We have studied the regulation of DNA ligase I gene expression in UV-C irradiated human primary fibroblasts. An increase of approximately 6-fold both in DNA ligase I messenger and activity levels was observed 24 h after UV treatment, when nucleotide excision repair (NER) is no longer operating. DNA ligase I induction is serum-independent and is controlled mainly by the steady-state level of its mRNA. The activation is a function of the UV dose and occurs at lower doses in cells showing UV hypersensitivity. No increase in replicative DNA polymerase alpha activity was found, indicating that UV i

We have previously shown that Mu restores an active DNA replication at non-permissive temperature in E. coli K12 ligts7 strains. In this paper we describe two new types of phage mutants for the Mu lig function. The Mu ligts mutants are conditional lethals defective for both integration and replication of DNA, unable to 'complement' the bacterial lig mutation; the map between B and lys. The mutants of the other type, on the other hand, are able to restore to a maximum level the activity impaired by the ligts7 mutation in the host. We suggest the hypothesis that the product of Mu lig gene could

We previously determined that ghrelin synthesis was downregulated after burn injury and that exogenous ghrelin retained its ability both to stimulate food intake and to restore plasma growth hormone levels in burned rats. These observations and the finding that anabolic hormones can attenuate skeletal muscle catabolism led us to investigate whether ghrelin could attenuate burn-induced skeletal muscle protein breakdown in rats. These studies were performed in young rats (50-60 g) 24 h after approximately 30% total body surface area burn injury. Burn injury increased total and myofibrillar prote

A pair of degenerate oligodeoxyribonucleotides was used to amplify, by the polymerase chain reaction (PCR), DNA fragments internal to genes encoding D-Ala:D-Ala ligase-related proteins of vancomycin-resistant (VmR) Enterococcus faecalis V583. Cloning and nucleotide sequencing of the PCR products indicated that fragments of two genes, designated vanB and ddl, were co-amplified. The vanB gene was previously shown to be present in Enterococcus strains expressing VanB-type VmR [Quintiliani Jr. et al., J. Infect. Dis. 8 (1993) 943-950]. The ddl gene was detected by Southern hybridization in all VmR

Thyroid transcription factor 1 (TTF1) regulates the tissue-specific expression of genes. However, the molecular regulation of TTF1 in thyroid normal and carcinoma cells has not been revealed. Here we identify 2 distinct ubiquitin E3 ligases that are responsible for TTF1 degradation in normal thyroid cells and carcinoma cells, respectively. Phorbol myristate acetate induced TTF1 protein degradation in the ubiquitin-proteasome system in both HTori3 thyroid follicular epithelial cells and follicular thyroid carcinoma 133 (FTC133) cells. Lysine 151 residue was identified as a ubiquitin acceptor si

Viral late domains are used by many viruses to recruit the cellular endosomal sorting complex required for transport (ESCRT) to mediate membrane scission during viral budding. Unlike the P(S/T)AP and YPX(1-3)L late domains, which interact directly with the ESCRT proteins Tsg101 and ALIX, the molecular linkage connecting the PPXY late domain to ESCRT proteins is unclear. The mammarenavirus lymphocytic choriomeningitis virus (LCMV) matrix protein, Z, contains only one late domain, PPXY. We previously found that this domain in LCMV Z, as well as the ESCRT pathway, are required for the release of

TRIM22 is a TRIM family E3 ubiquitin ligase whose expression is strongly induced by interferons. For many years, it has been identified primarily as an antiviral effector, a role that remains well validated. A growing body of cancer-related work now forces us to expand that view considerably. Depending on tumor type, upstream signaling context, and the specific substrates available in a given cell, TRIM22 can either constrain malignancy or actively promote it. In breast, colorectal, ovarian, endometrial, hepatocellular, cervical, and gastric cancers, as well as in certain lung tumor subsets, T

Cardiovascular disease, the leading cause of death worldwide, imposing an enormous economic burden on society. Histone deacetylase 6 (HDAC6), a member of the class IIb HDAC family, contains two tandem deacetylase domains. Recent study reported that the first deacetylase domain of HDAC6 exerts E3 ligase activity. It is known that HDAC6 participates in a variety of cellular activities including microtubule stability, epithelial homeostasis and autophagy by modulating its specific non-histone substrates. However, the role of HDAC6 and its selective inhibitors in cardiovascular disease remains unc

Singapore grouper iridovirus (SGIV) and red-spotted grouper nervous necrosis virus (RGNNV) are major pathogens that cause high mortality and substantial economic losses in grouper aquaculture. The role of ring finger protein 5 (RNF5), an E3 ubiquitin ligase, in the antiviral innate immunity of teleosts remains poorly understood. In this study, a homolog of the RNF5 gene was identified and functionally characterized from Epinephelus coioides, which was designated EcRNF5. The open reading frame (ORF) of EcRNF5 is 645 nucleotides in length, encoding a 214-amino acid protein that contains a charac

Acute myeloid leukemia (AML) is a challenging hematological malignancy characterized by poor clinical outcomes. This study investigates the role of MYCBP2 in AML progression, focusing on its interaction with KIF14 and its involvement in ubiquitin-mediated processes. We found that MYCBP2 is overexpressed in AML samples compared to normal tissues, with high expression correlating with adverse clinical outcomes. Knockdown of MYCBP2 using siRNA significantly inhibited cell proliferation and promoted apoptosis in MOLM-13 and HL-60 AML cell lines. Flow cytometry revealed that MYCBP2 knockdown leads

