Causal associations between Sjögren's syndrome and subtypes of endometrial cancer.

BACKGROUND: Endometrial cancer (EC) is a heterogeneous gynecologic malignancy with distinct histological subtypes and diverse etiological factors. While autoimmune diseases have been associated with cancer development, the causal relationship between Sjögren’s syndrome (SS) and EC remains unclear. In this study, we employed a Mendelian randomization (MR) approach to investigate this complex association. METHODS: We conducted a two-sample MR study using genome-wide association study summary statistics from European ancestry cohorts. Genetic instruments for SS were derived from the FinnGen conso

Causal relationship between type I diabetes mellitus and atrial fibrillation: A Mendelian randomization study.

BACKGROUND: Patients with type 1 diabetes mellitus have been at heightened risk for developing atrial fibrillation. We aimed to investigate whether this association is causal using Mendelian randomization. METHODS: Using publicly available genome-wide association studies data, we selected single nucleotide polymorphisms significantly associated with type 1 diabetes mellitus as instrumental variables. We employed inverse variance-weighted, weighted median, MR-Egger regression, simple mode, and weighted mode methods within a two-sample Mendelian randomization framework to assess the causal rela

Mendelian randomization analysis provides insights into the relationship between inflammatory bowel disease and skin cancer.

Limited and conflicting data have been available regarding the association between inflammatory bowel disease and skin cancer. It was hypothesized that inflammatory bowel diseases [Crohn's disease (CD) and ulcerative colitis (UC)] harbor a genetically increased risk of skin cancer [skin cutaneous melanoma (SKCM), basal cell carcinoma (BCC) and squamous cell carcinoma (SCC)] and performed two-sample mendelian randomization (MR) analysis using genome-wide association (GWAS) studies of European ancestry retrieved from FinnGen R8. The inverse variance weighted method was used to approximate MR eff

AQP4-ab-Positive Neuromyelitis Optica Spectrum Disorder Increases the Risk of Hydrocephalus: A Bidirectional Mendelian Randomization Study.

INTRODUCTION: Some observational studies indicated that AQP4-ab-positive neuromyelitis optica spectrum disorder (NMOSD) may predispose to hydrocephalus. However, the causal relationship between NMOSD and hydrocephalus remains elusive. We used bidirectional Mendelian randomization (MR) to examine the causal effect of AQP4-ab-positive NMOSD and hydrocephalus. METHODS: The exposure GWAS data used in this study were obtained from the GWAS Catalog, which included 132 AQP4-ab-positive patients and 1244 normal controls. The outcome GWAS data for hydrocephalus (N_case = 2455, N_control = 382,198) wer

Evidence for causal effects of sleep disturbances on risk for osteoarthritis: a univariable and multivariable Mendelian randomization study.

OBJECTIVE: To disentangle whether sleep disturbances have a causal effect on the risk of osteoarthritis (OA) using genetically based approaches. METHOD: We performed univariable and multivariable Mendelian randomization (MR) analyses using publicly released genome-wide association studies summary statistics to estimate the causal associations of sleep disturbances with OA risk. The inverse-variance weighted (IVW) method was utilized as primary MR analysis, whereas complementary methods including weighted median, weighted mode, MR-Egger regression, and MR pleiotropy residual sum and outlier (M

Neurodevelopment as a shared genetic etiology linking sleep-related phenotypes and psychiatric disorders: a genome-wide pleiotropic analysis.

Comorbidity and mutual transformation between psychiatric disorders (including autism spectrum disorder, attention-deficit/hyperactivity disorder, bipolar disorder, post-traumatic stress disorder, major depressive disorder, obsessive-compulsive disorder, schizophrenia, and anxiety disorders) and sleep disorders are common, with circadian rhythm disruption considered a potential biological basis, and neurodevelopmental abnormalities seemingly playing a key role. However, the extent to which shared genetic determinants contribute to these associations remains unclear. Extensive genetic correlati

Uncovering pleiotropic loci linking keratoconus and allergic diseases through integrative genomic analyses.

