Spatiotemporally Controlled Ultrasensitive Molecular Imaging Using a DNA Computation-Mediated DNAzyme Platform.

Programming ultrasensitive and stimuli-responsive DNAzyme-based probes that contain logic gate biocomputation hold great potential for precise molecular imaging. In this work, a DNA computation-mediated DNAzyme platform that can be activated by 808 nm NIR light and target c-MYC was designed for spatiotemporally controlled ultrasensitive AND-gated molecular imaging. Particularly, the sensing and recognition function of the traditional DNAzyme platform was inhibited by introducing a blocking sequence containing a photo-cleavable linker (PC-linker) that can be indirectly cleaved by 808 nm NIR lig…

Septal dysembryoplastic neuroepithelial tumor: a comprehensive clinical, imaging, histopathologic, and molecular analysis.

BACKGROUND: Dysembryoplastic neuroepithelial tumors (DNETs) are uncommon neural tumors presenting most often in children and young adults and associated with intractable seizures. Rare midline neoplasms with similar histological features to those found in DNETs have been described near the septum pellucidum and termed "DNET-like neoplasms of the septum pellucidum." Due to their rarity, these tumors have been described in just a few reports and their genetic alterations sought only in small series. METHODS: We collected 20 of these tumors for a comprehensive study of their clinical, radiologic

Molecular imaging with neural training of identification algorithm (neural network localization identification).

Superresolution localization microscopy strongly relies on robust identification algorithms for accurate reconstruction of the biological systems it is used to measure. The fields of machine learning and computer vision have provided promising solutions for automated object identification, but usually rely on well represented training sets to learn object features. However, using a static training set can result in the learned identification algorithm making mistakes on data that is not well represented by the training set. Here, we present a method for training an artificial neural network wi

Imaging H abstraction dynamics in crossed molecular beams: O(3P) + propanol isomers.

The crossed beam reaction dynamics of ground state O(3P) atoms with propanol isomers (1-propanol and 2-propanol) have been studied for the first time using the velocity map imaging technique. The hydroxypropyl radical products, generated from H-abstraction of the secondary and tertiary C-H groups of propanol isomers, were detected via single photon ionization at 157 nm under single collision conditions with collision energies of 8 and 10 kcal mol-1. Direct rebound dynamics were suggested by the angular distributions, which show overall sideways-backward scattering but more pronounced backward

Molecular imaging of mesothelioma with (99m)Tc-ECG and (68)Ga-ECG.

We have developed ethylenedicysteine-glucosamine (ECG) as an alternative to (18)F-fluoro-2-deoxy-D-glucose ((18)F-FDG) for cancer imaging. ECG localizes in the nuclear components of cells via the hexosamine biosynthetic pathway. This study was to evaluate the feasibility of imaging mesothelioma with (99m)Tc-ECG and (68)Ga-ECG. ECG was synthesized from thiazolidine-4-carboxylic acid and 1,3,4,6-tetra-O-acetyl-2-amino-D-glucopyranose, followed by reduction in sodium and liquid ammonia to yield ECG (52%). ECG was chelated with (99m)Tc/tin (II) and (68)Ga/(69)Ga chloride for in vitro and in vivo s

Small unilamellar vesicles: a platform technology for molecular imaging of brain tumors.

Molecular imaging enables the non-invasive investigation of cellular and molecular processes. Although there are challenges to overcome, the development of targeted contrast agents to increase the sensitivity of molecular imaging techniques is essential for their clinical translation. In this study, spontaneously forming, small unilamellar vesicles (sULVs) (30 nm diameter) were used as a platform to build a bimodal (i.e., optical and magnetic resonance imaging (MRI)) targeted contrast agent for the molecular imaging of brain tumors. sULVs were loaded with a gadolinium (Gd) chelated lipid (Gd-D…

Lymphatic drainage, CTV and molecular imaging in non-small cell lung cancer.

Prognosis for non-small cell lung cancer (NSCLC) patients who have locally advanced unresectable disease is poor for persistent thoracic disease and development of distant metastasis. In view of the poor rate of local control following conventional radiation therapy, there is a great need for methods to improve its efficacy. Three-dimensional conformal radiation therapy (3DCRT) is a high precision radiotherapy able to deliver higher doses with smaller doses to the surrounding normal tissues. However, the real problem is: "what do I want to treat?" This question is addressed based on the clinic

Biomarkers for Precision Prognosis in Prostate Cancer: Imaging, Molecular, and Integrated Approaches.

Prostate cancer (PCa) is predominantly an acinar adenocarcinoma arising from the prostatic glandular epithelium, with tumor grade assessed using the International Society of Urological Pathology (ISUP) Grade Group classification, reflecting the degree of glandular differentiation and underlying molecular heterogeneity. PCa exhibits wide clinical behavior heterogeneity, ranging from indolent disease to aggressive forms with poor outcomes. Accurate prognostic assessment is, therefore, essential for guiding treatment selection and monitoring disease progression. This review examines recent advanc

Imaging and Molecular Biomarkers of PFAS-Related Vascular Aging: A Narrative Review.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants increasingly associated with cardiovascular disease. Identifying early manifestations of vascular aging before the onset of overt disease is essential for improving cardiovascular risk stratification and prevention. Emerging evidence suggests that PFAS exposure contributes to early vascular and atherosclerotic alterations detectable by imaging techniques, including increased carotid intima-media thickness (CIMT), arterial stiffness, and endothelial dysfunction. In contrast, evidence for associations with coron

Atomic-Scale Imaging Reveals Polar-π Interactions in Two-Dimensional Molecular Superlattices.

Controlling coassembly of synthetic oligomers into binary superlattices at the atomic level is challenging. We report a strategy for programming polar-π interactions in oligomeric peptoids, a class of sequence-defined peptidomimetics, facilitating the formation of homogeneous two-dimensional (2D) superlattices. N-2-phenylethyl and N-(2-perfluorophenyl)ethyl side chains, similar in size, but with contrasting electrostatic characteristics, were introduced at defined sequence positions to generate favorable dipolar aromatic interactions. The resulting nanosheets exhibit different crystal motifs d

CD38-Targeted Molecular Imaging Probes for Multiple Myeloma: Advances, Challenges, and Opportunities.

Multiple myeloma (MM), the second most common hematologic malignancy, often presents with a gradual onset and minimal symptoms in its early stages, leading to frequent misdiagnosis and delays in treatment. In recent years, radionuclide-based molecular imaging has emerged as a pivotal tool in the noninvasive evaluation and clinical management of MM, particularly in assessing the expression of CD38─a transmembrane glycoprotein that is robustly expressed on approximately 80-100% of malignant plasma cells. Notably, clinical studies have revealed a negative correlation between CD38 expression level…

Hybrid CNN-Transformer Framework for Molecular Imaging-based Early Alzheimer's Disease Detection.

PURPOSE: Hybrid convolutional neural network (CNN)-transformer models are increasingly proposed for molecular imaging, but scan-level leakage and targets derived from the same image-quantification pipeline can overstate performance. We tested whether an image-only hybrid model could classify florbetapir PET amyloid positivity under participant-disjoint validation. PROCEDURES: The Dallas Lifespan Brain Study contributed 551 longitudinal PET scans from 295 participants; 152 scans (27.6%) were amyloid-positive using the released global standardized uptake value ratio (SUVR) threshold of 1.09.
