Development of Procamallanus saccobranchi (Nematoda: Camallanidae), a parasite of a freshwater fish in India.

The development of the nematode Procamallanus saccobranchi Karve, 1952, a parasite in the stomach of the fish Heteropneustes fossilis (Bloch), was studied in Mesocyclops crassus (Fischer) and Mesocyclops leuckarti (Claus). After being ingested by the copepods the nematode first-stage larvae penetrated into the haemocoel of the intermediate host; there they moulted twice (on days 3 and 5 p.i. at 28-30 degrees C) attaining the third, infective stage. The definitive host H. fossilis acquired infection by feeding on copepods harbouring infective-stage larvae; in the stomach of this definitive host

RNAi assay in the 28-spotted ladybeetle Henosepilachna vigintioctopunctata suggests nuclear receptor HR3 as a potential molecular target for pest control.

BACKGROUND: Nuclear hormone receptors play crucial roles in embryonic development, neural regulation, metabolism, and organogenesis in insec … ndle shape. CONCLUSION: These findings indicate that HvHR3 is a critical regulator of development, molting, and reproduction, underscoring its potential as a molecular target for RNA interference-based biopesticides aimed at controlling this pest. © 2025 Society of Chemical Industry.

Osmosis as nature's method for establishing optical alignment.

For eyes to maintain optimal focus, precise coordination is required between lens optics and retina position, a mechanism that in vertebrate … histicated camera-type eyes of the predatory larvae of Thermonectus marmoratus diving beetles. Upon molting, their eye tubes elongate in less than an hour, and osmotic pressure measurements reveal that this growth is preceded by a transient increase in hemolymph osmotic pressure. The medial occipital longitudinal tract supports early stage encoding of visuospatial information.

Visuospatial learning depends on the parahippocampal place area (PPA), a functionally heterogenous area which current visuospatial processing models place downstream from parietal cortex and only from area V4 of early visual cortex (EVC). However, evidence for anatomical connections between the PPA and other EVC areas is inconsistent, and these connections are not discussed in current models. Through a data-driven analysis based on diffusion MRI tractography, we present evidence that the PPA sits at the confluence of two white matter systems. The first conveys information from the retrosplenia

SLURP-1, an endogenous α7 nicotinic acetylcholine receptor allosteric ligand, is expressed in CD205(+) dendritic cells in human tonsils and potentiates lymphocytic cholinergic activity.

Immune cells often express various nicotinic ACh receptor (nAChR) subtypes, including α7 nAChRs, as well as mRNA encoding secreted lymphocyte antigen-6/urokinase-type plasminogen activator receptor-related peptide (SLURP)-1, an endogenous α7 nAChR allosteric ligand. We detected SLURP-1 immunoreactivity in CD205(+) dendritic cells (DCs) residing in human tonsils. Phytohemagglutinin (PHA, 10 μg/ml), a T cell activator, attenuated cell proliferation and increased the ACh content of MOLT-3 human leukemic T cells compared with the vehicle control. Requirement of the Caenorhabditis elegans RapGEF pxf-1 and rap-1 for epithelial integrity.

The Rap-pathway has been implicated in various cellular processes but its exact physiological function remains poorly defined. Here we show … n lethality. The cuticle secreted by pxf-1 mutants is disorganized and can often not be shed during molting. At later stages, hypodermal degeneration is seen and animals that reach adulthood frequently die with a burst vulva phenotype. Importantly, disruption of rap-1 leads to a similar, but less severe phenotype, which is enhanced by the simultaneous rem …

The terminal differentiation factor LIN-29 is required for proper vulval morphogenesis and egg laying in Caenorhabditis elegans.

Caenorhabditis elegans vulval development culminates during exit from the L4-to-adult molt with the formation of an opening through the adult hypodermis and cuticle that is used for egg laying and mating. Vulva formation requires the heterochronic gene lin-29, which triggers hypodermal cell terminal differentiation during the final molt. lin-29 mutants are unable to lay eggs or mate because no vulval opening forms; instead, a protrusion forms at the site of the vulva. We demonstrate through analysis of genetic mosaics that lin-29 is absolutely required in a small subset of lateral hypodermal s

Zinc is incorporated into cuticular "tools" after ecdysis: the time course of the zinc distribution in "tools" and whole bodies of an ant and a scorpion.

