Uncovering perturbations in human hematopoiesis associated with healthy aging and myeloid malignancies at single-cell resolution.

Early hematopoiesis is a continuous process in which hematopoietic stem and progenitor cells (HSPCs) gradually differentiate toward specific lineages. Aging and myeloid malignant transformation are characterized by changes in the composition and regulation of HSPCs. In this study, we used single-cell RNA sequencing (scRNA-seq) to characterize an enriched population of human HSPCs obtained from young and elderly healthy individuals. Based on their transcriptional profile, we identified changes in the proportions of progenitor compartments during aging, and differences in their functionality, as

White Matter Hyperintensities and Cognitive Impairment in Healthy and Pathological Aging: A Quantified Brain MRI Study.

BACKGROUND: Brain changes involving the white matter (WM), often an indication of cerebrovascular pathology, are frequently seen in patients with mild cognitive impairment (MCI) and Alzheimer disease (AD). Few studies have examined possible cognitive domain- or group-specific cognitive effects of WM pathology in old age, MCI, and AD. OBJECTIVE: Our purpose was to examine the relationship between WM hyperintensities (WMH), a typical marker for WM pathology, and cognitive functioning in healthy old age and pathological aging using quantified MRI data. METHODS: We utilized multidomain neuropsyc

Asymptomatic heart valve dysfunction in healthy middle-aged companion dogs and its implications for cardiac aging.

Heart disease is the leading cause of death in the USA, accounting for about one in every four deaths. Age is the greatest risk factor for heart disease in both people and dogs; however, heart disease is generally not considered as a major cause of morbidity or mortality in dogs. As part of the preliminary selection process for a veterinary clinical trial, 40 companion dogs with no history of cardiac pathology that were at least 6 years old and weighed at least 18 kg underwent a cardiac screening using Doppler echocardiography. Eleven dogs from this cohort were diagnosed with valvular regurgit

Diabetes mellitus Type II and cognitive capacity in healthy aging, mild cognitive impairment and Alzheimer's disease.

While diabetes mellitus (DM) Type II has repeatedly been linked to Alzheimer´s disease (AD) and mild cognitive impairment (MCI), longitudinal research is scarce and disease duration has not always been taken into account. In a birth cohort born between 1930 and 1932 we investigated the influence of DM Type II and disease duration on neuropsychological functioning (memory/learning, attention, verbal fluency, visuospatial thinking and abstract thinking) across 14 years. Subjects who developed MCI or AD performed significantly poorer on all neuropsychological tests applied. While significant main

Exploring opportunities for healthy aging among older persons with a history of homelessness in Toronto, Canada.

Within the areas of literature on both population aging and health and homelessness, little attention has been given to the opportunities and barriers to healthy aging among older persons with a history of homelessness. Set in the context of inner-city Toronto, Canada, this article reports on the findings from qualitative interviews with 29 formerly homeless older persons. The findings illustrate participants' experiences of positive health change since moving into a stable housing environment and the aspects of housing they perceive to have improved their health and wellbeing. The qualitative…

Effects of cardiorespiratory fitness on blood pressure trajectory with aging in a cohort of healthy men.

BACKGROUND: Although the trajectory of blood pressure (BP) with aging is well known, there is a lack of data on how cardiorespiratory fitness (hereafter referred to as fitness) affects age-associated changes in BP. OBJECTIVES: The objective of the study was to investigate whether fitness alters the aging-BP trajectory. METHODS: A cohort from the Aerobics Center Longitudinal Study totaling 13,953 men between 20 and 90 years of age who did not have hypertension, cardiovascular disease, or cancer completed 3 to 28 (mean of 3.8) follow-up medical examinations between 1970 and 2006. Fitness was m

SIRT7: an influence factor in healthy aging and the development of age-dependent myeloid stem-cell disorders.

Molecular alterations within the hematopoietic system influence cellular longevity and development of age-related myeloid stem-cell disorders like acute myeloid leukemia (AML) and chronic myeloid leukemia (CML). A reduced SIRT7-expression in aged murine hematopoietic stem cells (HSC) resulted in reduced longevity and increased proliferation. In this study we investigated age-related changes of SIRT7-expression in healthy humans and relevant pathomechanisms in AML and CML. SIRT7-expression in leukocytes of healthy people decreased in an age-dependent manner. Low SIRT7 mRNA levels were also dete

Exploratory Associations Between Height-for-Age and Gut Microbiota Composition in Healthy School-Aged Children.

Body mass index (BMI) is frequently used to investigate associations between gut microbiota and nutritional status in children, although it primarily reflects adiposity and may not fully capture broader aspects of physiological development. Height-for-age z-score (HAZ), an indicator of linear growth, may provide complementary information on microbiota-host interactions during childhood. This exploratory cross-sectional study investigated associations between anthropometric indicators and gut microbiota composition in a cohort of healthy Spanish school-aged children. Forty-five children aged 8-

The neuropsychology of healthy aging: the positive context of the University of the Third Age during the COVID-19 pandemic.

