Immunological markers of Langerhans' cells in mycosis fungoides skin lesions.

The patterns of two immunological markers of epidermal Langerhans' cells were compared in normal skin and mycosis fungoides (MF) skin lesions. A double-staining indirect immunofluorescence method was used. Cells suggestive of keratinocytes reacted with the anti-Ia antibodies in some cases of MF lesions, while no such pattern was seen with the thymocyte-antigen antibodies (OK T6). In other cases of MF, as in normal skin, the epidermal picture was mainly the same with the two markers. A great many of the dermal mononuclear cells in MF were reactive with the anti-Ia antibodies but only occasional

MHC class II invariant chain-adjuvanted viral vectored vaccines enhances T cell responses in humans.

Strategies to enhance the induction of high magnitude T cell responses through vaccination are urgently needed. Major histocompatibility complex (MHC) class II-associated invariant chain (Ii) plays a critical role in antigen presentation, forming MHC class II peptide complexes for the generation of CD4+ T cell responses. Preclinical studies evaluating the fusion of Ii to antigens encoded in vector delivery systems have shown that this strategy may enhance T cell immune responses to the encoded antigen. We now assess this strategy in humans, using chimpanzee adenovirus 3 and modified vaccinia A

Control of MHC class II gene expression in autoimmune, infectious, and neoplastic diseases.

The major histocompatibility complex (MHC) class II genes encode surface molecules that are required for presentation of antigenic peptides to helper T-cells. The concentration of these proteins on the surface of effector cells (antigen-presenting cells such as B-cells and macrophage) is one of the parameters affecting the intensity of the immune response. Many studies have thus focused their attention on the mechanisms that control the expression of class II genes, particularly in B-cells. The anatomy of MHC class II promoters has been dissected in detail, and many trans-acting factors and th

Gamma interferon production induced by antigens in patients with leprosy and American cutaneous leishmaniasis.

In this study, we measured gamma interferon production in mononuclear cell cultures from patients with diverse forms of leprosy and American cutaneous leishmaniasis. We studied patients with lepromatous, borderline lepromatous, borderline, and borderline tuberculoid forms of leprosy, as well as a Mitsuda-negative contact. In leishmaniasis we studied patients with localized cutaneous, mucocutaneous, and diffuse cutaneous forms of the disease. High correlation was observed between gamma interferon production and lymphocyte proliferation assays in both diseases. Resistant forms of both diseases s

HLA-DRw4 in pemphigus vulgaris patients in Israel.

Pemphigus vulgaris (PV) is relatively common in Jews. Three HLA antigens were significantly more frequent in 39 Israeli Jewish PV patients than in controls A26 - 59% vs 20%; Bw38 - 61% vs 20%; and DRw4 - 90% vs 38%. The joint occurrence of A26-Bw38-DRw4 was observed in 46% of PV patients and in 10% of controls. Similar results were recently reported for Jews in the Los Angeles area. Yet, when our patient sample was grouped into Ashkenazi and non-Ashkenazi Jews, it was evident that each of the three antigens had a higher frequency both in Ashkenazi patients and controls as compared to non-Ashke

What a rheumatologist needs to know about T cell receptor structure and function.

By understanding normal immune response, it has been possible to develop therapeutic strategies toward the treatment of autoimmune disease. The association of autoimmune disease with the major histocompatibility complex (MHC) class II gene products suggests that the inductive events in which the putative autoantigen is presented on the surface of antigen presenting cells in the context of the MHC class II gene products and is recognized by CD4(+) helper or inducer T cells form an interesting target for immunotherapeutic intervention. By understanding the structure/function relationships of T c

Aberrant expression of HLA-DR antigen on valvular fibroblasts from patients with active rheumatic carditis.

Immunofluorescence and immunoperoxidase staining was used to investigate the expression of Class II major histocompatibility antigens in myocardial tissue of 16 patients with acute rheumatic carditis. Aberrant expression of HLA-DR was examined using monoclonal anti-Ia antibodies and was detected on the valvular fibroblasts of those valves with ongoing active carditis. Sections of myocardial and valvular tissue from normal controls or from patients dying of other cardiac diseases did not express HLA-DR. The aberrant expression of HLA-DR on valvular fibroblasts could be important in triggering a

Detection of cytokine activated chondrocytes in arthritic joints from pigs infected with Erysipelothrix rhusiopathiae.

Chronic polyarthritis was induced in pigs by injection of Erysipelothrix rhusiopathiae and the in vivo activation of chondrocytes by cytokines was then investigated in the affected joints by immunocytochemistry. A polyclonal antiserum which recognises surface markers on in vitro interleukin 1 activated porcine chondrocytes was used to detect activated chondrocytes in all zones of the cartilage from diseased joints. In contrast, cartilage removed from an unaffected joint in the same animal showed no chondrocyte activation. Inflammatory synovial tissue removed from diseased joints and cocultured

MHC class I, MHC class II and intercellular adhesion molecule-1 (ICAM-1) expression in inflammatory myopathies.

We investigated the relationship between the MHC-I, MHC-II and intercellular adhesion molecule-1 (ICAM-1) expression on myofibres and the presence of inflammatory cells in muscle specimens of 18 patients with inflammatory myopathies (nine polymyositis, seven dermatomyositis, two inclusion body myositis). We observed MHC-I expression in muscle fibres, infiltrating mononuclear cells and endothelial cells in every specimen. In seven patients, some muscle fibres were MHC-II-positive for the DR antigen, while the DP and DQ antigens were absent. ICAM-1 expression, detected in seven patients, was fou

Combined imputation of HLA genotype and self-identified race leads to better donor-recipient matching.

