The Battle Between Japanese Encephalitis Virus and Host Innate Immune System: Evasion and Response.

Japanese encephalitis (JE) is a natural zoonotic disease caused by the Japanese encephalitis virus (JEV), which poses potential threats to human health and the pig farming industry. To establish infection, JEV must overcome the innate immune responses and complete its lifecycle in new hosts. Notably, the direct virus-induced neuronal cell death and an uncontrolled neuroinflammatory response jointly lead to the pathogenesis of JEV. In this review, we will focus on the innate immune response to JEV infection and the viral immune evasion strategies, such as escaping recognition or inhibiting the

Comparative Toll-like receptor profiling reveals breed-specific innate immune architecture following Newcastle disease vaccination.

The present study investigated the expression profiles of six TLR genes (TLR2A, TLR3, TLR4, TLR5, TLR7 and TLR21) in peripheral blood mononuclear cells (PBMCs) following genotype II ND vaccination across fifteen genetically diverse chicken populations representing exotic, indigenous, fancy, synthetic and broiler parent lines. Relative gene expression was quantified by qPCR, and receptor expression data were integrated into five novel composite immune indices, namely the Viral Recognition Index (VRI), Bacterial Recognition Index (BRI), Immune Diversity Score (IDS), Shannon TLR Diversity Index (

Pathogenesis of bronchopulmonary dysplasia: an immunological perspective.

Bronchopulmonary dysplasia (BPD), the most common chronic lung disease of prematurity, is at its mechanistic core an immune-developmental disease in which inflammatory injury disrupts normal lung maturation. In the United States, BPD remains among the most common morbidities of very preterm birth, affecting a substantial proportion of infants born before 32 weeks' gestation. These infants are born before alveolarization is complete, into an inflammatory environment that can impair ongoing lung development. This review advances a single thesis: the heterogeneous prenatal and postnatal insults t

Z-Nucleic Acid-Binding Protein (ZBP)1 (DAI/DLM1) Regulation in Antiviral Cell Death Pathways.

Metazoan organisms employ an array of overlapping innate and adaptive immune mechanisms to defend against infection and prevent disease. Z-nucleic acid-binding protein (ZBP)1 (also denoted DAI or DLM1) has emerged as a key nucleic acid (NA) sensor within mammalian cells responding to stress, infection or inflammation. Once engaged and activated by Z-nucleic acid, ZBP1 recruits receptor-interacting protein (RIP) kinase (RIPK)3 through RIP homotypic interaction motif (RHIM)-mediated interactions to promote regulated cell death and inflammatory signaling, with necroptosis representing a particula

Insights into the role of TLR21 in innate immunity of Japanese flounder (Paralichthys olivaceus).

As an essential immune receptor in Japanese flounder (Paralichthys olivaceus), Toll-like receptor 21(TLR21) plays a pivotal role in recognizing pathogen-associated molecular patterns (PAMPs) and activating innate immune signaling pathways. In this study, the functions of Japanese flounder TLR21 (PoTLR21) were studied. Expression pattern analysis revealed that Potlr21 was ubiquitously expressed in various tissues of Japanese flounder and induced following lymphocystis disease virus (LCDV) infection. PoTLR21 was subcellularly localized in cytoplasm with weak fluorescence signals detected on cell

[The investigations of the role of toll-like receptors (TLR) in host response to parasitic infection on the current background regarding TLR in mammals and the model nematode Caenorhabditis elegans].

Toll-like receptors (TLRs) are amongst the most highly conserved in the evolution of receptor family, being found in both immune and other cells. TLRs were observed in vascular endothelial cells, epithelial cells, microglia cells, adipocytes, and intestinal and renal cells. TLRs plays a key role in the innate immune response to a variety of pathogens. At present, very little is known about the role of TLRs in host defense against parasitic pathogen infections. The first study shows that TLRs contribute to both innate and adaptive immune responses following infection with protozoan parasite Lei

Recent developments in livestock and wildlife brucellosis vaccination.