Temozolomide (TMZ) resistance in glioblastoma (GBM) is often linked to high MGMT protein levels, but MGMT-deficient tumors can still acquire resistance, implicating MGMT-independent repair pathways. Here, we identify an oxidative stress-responsive LMO7-ALKBH2 ubiquitination mechanism that regulates ALKBH2 stability and modulates alkylation damage tolerance in GBM. The E3 ligase LMO7 directly binds ALKBH2 and promotes K48-linked polyubiquitination and proteasomal degradation of ALKBH2, with Lys7 serving as a major ubiquitin acceptor site. Oxidative stress enhances LMO7-dependent ALKBH2 ubiquiti

Amyotrophic lateral sclerosis is a fatal neurodegenerative disease characterized by the cytoplasmic mislocalization, aberrant phosphorylation and pathological aggregation of TDP-43, a nuclear RNA-binding protein essential for RNA metabolism. Despite its central involvement in ALS pathogenesis, the molecular mechanisms that govern TDP-43 proteostasis remain incompletely understood. Here, we investigated the role of LRSAM1, an E3 ubiquitin ligase, in regulating TDP-43 localization and degradation. Postmortem spinal cord tissues from five patients with sporadic amyotrophic lateral sclerosis and f

As the most common chronic liver disease, MASLD can progress to metabolic dysfunction-associated steatohepatitis (MASH) driven by accumulated metabolic and inflammatory stresses. We previously reported that liver ChREBPα protein is markedly downregulated in mouse models of diet-induced MASH and hepatotoxin-induced liver injury. Yet the impact of stress pathways on hepatocyte ChREBPα proteolysis has not been examined. Here, we show that a combined metabolic (palmitate, PA) and inflammatory (TNFα) stress signal promotes ubiquitination and proteasome-mediated degradation of ChREBPα in hepatocytes

E3 ubiquitin ligases include hundreds of members that can regulate the half-life of other proteins but also modulate their cellular localization and functions. While MARCH1 appears to target principally immune cell components, such as MHC class II molecules and the co-stimulatory molecule CD86, the repertory of its targets remains to be fully documented. Here, we adapted a proximity-dependent biotin identification (BioID)-based screening approach in live HEK293 cells. We transfected a fusion protein consisting of mouse MARCH1 linked to YFP at its N-terminus and to the biotin ligase of Aquifex

BACKGROUND AND AIMS: Metabolic associated fatty liver disease (MAFLD) is closely associated with metabolic disorders, including central obesity, dyslipidaemia, hypertension, hyperglycaemia and persistent abnormalities of liver function tests. Metabolic dysfunction-associated steatohepatitis (MASH) is a progressive form of MAFLD, characterized by chronic inflammation and accumulation of fat in liver tissue. Currently, no pharmacological interventions specifically tailored for MASH are approved. Signal transducer and activator of transcription 1 (STAT1) is the key transcription factor of the JAK

Mahogunin Ring Finger 1 (MGRN1) is a multifunctional E3 ubiquitin ligase with broad biological significance and belongs to a small group of membrane-tethered E3s capable of regulating signaling receptors at the plasma membrane. Studies in mice first revealed its physiological importance, as loss of Mgrn1 leads to a wide range of phenotypes, including abnormal pigmentation, congenital malformations, and neurodegeneration. Remarkably, MGRN1 localizes to multiple cellular compartments, including the plasma membrane, mitochondria, nucleus, and endo-lysosomal pathway. MGRN1 is also involved in seve

Targeted protein degradation (TPD), particularly proteolysis-targeting chimeras (PROTACs), has emerged as a powerful strategy for eliminating disease-associated proteins through the ubiquitin-proteasome system. However, current PROTACs rely heavily on a limited number of E3 ligases, mainly CRBN and VHL, due to the scarcity of suitable ligands. This review summarizes the development of classical small-molecule E3 ligase ligands and highlights emerging biological recruitment strategies involving antibodies, nanobodies, aptamers, peptides, and engineered proteins. These approaches expand the acce

The ubiquitin-proteasome system contributes to protein quality control, involving E3 ligases that ubiquitinate proteins and leading to their degradation. The dysregulation of protein degradation results in the abnormal accumulation of proteins and is implicated in the pathology of diverse diseases, making targeted protein degradation a promising therapeutic strategy. Here, we focus on RFFL, an endosome-associated RING E3 ligase involved in mitochondrial homeostasis and the clearance of misfolded cystic fibrosis transmembrane conductance regulator proteins. Using label-free quantitative mass sp

Epigenetic modulators of the histone deacetylase (HDAC) family control key biological processes and are frequently dysregulated in cancer. There is superior activity of HDAC inhibitors (HDACi) in patients with myeloproliferative neoplasms (MPNs) that carry the Janus kinase-2 point mutant JAK2V617F. This constitutively active tyrosine kinase activates signal-transducer-and-activator-of-transcription (STAT) transcription factors to promote cell proliferation and inflammatory processes. We reveal that the inhibition of HDAC1/HDAC2 with the clinically advanced HDACi romidepsin, the experimental HD
