Neddylation E2 enzyme UBE2M inhibits inflammatory lung tumor microenvironment by blocking the cGAS-STING signal upon KrasG12D activation.

UBE2M, also known as UBC12, is one of two neddylation conjugation enzymes. The in vitro cell culture work showed that UBE2M is essential for the growth and survival of cancer cells, but its in vivo role in tumorigenesis, particularly in the tumor microenvironment (TME), remains elusive. Here, we report that in a well-established KrasG12D lung tumor mouse model, Ube2m deletion caused an inflammatory TME, severe lung edema, and early death. Mechanistically, while KrasG12D activates MAPK and mTORC1 signals for proliferation, Ube2m deletion increases mitochondrial MOMP/mPTP and inhibits Parkin ned

Activation of pyruvate kinase as therapeutic option for rare hemolytic anemias: Shedding new light on an old enzyme.

Novel developments in therapies for various hereditary hemolytic anemias reflect the pivotal role of pyruvate kinase (PK), a key enzyme of glycolysis, in red blood cell (RBC) health. Without PK catalyzing one of the final steps of the Embden-Meyerhof pathway, there is no net yield of adenosine triphosphate (ATP) during glycolysis, the sole source of energy production required for proper RBC function and survival. In hereditary hemolytic anemias, RBC health is compromised and therefore lifespan is shortened. Although our knowledge on glycolysis in general and PK function in particular is solid,

Lipid peroxidation and phospholipase A2 activity in liposomes composed of unsaturated phospholipids: a structural basis for enzyme activation.

The effect of lipid peroxidation on membrane structure and phospholipase A2 activity was studied using liposomes composed of bovine liver phosphatidylcholine (PC) and phosphatidylethanolamine (PE). The phospholipids were mixed at set ratios and sonicated to yield small unilamellar vesicles. The liposome preparations were subjected to lipid peroxidation as induced by cumene hydroperoxide and hematin. Under these conditions, a sharp increase in lipid peroxidation was noted over a 30 min incubation period and was accompanied by loss of polyunsaturated fatty acids (PUFA). Liposomes enriched in PE

Effects of dietary zinc deficiency on the activity of enzymes associated with phase I and II of drug metabolism in Fischer-344 rats: activities of drug metabolising enzymes in zinc deficiency.

A study was undertaken to assay the various phase I and phase II drug metabolising enzymes in zinc deficiency. Male weanling Fischer rats were subjected to zinc deficiency for a period of 7 weeks. Zinc levels in the control and deficient diets were 30 mg and 1.1 mg/kg diet, respectively. At the end of the experimental period, the activities of various hepatic cytosolic and microsomal enzymes were estimated. It was observed that the activities of microsomal epoxide hydrolase (with benz(a)pyrene 4-5 oxide as substrate), uridine diphospho glucuronyl transferase (with 1-naphthol as substrate) and

MnO2@CeOx-GAMP radiosensitizer with oxygen vacancies depended mimicking enzyme-like activities for radiosensitization-mediated STING pathway activation.

Activation of the stimulator of interferon genes (STING) pathway by radiotherapy (RT) has a significant effect on eliciting antitumor immune responses. The generation of hydroxyl radical (·OH) storm and the sensitization of STING-relative catalytic reactions could improve radiosensitization-mediated STING activation. Herein, multi-functional radiosensitizer with oxygen vacancies depended mimicking enzyme-like activities was fabricated to produce more dsDNA which benefits intracellular 2', 3'-cyclic GMP-AMP (cGAMP) generation, together with introducing exogenous cGAMP to activate immune respons

Iron Heme Enzyme-Catalyzed Cyclopropanations with Diazirines as Carbene Precursors: Computational Explorations of Diazirine Activation and Cyclopropanation Mechanism.

The mechanism of cyclopropanations with diazirines as air-stable and user-friendly alternatives to commonly employed diazo compounds within iron heme enzyme-catalyzed carbene transfer reactions has been studied by means of density functional theory (DFT) calculations of model systems, quantum mechanics/molecular mechanics (QM/MM) calculations, and molecular dynamics (MD) simulations of the iron carbene and the cyclopropanation transition state in the enzyme active site. The reaction is initiated by a direct diazirine-diazo isomerization occurring in the active site of the enzyme. In contrast,

The mitochondrial amidoxime reducing component-from prodrug-activation mechanism to drug-metabolizing enzyme and onward to drug target.

The mitochondrial amidoxime-reducing component (mARC) is one of five known molybdenum enzymes in eukaryotes. mARC belongs to the MOSC domain superfamily, a large group of so far poorly studied molybdoenzymes. mARC was initially discovered as the enzyme activating N-hydroxylated prodrugs of basic amidines but has since been shown to also reduce a variety of other N-oxygenated compounds, for example, toxic nucleobase analogs. Under certain circumstances, mARC might also be involved in reductive nitric oxide synthesis through reduction of nitrite. Recently, mARC enzymes have received a lot of att

Novel Photo-Driven Activated Enzyme-Titanium Nanobiohybrids for Photocatalytic Applications.

