Bexarotene blocks calcium-permeable ion channels formed by neurotoxic Alzheimer's β-amyloid peptides.

The anticancer drug bexarotene has been shown to restore cognitive functions in animal models of Alzheimer's disease, but its exact mechanism of action remains elusive. In the present report, we have used a combination of molecular, physicochemical, and cellular approaches to elucidate the mechanisms underlying the anti-Alzheimer properties of bexarotene in neural cells. First of all, we noticed that bexarotene shares a structural analogy with cholesterol. We showed that cholesterol and bexarotene compete for the same binding site in the C-terminal region of Alzheimer's β-amyloid peptide 1-42

Epithelial immune response in Drosophila malpighian tubules: interplay between Diap2 and ion channels.

Systemic immune response via the Immune deficiency pathway requires Drosophila inhibitor of apoptosis protein 2 to activate the NF-κB transcription factor Relish. Malpighian tubules (MTs), simple epithelial tissue, are the primary excretory organs, performing additional role in providing protection to Drosophila against pathogenic infections. MTs hold a strategic position in Drosophila as one of the larval tissues that are carried over to adults, unlike other larval tissues that are histolysed during pupation. In this paper we show that Diap2 is an important regulator of local epithelial immun

Endogenous lipid-derived ligands for sensory TRP ion channels and their pain modulation.

Environmental or internal noxious stimuli excite the primary sensory nerves in our body. The sensory nerves relay these signals by electrical discharges to the brain, leading to pain perception. Six transient receptor potential (TRP) ion channels are expressed in the sensory nerve terminals and play a crucial role in sensing diverse noxious stimuli. Cation influx through activated TRP ion channels depolarizes the plasma membrane, resulting in neuronal excitation and pain. Natural and synthetic compounds have been found to act on these sensory TRP channels to alter the nociception. Evidence is

Characterization of acid-sensing ion channels in medium spiny neurons of mouse striatum.

Acid-sensing ion channels (ASICs) regulate synaptic activities and play important roles in neurodegenerative diseases. They are highly expressed in the striatum, where medium spiny neurons (MSNs) are a major population. Given that the properties of ASICs in MSNs are unknown, in this study, we characterized ASICs in MSNs of the mouse striatum. A rapid drop in extracellular pH induced transient inward currents in all MSNs. The pH value for half-maximal activation was 6.25, close to that obtained in homomeric ASIC1a channels. Based on psalmotoxin 1 and zinc sensitivity, ASIC1a (70.5% of neurons)

Effects of acamprosate on neuronal receptors and ion channels expressed in Xenopus oocytes.

BACKGROUND: Acamprosate (calcium acetylhomotaurinate) has proven to be a moderately effective pharmacological adjunct for the treatment of alcoholism. However, the central nervous system mechanism by which acamprosate reduces alcohol relapse remains unclear. Here we survey a number of metabotropic receptors, ligand-gated ion channels, and voltage-gated ion channels, to determine if acamprosate has actions at these sites in the central nervous system. METHODS: Xenopus oocytes were injected with cDNAs or cRNAs encoding metabotropic glutamate receptors 1 and 5, M1 muscarinic receptors, glycine a

Changes in cardiac myocyte morphology alter the properties of voltage-gated ion channels.

OBJECTIVE: The goal of this study was to determine if the properties of the transient outward potassium (I(to)), TTX-resistant sodium (I(Na)) and L-type calcium (I(Ca)) currents are altered during changes in cardiac cell shape. METHODS: Ventricular myocytes were isolated from 3- to 4-day-old neonatal rats and cultured on either non-aligned or aligned collagen thin gels. In contrast to the flat, stellar-shaped myocytes obtained when the cells are plated on non-aligned collagen gels, myocytes plated on aligned gels display an elongated, rod-like shape. Ion channel expression was measured using

Ion channels and ventricular arrhythmias: cellular and ionic mechanisms underlying the Brugada syndrome.

Brugada syndrome is characterized by ST segment elevation in the right precordial leads, V1-V3 (unrelated to ischemia or structural disease), normal QT intervals, apparent right bundle branch block, and sudden cardiac death, particularly in men of Asian origin. An autosomal dominant mode of inheritance with variable expression has been described. The only gene thus far linked to the Brugada syndrome is the cardiac sodium channel gene, SCN5A. The possible cellular and ionic basis for these features of the Brugada syndrome are discussed. Strong sodium channel block, among other modalities, has b

Ion permeation through light-activated channels in rhabdomeric photoreceptors. Role of divalent cations.

The receptor potential of rhabdomeric photoreceptors is mediated primarily by a Na influx, but other ions must also permeate through light-dependent channels to account for some properties of the photoresponse. We examined ion conduction in macroscopic and single-channel light-induced currents of slug and scallop photoreceptors. In the absence of Na, a fivefold change in extracellular K shifted the reversal voltage of the photocurrent (Vrev) by approximately 27 mV. Because the dependency of Vrev on [K]o was sub-Nernstian, and Vrev in each condition was more positive than Ek, some other ion(s)

Intracellular Na+ modulates the cAMP-dependent regulation of ion channels in the heart.