Allogeneic Hematopoietic Cell Transplantation (HCT) is a curative therapy for hematologic disorders and often requires human leukocyte antigen (HLA)-matched donors. Donor registries have recruited donors utilizing evolving technologies of HLA genotyping methods. This necessitates in-silico ambiguity resolution and statistical imputation based on haplotype frequencies estimated from donor data stratified by self-identified race and ethnicity (SIRE). However, SIRE has limited genetic validity and presents a challenge for individuals with unknown or mixed SIRE. We present MR-GRIMM "Multi-Race Gra

HLA-DR typing in 58 cases of rheumatoid arthritis.

The results of a previous study led us to believe that the association of HLA-DR4 and rheumatoid arthritis could be of particular interest in our region. In this work we typed 58 caucasoid patients in the Marseilles area by microlymphocytotoxicity assay in B-cells. HLA-DR4 was found to be at the same level as that observed by the authors in the 8th Workshop of histocompatibility (1980) (X2:32.36, P less than 0.001) and very high in the seropositive patients (61.5 per cent). No correlation with sex was observed. We again found a low frequency of HLA-DR4 (15.8 per cent) in our controls.

An HLA-DR negative acute leukaemia which stimulates MLC and CMC responses.

We describe an acute myelomonocytic leukaemia (E72) devoid of cell-surface HLA-DR antigens, but capable of inducing cellular responses. Leukaemia E72 induced proliferation of normal lymphocytes in primary mixed lymphocyte culture (MLC), which was only weakly inhibited by anti-DR sera. Depletion of a small percentage (congruent to 4%) of DR+ cells on a cell-sorter failed to abrogate the capacity of E72 to stimulate MLC and cell-mediated cytotoxicity (CMC) responses. We found that normal lymphocytes primed with E72 responded in secondary MLC to lymphocytes and leukaemias, suggesting that the lym

Processing of Listeria monocytogenes antigens and the in vivo T-cell response to bacterial infection.

Presentation of antigens to T lymphocytes is a critical step in the clearance of pathogens from their hosts and in the establishment of protective immunity. Several animal models have been developed to study this process, but few have been as informative as the murine immune response to Listeria monocytogenes infection. Herein we review the presentation of L. monocytogenes proteins by the MHC class I antigen-processing pathway and the in vivo T-cell response to these bacterial antigens. These studies demonstrate the following: 1) The size of a peptide-specific T-cell response does not correlat

Enhanced MHC Class-II Expression in Fibroblastic Reticular Cells Associates with Maturation.

Autoimmunity can be initiated by autoreactive T cells that escaped central and peripheral tolerance induction. Peripheral tolerance in lymph nodes (LNs) is maintained by fibroblastic reticular cells (FRCs) via self-antigen presentation in major histocompatibility complex (MHC) class II. FRCs can be divided into various subsets, with specific markers, functions, and locations. FRCs located in the T-cell zone (TRCs) can express genes for antigen presentation in MHC class-II, for example, H2-Ab1 and Cd74, as well as the immune inhibitory ligand Cd200. However, whether this can be linked to MHC cl

Experimental autoimmune uveitis (EAU): immunophenotypic analysis of inflammatory cells in chorio retinal lesions.

Using a panel of monoclonal antibodies to guinea pig leukocyte antigens we have examined the phenotype of inflammatory cells present in focal chorio-retinal lesions developing in guinea pigs during S-antigen induced experimental autoimmune uveitis (EAU). The earliest choroidal infiltrates of T cells appeared to be T helper (Th) cells but in choroidal foci of inflammatory cells and in early retinal lesions, where rod outer segment lysis had occurred, the presence of a Pan T negative, T cytotoxic/suppressor (T c/s) positive population of effector cells was identified. In areas of more extensive

MHC-chromosome dosage effects: evidence for increased expression of Ia glycoprotein and alteration of B cell subpopulations in neonatal aneuploid chickens.

Quantitative variation in the expression of MHC-encoded class II (Ia) glycoproteins has been associated with stages of lymphocyte development and a number of disease conditions. We have used an avian MHC dosage model to study the regulation of Ia expression and the effects of quantitative variation in membrane Ia on B-cell development. Lymphocyte membrane expression of Ia glycoprotein molecules and the frequency of small-versus-large lymphocytes were examined in trisomic line chickens containing either two (disomic), three (trisomic), or four (tetrasomic) copies of the microchromosome encoding

Expression of class II major histocompatibility complex molecules in renal tubular epithelial cells of canine kidneys affected with tubulointerstitial nephritis.

Class II major histocompatibility complex (MHC) products are important molecules on various antigen-presenting cells and induce a T cell-specific immune response. The distribution of class II MHC molecules in the normal canine kidneys of dogs with tubulointerstitial nephritis was investigated by using a sensitive immunocytochemical method. In the normal canine kidney, class II MHC molecules were detected in interstitial 'dendritic' cells. In cases of tubulointerstitial nephritis, however, the expression of class II MHC molecules extended to other renal elements such as the epithelial cells of
