BACKGROUND: The midbrain periaqueductal and periventricular gray (PAVG) region is important for pain and autonomic modulation. We have previously described changes in blood pressure dependent upon dorsal or ventral electrode positioning with PAVG deep brain stimulation (DBS), yet controversy exists about whether DBS acts via endogenous opioid release.
METHOD AND RESULTS: We combined local field potential (LFP) recording from PAVG DBS electrodes in humans with naloxone and saline infusions to determine whether dorsal and ventral PAVG DBS act through opioidergic or other mechanisms. Four awake

BACKGROUND: Bipolar disorder (BD) and major depressive disorder (MDD) have distinct pathophysiologies but similar depressive appearances. The present study aimed at the differentiation of the brain responses between BD and MDD patients. We hypothesized that different affective disorder patients may have distinct patterns of oscillatory cortical activities in response to negative emotional stimuli.
METHODS: Twenty BD patients, twenty MDD patients, and twenty age- and gender-matched healthy normal subjects were recruited. We adopted an implicit emotional task with facial image stimuli. The acqu

The human anterior prefrontal cortex (aPFC) is involved in regulating social-emotional behavior, presumably by modulating effective connectivity with downstream parietal, limbic, and motor cortices. Regulating that connectivity might rely on theta-band oscillations (4-8 Hz), a brain rhythm known to create overlapping periods of excitability between distant regions by temporally releasing neurons from inhibition. Here, we used MEG to understand how aPFC theta-band oscillations implement control over prepotent social-emotional behaviors; that is, the control over automatically elicited approach

The 40-Hz auditory steady-state response (ASSR) has been examined extensively because of its association with gamma-band oscillations and its potential as a biomarker for psychiatric disorders. However, most studies have focused particularly on neural activity at the driving frequency, with less focus on the temporal evolution of oscillations across other frequency bands during auditory stimulation. We aimed to elucidate the broadband oscillatory dynamics during 40-Hz click trains, focusing on stimulus onset and offset. Magnetoencephalography recordings were obtained from 30 healthy adults exp

Happiness is a subjective experience that can serve as the ultimate goal for humans. A recent study that employed resting-state functional magnetic resonance imaging (fMRI) reported that spontaneous fluctuation (fractional amplitude of low-frequency fluctuation: fALFF) in the precuneus is negatively associated with subjective happiness. However, little is known about the neural electrical correlates of subjective happiness, which can provide direct evidence of neural activity and insights regarding the underlying psychological, cellular, and neurotransmitter mechanisms. Therefore, we measured

Cognitive biotype in depression has long been associated with abnormalities in neural oscillations. Among them, gamma oscillations are widely observed correlates of cognitive dysfunction. However, whether gamma oscillations implement causal mechanisms of specific brain function in cognitive biotype of depression remains unclear. Depressed patients in remission were included in this study. Measurement and Treatment Research to Improve Cognition in Schizophrenia (MATRICS) Consensus Cognitive Battery (MCCB) was used to identify cognitive biotype. Here, we enrolled 141 individuals with stable depr

The present study sought to investigate how differences in trait empathy can influence emotional reactivity to a specific set of affective categories. Forty-one female students, divided in High (HE, n = 20) and Low (LE, n = 21) trait empathy, watched eight validated movie clips divided in four emotional categories (Erotic, Fear, Compassion and Neutral) while subjective evaluation of emotion and EEG gamma activity were recorded. Analysis of self-reports revealed that HE compared to LE exhibited an increased arousal level to all emotional clips. Concerning EEG data, the HE group showed a greater

This study examined the effects of stimulation targeting dorsolateral prefrontal cortex (DLPFC) on behavioral and neural oscillatory markers of proactive cognitive control in healthy adults. We hypothesized that active stimulation targeting the DLPFC would enhance proactive control compared to sham, leading to changes in the pattern of error rates and gamma-band power on the Dot Pattern Expectancy (DPX) task. We recorded EEG while participants completed the DPX, after receiving either 20 minutes of active DLPFC stimulation at 2 mA or sham stimulation in a counterbalanced within-participants de

BACKGROUND: The Default Mode Network (DMN) is severely compromised in several psychiatric and neurodegenerative disorders where plasticity alterations are observed. Glutamate and GABA are the major excitatory and inhibitory brain neurotransmitters respectively and are strongly related to plasticity responses and large-scale network expression.
OBJECTIVE: To investigate whether regional Glx (Glutamate + Glutamine) and GABA could be modulated within the DMN after experimentally-controlled induction of plasticity and to study the effect of intrinsic connectivity over brain responses to stimulati

BACKGROUND: Electroencephalography and magnetoencephalography (MEG) studies have identified alterations in gamma-band (30-80 Hz) cortical activity in schizophrenia and mood disorders, consistent with neural models of disturbed glutamate (and GABA) neuron influence over cortical pyramidal cells. Genetic evidence suggests specific deficits in GABA-A receptor function in schizoaffective bipolar disorder (SABP), a clinical syndrome with features of both bipolar disorder and schizophrenia. This study investigated gamma oscillations in this under-researched disorder.
METHOD: MEG was used to measure

