhibitors azide or cyanide, an initial partially reduced species is formed with absorption peaks at 415nm, 445nm and 605nm, which slowly gives rise to the final ;half-reduced' species in whose spectrum the 415nm peak has disappeared and a new absorption is seen at 430-435nm. 2. In the absence of reductant, cyanide forms an initial complex with the enzyme with a spectrum similar to that of the uncombined form, which slowly chang

Cytochrome oxidase repair during treatment of copper deficiency: relation to mitochondrial turnover.

The repair of cytochrome oxidase depletion during the treatment of copper deficiency was studied in the rat. The purpose of this study was to distinguish the role of new cell production from the possibly more specific role of mitochondrial turnover in determining the rate of this repair. In rats on a copper-deficient regimen until 2.5-3 months of age, activities of cytochrome oxidase expressed as per cent of control were as follows: skeletal muscle (quadratus lumborum), 18%; heart, 27%; liver, 34%; and intestinal mucosa, 34%. After 2-3 days of dietary supplementation with cupric acetate, repai

SQR mediates therapeutic effects of H2S by targeting mitochondrial electron transport to induce mitochondrial uncoupling.

Hydrogen sulfide (H2S) is a gasotransmitter and a potential therapeutic agent. However, molecular targets relevant to its therapeutic actions remain enigmatic. Sulfide-quinone oxidoreductase (SQR) irreversibly oxidizes H2S. Therefore, SQR is assumed to inhibit H2S signaling. We now report that SQR-mediated oxidation of H2S drives reverse electron transport (RET) at mitochondrial complex I, which, in turn, repurposes mitochondrial function to superoxide production. Unexpectedly, complex I RET, a process dependent on high mitochondrial membrane potential, induces superoxide-dependent mitochondri

First molecular confirmation of Spirometra mansoni (Cestoda: Diphyllobothriidea) in Indonesian cobras (Naja sputatrix and Naja sumatrana).

BACKGROUND: Cobras (Naja spp.) are widely consumed and traded in Southeast Asia, particularly in Indonesia, yet their role in the transmission of zoonotic cestodes of the genus Spirometra remains unresolved at the species level. Accurate molecular identification is essential to clarify host associations, population connectivity, and potential public health implications. METHODS: Plerocercoids were collected from Naja sputatrix (East Java) and Naja sumatrana (West Sumatra). Specimens were characterised morphologically and subjected to mitochondrial cytochrome c oxidase subunit I (cox1) sequenc

Larval parasite occurrence in two killifish species, Argolebias guarani and Argolebias adrianae (Cyprinodontiformes: Rivulidae) inhabiting two ephemeral ponds.

Ephemeral ponds provide unique opportunities to study host-parasite interactions under contrasting environmental conditions. We assessed the occurrence of larval parasites in two closely related rivulid fishes, Argolebias guarani and Argolebias adrianae, inhabiting geographically proximate but hydrologically isolated ponds. A total of 121 individuals of A. guarani and 30 of A. adrianae were examined. Parasites were recorded only in A. guarani, including a nematomorph larva, a proteocephalidean metacestode, a telorchiid metacercaria, and a diplostomid metacercaria, whereas A. adrianae showed no

Aedes (Ochlerotatus) amateuri Ortega and Zavortink (Diptera: Culicidae) in the Madrean Sky Islands of Arizona, USA: a new country record.

The mosquito community of the Madrean Sky Islands of southeastern Arizona is diverse and distinct with several species that are endemic or at their northern distributional limits. We collected mosquitoes across many of southeastern Arizona's Sky Island mountain ranges. Here, we present new collection records supported by morphological and molecular evidence that indicate the presence of Aedes (Ochlerotatus) amateuri in at least two Sky Islands ranges, the Santa Rita Mountains and the Dragoon Mountains. Aedes amateuri was described in 2019, previously known only from the type locality in Nuevo

Geographic distribution and environmental associations of Psorophora mexicana (Diptera: Culicidae) in the Yucatan Peninsula, Mexico.