The cAMP-dependent regulation of ion channels was studied by using the whole-cell configuration of the patch clamp technique. In isolated cardiac ventricular myocytes, the beta-adrenergically regulated Cl- current (ICl) exhibited an unusual dependence on Na+, such that replacement of extracellular Na+ with compounds such as tetramethylammonium, choline, Tris, or N-methyl-D-glucamine resulted in a reduction in current amplitude without changing the reversal potential. Replacement of extracellular Na+ with tetramethylammonium also reduced the magnitude of the beta-adrenergically enhanced Ca2+ cu

Blockade by neurotransmitter antagonists of veratridine-activated ion channels in neuronal cell lines.

The voltage-dependent Na+ ionophore of various neuronal cells is permeable not only to Na+ ions but also to guanidinium ions. Therefore, the veratridine- (or aconitine-)stimulated influx of [14C]guanidinium in neuroblastoma x glioma hybrid cells was measured to characterize the Na+ ionophore of these cells. Half-maximal stimulation of guanidinium uptake was seen at 30 microM veratridine. At 1 mM guanidinium, the veratridine-stimulated uptake of guanidinium was lowered to 50% by approximately 60 mM Li+, Na+, or K+ and by a few millimolar Mn2+, Co2+, or Ni2+. The basal, as well as the veratridin

Ion channels and membrane potential in stimulus-secretion coupling in adrenal medulla cells.

The role of Na+ channels and membrane potential in stimulus secretion coupling in adrenal medulla cell cultures was investigated. Veratridine, aconitine, batrachotoxin (BTX), and scorpion venom, which increase the flux of ions through tetrodotoxin(TTX)-sensitive Na+ channels, all evoke secretion of catecholamines that is blocked by TTX. TTX partially inhibits secretion induced by low concentrations of nicotine in Locke's solution but has no effect on high concentrations of nicotine (20 microM). In Ca2+-sucrose media TTX has no effect on secretion at either high or low concentrations of nicotin

Model topology determines parameter accessibility in Markov models of voltage-gated ion channels.

Markov models are widely used to describe the gating kinetics of voltage-gated ion channels, but increasing model complexity can introduce parameters that are difficult to constrain from macroscopic measurements. In this study, global variance-based Sobol sensitivity analysis was used to examine how model topology, stimulation protocol, and parameter uncertainty shape the accessibility of kinetic parameters in voltage-gated ion channel models. Progressively more complex Markov schemes were analyzed, beginning with a two-state closed-open model, extending to a three-state linear closed-closed-o

Systematic Profiling Reveals Propensity of Human Ion Channels for Liquid-Liquid Phase Separation.

Liquid-liquid phase separation (LLPS), a fundamental biophysical process driven by multivalent weak interactions, regulates diverse cellular physiological functions. Ion channels are critical transmembrane protein complexes governing electrophysiological signaling, yet their phase separation characteristics remain largely uncharacterized. Here, we systematically assessed the phase-separation propensity of 140 human ion-channel proteins from the PhaSePred database, and 69 human ion channel proteins were predicted to have a high propensity for LLPS (prediction score ≥ 0.5). Based on the self-ass

Beyond Ion Channels: Emerging Roles of FGF12 in Cellular Regulation and Cancer Progression.

Fibroblast growth factor 12 (FGF12), a member of the intracellular fibroblast growth factor homologous factor (iFGF) subfamily, has been widely studied for its role in the modulation of voltage-gated ion channels. However, recent studies suggest that FGF12 possesses various cellular functions beyond ion channel regulation, particularly in cancer progression. Accumulating evidence indicates that the upregulation of FGF12 is associated with tumor survival, therapeutic resistance, and poor prognosis through signaling pathways independent of its canonical ion channel interactions. This review summ

Mechanosensitive TMEM63 Ion Channels: Structure, Gating Mechanism, and Emerging Physiological and Pathological Roles.

The TMEM63 family of mechanosensitive ion channels has recently emerged as a distinct group of mammalian force sensors evolutionarily related to plant OSCA channels. Structural and functional studies establish TMEM63s as monomeric, high-threshold mechanotransducers featuring an extended IL2 domain, a hydrophobic latch at the membrane interface, and a lateral lipid-accessible pore that may enable both ion conduction and lipid scrambling. These features define a unique gating mechanism, distinct from other mechanosensitive ion channels. Growing genetic and animal evidence indicates that TMEM63s

Mechanosensitive Ion Channels and Neuroplasticity in Tuina Therapy: Molecular Mechanisms and Therapeutic Implications.

Tuina is a traditional Chinese manual therapy distinguished from generic massage by its acupoint- and meridian-based manipulations, and it is commonly used in the management of neurological and musculoskeletal disorders, although its underlying biological mechanisms remain incompletely defined. This review summarizes current experimental and clinical evidence on the neurobiological processes associated with Tuina therapy, with a focus on pain modulation and neural repair. Available evidence suggests that mechanical stimulation during Tuina may engage mechanosensitive ion channels, including tr

Ion Channels and Dry Eye Disease: From Physiological Functions to Targeted Therapeutic Mechanisms.

Dry eye disease, as an ocular surface disorder, affects millions of people globally and has consequently become one of the most prevalent conditions encountered in ophthalmic clinics. The pathogenesis of dry eye involves several key factors, including tear film instability, hyperosmolarity, chronic ocular surface inflammation, and neurosensory abnormalities. The ocular surface expresses a diverse array of ion channels, which play essential physiological roles in maintaining tear film stability, regulating osmotic pressure, balancing ion transport, and modulating inflammatory responses.