Live attenuated brucellosis vaccines have been available for protecting domestic livestock against Brucella melitensis and B. abortus for more than 60 years. Current vaccines are effective in preventing abortion and transmission of brucellosis, but poor at preventing infection or seroconversion. In addition, they can induce abortions in pregnant animals and are infectious to humans. It can be argued that current vaccines were developed empirically in that the immunological mechanism(s) of action were not determined. Current knowledge suggests that both the innate and adaptive immune responses

Macrophage and Dendritic Cell Activation and Polarization in Response to Coccidioidesposadasii Infection.

Coccidioidomycosis is a fungal, respiratory disease caused by Coccidioides immitis and Coccidioides posadasii. The host immune responses that define disease outcome during infection are largely unknown, although T helper responses are required. Adaptive immunity is influenced by innate immunity as antigen-presenting cells activate and educate adaptive responses. Macrophage and dendritic cell (DC) recognition of pathogen surface molecules are critical for Coccidioides clearance. We characterize the broad innate immune responses to Coccidioides by analyzing macrophage and dendritic cell response

Translating innate response into long-lasting antibody response by the intrinsic antigen-adjuvant properties of papaya mosaic virus.

Identifying the properties of a molecule involved in the efficient activation of the innate and adaptive immune responses that lead to long-lasting immunity is crucial for vaccine and adjuvant development. Here we show that the papaya mosaic virus (PapMV) is recognized by the immune system as a pathogen-associated molecular pattern (PAMP) and as an antigen in mice (Pamptigen). A single immunization of PapMV without added adjuvant efficiently induced both cellular and specific long-lasting antibody responses. PapMV also efficiently activated innate immune responses, as shown by the induction of

Synthetic RNAs Mimicking Structural Domains in the Foot-and-Mouth Disease Virus Genome Elicit a Broad Innate Immune Response in Porcine Cells Triggered by RIG-I and TLR Activation.

The innate immune system is the first line of defense against viral infections. Exploiting innate responses for antiviral, therapeutic and vaccine adjuvation strategies is being extensively explored. We have previously described, the ability of small in vitro RNA transcripts, mimicking the sequence and structure of different domains in the non-coding regions of the foot-and-mouth disease virus (FMDV) genome (ncRNAs), to trigger a potent and rapid innate immune response. These synthetic non-infectious molecules have proved to have a broad-range antiviral activity and to enhance the immunogenici

A Comprehensive Review on Crustaceans' Immune System With a Focus on Freshwater Crayfish in Relation to Crayfish Plague Disease.

Freshwater crayfish immunity has received great attention due to the need for urgent conservation. This concern has increased the understanding of the cellular and humoral defense systems, although the regulatory mechanisms involved in these processes need updating. There are, however, aspects of the immune response that require clarification and integration. The particular issues addressed in this review include an overall description of the oomycete Aphanomyces astaci, the causative agent of the pandemic plague disease, which affects freshwater crayfish, and an overview of crustaceans' immun

Stub1 promotes chaperone-mediated autophagy to suppress antiviral immunity.

The E3 ubiquitin ligase Stub1 (CHIP) is a core regulator of protein homeostasis and antiviral innate immunity, with established roles in targeting RIG‑I and MAVS for proteasomal or autophagic degradation. Here, we summarize our recent work revealing that Stub1 negatively regulates type I interferon (IFN‑I) production by driving chaperone‑mediated autophagy (CMA)-dependent degradation of TBK1. Stub1 directly interacts with TBK1 and catalyzes K27‑linked polyubiquitination at lysine 344 (K344) of TBK1, which enables recognition by the CMA chaperone HSC70 via a conserved KFDKQ CMA recognition moti

Transcriptomics reveals different response mechanisms of Litopenaeus vannamei hemocytes to injection of Vibrio parahaemolyticus and WSSV.

As the most important cultural crustacean species worldwide, studies about Pacific white shrimp (Litopenaeus vannamei) have received more attention. It has been well-documented that various pathogens could infect L. vannamei, resulting in huge economic losses. The studies about the responding mechanism of L. vannamei to sole pathogens such as Vibrio parahaemolyticus and white spot virus (WSSV) have been extensively reported, while the studies about the differently responding mechanisms remain unclear. In the present study, we identified the differently expressed genes (DEGs) of L. vannamei hem

Harnessing trained immunity for next-generation vaccines: from rational adjuvant design to self-adjuvanting antigens.

