Subunits of the epithelial sodium channel family are differentially expressed in the retina of mice with ocular hypertension.

Glaucoma is a prevalent cause of blindness, resulting in the apoptotic death of retinal ganglion cells and optic nerve degeneration. The disease is often associated with elevated intraocular pressure, however, molecular mechanisms involved in ganglion cell death are poorly understood. To identify proteins contributing to this pathological process, we analysed the retinal gene expression of DBA/2J mice that develop an elevated intraocular pressure by the age of 6 months with subsequent ganglion cell loss. In this study, we identified subunits of the epithelial sodium channel (ENaC) family that

Stereotypic behaviors in mice selectively bred for high and low methamphetamine-induced stereotypic chewing.

RATIONALE: At high doses, methamphetamine produces repetitive stereotypic behaviors, and the degree to which this occurs is heritable. OBJECTIVES: Mice of a B6D2F2 genetic background were selectively bred for four generations for high (HMA) and low (LMA) numbers of stereotyped chewing episodes measured for 1 min at 33 min post-injection following 10 mg/kg methamphetamine (changed to 7 mg/kg for the high line and 15 mg/kg for the low line in the third selected generation to avoid ceiling and floor effects, respectively). We sought to determine whether stereotypic behaviors other than number of

Seasonal dependency of the effects of experimental stressors on tumor metastasis in mice bearing Lewis lung carcinoma.

Increasing evidence indicates that the application of stressor paradigms in experimental animals affects tumor incidence and progression. However, a high heterogeneity appears both for the animal-tumor system used and for the characteristics of the stressor employed. A high variability was observed also with the application of rotational stress, a carefully and widely characterized mild psychological stressor, to mice bearing Lewis lung carcinoma. The aim of this work has been therefore to examine the possible seasonal dependency of the effects of experimental stressors (rotational stress, for

In vitro and in vivo studies of the effect of artemether on Schistosoma mansoni.

To determine whether artemether, a derivative of the antimalarial agent qinghaosu, is therapeutically active against Schistosoma mansoni, we determined the in vitro, in vivo, and histopathologic effects of the drug on S. mansoni worms. In vitro, toxic effects of artemether on S. mansoni were not seen at concentrations of less than 100 micrograms/ml. However, in vivo, 30 and 50% reductions in the lengths of male and female worms, respectively, were observed 14 days after treatment. By 56 days worm dimensions had returned to control values. Similar reversible effects on male testes and female ov

Changes of lipid-bound sialic acid in the plasma of mice bearing benzpyrene-induced tumors.

The level of lipid-bound sialic acid (LSA) was followed in inbred C57Bl/6 mice bearing tumors induced by benzpyrene at a dose of 20 mg per kg of body weight. In the plasma of mice with macroscopically ascertained tumors, the level of LSA was increased significantly. The LSA level was also significantly increased in the plasma of suspect mice i.e., in a period when the tumor was not yet macroscopically ascertained, however, when a change was observed in the motion of the hind limb of mice to which benzpyrene was administered.

Genetic effects on male mouse kidney glucuronidase activity.

Kidney beta-glucuronidase activity in C57BL/K1 and DBA/2/K1 male mice differes about tenfold, C57 giving low and DBA high values. Another C57 subline, C57BL/6J, has slightly higher activity than C57BL/K1. There is an association between the kidney glucuronidase activity and coat color determined by the buff locus, which indicates that part of the variation is due to differences at the Gur locus. The bf allele per se raises the activity of the enzyme. The backcross distributions give evidence that at least one more locus is involved.

Effects of temporary access to environmental enrichment on measures of laboratory mouse welfare.

Laboratory mice are typically housed in "shoebox" cages with limited opportunities to engage in natural behaviour. Temporary access to environments with increased space and complexity (playpens) may improve mouse welfare. Previous work by our group has shown that mice are motivated to access and use these environments, but it is unknown how other aspects of welfare are impacted. Female C57BL/6J, BALB/cJ, and DBA/2J mice (n = 21; 7 mice per strain) were housed in mixed-strain trios and given temporary access to a large playpen with their cage mates three times per week. Control mice (n = 21; 7

Sex differences in acute delta-9-tetrahydrocannabinol (Δ9-THC) response and tolerance as a function of mouse strain.

Cannabinoids are increasingly used to alleviate pain; however, tolerance to their antinociceptive effects, including those of delta-9-tetrahydrocannabinol (Δ9-THC), may limit their therapeutic utility. With more women than men using medical cannabis for pain relief, it is crucial to understand how sex influences cannabinoid-mediated antinociception and tolerance. Though studies in rats consistently find females are more sensitive to the acute antinociceptive effects of cannabinoids, our work with mice consistently finds the converse. The present study examined whether our observed sex differen

Inhibition of Aberrant DNA Re-methylation Improves Post-implantation Development of Somatic Cell Nuclear Transfer Embryos.

