Autoimmune Disease in Primary Immunodeficiency: At the Crossroads of Anti-Infective Immunity and Self-Tolerance.

The association of autoimmunity and primary immunodeficiency suggests the existence of mechanistic links between development of the various elements of the immune system and the maintenance of self-tolerance. In this review, various monogenic primary immunodeficiencies (PID) are systematically explored, with a specific focus on the impact of these genetic lesions on tolerance, correlating these defects in tolerance with clinical autoimmune and inflammatory syndromes seen in these PIDs. Common variable immunodeficiency (CVID) is explored, and areas are highlighted in which findings in monogenic

ADP-ribosylation controls the outcome of tolerance or enhanced priming following mucosal immunization.

Accumulating evidence suggests that the dichotomy between tolerance and active IgA immunity in mucosal immune responses is regulated at the APC level. Therefore, immunomodulation of the APC could be an effective mechanism to control the two response patterns. In this study, we demonstrate that ADP-ribosylation controls the outcome of tolerance or active effector T cell immunity to an internal peptide p323-339 from OVA inserted into the cholera toxin (CT)-derived CTA1-OVA-DD adjuvant. We found that a single point mutation, CTA1R7K-OVA-DD, resulting in lack of enzymatic activity, promoted peptid

Passive transfer of Theileria equi antibodies to neonate foals of immune tolerant mares.

Equine babesiosis, a tick transmitted haemoprotozoan disease caused by Theileria equi is globally distributed and responsible for heavy economic losses to the equine husbandry. Equids reared in endemic areas usually pick up infection at an early age and become immune tolerant throughout their life span. We studied the level of passively transferred antibodies in neonate foals born from pre-immuned mares. Latently T. equi infected pre-immuned pony and donkey mares (three each) were selected and T. equi antibody titres in neonates was monitored till 90 days post foaling (DPF) by applying Dot-ELI

Covalent modification as a mechanism for the breakdown of immune tolerance to pyruvate dehydrogenase complex in the mouse.

The autoimmune liver disease primary biliary cirrhosis (PBC) is characterized by the breakdown of normal immune self tolerance to pyruvate dehydrogenase complex (PDC). How tolerance is broken to such a central and highly conserved self antigen in the initiation of autoimmunity remains unclear. One postulated mechanism is that reactivity arises to an altered form of self antigen with subsequent cross-reactivity to native self. In this murine study, we set out to examine whether sensitization with a covalently modified form of self PDC can give rise to the pattern of breakdown of B-cell and T-ce

Association of anti-idiotypic antibodies with immune tolerance induction for the treatment of hemophilia A with inhibitors.

BACKGROUND AND OBJECTIVES: Hemophilia A patients with inhibitors can be treated effectively with immune tolerance induction therapy (ITI). One of the underlying mechanisms of ITI is conceived to be a neutralizing activity of anti-idiotypic antibodies on inhibitors. The goal of the present study was to develop an uncomplicated method for assessing antiidiotypic antibodies and to prove the advent of anti-idiotypic antibodies in ITI. DESIGN AND METHODS: We studied a total of 26 plasma samples obtained from 9 hemophilic inhibitor patients who were treated with ITI. The samples were investigated w

Tracking the role of Aire in immune tolerance to the eye with a TCR transgenic mouse model.

Roughly one-half of mice with partial defects in two immune tolerance pathways (AireGW/+Lyn-/- mice) spontaneously develop severe damage to their retinas due to T cell reactivity to Aire-regulated interphotoreceptor retinoid-binding protein (IRBP). Single-cell T cell receptor (TCR) sequencing of CD4+ T cells specific for a predominate epitope of IRBP showed a remarkable diversity of autoantigen-specific TCRs with greater clonal expansions in mice with disease. TCR transgenic mice made with an expanded IRBP-specific TCR (P2.U2) of intermediate affinity exhibited strong but incomplete negative s

Antiviral therapy in hepatitis B virus-infected with immune-tolerant: A meta-analysis.

To access the efficacy of antiviral therapy in patients of HBV-infected with immune-tolerant. We conducted a meta-analysis search of the Cochrane Library, PubMed, ClinicalTrials.gov, Web of science, and EMBASE on through August 2021. We combined the data by means of a random-effect DrSimonian-Laird model and calculated risk ratios (RRs) for the outcomes of hepatitis B surface antigen (HBsAg) loss, hepatitis B e antigen (HBeAg) seroconversion, HBV deoxyribonucleic acid (DNA) negative conversion rate, and the risk for hepatocellular carcinoma (HCC) and cirrhosis. An extensive literature search i

Inducible mechanisms of disease tolerance provide an alternative strategy of acquired immunity to malaria.

Immunity to malaria is often considered slow to develop but this only applies to defense mechanisms that function to eliminate parasites (resistance). In contrast, immunity to severe disease can be acquired quickly and without the need for improved pathogen control (tolerance). Using Plasmodium chabaudi, we show that a single malaria episode is sufficient to induce host adaptations that can minimise inflammation, prevent tissue damage and avert endothelium activation, a hallmark of severe disease. Importantly, monocytes are functionally reprogrammed to prevent their differentiation into inflam

Oral Tolerance to Environmental Mycobacteria Interferes with Intradermal, but Not Pulmonary, Immunization against Tuberculosis.

