The Role of BmTMED6 in Female Reproduction in Silkworm, Bombyx mori.

Transmembrane emp24 domain (TMED) proteins have been extensively studied in mammalian embryonic development, immune regulation, and signal t … n. We find that reducing TMED6 expression significantly decreases the number of eggs laid by female moths, leading to an accumulation of unlaid eggs in the abdomen. Moreover, downregulation of BmTMED6 leads to a decrease in the expression of BmDop2R1 and BmDop2R2, while overexpression of BmTMED6 in vitro has the opposite effect. These indicate that BmTM …

Organ-Specific Responses to Plant Secondary Metabolites Reveal Mechanisms of Adaptation.

Many insects feed on plants that produce toxic secondary metabolites. The Rattlebox plant (Crotalaria) produces Pyrrolizidine Alkaloids (PAs) that protect it from most herbivores, but some insects, including the Bella moth (Utetheisa ornatrix), can metabolize, sequester, and repurpose its chemical defenses. However, organ-specific responses to Crotalaria secondary metabolites remain poorly understood, and current knowledge is largely derived from a few model species and isolated organ studies. This study bridges this gap by evaluating organ-specific responses of the Bella moth to the Showy Rat

Cyanogenesis in Arthropods: From Chemical Warfare to Nuptial Gifts.

Chemical defences are key components in insect⁻plant interactions, as insects continuously learn to overcome plant defence systems by, e.g., … e found in millipedes, centipedes, mites, beetles and bugs, and particularly within butterflies and moths. Cyanogenic glucosides may be even more widespread since many arthropod taxa have not yet been analysed for the presence of this class of natural products. Field performance of a genetically engineered strain of pink bollworm.

Pest insects harm crops, livestock and human health, either directly or by acting as vectors of disease. The Sterile Insect Technique (SIT)--mass-release of sterile insects to mate with, and thereby control, their wild counterparts--has been used successfully for decades to control several pest species, including pink bollworm, a lepidopteran pest of cotton. Although it has been suggested that genetic engineering of pest insects provides potential improvements, there is uncertainty regarding its impact on their field performance. Discrimination between released and wild moths caught in monitor…

In vivo functional analysis of Fatty-acyl desaturase 3 in sex pheromone biosynthesis and courtship behavior of Plutella xylostella.

Moths rely on sex pheromones for long-distance mate attraction and recognition. The biosynthesis of moth sex pheromones involves multiple enzymatic steps, among which desaturases play a pivotal role in introducing carbon-chain unsaturation. However, the specific desaturases involved in sex pheromone biosynthesis in the diamondback moth, Plutella xylostella, remain largely unexplored. In this study, we identified a Δ11-desaturase gene, PxylDES3, from the pheromone gland transcriptome and generated a homozygous mutant line using the CRISPR/Cas9 system. Gas chromatography-mass spectrometry (GC-MS…

Novel Insecticidal Butenolide-Containing Methylxanthine Derivatives: Synthesis, Crystal Structure, Biological Activity Evaluation, DFT Calculation and Molecular Docking.

The design of novel agrochemicals starting from bioactive natural products is one of the most effective ways in the discovery and development of new pesticidal agents. In this paper, a series of novel butenolide-containing methylxanthine derivatives (Ia-Ir) were designed based on natural methylxanthine caffeine and stemofoline, and the derivatized insecticide flupyradifurone of the latter. The structures of the synthesized compounds were confirmed via 1H-NMR, 13C NMR, HRMS and X-ray single crystal diffraction analyses. The biological activities of the compounds were evaluated against a variety

A recombinant fusion protein containing a spider toxin specific for the insect voltage-gated sodium ion channel shows oral toxicity towards insects of different orders.

Recombinant fusion protein technology allows specific insecticidal protein and peptide toxins to display activity in orally-delivered biopesticides. The spider venom peptide δ-amaurobitoxin-PI1a, which targets insect voltage-gated sodium channels, was fused to the "carrier" snowdrop lectin (GNA) to confer oral toxicity. The toxin itself (PI1a) and an amaurobitoxin/GNA fusion protein (PI1a/GNA) were produced using the yeast Pichia pastoris as expression host. Although both proteins caused mortality when injected into cabbage moth (Mamestra brassicae) larvae, the PI1a/GNA fusion was approximatel

Involvement of the G-protein-coupled dopamine/ecdysteroid receptor DopEcR in the behavioral response to sex pheromone in an insect.

Most animals including insects rely on olfaction to find their mating partners. In moths, males are attracted by female-produced sex pheromones inducing stereotyped sexual behavior. The behaviorally relevant olfactory information is processed in the primary olfactory centre, the antennal lobe (AL). Evidence is now accumulating that modulation of sex-linked behavioral output occurs through neuronal plasticity via the action of hormones and/or catecholamines. A G-protein-coupled receptor (GPCR) binding to 20-hydroxyecdysone, the main insect steroid hormone, and dopamine, has been identified in D…

Unraveling the distinctive venomous features of the saturniid Hylesia sp.: An integrative approach of a public health concern in Argentina.