Somatic cell nuclear transfer (SCNT) enables cloning of differentiated cells by reprogramming their nuclei to a totipotent state. However, successful full-term development of SCNT embryos is a low-efficiency process and arrested embryos frequently exhibit epigenetic abnormalities. Here, we generated genome-wide DNA methylation maps from mouse pre-implantation SCNT embryos. We identified widespread regions that were aberrantly re-methylated, leading to mis-expression of genes and retrotransposons important for zygotic genome activation. Inhibition of DNA methyltransferases (Dnmts) specifically

Stat1 deficiency exacerbates carditis but not arthritis during experimental lyme borreliosis.

Activation of the transcription factor Stat1 by interferon-gamma (IFN-gamma) is an important step in the development of antimicrobial effector mechanisms against many bacterial pathogens. Susceptibility to murine Lyme arthritis has been correlated with the production of several proinflammatory cytokines, especially IFN-gamma. To determine the role of IFN-mediated effector mechanisms in the development of Lyme borreliosis, we infected Stat1-deficient mice on both resistant (DBA), and susceptible (C3H) genetic backgrounds. Arthritis in Stat1(/) mice was similar to that of wild-type controls in b

Robust Axonal Regeneration Occurs in the Injured CAST/Ei Mouse CNS.

Axon regeneration in the CNS requires reactivating injured neurons' intrinsic growth state and enabling growth in an inhibitory environment. Using an inbred mouse neuronal phenotypic screen, we find that CAST/Ei mouse adult dorsal root ganglion neurons extend axons more on CNS myelin than the other eight strains tested, especially when pre-injured. Injury-primed CAST/Ei neurons also regenerate markedly in the spinal cord and optic nerve more than those from C57BL/6 mice and show greater sprouting following ischemic stroke. Heritability estimates indicate that extended growth in CAST/Ei neurons

Injectable butyrate-prodrug micelles induce long-acting immune modulation and prevent autoimmune arthritis in mice.

Short chain fatty acid (SCFAs), such as butyrate, have shown promising therapeutic potential due to their immunomodulatory effects, particularly in maintaining immune homeostasis. However, the clinical application of SCFAs is limited by the need for frequent and high oral dosages. Rheumatoid arthritis (RA) is characterized by aberrant activation of peripheral T cells and myeloid cells. In this study, we aimed to deliver butyrate directly to the lymphatics using a polymeric micelle-based butyrate prodrug to induce long-lasting immunomodulatory effects. Notably, negatively charged micelles (Neg-

Genetic background modulates behavioral impairments in R6/2 mice and suggests a role for dominant genetic modifiers in Huntington’s disease pathogenesis.

Variability and modification of the symptoms of Huntington’s disease (HD) are commonly observed in both patient populations and animal models of the disease. Utilizing a stable line of the R6/2 HD mouse model, the present study investigated the role of genetic background in the onset and severity of HD symptoms in a transgenic mouse. R6/2 congenic C57BL/6J and C57BL/6J×DBA/2J F1 (B6D2F1) mice were evaluated for survival and a number of behavioral phenotypes. This study reports that the presence of the DBA/2J allele results in amelioration or exacerbation of several HD-like phenotypes character

Different pathological phenotypes of autoimmune gastritis induced by neonatal thymectomy between BALB/c and (BALB/c x DBA/2) F1 mice: role of eosinophils in hypertrophic autoimmune gastritis.

BACKGROUND: A unique pathological feature of murine autoimmune gastritis (AIG) is its pronounced mucosal hypertrophy, which is different from human type A chronic atrophic gastritis with pernicious anemia. The aim of this study was to clarify the mechanism of gastric hypertrophy in murine AIG, especially in relation to inflammatory cells infiltrating the gastric mucosa. METHODS: Neonatally thymectomized (NTx) BALB/c and (BALB/c x DBA/2) F1 mice with gastritis were examined histologically and serologically. The T-helper (Th1/Th2) immune balance in the spleen was evaluated by intracellular cyto

The effects of the anti-enteroviral agent PTU-23 on immune response in normal mice.

The anti-enteroviral agent PTU-23 (N-phenyl-N'-3-hydroxyphenyl carbamide) was tested for an effect on humoral and cellular immune response in normal BD2F1 hybrid mice. Humoral immune response was assessed by hemolytic plaque technique and cellular immunity--by the delayed type hypersensitivity reaction and by the rate of activated lymphocytes in the mesenteric lymph nodes. PTU-23 was administered subcutaneously in doses of 200, 400 and 800 mg/kg body weight during 6 consecutive days. The immune response was recorded on day 1, 7 and 14 after the treatment. The changes observed in humoral and ce

Interactions of Thermoactinomyces vulgaris and mouse alveolar macrophages. Loss of mitogenic activity and release of suppressive prostaglandins and interleukin 1.

Interactions of mouse alveolar macrophages from three different inbred strains of mice and Thermoactinomyces vulgaris, a microbe associated with Farmer's lung disease, were studied. Alveolar macrophages were found to abolish the mitogenic activity of T. vulgaris. A prostaglandin synthesis inhibitor indomethacin, could not restore the activity. Alveolar macrophage supernatants generated by T. vulgaris treatment exerted strong suppression in secondary concanavalin A-induced lymphocyte transformation. Indomethacin partly relieved the suppression but a histamine 2 receptor blocker, cimetidine, had