Keratoconus (KC) is a progressive corneal disease increasingly linked to immune dysregulation. Epidemiological studies have suggested comorbidity between KC and allergic diseases such as atopic dermatitis (AD), allergic conjunctivitis (AC), allergic rhinitis (AR), and allergic asthma (AA), yet their shared genetic mechanisms remain unclear. To investigate the shared genetic architecture between KC and four common allergic conditions, we integrated genome-wide association study (GWAS) summary statistics using a multi-omics framework. We applied linkage disequilibrium score regression (LDSC) and

A Novel One-Sample Mendelian Randomization Approach for Count-Type Outcomes That Is Robust to Correlated and Uncorrelated Pleiotropic Effects.

We propose two novel one-sample Mendelian randomization (MR) approaches to causal inference from count-type health outcomes, tailored to both equidispersion and overdispersion conditions. Selecting valid single-nucleotide polymorphisms (SNPs) as instrumental variables (IVs) poses a key challenge for MR approaches, as it requires meeting the necessary IV assumptions. To bolster the proposed approaches by addressing violations of IV assumptions, we incorporate a process for removing invalid SNPs that violate the assumptions. In simulations, our proposed approaches demonstrate robustness to the v

Investigating The Bidirectional Relationship Between Low Back Pain, Hip Pain, Knee Pain, And Gait Abnormalities: A Mendelian Randomization Study.

Gait abnormalities are associated with altered lower-limb biomechanics and functional impairment in musculoskeletal disorders. Although low back pain, hip pain, and knee pain have been linked to impaired gait in observational studies, their causal relationships remain unclear. We conducted a bidirectional Mendelian randomization (MR) study to assess potential causal associations between site-specific pain and gait abnormalities. Independent single nucleotide polymorphisms from large-scale genome-wide association studies were used as genetic instruments for low back pain, hip pain, and knee pai

Causal relationships between oral-gut microbiome and bone neoplasm-related phenotypes: Insights from bidirectional Mendelian randomization.

The human oral and gut microbiota are the 4 largest microbial communities in the body and play crucial roles in maintaining homeostasis and influencing disease. Observational studies have suggested links between these microbiota and bone neoplasm-related phenotypes, but establishing causality has been challenging due to confounding factors and reverse causality. We conducted a bidirectional, 2-sample Mendelian randomization (MR) study to investigate evidence consistent with a potential causal association between the saliva and gut microbiota and various bone neoplasm-related phenotypes. Association between antibody-mediated immune responses and gastroesophageal reflux disease: Evidence from genetic studies.

Studies have proven an association between the specific antibody-mediated immune response and gastroesophageal reflux disease (GERD); however, the exact causal associations remain unclear. Our research aims to evaluate the causal relationships between the genetic susceptibility to 46 antibody-mediated immune responses and GERD, with the bidirectional two-sample Mendelian randomization (TSMR) and to explore the mediating effects of 91 circulating inflammatory cytokines by a two-step MR analysis. Multiple MR analysis methods, including inverse variance weighted, MR-Egger, weighted mode, weighted

Causal relationship between body mass index and risk of juvenile idiopathic arthritis: A 2-sample Mendelian randomization study.

The association between body mass index (BMI) and juvenile idiopathic arthritis (JIA) has been suggested, but the causal relationship remains unclear. Mendelian randomization (MR) offers a tool to address this causal question using genetic data. This study aims to investigate the causal relationship between BMI and JIA, providing genetic evidence to inform clinical prevention and treatment strategies. A total of 35 BMI-related single nucleotide polymorphisms were included as instrumental variables. Inverse variance weighting analysis revealed a significant positive association between increase

Integrative genomic characterization of the FADS1-2-3 mood disorder risk locus: implications for the role of polyunsaturated fatty acids.

BACKGROUND: Polyunsaturated fatty acids (PUFA) have been implicated in mood disorder pathophysiology, with Mendelian randomisation studies suggesting a causal role. However, much of this evidence centres on the highly pleiotropic FADS1-2-3 gene cluster, raising concerns about horizontal pleiotropy and uncertainty regarding the true effector gene(s). To assess the validity of these concerns, we performed integrative genetic characterization of the FADS1-2-3 mood disorder risk locus. METHODS: We analysed summary statistics from the largest (up to n = 4 million) genome-wide association studies (

Mapping the immune-genetic architecture of Epstein-Barr virus-related phenotypes and multiple sclerosis through a single-cell genetic framework for target prioritization and pharmacologic hypothesis generation.

