Lymphocytes and B-cell abnormalities in patients with common variable immunodeficiency (CVID).

BACKGROUND AND AIMS: Common variable immunodeficiency (CVID) is a primary antibody deficiency characterised by decreased antibody production and low or normal B-cell numbers. To elucidate the clinical and immunological heterogeneity of CVID, we studied 16 patients diagnosed with CVID. METHODS: We analysed B, T and NK cell populations. We also assessed CD27 expression to define B-cell subsets and examined the expression of molecules important in B-cell proliferation and differentiation, such as the transmembrane activator and CALM interactor (TACI), inducible costimulator (ICOS), CD154 and CD4

Calcineurin inhibitors suppress cytokine production from memory T cells and differentiation of naïve T cells into cytokine-producing mature T cells.

T cells have been classified as belonging to the Th1 or Th2 subsets according to the production of defining cytokines such as IFN-γ and IL-4. The discovery of the Th17 lineage and regulatory T cells shifted the simple concept of the Th1/Th2 balance into a 4-way mechanistic pathway of local and systemic immunological activity. Clinically, the blockage of cytokine signals or non-specific suppression of cytokine predominance by immunosuppressants is the first-line treatment for inflammatory T cell-mediated disorders. Cyclosporine A (CsA) and Tacrolimus (Tac) are commonly used immunosuppressants f

Development of acquired immunity to Salmonella.

Salmonella enterica serovar Typhi (S. typhi) causes human typhoid fever, a serious and widespread disease in developing countries. Other Salmonella serovars are associated with food-borne infections. The recent emergence of multi-drug-resistant Salmonella strains highlights the need for better preventive measures, including vaccination. The available vaccines against Salmonella infection do not confer optimal protection. The design of new Salmonella vaccines must be based on the identification of suitable virulence genes and on knowledge of the immunological mechanisms of resistance to the dis

Sequential infection reprograms the immune landscape to shape future responses.

Mouse models have been instrumental in defining immune mechanisms but often fail to capture the complexity of human immunity, limiting clinical translation. A major limitation is the immunological immaturity of specific pathogen-free (SPF) mice relative to pathogen-experienced adult humans. Here, we use a sequential infection (SI) model that recapitulates cumulative pathogen exposure and define its impact on immune composition and function. Beyond the previously reported expansion of memory T cells, SI induced durable, system-wide remodeling across lymphoid and non-lymphoid tissues, reshaping

Lymphocyte Perturbations in Malawian Children with Severe and Uncomplicated Malaria.

Lymphocytes are implicated in immunity and pathogenesis of severe malaria. Since lymphocyte subsets vary with age, assessment of their contribution to different etiologies can be difficult. We immunophenotyped peripheral blood from Malawian children presenting with cerebral malaria, severe malarial anemia, and uncomplicated malaria (n = 113) and healthy aparasitemic children (n = 42) in Blantyre, Malawi, and investigated lymphocyte subset counts, activation, and memory status. Children with cerebral malaria were older than those with severe malarial anemia. We found panlymphopenia in children

Loss of IRF-4-binding protein leads to the spontaneous development of systemic autoimmunity.

IFN regulatory factor 4-binding (IRF-4-binding) protein (IBP) is a novel type of activator of Rho GTPases that is recruited to the immunological synapse upon TCR stimulation. Here we demonstrate that loss of IBP leads to the spontaneous development of a systemic autoimmune disorder characterized by the accumulation of effector/memory T cells and IgG+ B cells, profound hypergammaglobulinemia, and autoantibody production. Similar to human SLE, this syndrome primarily affects females. T cells from IBP-deficient mice are resistant to death in vitro as well as in vivo and exhibit selective defects

Reshape memory T cell landscape in CML patients' blood by tyrosine kinase inhibitors.

BACKGROUND: CML, a blood cancer characterized by excessive myeloid cell proliferation, stems from the BCR-ABL1 gene fusion. This research aimed to uncover why leukemic stem cells resist treatment and explore innovative immune-based therapies to improve patient outcomes. METHODS: This study investigated the immunologic phenotypes (CD8, CD27, CD197, and CD45RA) of T cell subsets (TN, TCM, TEM, and TEF) in the peripheral blood of chronic myeloid leukemia patients in the chronic phase who achieved a complete molecular response to tyrosine kinase inhibitors. The treatment-free remission (TFR) pati

Phenotypic characterization of peripheral T cells and their dynamics in scrub typhus patients.

BACKGROUND: Scrub typhus, caused by Orientia tsutsugamushi infection, is one of the main causes of febrile illness in the Asia-Pacific region. Although cell-mediated immunity plays an important role in protection, little is known about the phenotypic changes and dynamics of leukocytes in scrub typhus patients. METHODOLOGY/PRINCIPAL FINDINGS: To reveal the underlying mechanisms of immunological pathogenesis, we extensively analyzed peripheral blood leukocytes, especially T cells, during acute and convalescent phases of infection in human patients and compared with healthy volunteers. We observ

Mechanistic insights into the impairment of memory B cells and antibody production in the elderly.

It is well established that immunologic memory generated early in life can be maintained into old age and mediate robust anamnestic antibody responses. Little is known, however, about the initiation of memory B cells in the elderly. We have conducted a prospective analysis of the quantities and functionalities of antigen-specific B cell responses and its association with the functional helper CD4(+)T cell responses. The ability of naïve B cells from old (60-80 years) and young (20-31 years) humans to establish functional memory was examined following primary and booster vaccination with an ina

Vigorous response of human innate functioning IgM memory B cells upon infection by Neisseria gonorrhoeae.

