[Gerstmann-Sträussler-Scheinker disease. Pathologal and genetic study].

Gerstmann-Sträussler-Scheinker's disease is a familial spongiform encephalopathy whose pathological hallmark is the existence--especially in the cerebellum--of numerous amyloid plaques. We report here the third clinicopathological case in a French family. Brain tissue from one of its members--initially described as familial Creutzfeldt-Jakob's disease--has been reported as successfully inoculated to monkeys. We present the currently accumulating data favouring the hypothesis of a common etiology for familial Creutzfeldt-Jakob's disease and Gerstmann-Sträussler-Scheinker's disease. The familial

Prion-induced ferroptosis is facilitated by RAC3.

Prions are infectious agents that initiate transmissible spongiform encephalopathies, causing devastating neuronal destruction in Creutzfeldt-Jakob and Kuru disease. Rapid cell death depends on presence of the endogenous prion protein PrPC, but its mechanistic contribution to pathogenesis is unclear. Here we investigate the molecular role of PrPC, reactive oxygen species and lipid metabolism in ferroptosis susceptibility, a regulated cell death process characterized by lipid peroxidation. We discover that elevated expression of the cellular prion PrPC creates a relaxed oxidative milieu that fa…

Induction of antibodies against human prion proteins (PrP) by DNA-mediated immunization of PrP0/0 mice.

Prion diseases are neurodegenerative disorders, affecting humans and animals. The human diseases include kuru, Creutzfeldt-Jakob disease (CJD), Gerstmann-Sträussler-Scheinker syndrome (GSS), and fatal familial insomnia (FFI). To generate monospecific antisera against human prion proteins we have immunized mice with DNA coding for different human prion proteins. We constructed immunization vectors expressing individual genotypes of either the cellular prion gene (PRNP) or mutant forms under appropriate promoters. This approach avoids the preparation of infectious material for immunization. To c…

[Bovine spongiform encephalopathy: a review].

Transmissible Spongiform Encephalopathies are a group of chronic, always fatal diseases affecting the central nervous system of humans and animals. They occur in all species and are probably caused by agents called prions. In this minireview, a first part provides an overview of the various disease forms, a second part is devoted to the molecular biology of transmissible spongiform encephalopathies, and a last part deals with the specific problems of the Bovine Spongiform Encephalopathy.

Extensive cortical spongiform changes with cerebellar small amyloid plaques: the clinicopathological case of MV2K+C subtype in Creutzfeldt-Jakob disease.

We report a clinical case report of the MV2K+C subtype of sporadic Creutzfeldt-Jakob disease (sCJD). The patient was a 72-year-old woman who … for the typical MV2 subtype of sCJD. The other intermediate band might exist in the cerebellum with kuru plaques. Therefore, small amyloid plaques in the cerebellum can be crucial for MV2K+C subtype.

Sod1 deficiency reduces incubation time in mouse models of prion disease.

Prion infections, causing neurodegenerative conditions such as Creutzfeldt-Jakob disease and kuru in humans, scrapie in sheep and BSE in cattle are characterised by prolonged and variable incubation periods that are faithfully reproduced in mouse models. Incubation time is partly determined by genetic factors including polymorphisms in the prion protein gene. Quantitative trait loci studies in mice and human genome-wide association studies have confirmed that multiple genes are involved. Candidate gene approaches have also been used and identified App, Il1-r1 and Sod1 as affecting incubation t…

Western blot mapping of disease-specific amyloid in various animal species and humans with transmissible spongiform encephalopathies using a high-yield purification method.

SAF-protein, an amyloid, is the main constituent of scrapie-associated fibrils (SAF) and a specific marker for transmissible spongiform encephalopathies (TSE). Using an improved extraction method and Western blot detection, the disease-specific amyloid was found in various parts of the central nervous system of hamsters orally infected with scrapie, of squirrel monkeys orally infected with kuru, sporadic Creutzfeldt-Jakob disease (CJD) and scrapie, of human patients with sporadic CJD, of a sheep with natural scrapie and of a cow with bovine spongiform encephalopathy (BSE). In human CJD samples…

Comparative sequence analysis, in vitro expression and biosynthesis of mouse PrP.