BACKGROUND: Multiple sclerosis (MS) is a severe neuroinflammatory disease causing substantial long-term disability. Strong epidemiologic evidence links Epstein-Barr virus (EBV) exposure with MS risk, but genetic evidence for immune target prioritization in EBV-related phenotypes remains limited. METHODS: We integrated single-cell cis-eQTL data from 14 immune cell types with GWASs of an EBV-related clinical phenotype and MS using a single-cell Mendelian randomization framework with colocalization analyses. Candidate eGenes were evaluated in independent cohorts. For multi-SNP instruments, we pe

Genome-Wide Pleiotropy Analysis Identifies Shared and Opposing Pathways Influencing Coronary Artery Disease and Cancer.

BACKGROUND: Coronary artery disease (CAD) and cancer are 2 leading global causes of mortality, with shared modifiable risk factors, yet the genetic and molecular mechanisms underlying their comorbidity remain poorly understood. METHODS: We performed a genome-wide pleiotropy analysis to identify shared genetic mechanisms across CAD and 4 common cancers that share modifiable risk factors with CAD (breast, colorectal, lung, prostate). RESULTS: Using genome-wide pleiotropy and colocalization analysis, we identified 60 colocalized susceptibility loci shared by CAD and site-specific cancer, of whi

Investigating Pleiotropy Between Depression and Autoimmune Diseases Using the UK Biobank.

BACKGROUND: Epidemiological studies report increased comorbidity between depression and autoimmune diseases. The role of shared genetic influences in the observed comorbidity is unclear. We investigated the evidence for pleiotropy between these traits in the UK Biobank (UKB). METHODS: We defined autoimmune and depression cases using hospital episode statistics, self-reported conditions and medications, and mental health questionnaires. Pairwise comparisons of depression prevalence between autoimmune cases and controls, and vice versa, were performed. Cross-trait polygenic risk score (PRS) ana

The causality between chronic rhinosinusitis and inflammatory bowel disease: A bidirectional 2-sample Mendelian randomization analysis.

Prior observational studies suggested an association between chronic rhinosinusitis (CRS) and inflammatory bowel disease (IBD), but causality remains unclear. This study employed a bidirectional 2-sample Mendelian randomization (MR) analysis to investigate potential causal relationships. Genetic variants associated with CRS and IBD (including Crohn's disease and ulcerative colitis) were sourced from the IEU Open genome-wide association study project. Primary analysis used inverse-variance weighted, supplemented by weighted median and MR-Egger methods. Sensitivity analyses included Cochran's Q

Divergent associations between adiposity and hernia subtypes in European populations: A Mendelian randomization study.

Obesity is a suspected risk factor for hernia, but observational studies are susceptible to confounding and reverse causation. We aimed to investigate the causal effects of general and central adiposity on 5 hernia subtypes using Mendelian randomization (MR). Two-sample MR analyses were conducted using genetic instruments for 7 adiposity traits from the UK Biobank and hernia outcome data (diaphragmatic, femoral, inguinal, umbilical, and ventral hernias) from the Finn Gen consortium. The inverse-variance weighted method was the primary analysis, complemented by MR Egger and weighted median meth

Association of gestational diabetes mellitus with intrahepatic cholestasis of pregnancy: A 2-sample Mendelian randomization analysis utilizing FinnGen consortium data.

Gestational diabetes mellitus (GDM) is a common metabolic disorder during pregnancy and a significant risk factor for maternal and neonatal complications. Although management strategies have improved, early identification of high-risk cases remains challenging. This study aimed to clarify whether GDM exerts a causal effect on intrahepatic cholestasis of pregnancy (ICP), a hepatic disorder associated with adverse perinatal outcomes. We performed a 2-sample Mendelian randomization (MR) analysis using summary data from the genome-wide association study of the FinnGen consortium. Genetic variants