This work reports the development of innovative enzyme-titanium nanobiohybrids synthesized via a protein-assisted approach to obtain efficient and sustainable photocatalysts for environmental remediation. By addressing the limitations of conventional TiO2 nanoparticle synthesis, this strategy enables controlled material properties under milder, potentially scalable conditions for enhanced ROS-driven degradation of persistent dye pollutants. This work employs a bio-assisted synthesis approach using β-glucosidase as a protein scaffold, TiCl4 as the titanium precursor, and H2O2 in bicarbonate buf

Multidomain annotation of carbohydrate-active enzymes beyond CAZy domains with GeneHunt2.

Carbohydrate-active enzymes (CAZymes) are central to carbohydrate metabolism, yet their functional annotation is typically restricted to catalytic CAZy domains, overlooking the broader multidomain architectures in which these domains operate. Here, I present GeneHunt2, a scalable framework for multidomain annotation of CAZymes that integrates curated HMM profiles from dbCAN and Pfam into a unified, deduplicated database, enabling systematic identification of both CAZy and non-CAZy domains. After confirming the robust recovery of CAZy domain assignments using GeneHunt2, I investigated the detai

Structural determinants for FAT10 activation and transfer from UBA6 to E2 enzymes.

Attachment of the ubiquitin-like protein (UBL) FAT10 onto substrates targets them for proteasomal degradation. Like ubiquitin, FAT10 is activated by the E1 enzyme UBA6 then transferred to E2 enzymes, but mechanisms controlling ubiquitin versus FAT10 activation by UBA6 and FAT10 transfer onto E2s remain unclear. Using cryo-EM, we visualise all stages of FAT10 E1-E2 handover: adenylation, thiolation and transthiolation. We find that FAT10 monopolises UBA6 by out-competing ubiquitin for thiolation and blocking the adenylation domain, preventing further UBL recruitment and promoting FAT10 signalli

Skin friendly anti-photoaging semiconductor-nanozymes with dual enzyme activities.

Skin photoaging, predominantly induced by UV light exposure, is a significant external factor to the aging process of the skin. Current anti-photoaging strategies primarily utilize semiconductor materials (e.g. TiO2, ZnO) as UV light absorbers. However, these materials also generate reactive oxygen species (ROS) during UV absorption, potentially causing oxidative damage to skin cells. The development of UV absorption semiconductors with limited ROS generation is highly desired. Herein, inspired by the enzyme-mediated ROS clearance in living organisms, we propose to employ antioxidant nanozyme

pH Controlled Activation and Stabilization of Enzymes Using Responsive Polymer-Bioconjugates.

Stimuli-responsive bioconjugates are developed to control enzymatic activity and stability using a pH-responsive polymer based on N,N-dimethyl aminopropylacrylamide. Prior work has shown that conjugating N-isopropylacrylamide (NIPAm) polymer to theCandida antarctica lipase B (CalB) enzyme has an inhibitory effect. Due to the similar hydrophobicity to the substrate of CalB, NIPAm polymer may block the enzyme's catalytic site, thereby inhibiting the enzyme's activity. This study shows that conjugating pDMAPA DP40-b-NIPAm DP10 to CalB has a pH-responsive effect on the catalytic property of CalB,

Spectroscopic definition of ferrous active sites in non-heme iron enzymes.

Non-heme iron enzymes play key roles in antibiotic, neurotransmitter, and natural product biosynthesis, DNA repair, hypoxia regulation, and disease states. These enzymes had been refractory to traditional bioinorganic spectroscopic methods. Thus, we developed variable-temperature variable-field magnetic circular dichroism (VTVH MCD) spectroscopy to experimentally define the excited and ground ligand field states of non-heme ferrous enzymes (Solomon et al., 1995). This method provides detailed geometric and electronic structure insight and thus enables a molecular level understanding of catalyt

Engineering C-C Bond Cleavage Activity into a P450 Monooxygenase Enzyme.

The cytochrome P450 (CYP) superfamily of heme monooxygenases has demonstrated ability to facilitate hydroxylation, desaturation, sulfoxidation, epoxidation, heteroatom dealkylation, and carbon-carbon bond formation and cleavage (lyase) reactions. Seeking to study the carbon-carbon cleavage reaction of α-hydroxy ketones in mechanistic detail using a microbial P450, we synthesized α-hydroxy ketone probes based on the physiological substrate for a well-characterized benzoic acid metabolizing P450, CYP199A4. After observing low activity with wild-type CYP199A4, subsequent assays with an F182L muta

Regulatory role of acetylation on enzyme activity and fluxes of energy metabolism pathways.

BACKGROUND: Most of the enzymes involved in the central carbon metabolism are acetylated in Lys residues. It has been claimed that this covalent modification represents a novel regulatory mechanism by which both enzyme/transporter activities and pathway fluxes can be modulated. METHODS: To establish which enzymes are regulated by acetylation, a systematic experimental analysis of activities and acetylation profile for several energy metabolism enzymes and pathway fluxes was undertaken in cells and mitochondria. RESULTS: The majority of the glycolytic and neighbor enzymes as well as mitochond

Interaction of artemisinin protects the activity of antioxidant enzyme catalase: A biophysical study.

The major antioxidant enzyme catalase is downregulated and the enzyme activity is compromised in various disease conditions such as malarial and cancer. Hence, the restoration and protection of catalase is a promising therapeutic strategy in disease management. In the present study, for the first time we have demonstrated the protective role of well-known anti-malarial drug Artemisinin (ART) on the time and temperature-induced degradation of bovine liver catalase (BLC) activity. The findings at different time intervals and at higher temperature showed the protective role of ART on BLC activity
