Mechanism and effects of Zearalenone on mouse T lymphocytes activation in vitro.

Zearalenone (ZEA) is particularly toxic to the female reproductive system. Nevertheless, the effect of ZEA on the immune system is still not fully understood. The following study investigates the effects and mechanism of ZEA on mouse T cell activation in vitro. Briefly, T lymphocytes were extracted from primary splenic lymphocyte in mice, activated by concanavalin A, and then were exposed to different concentrations of ZEA for a certain period of time. Flow cytometry was used to detect the expression of activating and co-stimulatory molecules, and the secretion of cytokines in T cells at vario

In Situ Activation of Pituitary-Infiltrating T Lymphocytes in Autoimmune Hypophysitis.

Autoimmune hypophysitis (AH) is a chronic inflammatory disease characterized by infiltration of T and B lymphocytes in the pituitary gland. The mechanisms through which infiltrating lymphocytes cause disease remain unknown. Using a mouse model of AH we assessed whether T lymphocytes undergo activation in the pituitary gland. Infiltrating T cells co-localized with dendritic cells in the pituitary and produced increased levels of interferon-γ and interleukin-17 upon stimulation in vitro. Assessing proliferation of CD3- and B220-postive lymphocytes by double immunohistochemistry (PCNA-staining) a

Impaired lymphocyte trafficking in mice deficient in the kinase activity of PKN1.

Knock-in mice lacking PKN1 kinase activity were generated by introducing a T778A point mutation in the catalytic domain. PKN1[T778A] mutant mice developed to adulthood without apparent external abnormalities, but exhibited lower T and B lymphocyte counts in the peripheral blood than those of wild-type (WT) mice. T and B cell development proceeded in an apparently normal fashion in bone marrow and thymus of PKN1[T778A] mice, however, the number of T and B cell counts were significantly higher in the lymph nodes and spleen of mutant mice in those of WT mice. After transfusion into WT recipients,

Circulating B-lymphocytes as potential biomarkers of tuberculosis infection activity.

Accurate biomarkers of Mycobacterium tuberculosis infection activity would significantly improve early diagnosis, treatment and management of M. tuberculosis infection. We hypothesised that circulating B-lymphocytes may be useful biomarkers of tuberculosis (TB) infection status in highly TB-endemic settings. Ex-vivo and in-vitro mycobacteria-specific B-cell ELISPOT assays were used to examine the plasmablast (PB) and memory B-cell (MBC) responses in the peripheral blood of adult, healthy, community controls (n = 151) and of active TB patients (n = 48) living in Uganda. Frequencies of mycobacte

Local activation and systemic dysregulation of T lymphocytes in Sjögren's syndrome.

T cells are implicated in both local and systemic pathophysiology of primary Sjögren's syndrome (PSS). Lymphocytic infiltrates in exocrine glands are dominated by CD4+ T cells, some contributing to ectopic lymphoid tissue, others, unusually, exhibiting cytotoxic potential. Cytokine secretion patterns are complex, with Th1 and Th17 components implicated in pathology. Circulating T cells exhibit phenotypes consistent with hyperactivation, cytokine imbalance, and homeostatic alterations; CD4 lymphopenia is recognized as a risk factor for developing lymphoma. Evidence of oligoclonal expansion is f

Homing commitment of lymphocytes activated in the human gastric and intestinal mucosa.

BACKGROUND: Gastric infection with the human pathogen Helicobacter pylori results in a large accumulation of IgA and IgM secreting cells in the gastric mucosa. The molecular mechanisms resulting in B cell migration to the gastric mucosa in H pylori infection are however not known. AIMS: To examine expression of the mucosal homing receptor integrin alpha4beta7 and the homing receptor for secondary lymphoid tissues, L-selectin, on lymphocytes activated by gastric, intestinal, or systemic antigens. Furthermore, to examine gastric expression of the mucosal addressin cellular adhesion molecule 1 (

Macrophage-lymphocyte interactions mediate anti-Burkholderia pseudomallei activity.

The mechanisms involved in the pathogenesis of melioidosis, caused by the intracellular bacterium Burkholderia pseudomallei, are unclear. C57BL/6 mice are resistant to infection, while BALB/c mice are highly susceptible. Previous studies have demonstrated that peritoneal exudate cell preparations enriched for macrophages are capable of effectively eliminating intracellular pathogens. In this study we present evidence showing that interaction of macrophages with lymphocytes is necessary for efficient anti-B. pseudomallei activity.

[The role of the killer activity of the peripheral blood lymphocytes in the pathogenesis of rheumatism].

A total of 49 patients with rheumatism running a sluggish course, were examined, as were those with decompensated tonsillitis, systemic lupus erythematosus, and 20 essentially healthy subjects. In inactive rheumatism an increase in the killer activity of lymphocytes and augmentation of amounts of Fc+lymphocytes were revealed, which values were significantly different from those in the other groups. The detected changes in the activity of lymphocytes may serve as differential-diagnostic criteria.

Proliferative activity of epidermal and dermal T lymphocytes in mycosis fungoides.