An understanding of the developmental course of specialized accumulations in the cuticular "tools" of arthropods will give clues to the chemical form, function and biology of these accumulations as well as to their evolutionary history. Specimens from individuals representing a range of developmental stages were examined using MeV - Ion microscopy. We found that zinc, manganese, calcium and chlorine began to accumulate in the mandibular teeth of the ant Tapinoma sessile after pre-ecdysial tanning, and the zinc mostly after eclosion; peak measured zinc concentrations reached 16% of dry mass. Reduced 20-Hydroxyecdysone Signaling Modulates Antifungal Immune Responses and Growth in Lymantria dispar Against Beauveria bassiana.

20-Hydroxyecdysone (20E) is the active form of the steroid hormone ecdysone and is responsible for regulating insect development, molting, and metamorphosis. Infection with the entomopathogenic Beauveria bassiana enhances innate immunity and reduces 20E titers in Lymantria dispar. To further explore the role of the 20E signaling pathway in antifungal immunity, we combined exogenous hormone assays, proteomic profiling, and RNAi-mediated knockdown. In this study, exogenous hormone assays revealed that 20E accelerated the death of B. bassiana-infected larvae, whereas juvenile hormone (JH) markedl…

Effects of fasting on the interplay between temperature and Trypanosoma cruzi infection on the life cycle of the Chagas disease vector Rhodnius prolixus.

Rhodnius prolixus, a triatomine insect, is one of the most important vectors of Chagas disease in South America. Its interaction with the parasite Trypanosoma cruzi, the causative agent of this disease, is known to be deeply affected by ambient temperature and the nutritional status of the insect vector. In this study, we investigated how starvation affects the life cycle of R. prolixus and the population dynamics of the parasite inside the intestine of the vector at four temperatures ranging from 24°C to 30°C. The weights and molting times of chronically infected and uninfected insects were m…

Identification of potential vaccine and drug target candidates by expressed sequence tag analysis and immunoscreening of Onchocerca volvulus larval cDNA libraries.

The search for appropriate vaccine candidates and drug targets against onchocerciasis has so far been confronted with several limitations due to the unavailability of biological material, appropriate molecular resources, and knowledge of the parasite biology. To identify targets for vaccine or chemotherapy development we have undertaken two approaches. First, cDNA expression libraries were constructed from life cycle stages that are critical for establishment of Onchocerca volvulus infection, the third-stage larvae (L3) and the molting L3. A gene discovery effort was then initiated by random e…

Control of Drosophila wing size by morphogen range and hormonal gating.

The stereotyped dimensions of animal bodies and their component parts result from tight constraints on growth. Yet, the mechanisms that stop growth when organs reach the right size are unknown. Growth of the Drosophila wing-a classic paradigm-is governed by two morphogens, Decapentaplegic (Dpp, a BMP) and Wingless (Wg, a Wnt). Wing growth during larval life ceases when the primordium attains full size, concomitant with the larval-to-pupal molt orchestrated by the steroid hormone ecdysone. Here, we block the molt by genetically dampening ecdysone production, creating an experimental paradigm in

Stress responses in tropical sparrows: comparing tropical and temperate Zonotrichia.

Seasonal modulation of the adrenocortical response appears to be ubiquitous in mid- to high-latitude vertebrates but has not been investigated in tropical vertebrates. Previous studies demonstrate that temperate passerines show seasonality in corticosterone secretion and corticosteroid binding globulin capacities. We examined seasonal and sex differences in the stress response in an equatorial population of Zonotrichia capensis, the only Zonotrichia that breeds in the tropics, and compared the results with those of northern Zonotrichia. Seasonal differences in tropical Zonotrichia would presum

Release of platelet-activating factor (PAF)-like material from human lymphoid cell lines.

The release of platelet-activating factor (PAF)-like material was evaluated from eight B and T human leukemic cell lines and from three lymphoblastoid cell lines obtained after Epstein-Barr virus (EBV) transformation. All cell lines were stimulated with ionophore A23187, phytohemagglutinin (PHA), or both either in the presence of or without acetyl coenzyme A (AcCoA). After appropriate stimuli, all cell lines tested released PAF-like material, irrespective of the surface phenotype and of the level of differentiation. Some cell lines released PAF-like material after stimulation with A23187 (Molt
