Sickle hemoglobin aggregation: a new class of inhibitors.

A number of tri- and tetrapeptides have been found to inhibit aggregation and gelation of deoxygenated sickle cell hemoglobin. These inhibitors have hydrophobic phenylalanine residues at one end and hydrogen bonding lysine or arginine side chains at the other end. The backbone is not very specific. The inhibitors do not modify the oxygen carrying properties of hemoglobin. When the inhibitor and sickle hemoglobin are put inside reconstituted cells, the erythrocytes do not sickle upon deoxygenation. Compounds of this type may develop useful agents in the therapy of sickle cell anemia.

Immunohistological analysis of intravascular agglutination in a case of ABO incompatible blood transfusion by means of mixed cell agglutination reaction.

Applicability of the mixed cell agglutination reaction (MCAR of Davidsohn) in histological examination was tested in a case of ABO incompatible blood transfusion, who had died of severe head injury two days after an accident. Massively disseminated intravascular aggregation of erythrocytes was identified as immune agglutination by the modified method of MCAR (ISHIYAMA & OKADA) with precise knowledge of topographic isoantigen localization in tissues. The immune specific agglutination mimics intravascular changes such as hyperemia, stagnation and hemorrhage derived from various circulatory distu

Hemorheological abnormalities in stable angina and acute coronary syndromes.

The pathophysiological abnormalities of stable angina (SA) and acute coronary syndromes (ACS) may, in part, be promoted by fluid forces associated with local blood flow and hence by the rheological properties of blood. This study evaluated several hemorheological parameters in 16 healthy controls and in 16 SA, 18 unstable angina (UA) and 19 acute myocardial infarct (AMI) patients; all patients underwent diagnostic angiography following blood sampling. Rheological measurements included whole blood viscosity, plasma viscosity and RBC aggregation via erythrocyte sedimentation rate (ESR) and Myren

Increased blood viscosity in iron-depleted elite athletes.

Since iron deficiency is associated with abnormal erythrocyte rheology, we investigated relationships between plasma ferritin and blood rheology in 36 male elite sportsmen (age: 22.38+/-0.9 years). On the whole, ferritin was negatively correlated with blood viscosity (r = -0.36, p < 0.05). When 23 subjects with low ferritin levels suggesting mild iron deficiency were compared with 13 matched sportsmen with normal ferritin levels, iron-deficient sportsmen were shown to have a higher blood viscosity at 1000 s(-l) (3.17+/-0.09 vs. 2.85+/-0.06 mPas, p < 0.05), explained by a higher plasma viscosit

[Rheologic particulars of venous flow. Physiopathologic consequences].

Hemorheological parameters must be taken into account for a better knowledge of venous flow properties. The postcapillary venules are the region of lowest shear rates and therefore the region of highest whole blood viscosity. Red cell aggregation plays a major role in blood viscosity, especially at low shear rate. This microrheological parameter can be increased in pathological circumstances, including a low capillary flow, venular insufficiency, elevated hematocrit, high levels of acute phase proteins. At the level of the capillaries, microcirculatory stasis leads to a low oxygen supply and c

Activation of the alternative pathway of complement by human serum IgA.

In order to study the activation of complement by soluble aggregates of human polyclonal serum IgA, lysis of sheep erythrocytes (E) coated with several IgA preparations was used as a model. A complement nonactivating monoclonal mouse IgG1 against IgA was used to coat the cells. IgA, isolated from normal human serum, was aggregated by either N-succinimidyl 3-(2-pyridyldithio)propionate (SPDP), glutaraldehyde, carbodiimide or heating. Depending on the size of the aggregates, and on the method of aggregation, E coated with aggregated IgA (E gamma 1.AIgA) could be lysed. The alternative pathway of

[Immunocytochemical identification of AB0 incompatible erythrocytes following fatal transfusion reaction].

A patient with blood group O died 8 h after an accidental transfusion of one unit of A1 erythrocytes. The admixture of group A red cells was not detected during postmortem serology. Paraffin-embedded autopsy material was studied by the indirect immunoperoxidase technique using monoclonal antibodies. Group A red cells were easily identified as circulating agglutinates in the larger vessels, in the capillary vessels of all organs examined, and as closely packed cell aggregations in the sinuses of the spleen. However, there was no clear evidence of erythrophagocytosis in the spleen or in liver.

Alpha-synuclein is increased in erythrocytes in parkinson's disease cases.

Idiopathic Parkinson's disease (iPD) is the second most common neurodegenerative disease after Alzheimer's disease (AD). Mutations in the SCNA gene, which encodes the protein alpha synuclein (α-syn), are associated with familial forms of Parkinson's disease (PD). Additionally, Lewy bodies (LBs) rich in α-synuclein are a hallmark of idiopathic Parkinson's disease (iPD) pathology. Unlike AD, there are no effective blood-based diagnostic assays for iPD. Recent studies show that measures of misfolded α-syn in cerebrospinal fluid (CSF) and skin biopsies reflect the diagnosis of iPD. The presence of

Red blood cell aggregation, aggregate strength and oxygen transport potential of blood are abnormal in both homozygous sickle cell anemia and sickle-hemoglobin C disease.