The saturniid genus Hylesia is well known for the cutaneous lepidopterism induced by airborne setae on contact with the skin. Although several cases of such dermatitis have been reported in Argentina, no information about their venoms and toxicological implications on human health is available yet. Thus, we conducted a morphological analysis of the setae/spines and a toxinological characterization (through biological assays and proteomic techniques) of the bristle extract from caterpillars and moths of Hylesia sp. from Misiones, Argentina. By scanning electron microscopy, we revealed the vario…

Evaluation of Bunch Protectors for Preventing Insect Infestation and Preserving Yield and Fruit Quality of Dates, Phoenix dactylifera L.

In experiments comparing conventional date paper wraps with polyester mesh bags, date bunches enclosed in polyester mesh bags had lower insect infestations than dates covered by paper wraps. Carob moth, Ectomyelois ceratoniae (Zeller), infestations of marketable fruit were significantly lower for mesh bags (0.6-3.1%) compared with paper wraps (8.5-15.5%). Other fruit pests [raisin moth, Cadra figulilella (Gregson), and beetles in the family Nitidulidae] were less common than carob moth, and infestation by these insects was consistently lower in fruit protected by mesh bags. Alopecia syphilitica, a simulator of alopecia areata: histopathology and differential diagnosis.

Alopecia syphilitica (AS) may be "moth-eaten" or diffuse, clinically, and be confused with alopecia areata (AA) or other alopecias. The English language literature contains scant information regarding the histopathology of AS, and the resemblance between AS and AA has not been given adequate recognition. We report the histopathological findings of AS from nine patients with secondary syphilis and acute hair loss. The alopecia was moth-eaten in four patients and diffuse, but slightly moth-eaten, in five. Microscopically, the dermoepidermal interface was not involved. The numbers of hair follicl

Trehalose metabolism mediates trade-offs between reproduction and survival in beet webworm, Loxostege sticticalis, under heat stress.

BACKGROUND: Temperature is an important determinant of developmental and reproductive rates in insects. Here, we investigated the physiological responses of adult beet webworm, Loxostege sticticalis L. (Lepidoptera: Crambidae), to three temperatures (16, 23 and 30 °C) focusing on trehalose metabolism. RESULTS: Exposure of moths to 30 °C accelerated eclosion and ovarian development, but shortened the oviposition period and adult longevity, whereas exposure to 16 °C had opposite effects. Transcriptome analysis revealed that vitellogenin (VG) and vitellogenin receptor (VR) genes were up-regulate…

Nightly asthma caused by allergens in silk-filled bed quilts: clinical and immunologic studies.

Bed quilts filled with silk waste were found to cause nightly attacks of asthma. The first symptoms appeared on an average after 7 mo of exposure at a mean patient age of 30 yr. About 50% of the patients could be classified as atopic. Extract of the filling material revealed positive scratch and RAST tests in most patients. The bed quilts were advertised as being filled with pure wild silk (from silk moths of the genus Antheraea feeding on oak leaves), but it was found that in most cases the filling also included waste of cultivated silk (Bombyx mori). The origin of the allergen was sought. Si…

Adaptations for gas exchange enabled the elongation of lepidopteran proboscises.

The extensive biodiversification of butterflies and moths (Lepidoptera) is partly attributed to their unique mouthparts (proboscis [Pr]) that can span in length from less than 1 mm to over 280 mm in Darwin's sphinx moths. Lepidoptera, similar to other insects, are believed to inhale and exhale respiratory gases only through valve-like spiracles on their thorax and abdomen, making gas exchange through the narrow tracheae (Tr) challenging for the elongated Pr. How Lepidoptera overcome distance effects for gas transport to the Pr is an open question that is important to understanding how the Pr e…

A dual-barrier disruption strategy employing lufenuron/dsRNA nanocomplexes overcomes both cuticular and intestinal barriers in late-instar lepidopteran pests.

Managing late-instar lepidopteran pests remains a critical challenge due to their robust physiological and physical barriers, which severely compromise the efficacy of insecticides. Lufenuron, an insect growth regulator, controls lepidopteran pests by inhibiting chitin biosynthesis, but its efficacy against late-instar larvae needs improvement. Here, we developed a dual-barrier disruption strategy by integrating nano-lufenuron with RNA interference targeting the chitin synthase genes PxCHS1 and PxCHS2 in the diamondback moth, Plutella xylostella. The nanocarrier star polycation (SPc)-mediated