Older adults have been reported to have increased susceptibility to the adverse effects of SARS-CoV-2 infection, such as fatal outcomes, cognitive decline, and changes in physical and/or mental health. However, few studies have examined neuropsychological changes by comparing measurements before and during the pandemic in healthy older people. In addition, no longitudinal studies have examined whether older adults may have responded positively to the pandemic. We examined these issues through a 2-year neuropsychological study before and during the pandemic period. Results showed that scores be

The effect of context and age on functional reach performance in healthy adults aged 21 to 94 years.

BACKGROUND: Activities of daily living require an individual to exhibit dynamic standing balance, while reaching beyond arm's length under varying contexts that involve an object to reach toward or grasp. Research shows that object context impacts the kinematics of reaching; however, there has been less research regarding the effect of context on functional reach (FR) ability. The purpose of the present study was to assess FR ability under 3 conditions between 3 age groups: (1) traditional FR, (2) reaching to an object (object present FR), and reaching to grasp an object (FR to grasp). The variability of cell-derived microparticles and the age of healthy blood donors.

INTRODUCTION: Cell-derived microparticles that promote coagulation can lead to transfusion-related complications. Although age-dependent changes in hemostasis are known, the impact of donor age on microparticle concentration variability remains largely unexplored. We sought to determine microparticle concentrations and investigate their relationship with donor age. METHODS: Whole-blood samples were collected from volunteers aged 17 to 60 years using K3EDTA as an anticoagulant. Donors were allocated to 1 of 5 age groups. Flow cytometric analysis and counting beads were used to determine microp

Corneal Nerve Fiber Morphology and Biological Age in Healthy Adults.

Background/Objectives: Corneal confocal microscopy (CCM) quantifies subbasal corneal nerve fibers noninvasively and may inform peripheral neuroaging. PhenoAge is a validated clinical measure of biological aging linked to morbidity and mortality risk and therefore provides a geriatric-relevant index of systemic aging. We aimed to assess corneal nerve morphology in clinically healthy adults and determine whether CCM-derived parameters are associated with biological age (PhenoAge) beyond chronological age. Methods: Eighty-four healthy volunteers (22-89 years) underwent CCM. PhenoAge was calculate

Healthy Ageing Through Yoga: Bridging Prevention, Function, and Rehabilitation.

Population ageing is a growing global challenge that requires strategies aimed not only at increasing lifespan but also at preserving health, independence, and quality of life. Healthy ageing emphasizes the maintenance of physical, mental, and social well-being, with particular focus on sustaining functional ability throughout later life. Yoga, an ancient mind-body practice integrating physical postures, breathing techniques, and meditation, has gained increasing recognition as a potential contributor to these goals. This editorial explores the role of yoga in promoting healthy ageing through

Oxidative stress markers in different age ranges of healthy horses.

Oxidative stress results from an imbalance between oxidants and antioxidants, impairing cellular functions and contributing to diseases and aging. Despite the growing interest in oxidative stress, studies specifically evaluating the influence of age on laboratory and oxidative stress parameters in clinically healthy horses remain limited. Thus, this study aimed to evaluate the influence of age on laboratory and oxidative stress parameters in clinically healthy horses. A total of 152 clinically healthy horses (80 females and 72 males) were selected, including young horses aged 3 to 48 months (n

Measuring and monitoring the quality of dying in the UN Decade of Healthy Ageing.

WHO aims to identify metrics to monitor the quality of dying, complementing those indicators proposed under the UN Decade of Healthy Ageing. However, the proposed criteria for a good death are contentious. Needs and priorities vary between individuals and their carers, across conditions, over time, and across communities and cultures. Monitoring should also consider sudden or rapid deaths and assisted dying. Fundamental challenges in data collection include who reports, over what timeframe, and when. This Personal View explores these challenges, identifying potentially measurable indicators an

Multidimensional associations between nutrient intake and healthy ageing in humans.

BACKGROUND: Little is known about how normal variation in dietary patterns in humans affects the ageing process. To date, most analyses of the problem have used a unidimensional paradigm, being concerned with the effects of a single nutrient on a single outcome. Perhaps then, our ability to understand the problem has been complicated by the fact that both nutrition and the physiology of ageing are highly complex and multidimensional, involving a high number of functional interactions. Here we apply the multidimensional geometric framework for nutrition to data on biological ageing from 1560 ol

Healthy Human Laryngopharyngeal Sensory Innervation Density Correlates with Age.

OBJECTIVE: Somatosensory feedback from upper airway structures is essential for swallowing and airway defense but little is known about the identities and distributions of human upper airway neurons. Furthermore, whether sensory innervation modifies with aging is unknown. In this study, we quantify neuronal and chemosensory cell density in upper airway structures and correlate with age. METHODS: Participants underwent biopsies from base of tongue, lateral and midline pharyngeal wall, epiglottis, and arytenoids (N = 25 13 female/12 male; 20-80 years, mean 51.4 years without clinical diagnosis