A number of studies have indicated that an endogenous brain protein, PrP, is associated with transmissible agents causing spongiform encephalopathies such as scrapie, kuru, and Creutzfeldt-Jakob disease. It has been proposed that PrP derived from scrapie brain is the scrapie agent itself. To test directly whether the PrP mRNA in scrapie brain tissue can encode the scrapie agent, we expressed PrP cDNA cloned from scrapie-infected mouse brain in vitro. The expressed PrP did not transmit scrapie to susceptible mice. Thus either PrP is not the scrapie agent, or the expressed PrP requires additiona…

Distinct transmissibility features of TSE sources derived from ruminant prion diseases by the oral route in a transgenic mouse model (TgOvPrP4) overexpressing the ovine prion protein.

Transmissible spongiform encephalopathies (TSEs) are a group of fatal neurodegenerative diseases associated with a misfolded form of host-encoded prion protein (PrP). Some of them, such as classical bovine spongiform encephalopathy in cattle (BSE), transmissible mink encephalopathy (TME), kuru and variant Creutzfeldt-Jakob disease in humans, are acquired by the oral route exposure to infected tissues. We investigated the possible transmission by the oral route of a panel of strains derived from ruminant prion diseases in a transgenic mouse model (TgOvPrP4) overexpressing the ovine prion protei…

Discriminant analysis of prion sequences for prediction of susceptibility.

Prion diseases, including ovine scrapie, bovine spongiform encephalopathy (BSE), human kuru and Creutzfeldt-Jakob disease (CJD), originate from a conformational change of the normal cellular prion protein (PrP(C)) into abnormal protease-resistant prion protein (PrP(Sc)). There is concern regarding these prion diseases because of the possibility of their zoonotic infections across species. Mutations and polymorphisms of prion sequences may influence prion-disease susceptibility through the modified expression and conformation of proteins. Rapid determination of susceptibility based on prion-seq…

[Human spongiform encephalopathies. Diseases caused by prions].

Spongiform encephalopathies or prion diseases are common denominators for a group of diseases, all fatal, which show characteristic neuropathological changes. In man the group includes four diseases: kuru, Creutzfeldt-Jakob's disease (CJD), Gerstmann-Sträussler-Scheinker's disease (GSS) and fatal familial insomnia (FFI). In animals it comprises the following six: scrapie (sheep and goats), transmissible mink encephalopathy (mink), chronic wasting disease (mule/elk), exotic ungulate encephalopathy (antelopes) and feline and bovine spongiform encephalopathy (cats and cattle). The diseases can be…

[Slow virus infection in the field of obstetrics and gynecology--with special reference to HBV, HTLV-1 and HCV].

The slow virus infection (SVI) established by Gajdusek DC in 1964 has been known to involve not only Kuru or Creutzfeldt-Jakob disease but also hepatitis B virus (HBV) infection and very recently human T-cell lymphotropic virus type 1 (HTLV-1) or hepatitis C virus (HCV) infection. These all viruses potentially develop serious, irreversible disease, ie, hepatoma or adult T-cell leukemia, after long latent periods. HBV, HTLV-1 and HCV can be transmitted vertically from carrier mothers to their offspring, and therefore, are serious SVIs in the field of obstetrics. HB immunoglobulin (HBIG) and HB …

From slow virus to prion: a review of transmissible spongiform encephalopathies.

Spongiform encephalopathies include seven neurodegenerative diseases: three in man (Creutzfeldt-Jakob disease, Gerstmann-Sträussler-Scheinker disease and kuru) and four in animals (scrapie, mink encephalopathy, bovine spongiform encephalopathy and chronic wasting disease in deer and elks). They are all transmissible to a variety of species, and man-to-man propagation of the diseases in the form of iatrogenic transmission has been well-documented. The infectious agent is highly unusual and the pathogenesis of infection remains controversial. The term prion was introduced to describe the protein…