BACKGROUND: Recent evidence suggests that red blood cell aggregation and the ratio of hematocrit to blood viscosity (HVR), an index of the oxygen transport potential of blood, might considerably modulate blood flow dynamics in the microcirculation. It thus seems likely that these factors could play a role in sickle cell disease. DESIGN AND METHODS: We compared red blood cell aggregation characteristics, blood viscosity and HVR at different shear rates between sickle cell anemia and sickle cell hemoglobin C disease (SCC) patients, sickle cell trait carriers (AS) and control individuals (AA).

Digital ischemia and gangrene due to red blood cell aggregation induced by acquired dysfibrinogenemia.

Digital gangrene was observed in a patient who had angiographic findings of digital arterial occlusion. The patient's blood showed a marked red blood cell aggregation with rouleaux formation in long chains, which could not be dispersed at shear rates up to 200 sec-1. Studies of the patient's blood revealed the presence of an abnormal fibrinogen capable of aggregating normal red blood cells. This fibrinogen was found by Raman spectroscopy to have an increased alpha-helical content, whereas the beta-sheet content was decreased. Defibrinogenation therapy with ancrod resulted in a dramatic symptom

Purification and some properties of hemagglutinating protein mutina from bushmaster Lachesis muta snake venom.

Lachesis muta snake venom induced aggregation of bromelain sensitized human erythrocytes at a concentration of 1 mg/ml. The hemagglutinating protein was purified by DEAE-Sephadex A-50 column chromatography. Polyacrylamide gel electrophoresis revealed at least three bands, whereas SDS electrophoresis in the presence of 2-mercaptoethanol showed a single one. Isoelectric focusing revealed hemagglutinating activity in the range of pH 3-8. The maximum peak (mutina) at pH 5.5. This fraction was active in agglutinating human RBC of types A, B, O Rh (+) and B, O Rh (-). One mM EDTA and 1 mM Ca++ did n

Pharmacobiological management of hemostasis within clinical backgrounds via Ankaferd hemostat (Ankaferd blood stopper).

Ankaferd hemostat (Ankaferd blood stopper [ABS], Istanbul, Turkey) is a hemostatic agent affecting red blood cell-fibrinogen interactions. ABS has been traditionally used in Anatolia as a hemostatic agent for centuries. ABS contains a standardized combination of the plants namely Glycyrrhiza glabra, Thymus vulgaris, Alpinia officinarum, Vitis vinifera, and Urtica dioica. The hemostatic effect of ABS depends upon the quick promotion of a protein network, particularly fibrinogen gamma, in relation to the erythrocyte aggregation. The aim of this review is to indicate pharmacobiological basis and …

[Clinical implications of impaired microcirculation and hemodynamics in acute respiratory viral infections and their pharmacological correction].

Microcirculation, hemocoagulation and blood viscosity were studied in 377 of 1033 inpatients with influenza and other acute respiratory viral infections. The new data show that it is feasible to improve the course, outcomes and results of the treatment. Microcirculation disorders persist throughout the disease, but in convalescence, intravascular alterations are leading. The microcirculatory changes manifest themselves with advanced erythrocyte aggregation, activation of vascular-platelet and plasmic links of hemostasis, and high blood viscosity at low shift velocities. In the presence of conc…

Evaluation of anionic polymer-coated siRNA lipoplexes prepared using a modified ethanol injection method.

Cationic liposomes are useful carriers for delivering small interfering RNA (siRNA). In our previous study, a simple and efficient modified ethanol injection (MEI) method was developed for the preparation of cationic siRNA lipoplexes. However, non-specific interactions between cationic lipoplexes and biological components, including erythrocytes, induce aggregation, and thus, siRNA is not delivered to the target site after intravenous injection. In the present study, an anionic polymer coating was applied to cationic lipoplexes prepared using the MEI method, and their gene knockdown effect and

Hemorheological background of acetylsalicylic acid resistance.

BACKGROUND: Pathologic hemorheological parameters and increased platelet aggregation in association with other risk factors significantly increase the possibility of the development of ischemia. Acetylsalicylic acid (ASA) is an effective antithrombotic agent, which prevents a variety of cardiovascular diseases. OBJECTIVES: The aim of our present study was to compare the hemorheological parameters of patients with effective platelet inhibition by ASA to those with ineffective one. METHODS: 2045 patients taking 100 mg ASA daily were involved in our study (1255 males, mean age: 63+/-11 yrs, 790

Neuronal hemoglobin affects dopaminergic cells' response to stress.

Hemoglobin (Hb) is the major protein in erythrocytes and carries oxygen (O2) throughout the body. Recently, Hb has been found synthesized in atypical sites, including the brain. Hb is highly expressed in A9 dopaminergic (DA) neurons of the substantia nigra (SN), whose selective degeneration leads to Parkinson's disease (PD). Here we show that Hb confers DA cells' susceptibility to 1-methyl-4-phenylpyridinium (MPP+) and rotenone, neurochemical cellular models of PD. The toxic property of Hb does not depend on O2 binding and is associated with insoluble aggregate formation in the nucleolus. Neur

Effects of plasma donation on blood rheology and peripheral vascular function.

Plasma donation, the removal of plasma and reinfusion of cellular components, is widely used to produce plasma-derived products; however its physiological effects on blood rheology and peripheral vascular function remain poorly understood. This study investigated the effects of plasma donation (Investigation 1) on blood viscosity, red blood cell (RBC) deformability and aggregation, and brachial artery reactive hyperaemia flow-mediated dilatation (RH-FMD), and compared these responses with those induced by frusemide-mediated haemoconcentration (40 mg; Investigation 2) to isolate the independent