Psorophora mexicana (Bellardi) is a rare mosquito species with limited information on its taxonomy, geographic distribution, and ecological preferences. In Mexico, presence records are scarce and largely outdated, particularly in the Yucatan Peninsula. Here, we document the current distribution of Ps. mexicana based on collections conducted between 2017 and 2025 across 14 localities in Campeche, Quintana Roo, and Yucatan states, including the first confirmed record for Quintana Roo. Specimens were identified morphologically and genetically using the mitochondrial region cytochrome c oxidase I

Molecular detection of filarial parasites in black flies (Diptera: Simuliidae) in Laos.

Filarial parasites cause diseases in humans and other animals and are transmitted primarily by hematophagous Diptera, including black flies (Simuliidae). However, knowledge of the vectorial role of black flies in transmitting filarial parasites in tropical Asia is limited. We used molecular approaches to detect filarial parasites in two common black flies, Simulium asakoae and S. khelangense, in Laos. Among 483 black flies, 15 were infected with filarial parasites. Based on sequence similarity analyses of the COI and 12S rDNA genes, the parasites were closely related to two filarial taxa: one

Anopheles species complexes from Guyana and Venezuela malaria endemic areas using COI sequences.

BACKGROUND: Effective interventions for controlling Anopheles malaria vectors depend on accurately identifying the species. Venezuela has the highest malaria prevalence in the region of the Americas; however, there is very limited knowledge about the vectors, especially regarding species complexes and their distribution. OBJECTIVES: To conduct molecular taxonomy and phylogenetic analysis of the Albitarsis, Nuneztovari, and Oswaldoi complexes from malaria-endemic regions of Guyana and Venezuela using mtCOI sequences. METHODS: Anopheles were collected in Guyana (Potaro-Siparuni Region) and Ven

Cryptic diversity and recent diversification in the Pyrrhulina australis species complex (Characiformes, Lebiasinidae) revealed by mitochondrial DNA.

Pyrrhulina australis Eigenmann & Kennedy, 1903, is widely distributed across the main hydrographic basins of South America. The taxonomic integrity of this species is nevertheless challenged by the lack of clear diagnostic characters, its marked geographic variation and the morphological overlap with its congeners. In this context, the present study investigated the genetic diversity of the P. australis group (P. australis, Pyrrhulina marilynae and Pyrrhulina vittata), using sequences of the mitochondrial Cytochrome Oxidase I gene to delimit evolutionary lineages and provide insights into biog

Fall-like acclimation alters mitochondrial function and substrate use in freeze-tolerant crickets.

Acclimation to low temperatures in insects involves complex metabolic adjustments to ensure survival. Notably, cryoprotectants including glycerol, trehalose and proline can accumulate in insects to promote freeze tolerance. Interestingly, these metabolites can also be converted to metabolic fuels sustaining mitochondrial ATP production. This study investigates the metabolic shifts in the freeze-tolerant cricket Gryllus veletis during fall-like acclimation, focusing on mitochondrial function and its possible link with cryoprotectant accumulation. We hypothesized that fall-like acclimation promo

Review of the Progamotaenia macropodis (Cestoda: Anoplocephalidae) species complex from Australian macropodid marsupials, with description of P. mollicula sp. nov.

Genetic evidence indicates that Progamotaenia macropodis Beveridge, 1976 (Cestoda: Anoplocephalidae), found in various macropodid host species, is a complex of cryptic species. However, the genetic data are incomplete, and no morphological re-appraisal of the species has been undertaken since its original description. Here, additional mitochondrial cytochrome c oxidase I sequence data have been added from the type host, Macropus giganteus, as well as other host species, and a morphological study of all available material undertaken. A new species, Progamotaenia mollicula sp. nov., is erected f

Reports of Microthoracius mazzai (Phthiraptera: Anoplura) infestations in alpaca (Vicugna pacos) herds from Austria and Germany.

Sucking lice (Phthiraptera: Anoplura) constitute obligate and permanent insect ectoparasites of mammals and exhibit a hematophagous lifestyle. In South American camelids, three species of the genus Microthoracius have been described, however, reports outside South America are scarce. Here we describe infestations with Microthoracius mazzai in three alpaca (Vicugna pacos) herds from Austria (Styria and Salzburg), and Germany (Bavaria). Overall, lice infestations were detected in eight animals. Four of them exhibited a massive generalized infestation. Lice specimens were identified as M. mazzai