Neisseria gonorrhoeae, the cause of the sexually transmitted infection gonorrhea, elicits low levels of specific Ig that decline rapidly after the bacteria are cleared. Reinfection with the same serovar can occur, and prior gonococcal infection does not alter the Ig response upon subsequent exposure, suggesting that protective immunity is not induced. The mucosal Ig response apparent during gonorrhea does not correlate with that observed systemically, leading to a suggestion that it is locally generated. In considering whether N. gonorrhoeae directly influences B cells, we observed that gonoco

T follicular helper cell development during malaria.

Malaria is a life-threatening disease affecting more than 200 million people worldwide. CD4 T helper 1 (Th1) and T follicular helper (Tfh) cells are critical for stimulating phagocyte activation and humoral immunity, yet sterilizing, protective immune memory responses rarely develop. Malaria induces unique physiologic changes in the infected host that may give rise to nonconventional Tfh subsets and inefficient memory responses. While our understanding of the differentiation of conventional Tfh cells induced by Plasmodium infection is growing, how and whether malaria-associated environmental c

Profiles of B-cell subsets in immunologically stable renal allograft recipients and end-stage renal disease patients.

BACKGROUND: Post-transplantation pharmacotherapies typically employ combinations of immunosuppressive agents that have been designed for targeted inhibition of T-cells and T-cell subsets. Studies of acute and chronic effects of clinically employed immunosuppressive agents on B-cells and B-cell subsets are significantly fewer in number and warrant further investigation. Accordingly, the goal of the present cross-sectional study is to functionally evaluate differences of B-cell subsets in patients with end-stage renal disease (ESRD) and immunologically stable renal transplant patients. Immune Networks and Therapeutic Targeting of iNKT Cells in Cancer.

One of the primary goals in tumor immunotherapy is to reset the immune system from tolerogenic to immunogenic - a process in which invariant natural killer T (iNKT) cells are implicated. iNKT cells develop in the thymus and perform immunosurveillance against tumor cells peripherally. When optimally stimulated, iNKT cells differentiate and display more efficient immune functions. Some cells survive and act as effector memory cells. We discuss the putative roles of iNKT cells in antitumor immunity, and posit that it may be possible to develop novel therapeutic strategies to treat cancers using i

Lipid metabolic regulation of pathogenic type 2 immunity in the airway.

Immune memory is central to host protection against pathogens and contributes to the pathogenesis of chronic inflammatory diseases. Beyond canonical cytokines and antigenic stimuli, metabolic signals have emerged as pivotal regulators of T-cell activation, differentiation, and memory formation. However, the mechanisms by which metabolic and tissue-derived cues imprint pathogenic features on T cells remain poorly understood. Dysregulation of type 2 immune memory is a major driver of chronic inflammation in allergic diseases. A distinct subpopulation of memory T helper 2 (Th2) cells that express

Elephant in the room: natural killer cells don't forget HIV either.

PURPOSE OF REVIEW: Like elephants (and T cells), accumulating evidence suggest natural killer (NK) cells never forget. The description of adaptive or memory NK cells, which can be induced by HIV/SIV infections and vaccines and associated with protective effects in persons with HIV (PWH), has dramatically increased the interest in leveraging NK cells to prevent HIV infection or suppress HIV reservoirs. However, harnessing their full antiviral potential has been hindered by an incomplete understanding of mechanisms underlying adaptive NK cell development and infected cell recognition. Herein, we

Lymphocyte transformation to Legionella pneumophila.

Lymphocytes from seventeen subjects with previously diagnosed Legionnaires' disease and 63 subjects without a history of Legionellosis were studied in a transformation assay with a crude extract of Legionella pneumophila. In the lymphocyte transformation assay the mean stimulation index was 48+/-14 in the Legionnaires' disease group and 18+/-13 (p less than 0.001) in the control group. Endotoxin from Escherichia coli did not stimulate the cultures. Extracts from multiple other gram negative bacilli had minimal stimulation which was similar in lymphocytes from both groups. Lymphocytes from 2/3

Alternative B Cell Differentiation During Infection and Inflammation.

Long-term protective immunity to infectious disease depends on cell-mediated and humoral immune responses. Induction of a strong humoral response relies on efficient B cell activation and differentiation to long-lived plasma cells and memory B cells. For many viral or bacterial infections, a single encounter is sufficient to induce such responses. In malaria, the induction of long-term immunity can take years of pathogen exposure to develop, if it occurs at all. This repeated pathogen exposure and suboptimal immune response coincide with the expansion of a subset of B cells, often termed atypi

Natural killer cells in patients with allergic diseases.

Natural killer (NK) cells not only exert cytotoxic activity against tumor cells or infected cells but also act to regulate the function of other immune cells through secretion of cytokines and chemokines or cell contact-dependent mechanisms. NK cells are able to polarize in vitro into 2 functional distinct subsets, NK1 or NK2 cells, which are analogous to the T-cell subsets TH1 or TH2. In addition, a regulatory NK cell subset has been described that secretes IL-10, shows antigen-specific T-cell suppression, and suppresses IgE production. Although it has been demonstrated that NK cells play imp

CD4 T cell phenotype after AS01-adjuvanted immunization is shaped by prior antigen or pathogen exposure.

CD4 T-cell responses are important for the protection conferred by many infectious disease vaccines and can be quantitatively and qualitatively improved by adjuvants. The AS01 adjuvant is present in critical licensed or candidate vaccines against, amongst others, zoster, respiratory syncytial virus, malaria and tuberculosis. During their clinical development, these vaccines have been evaluated in populations ranging from immunologically naïve to strongly pathogen-primed individuals. We dissected the vaccine-induced CD4 T-cell responses across these studies, to elucidate how pre-existing immune