BACKGROUND: Mycosis fungoides (MF) is histologically characterized by an accumulation of T lymphocytes (T cells) in both epidermis and dermis. However, the mechanism of the formation of Pautrier's microabscess or the maintenance of the epidermal T cells has not yet been clarified. OBJECTIVE: The rate of proliferation of these T cells was investigated. METHODS: The formalin-fixed, paraffin-embedded sections from 9 cases of patch/plaque stage MF were immunostained with proliferating cell nuclear antigen (PCNA). RESULTS: The PCNA-positive T cell nuclei were found both in the epidermis (includi

Changes in free cytoplasmic magnesium following activation of human lymphocytes.

Activation of lymphocytes with 10 microM ionomycin leads to a rapid increase in the concentration of free cytoplasmic calcium ([Ca2+]i) and, at a slower rate, also to an increase in the cytoplasmic free magnesium concentration ([Mg2+]i). The ionomycin-induced Mg(2+)-mobilization response is dependent on the influx of extracellular Ca2+. After receptor-mediated lymphocyte activation, induced by mitogens or anti-receptor antibodies, a Mg(2+)-mobilization response does occur in a small fraction of the cells. Simultaneous measurement of [Ca2+]i and [Mg2+]i in individual cells showed that the recep…

Lymphocyte and macrophage subsets in active and inactive lesions of lichen planus.

Lichen planus (LP) is a mucocutaneous disease for which the etiology and pathogenesis are poorly understood. We performed an immunohistochemical study on formalin-fixed tissue sections of 10 cases of LP using subsets of antibodies to lymphocytes (LCA, CD3, OPD4-CD4, L26, LN1 and Leu-7), and monocyte-macrophages [lysozyme, KP1-Mac, Factor XIIIa (FXIIIa) and S-100 protein]. Six cases showed typical histological features of active LP, two cases showed features of active and inactive LP, and two cases showed only inactive LP. In active LP, scattered T cells (CD3+ and pan T cells) were present in t

Relationship between CD5+ B lymphocytes and the activity of systemic autoimmunity.

We studied the relationship between CD5+ B cells and the activity of the disease process in patients with autoimmune diseases. In rheumatoid arthritis (RA), levels of CD5+ B cells were associated with autoantibody production as determined by serum rheumatoid factor and antinuclear antibodies. In addition, CD5+ B cells were significantly correlated with C-reactive protein, and data from longitudinal studies showed a marked influence of corticosteroid treatment on numbers of CD5+ B cells. Patients with systemic lupus erythematosus (SLE) had slightly elevated levels of CD5+ B cells as compared wi

Activation of peripheral blood lymphocytes in patients with primary Sjögren's syndrome.

The peripheral blood from 15 patients with primary Sjögren's syndrome and from 15 matched controls was examined for the presence of activated T and B lymphocytes, by using monoclonal antibodies directed to interleukin-2 (IL-2) and transferrin receptors, and to HLA-DR determinants. The number of circulating positive-T cells was significantly greater in the patients than in the controls, irrespective of disease activity. There were more of the CD8 cells than of the CD4 cells that expressed IL-2 receptors. There was a small but significant increase in activated B cells in the patients, since this

Human T-lymphocyte-derived megakaryocyte colony-stimulating activity.

Conditioned medium from a T-lymphoblastic cell line (Mo) contains a number of well-characterized hemopoietins. In this paper we demonstrate that Mo cells also release a factor(s) able to stimulate the growth and the differentiation of megakaryocytic progenitors into large-size pure megakaryocytic colonies in plasma clot cultures. Comparison with other sources of human-active hemopoietins shows that Mo-conditioned medium performs better than others, especially for the megakaryocytic lineage. The factor(s) shows strong similarities with human Meg-CSF obtained from a thrombocytopenic patient's pl

Products of activated lymphocytes stimulate Schwann cell mitosis in vitro.

Culture of Schwann cells and endoneurial fibroblasts from newborn rat sciatic nerves in the presence of supernatants obtained from concanavalin A (Con-A)-stimulated rat mononuclear cells resulted in proliferation of both cell types. Con-A did not induce Schwann cell or fibroblast proliferation. Supernatant from a Gibbon T-cell lymphoma and chromatographically purified rat interleukin-2 (IL-2) induced fibroblast but not Schwann cell proliferation, and cloned human IL-2 did not induce proliferation of either cell type. Helper and suppressor activities of lymphocyte subsets on antithyroglobulin production in vitro.

We studied the regulatory activities of T cells on specific antithyroglobulin (anti-Tg) and nonspecific immunoglobulin secretion in cultures of peripheral blood lymphocytes (PBL) of five patients with autoimmune thyroid diseases and high levels of serum anti-Tg. PBL were separated into a non-T population, including B-cells and monocytes, and a T-cell population by rosetting with sheep red cells. T-Cells were further separated into T helper (Th) and T suppressor (Ts) subsets by a panning technique using the monoclonal antibodies anti-Leu 3a and anti-Leu 2a, respectively. The three sets of cells

Acetylcholinesterase (AchE) activity of lymphocytes in chronic lymphoid leukemia (CLL).

The specific AchE (EC 3.1.1.7) activity of lymphocytes from the peripheral blood of normal donors was determined. On Leukopak filter the isolated T lymphocytes showed activity, whereas in stepwise Percoll gradient, population TLD displayed enzyme activity. The THD and B cells were inactive. [Szelényi J.