Cooperation, productivity, and cohesion in human societies depend on altruism, the tendency to share resources with others even though this is costly. While altruism is a widely shared social norm, people vary strongly in their inclination to behave altruistically, in particular across situations with different types of inequality in resource distribution. What neurobiological factors underlie this variability? And can these be targeted by interventions to enhance altruistic behavior? Here, we build on electroencephalography (EEG) evidence that altruistic choices during disadvantageous inequal

Fast periodic visual stimulation (FPVS) combined with magnetoencephalography (MEG) provides a powerful approach for extracting frequency-specific brain responses from perceptual processing areas. However, its application in cognitive paradigms, particularly those addressing visuospatial attention, remains underexplored. This study developed an FPVS-based line bisection paradigm to probe visuospatial cognition during visual exploration of the line center using eye movements without manual responses. Line stimuli were spatially modulated at high-gamma frequencies (70-90 Hz), centered at 80 Hz. S

The Action Observation Network (AON) is a large-scale brain network that supports the perceptual encoding and recognition of actions performed by others. The identification of the nodes of the human AON has been clarified over the past 30 years thanks to the high spatial resolution of neuroimaging techniques. The temporal dynamics underpinning their activations is in contrast still unsettled, because of methodological constraints. Here we investigate the timing of the AON components by intracranially recording gamma-band oscillations from 23 drug-resistant epileptic patients during the observa

Growing evidence links amyloid accumulation to altered gamma activity, positioning gamma oscillations as a promising biomarker for Alzheimer's pathology. We examined resting state gamma activity in early-onset Alzheimer's disease (EOAD) patients confirmed by CSF biomarkers. We aim to provide a holistic view of gamma activity patterns in EOAD by integrating structural and functional neuroimaging. In this cross-sectional study, we compared gamma power and coherence-total gamma (30-48 Hz), and sub-bands (gamma-1: 30-35 Hz; gamma-2: 35-40 Hz; gamma-3: 40-48 Hz)- alongside gray matter volumes in EO

During spatial learning, subjects progressively adjust their navigation strategies as they acquire experience. The medial prefrontal cortex (mPFC) supports this operation, for which it may integrate information from distributed networks, such as the hippocampus (HPC) and the posterior parietal cortex (PPC). However, the mechanism underlying the prefrontal coordination with HPC and PPC during spatial learning is poorly understood. Here we show that during navigation trials, mice displayed two sequential behavioral stages: searching and exploration. Exclusively during searching, mice gradually i

Finely tuned gamma (FTG) oscillations from the subthalamic nucleus (STN) and cortex in Parkinson's disease (PD) patients undergoing deep brain stimulation (DBS) are often associated with dyskinesia. Recently it was shown that DBS entrains gamma activity at 1:2 of the stimulation frequency; however, the functional role of this signal is not yet fully understood. We recorded local field potentials from the STN in 19 chronically implanted PD patients on dopaminergic medication during DBS, at rest, and during repetitive movements. Here we show that high-frequency DBS induced 1:2 gamma entrainment

Deep brain stimulation (DBS) significantly alleviates symptoms in various neurological disorders. Current research focuses on developing programmed stimulation protocols for customization to individual symptoms. However, the therapeutic mechanism of action of programmed DBS (pDBS) is poorly understood. We previously demonstrated that pDBS in the ventral tegmental area (VTA) normalizes molecular and behavioral abnormalities in the Flinders Sensitive Line (FSL) rat model for depression. Herein, we examined the effect of a short-duration, low-frequency DBS template on local field potential (LFP)

Non-pharmacological treatment (NPT) improves cognitive functions and behavioural disturbances in patients with dementia, but the underlying neural mechanisms are unclear. In this observational study, 21 patients with dementia received NPTs for several months. Patients were scanned using magnetoencephalography twice during the NPT period to evaluate NPT effects on resting-state brain activity. Additionally, cognitive functions and behavioural disturbances were measured using the Mini-Mental State Examination (MMSE-J) and a short version of the Dementia Behaviour Disturbance Scale (DBD-13) at th

Hippocampal theta and gamma oscillations degenerate early in Alzheimer's disease (AD), and may be a critical pathogenic factor and therapeutic target for AD. Deep brain stimulation (DBS) improves abnormal theta and gamma oscillations in AD; however, how these oscillations dynamically change after stimulation remains unclear. Exploring the prolonged neuroregulatory effects of DBS is essential for optimizing parameters and treatment strategies. Therefore, we investigated the sustained changes in the theta and gamma oscillations of the hippocampal cornu ammonis 1 region induced by acute DBS of th

Theta-gamma transcranial alternating current stimulation (TG tACS) over primary motor cortex (M1) can improve ballistic motor performance in young adults, but the effect on older adults is unknown. This study investigated the effects of TG tACS on motor performance and M1 excitability in 18 young and 18 older adults. High-definition TG tACS (6 Hz theta, 75 Hz gamma) or sham tACS was applied over right M1 for 20 min during a ballistic left-thumb abduction motor training task performed in two experimental sessions. Motor performance was quantified as changes in movement acceleration during and u