INTRODUCTION: The paradigm of trained immunity (TI), a de facto innate immune memory mediated by metabolic and epigenetic reprogramming of innate leukocytes, has expanded the conceptual framework for rational vaccine adjuvant design. Moving beyond conventional empiricism, TI offers a mechanistic basis that may enhance protection against certain heterologous pathogens, particularly in specific conditions. AREAS COVERED: This review provides a comprehensive analysis of next-generation adjuvants engineered to induce TI. We outline the core mechanistic foundations encompassing the PI3K/Akt/mTOR/H

Glycolipid sensing and innate immunity in paracoccidioidomycosis.

Distinct glycolipid profiles are described in microorganisms, which have been shown to modulate the innate immune system. We tested the hypothesis that glycosphingolipids from Paracoccidioides brasiliensis have immunomodulatory properties on monocytes and dendritic cells of two groups of healthy individuals, one cured of paracoccidioidomycosis in the past (CUR-I) and the other nonexposed to P. brasiliensis (HNE-I). Two classes of glycosphingolipids purified from yeast cells were evaluated: a neutral glycosphingolipid, monohexosylceramide (CMH), and acidic glycosylinositolphosphorylceramides (G

Gene family clustering identifies functionally associated subsets of human in vivo blood and tonsillar dendritic cells.

Human dendritic cells (DCs) are a distinct but heterogeneous lineage of APCs operating as the link between innate and adaptive immune responses, with the function to either maintain tolerance or trigger immunity. The DC lineage consists of several subpopulations with unique phenotypes; however, their functional characteristics and transcriptional similarities remain largely unknown. To further characterize the phenotypes and transcriptomes of the subsets, we purified myeloid CD16(+), blood DC Ag 1(+) (BDCA1(+)), and BDCA3(+) DC populations, as well as plasmacytoid CD123(+) DCs, from tonsillar

Innate immune response and hepatic inflammation.

Inflammation is a pathogenic component of various types of acute and chronic liver diseases, and it contributes to progressive liver damage and fibrosis. Cells of the innate immune system initiate and maintain hepatic inflammation though mediator production as a result of their activation by pathogen-derived products recognized by pattern recognition receptors. Innate immune cells, particularly dendritic cells, have a pivotal role in sensing pathogens and initiating adaptive immune responses by activation and regulation of T-lymphocyte responses. Although the liver provides a "tolerogenic" imm

Engineering plant tandem kinase immune receptors expands effector recognition profiles.

Plant intracellular immune receptors are widely deployed in breeding to protect crops from disease. In addition to nucleotide-binding leucine-rich repeat receptors (NLRs), tandem kinase proteins (TKPs) have recently emerged as an important family of immune receptors within staple cereal food crops, but how TKPs recognize effectors and whether they are amenable to engineering is essentially unknown. Here, we show that the barley and wheat TKPs Rmo2 and Rwt7 recognize different blast fungus effectors via their integrated HMA domains using different protein interfaces with nanomolar binding affin

Immune Biology and Persistence of Helicobacter pylori in Gastric Diseases.

Helicobacter pylori is a prevalent pathogen, which affects more than 40% of the global population. It colonizes the human stomach and persists in its host for several decades or even a lifetime, if left untreated. The persistent infection has been linked to various gastric diseases, including gastritis, peptic ulcers, and an increased risk for gastric cancer. H. pylori infection triggers a strong immune response directed against the bacterium associated with the infiltration of innate phagocytotic immune cells and the induction of a Th1/Th17 response. Even though certain immune cells seem to b

Modulation of Host Innate Immune Response by Highly Pathogenic Human Coronaviruses during Viral Infection.

Highly pathogenic human coronaviruses, including SARS-CoV, SARS-CoV-2, and MERS-CoV have emerged as significant public health threats due to their ability to cause widespread outbreaks and pandemics. These viruses induce dysregulated inflammatory responses, typified by cytokine storms that drive extensive tissue damage in pulmonary and extrapulmonary systems, leading to acute respiratory distress syndrome (ARDS) and multi-organ failure. These pathological outcomes are driven by sophisticated mechanisms that manipulate host immune pathways and evade innate and adaptive immune surveillance. The