Bacille Calmette-Guérin (BCG) is currently the only approved vaccine against tuberculosis (TB) and is administered in over 150 countries worldwide. Despite its widespread use, the vaccine has a variable protective efficacy of 0-80%, with the lowest efficacy rates in tropical regions where TB is most prevalent. This variability is partially due to ubiquitous environmental mycobacteria (EM) found in soil and water sources, with high EM prevalence coinciding with areas of poor vaccine efficacy. In an effort to elucidate the mechanisms underlying EM interference with BCG vaccine efficacy, we expos

Immune function alterations in mice tolerant to delta9-tetrahydrocannabinol: functional and biochemical parameters.

We studied the effect of acute (1 h) or chronic exposure (7 and 14 days) to delta9-tetrahydrocannabinol (delta9-THC) on immune parameters in male Swiss mice. One hour after a dose of 10 mg/kg s.c., the splenocyte proliferative response to ConA and NK activity were not inhibited, but there was a significant decrease in the production of IL-2. After 7 days of treatment, when mice were tolerant to delta9-THC-induced analgesia, these functional parameters were strongly inhibited and there was a persistent reduction in IL-2 and IFNgamma. With 14 days exposure to the drug, splenocyte proliferation w

The activity of T and B lymphocytes in immunity and tolerance to the lymphocytic choriomeningitis virus in mice.

Treatment with anti-theta serum and the Wigzell column technique for cell separation was employed to study the separate functions of the B and T lymphocytes in the late states of immunity to the LCM virus in mice. The cell preparations examined were mixtures of spleen and lymph node cells from immune mice. The results revealed that the anti-viral effect of such cells after transfer to virus carriers was unimpaired in T cell-enriched and B cell-deprived cell preparations. The anti-viral effect was also retained in cell preparations deprived so much of B cells that no antibody was produced in th

Key molecules and functional subsets of regulatory T cells in maternal-fetal immune tolerance: Recent advances.

The establishment and maintenance of maternal-fetal immune tolerance constitutes a fundamental immunological adaptation essential for a successful pregnancy. Regulatory T cells (Tregs) are central orchestrators of this process, actively suppressing effector responses and fostering a tolerogenic microenvironment at the maternal-fetal interface. This review systematically synthesizes recent advances in deciphering the specialized molecular signature and functional heterogeneity of decidual Tregs. We first delineate the molecular profile of CD4⁺ Tregs, emphasizing critical inhibitory receptors-in

Nematode gut microbiota confers cadmium tolerance through microbial-dependent activation of detoxification and innate immune pathways.

INTRODUCTION: Soil heavy metal pollution, especially cadmium (Cd) contamination, represents a serious threat to ecosystem health and agricultural sustainability. Understanding how soil organisms and their associated microbiota respond to such stress is crucial for developing bioremediation strategies. OBJECTIVES: This study aimed to investigate the response of the gut microbiota in the model soil nematodeCaenorhabditis elegansunder Cd(II) stress, identify key bacterial taxa involved in metal detoxification, and elucidate the underlying mechanisms of microbe-mediated host resistance. METHODS:

The role of Astragalus membranaceus and its derivatives in digestive system diseases: reconstruction of immune tolerance.

The gastrointestinal tract, a vital immune organ in the human body, serves as the primary site of direct contact with external antigens. Maintaining immune tolerance is essential for preventing the onset and progression of digestive system diseases. Autoimmune diseases and tumors represent the overactivation and evasion of the immune system, respectively, and the imbalance of "immune tolerance" is the common core mechanism underlying both conditions. The function of traditional Chinese medicine in regulating the immune function of the digestive system has attracted wide attention, especially i

FGF21 induced by endoplasmic reticulum stress maintains medullary thymic epithelial cell function and central immune tolerance.

Medullary thymic epithelial cells (mTECs) establish central immune tolerance by expressing diverse tissue-restricted antigens and eliminating self-reactive thymocytes. Here, we show that fibroblast growth factor 21 (FGF21), although predominantly produced in the liver, is also expressed locally by mature mTECs and contributes to central tolerance. Fgf21-deficient mice exhibited exacerbated peripheral autoimmune responses. FGF21 supported the number and function of mTECs and promoted clonal deletion in cooperation with thymic dendritic cells. In mature mTECs, endoplasmic reticulum stress induce

CCR2-Guided Treg Exosome-Gas6 Hybrid Synergistically Reprograms Macrophage Inflammation and Immune Tolerance in Gouty Arthritis.

Gouty arthritis (GA) is a recurrent inflammatory joint disease initiated by monosodium urate (MSU) crystal deposition and driven by progressive immune dysregulation. The collapse of immune tolerance, together with persistent synovial inflammation and pathogenic cytokine imbalances, jointly exacerbates joint injury. Here, we design a bioresponsive therapeutic platform composed of regulatory T cell (Treg)-derived exosomes co-loaded with growth arrest-specific protein 6 (Gas6), engineered to preferentially home to inflamed joints via CCR2-guided chemotactic targeting. The two components of the hy

Nanoparticle-enabled CD40 silencing programs immune tolerance via a DC-Treg-Breg axis in autoimmune myocarditis.

Autoimmune myocarditis is a severe inflammatory heart disease that can progress to dilated cardiomyopathy and heart failure, while disease-modifying therapies remain limited. Dysregulated activation of antigen-presenting cells, particularly dendritic cells (DCs), disrupts immune tolerance and drives pathogenic cardiac inflammation. Here, we report a nanoparticle-enabled CD40 silencing strategy for immune tolerance programming in experimental autoimmune myocarditis. Using a polymer-lipid hybrid siRNA formulation, we achieved sustained CD40 silencing in myeloid antigen-presenting cells and induc
